Abstract

The objective of this study was to determine the minimum concentration of extract in inhibiting motile motile aeromonad septicaemia. The material used was the stems and leaves of the plant P. grandiflora as much as 10 g. The crushed simplicia was then macerated with aquades with a ratio of 1:10,  then filtered with filter paper and, put in an Erlenmeyer, and evaporated in an incubator at temperature of 30°C. The sensitivity test was performed by placing a paper disc containing extract on the agar medium that had been scratched with bacterial colonies. Clear zone formed around the paper disc was compared with standard classification table for clarifying the inhibition level.  The results showed that stem and leaf extract had inhibition zone against Aeromonas hydrophila bacteria that caused motile aeromonad septicaemia. The best result was obtain at concentration of extract 3.25 mg / L) with an average diameter of 22.67 mm and for leaf extract had a moderate to strong zone for all treatments. These proved that stem and leaf extract of P. grandiflora plant had antibacterial activity and can be used as control of MAS disease caused by A. hydrophila bacteria. Abstract

The objetive of research was to examine the effect of yeast cell Saccharomyces cereviciae on non-specific immune response of nile tilapia, Oreochromis niloticus. Juveniles weighing 31.83 g in average were cultured in five concrete tanks with a density of 30 fish per tank. The fish were fed pellet supplemented with yeast cell at five different doses (0, 10, 20, 30, 40 g/kg pellet) for three consecutive weeks as much as 4%/bb/day, twice daily. At the end of feeding, three fish from each tank were sampled to measure the immune parameters namely total leucocyte count and phagocytosis activity of phagocyte cells. Research results showed that after three weeks of feeding, total leucocyte and phagocytosis activity of fish fed pellet supplemented with yeast cells were significantly different as compared to those of control fish.  The highest total leucocyte and phagocytosis activity were achieved in fish fed pellet supplemented with 10 g of yeast cells/kg pellet.  It was concluded that the use of yeast cells for three weeks could enhance the nonspecific immune response of nile tilapia. Abstract

This study aimed to identify the bacterial disease in cultured eel elvers (Anguilla marmorata). The samples used for this study were taken from the Tatelu Freshwater Aquaculture Center. The samples taken were elvers (juvenile life stage of A. marmorata)  which showed clinical symptoms of bacterial infection such as bleeding in the chest, abdomen, and the base of the fin, passive movement, weakness, and loss of balance of the body, loss of appetite, some wounds on back fin, chest, and tail, and the body was not slippery/shiny. Three elvers with the size of 4-6 cm, and weigh about 0.2 g were taken as samples. The organ chosen to be targeted for isolation was head kidney. Each sample was isolated twice as replicates and streaked on Triptic Soy Agar (TSA) medium. First replicates were identified in the Tatelu Health Lab of Freshwater Aquaculture, and the second replicates were identified in North Sulawesi Provincial Health Lab. Bacterial identification was done through a series of observations and morphology of bacterial colonies through gram stain, followed by biochemical tests through oxidase test, catalase test, Triple Sugar Iron Agar (TSIA) test, H2S production test, motility test, O/F test, and Simmons Citrate Agar (SCA) test. Water quality parameters that were measured included temperature, disolve oxygen, pH, and ammonia. Based on identification of bacteria in cultured elvers in Tatelu Freshwater Aquaculture Center, the elver samples had been infected with bacterial  Aeromonas hydrophila. Abstract

This study was conducted to determine the effect of yeast as a counterweight of fibrous raw material in Tilapia  feed formulations on growth and feed efficiency value. The research results found that the biggest absolute growth of Tilapia fry occurred in treatment A: 5% yeast (9,2 g),  followed by  C: 15% yeast (8,6 g), D: 20% yeast (7,14 g), and B: 10% yeast (5,03 g). The highest feed efficiency value occurred in treatment A (23,75%), followed by C (22,45%), D (18,69%) and B (13,96%). The parameter of water quality measured during experiment included temperature 26-28o C and pH  7. The range value of water quality in each treatment was considered to be proper for the living condition of fish fry. Keywords: yeast, Oreochromis niloticus, growth, feed efficiency

Abstract

The aim of this research was to study the effect of Binahong extract on the growth of Nile tilapia. A number of 225 fish measuring 10-12 cm in length with an average weight of 15 g was obtained from Fishery Development Center and Training, Tateli. The fish were put  in oxygenated plastic bags and transported to Laboratory of Aquaculture Technology. The fish was kept in aquarium equipped with an aerator with a density of 15 fish per aquaria. The test material used was the stem of Binahong plant stem, macerated with alcohol 70% for 3 days. The extract was then filtered with whatman filter paper and extract was concentrated using a Rotary Evaporator. The extract was further diluted to obtain  concentrations of 12.5, 25, 50, and 100 mg / mL Binahong  extract as treatment was given to the fish after mixed first into the pellet. The fish was fed with treatment feed for 30 days. Control feed was not supplemented with binahong extract. Growth data was measured at day 30 after treatment. The results showed that the extract of binahong had significant effect on fish growth (p <0.01) with the best result achieved at concentration of 100 mg/mL. In conclusion, supplementation of  binahong extract into fish feed could improve growth of  fish in which the best growth was obtained in fish fed diet containing 100 mg/mL of binahong extract.Keywords: Medicinal plants, tilapia, Anredera cordifolia, growth, maceration

Abstract

The use of trash fish ( Treatment A ), trash fish + cassava( Treatment B), pellets+ trash fish ( Treatment C ) and a mixture of trash fish + cassava + pellets ( Treatment D ) as feed had been conducted to evaluate its effect on growth of Caranx sexfasciatus. This research was carried out inTelengen Bay waters, Central Tabukan District, Sangihe Islands Regency. Fish with weighing 60,9 -62,9 g were distributed in 12 net cages measuring 3x1x1 m with the density of 5 fish each. Fish were fed three times a day for seven weeks. Fish weight was measured every week. At the end of experiment, the individual weight of fish ranged from 168,7 to 222,8 g in with the highest average weigth gain was achieved in treatment A namely 222.8 g (354.2%)), followed by treatment C 178.0 g (286.6 %) treatment D 170.08 g (280.5%), and treatment B as much as 168.7 g (274.3%). Statistical analysis displayed that weight gain of fish in treatment A was significantly different as compared to that of treatments B, C, and D. There was no significant differencesbetween treatment B, C, and D. Food conversion ratio of treatment A was significantly different compared to other treatments. Food conversion ratio of treatment A was 2.80, B 4.25, C 3.70, D 3.88. as conclusion, the use of trash fish without supplementaion with others ingradients resulted in the highest absolute and relatif growth of fish and the lowest food conversion ratio of 2.80 as compared to other treatments. Abstract

This paper was the result of a feasibility analysis of Talengen Bay Waters for aquaculture of Kappaphycus alvarezii. Feasibility analysis of water was based on the environmental data and water quality parameters of Talengen Bay. One mistake in the development of aquaculture is environmentally unsuitable. In order to develop aquaculture properly, appropriate water conditions data were required. Suitable aquatic environments are required by organisms for their survival because they are related to the patterns and behavior of the biota. In connection with the problems of mariculture at Talengen Bay, several efforts were required to overcome them. One of the efforts was made through this study that was assessment of appropriate location for the cultivation of seaweed. Talengen Gulf was located in  in the territory of Kampung  Talengen and  Kampung Bunga Lawang, Central District of Tabukan, Sangihe Islands Regency, North Sulawesi. Laboratory analysis was conducted at the Laboratory of Industrial Baristand Manado. Determination of research stations done intentionally (purposive sampling). Based on the results obtained it could then be concluded that the condition of the aquatic environment and water quality parameters in the Talengen Bay were categorized as suitable for seaweed cultivation K. alvarezii. Abstract

The purpose of this research was to know and evaluate the condition of aquatic environment and water quality parameters for cultivation of seaweed Ulva sp. This research was conducted on North Sulawesi Marine Education Center (NSMEC). NSMEC is planned to be built at Marine Field Station FPIK UNSRAT located in Likupang Timur Kabupaten Minahasa Utara. The waters of NSMEC development area are geographically located at 1040.437 'LU and 12504.499' BT. The determination of 4 stations was done by purposive sampling which was considered to represent the condition of waters. Coordinate stations were recorded with GPS help. The data were collected for 14 days every 6:00 pm, at 12:00 pm and 17:00 pm. The observation of environmental conditions was carried out for protection factor and substrate of water base, while water quality parameter measured in situ included depth, brightness, temperature, dissolved oxygen (DO) salinity, pH and current velocity. Phosphate, nitrate and total suspended solid (TSS) were measured. Tide measurements were measured every hour for 24 hours. Data analysis used conformity matrices that included scores and weights for the determination of conformity classes. Class suitability was used to describe the suitability of seaweed. The results showed that the waters of the NSMEC development zone were in class S1 meaning very suitable for seaweed cultivation location. The analysis results of each station found that 4 stations were very suitable. As conclusion, water environment and water quality parameters of the waters of North Sulawesi Marine Education Center development area located at Marine Field Station FPIK UNSRAT were categorized as very suitable for seaweed cultivation, Ulva sp.Keywords: Ulva sp., feasibility analysis, water quality, location suitability

Abstract

This study was conducted to determine the effect of different doses of ovaprim on latency time of spawning, hatching rate, and survival of fish larvae (Pangasianodon hiphopthalmus). Experimental design used was Randomized Complete Design (CRD) with four treatments, each with three replication. The treatments including 0 ml, 0.3 ml, 0.6 ml and 0.9 ml ovaprim per kg of body weight of fish. Data collected consisted of latency time of spawning, hatching rate, and survival rate of larvae. The results showed that treatment with different doses of ovaprim hormones resulted in a significant influence on latency time of spawning, egg hatchability and survival rate of larvae. It was found that 0.6 ml ovaprim could increase the latency time of spawning, egg hatchability and survival rate of larvae. The average latency time of larvae was 529 minutes, hatching rate was 80,86% and survival rate of larvae was 83,33%.

Abstract

The research was aimed to find out the optimum dose of feeding regime             to ensure the maximum growth of carp-seeds, Cyprinus carpio, reared in backyard pond with recirculation system. The experiment was carried out in Randomized Block Design with 3 different dose of feeding regimes as treatments, which were 3%, 4% and 5% of the total body weight per day.  Each treatment was triplicate and each repetition functioned as group of fish with different weight.  There were 3 groups which were, fish weighing 1.5–2.4 gram (Group-1), fish weighing 2.5 – 3.4 gram (Group-2) and fish weighing 3.5 – 4.8 gram (Group-3).  Each experimental unit was composed of 8 tested fish, placed in a netting-cage bag measuring 60×30×60 cm (l×w×h) with a mesh-size of 4 mm. Each group was composed of 24 tested fish, hence there were 72 tested fish in total.  Fish were fed twice a day at 07:00 and 17:00.  Tested fish were weighed every week, and the fish weight data were converted into FCR, absolute, relative and daily growth rate. Data were statistically analyzed using JMP statistic-program.  The results showed that, there was no significant difference in absolute, relative and daily growth rate among fish fed 3%, 4% and 5% of total body weight per day.   The best FCR was performed by fish fed 3% of total body weight per day.   Hence, this experiment result suggests that, carp reared in backyard pond with recirculation system should be fed with a dose of 3% of the total body weight each day, as fish might grow well and at the same time the amount of uneaten food, feces and metabolic wastes is minimized at this dose of feeding.
