Abstract

The aim of this research is to determine the influence of jamu Madura “Empot Super” (JMES) on the vaginal epithelium thickness of Rattus norvegicus in vivo. This research is kind of “true experimental-post test only control group design”. The rats were given drinking JMES once daily PS (Per-Sonde) for a month, then the vagina was taken to be sample for HE colouring. The sample was observed by the binocular microscope (100 times magnification) to identify the changes in the thickness of their vaginal epithelium. Calculation of the vaginal epithelium thickness was counted on the 10 field of view chosen randomly by the blind method. The result show that the vaginal epithelium thickness increased with dose 0,17mg/BW, 0,34mg/BW, and 0,68mg/BW of JMES compared with negative control group. But, the vaginal epithelium thickness decrease at the dose 0,51mg/BW compared with negative control group. Keywords: Jamu Madura “Empot Super” (JMES), vaginal epithelium thickness, white mice (Rattus norvegicus), In Vivo study

Abstract

Students as well as the next intellectual generation in society is not limited to academic obligations in the campus environment, but also should be useful in a variety of other functions in society. Students are also required to participate in community service through various forms of devotion. In this community service, lecturers with students to jointly work devotion in advancing knowledge and skills Tengger tribe in Ngadas Village District of Poncokusumo in developing the potential of medicinal plants and agricultural products based ethnopharmacy towards the creation of an independent village. Ngadas village is village with abundant natural resources, especially medicinal plants and agricultural products. Medicinal plants rare owned by the village is pronojiwo (Euchresta horsfieldii), pulosari (Alyxia reinwardtii), sintok (Cinnamomum sintoc), purwoceng (Pimpinella pruatjan), krangean (Litsea cububa Pers), flour muscles (Borreria laevis Griseb.), guava wer (Prunus persica Bl.), dringu (Acorus calamus L.). Medicinal plants have the potential to be developed and produced in large quantities in this village. Natural wealth of agricultural products is also very abundant in this village include potato, cassava, sweet potatoes and celery. But not taken into ready to eat foods products. Performed community service on 19 November 2016 in the village hall Ngadas. The activities carried out in the form of workshops begins with exposure of the material on the benefits and applications of potential medicinal plants that become local wisdom in Tengger tribe and then followed by demo making products. The product are rub medicine guava wer,  muscle flour, starfruit herbal drinks and bajigur. Participants consisted of PKK Ngadas village. Good response shown by them with great enthusiasm when exposure of the material provided and when making products. In the future, this mentoring program to be developed into entrepreneurial activities of local communities to improve their welfare.

Abstract

Chrysanthemum cinerariifolium (C. cinerariifolium) is an empirically proven medicinal plant that has anticancer activities. This study aimed to profile metabolites, and cytotoxic activity of root, leaf, stem, and root extracts C. cinerariifolium on T47D cells and to determine the correlation of metabolite content with cytotoxic activity. The metabolite profile was carried out using UPLC-QToF-MS / MS, and cytotoxic activity was carried out using the MTT method. The results obtained in the form of a chromatogram were processed with the application Masslynk so that a metabolite profile data obtained. The data is then analyzed statistically using Principal Component Analysis (PCA). The results obtained on metabolite profiling showed that there were differences in metabolite profiles in the roots, stems, leaves, and chrysanthemums. The characteristic compounds in the flower section are D - (-) - Morphine and in the leaf part of genistein  and N - [(5-Chloro-1,2,3-thiadiazol-4-yl) methyl] -1- (2-isopropyl-4 -methyl-1,3-thiazol-5-yl) -N-methyl ethanolamine. The major compounds in the roots, stems, and leaves are Orphenadrine with successive percentages of 9.11%, 10.16%, and 3.24%, and the major compounds in the flower section are D - (-) - Morphine with a percentage of 10, 86%. Furthermore, the results of the cytotoxic activity test showed differences in anticancer cytotoxic activity in the parts of the flower, leaf, stem, and root of C. cinerariifolium. Besides, there is a relationship between the metabolite content and anticancer cytotoxic activity of each part of C. cinerariifolium. The higher the level of orphenadrine compounds in the plant, the higher the potential for anticancer. Keywords: C. cinerariifolium, UPLC-QToF-MS/MS, T47D cells.

Abstract

Abortion is still a controversial problem in Indonesian society recently. Statistical data which are revealed by National Board for family Planning Coordination show that around 2.000.000 cases of abortion practiced in Indonesia every year. The debate on its legality to be practiced, particularly the four schools of sect, agree to put abortion as an unlawful practice. Therefore, abortion generally is a cruel action, against humanity valuesand it also against law and religious tenets. However, regardless all those aspects, its legal status should be studied specifically and deeply because it is not a single action .This sort of action has various kinds of action based on its conditional consideration. This writing aims at discussing abortion by looking at Indonesian legal regulation through criminal law and health regulation.

Abstract

Maraknya kasus kekerasan seksual pada anak (child abuse) yang dilakukan oleh orang-orang terdekat anak termasuk keluarga menunjukkan pentingnya pemahaman akan pendidikan seks usia dini. Masalah pendidikan seks kurang diperhatikan orang tua pada masa kini sehingga mereka menyerahkan semua pendidikan termasuk pendidikan seks pada sekolah. Padahal yang bertanggungjawab mengajarkan pendidikan seks pada anak usia dini adalah orang tua, sedangkan sekolah hanya sebagai pelengkap dalam memberikan informasi kepada si anak. Hal ini menunjukkan, bahwa peranan orang tua, terutama ibu sangat strategis dalam mengenalkan pendidikan seks sejak dini kepada anak-anak mereka.Kata Kunci: Ibu, pendidikan seks, anak usia dini

Abstract

Dayak onion (Eleutherine palmifolia (L.) Merr) is a typical plant in Central Kalimantan. This plant has been passed down for generations by Indonesian people for various treatments. The potential of dayak onions that have various pharmacological activities needs to be increased in their use as modern medicinal ingredients. The purpose of this literature study is to review the various uses, the secondary metabolites content, and the action mechanism as an anticancer both in vitro, in vivo, and in silico. The results of this literature study show that traditionally the Indonesian people use the bulbs for the treatment of breast cancer, colon cancer, hypertension, diabetes mellitus, stroke, fever, dysuria, intestinal inflammation, dysentery, boils, cysts, prostate, lowering cholesterol and triglycerides, breastfeeding and sexual disorders. In the empiric treatment of cancer, this plant is used by drying the bulbs and chewing it. Dayak onion bulbs (Eleutherine palmifolia (L.) Merr) are known to contain flavonoid compounds wich isoliquiritigenin, polyphenols group wich oxyresveratrol and naphtoquinon group, and its derivatives such as elecanacine, eleutherine, eleutherol, eleutherinol, eleutherinon, eleuthoside B, eletherinoside A which has anti-cancer activity. Pre-clinical studies with in vitro and in vivo mechanism showed that dayak onion bulbs (Eleutherine palmifolia (L.) Merr) extracts have pharmacological activities, which are anti-cancer that can inhibit cell signaling through apoptosis and cell cycle arrest. Also, the mechanism of in silico showed anti-cancer activity from the inhibition of VHR receptors, BCL-2 receptors, VEGFR-2 receptors and alpha estrogen receptors (ERα).

Abstract

Ethanol extract 96% of Chrysanthemum cinerariifolium (Trev.) Vis leaves are proven to have anticancer activity. However, these compounds have low solubility in water and fat. To improve the bioavailability of preparations, the development of drug design in the form of Nanostructured Lipid Carrier (NLC) is carried out.The aim this study was to determine the effect of differences in lipid concentration of Monostearin and Oleic Acid in the formulation of the NLC system of Chrysanthemum cinerariifolium (Trev.) Vis leaves, which produced good physicochemical characteristics including organoleptic characteristics, pH, viscosity, particle size, and drug entrapment efficiency. Making is done using the High Shear Homogenization method. Organoleptic tests showed ideal results for the Nanostructured Lipid Carrier (NLC) system, the resulting pH values in formula 1 (7.0 ± 0.12), formula 2 (6.7 ± 0.17) and formula 3 (6.8 ± 0.2)), the viscosity value of formula 1 (55.66 ± 2.84 cPs), formula 2 (28.86 ± 3.91 cPs), and formula 3 (28.57 ± 16.85 cPs). Particle size of formula 1 (5530 ± 320.47 nm), formula 2 (5337 ± 671.44 nm) and formula 3 (4676 ± 2215.75 nm)). The entrapment efficiency value of formula 1 drugs (33.55%), formula 2 (38.77%), formula 3 (83.75%). Based on the characterization result, the best formula was obtained at a lipid concentration of 10%, which was used for the formula of the NLC system of Chrysanthemum cinerariifolium (Trev.) Vis leaves for oral anticancer preparations

Abstract

Sambiloto (Andrographis paniculata Nees) contains andrographolid compound which has potentialas an anticancer. The difference of growth location has affected the distinction of metabolite content. Thisstudy aims to determine the profile of anticancer activities of A. paniculata extract from several locations inEast Java. The extracts from several locations were tested for activity on T47D cancer cell using MTT assaymethod. Thus, the result of this study describes that A. paniculata extract from several different locations doesnot provide significantly different anticancer activities with the amount of (p>0.05). Abstract

Benalu belimbing (Macrosolen cochinensis) and onion sabrang / onion dayak (Eleutherine palmifolia (L) Merr.) is a traditional medicinal plant that has been used by the community as a cancer drug from generation to generation. This study aims to prove scientifically about the anticancer potential from combination of both plants. Anticancer potential of the plant has been proven by testing the extract both in single form and in combination with Hela cells. While to know the selectivity of the extract to HeLa cell then extract is tested on normal cell Vero cell line. Furthermore, a combination test of both extracts was done using variation dose under IC50. The method used to determine anticancer activity was the MTT method. The results obtained in this study showed that ethanolic extract onion sabrang selectively able to inhibit the growth of cervical cancer cells HeLa on IC50: 40.36 μg / ml with 4.06 for the SI (selectivity index) value. While ethanolic extract of benalu belimbing bselectively can inhibit the growth of cervical cancer cells HeLa on IC50: 217.72 μg / ml with 5.77 for the SI (selectivity Index) value. The combination test of sabrang onion extract and extract of benalu belimbing showed that the combination was highly synergistic in combination dose of EBS 22.62 μg / ml and EBB 23.3 ug / ml with combination index 0.15. Whereas the combination doses having synergistic effects on combination doses 23.3 μg / ml for EBB and 11.31 μg / ml for EBS; 23.3 μg / ml EBB and 16.96 μg / ml EBS with a consecutive combination index; 0.51 and 0.64. From these results it can be concluded that the onion sabrang extract has anticancer effect on HeLa cells and selectively killing HeLa cells because the value of selectivity index is greater than 3. The result of the combination test of both extracts shows that the combination dose having strong synergistic effect, so that combination can be recommended for further research.

Abstract

Chrysanthemum cinerariifolium (C.cinerariifolium) is a plant of the Asteraceae family, which has been applied by the community as an ornamental plant and traditional medicine. In this study, the effect of C. cinerariifolium leaves extract on inhibition of cell cycle and induction of apoptosis in T47D breast cancer cells was tested and compared to the standard chemotherapy agent. The citotoxic activity of C. cinerariifolium leaves extract against T47D cancer cells and Vero normal cells was tested by MTT method. Profile of apoptosis and cell cycle were observed by flow cytometry method. Based on chemical compounds profil which is tested used TLC showed that C.cinerariifolium leaves extracts contained flavonoid and terpenoid chemical compounds. The result of cytotoxic test showed that leaves extract of C. cinerariifolium was able to inhibit the growth of T47D cancer cell at IC50 418.8μg/mL. Doxorubicin, extracted from Streptomyces peucetius used as treatment in several cancers including breast cancer. Doxorubicin could inhibit the growth of T47D cancer cells in 115.1μg/mL. The results of cell cycle analysis showed that the C. cinerariifolium leaves extract inhibited cell cycle in G0-G1 and S phase, whereas doxorubicin was able to inhibit cell cycle in G0-G1 phase but experienced cell accumulation in G2-M phase. The percentage of apoptosis in cycle was showed in M1 (sub G1) and M5 (multinuclear) phase which treatment of C. cinerariifolium leaves extract was higher than doxorubicin. Therefore, C. cinerariifolium leaves extract has potential activity as anticancer agent causes inhibition of cell cycle and induction apoptosis.
