Abstract

Abstract

Abstract

This study aims to describe the management of used culture media waste and contaminated materials, and to assess the understanding of laboratory staff and interns regarding ethical principles in plant tissue culture laboratories. A descriptive qualitative method was applied using observation, questionnaires, and literature review. The results indicate that most respondents have a good understanding, reflected in their agreement on the importance of sterilization, waste segregation, use of personal protective equipment, and implementation of standard operating procedures (SOP). Respondents are also aware of the risks of laboratory waste to health and the environment. Ethical practices are recognized as part of professional responsibility, but require improvement through training, supervision, and consistent SOP implementation.

Abstract

The management of hazardous and toxic waste (B3) in health laboratories remains a challenge, particularly inensuring regulatory compliance while consistently applying bioethical principles. This issue is important becauseimproper waste management can negatively impact the environment and public health. Therefore, this study aimsto analyze the B3 waste management system, its compliance with regulations, and the implementation of bioethicalprinciples at the Regional Health Laboratory (Labkesda) of Bekasi City. This study employed a descriptivequalitative method with a case study approach. Data were collected through interviews, observations, anddocumentation involving two informants directly engaged in waste management. In addition, the implementationof bioethics was assessed using a Likert scale with ten indicators. The results show that B3 waste management hasbeen carried out in a structured manner, including identification, containment, transportation, temporary storage, and processing by licensed third parties, thus complying with applicable regulations. Furthermore, theimplementation of bioethical principles achieved an average score of 4.8 out of 5, indicating a very good category. However, an obstacle was identified in the form of delays in waste collection by third parties. In conclusion,Labkesda Bekasi City has been able to manage B3 waste effectively and sustainably, although improvements inexternal coordination are still needed

Abstract

Food safety and environmental quality are critical aspects in maintaining public health, particularly in fishery products and water sources that are vulnerable to contamination by pathogenic microorganisms. This study aims to identify the presence of Escherichia coli in water samples and Salmonella in tilapia (Oreochromis niloticus), as well as to analyze the use of Compact Dry media based on ISO/IEC 17025 and the implementation of bioethical principles in microbiological testing. This research employed a descriptive laboratory approach conducted at the BPPMHKP Medan I Laboratory using Compact Dry media based on AOAC methods. The samples consisted of lake water and tilapia, which were tested through enrichment, inoculation, incubation, and colony observation stages. The results showed no growth of Escherichia coli colonies on Compact Dry EC media and no Salmonella colonies on Compact Dry SL media, indicating that all samples were negative for the tested bacteria. The use of Compact Dry media in this study complied with ISO/IEC 17025 standards, as indicated by systematic procedures, the use of validated media, and laboratory quality control. Furthermore, bioethical principles, including scientific integrity, responsibility, biosafety, and accountability, were applied throughout all stages of testing. Therefore, this study demonstrates that the use of validated media and the application of bioethical principles play an important role in ensuring valid, reliable, and accountable laboratory results.
