Abstract

Pharmaceutical Grade Dextrose Monohydrate from Manihot ecsulenta, Metroxylon sagu, Zea mays, Oryza sativa, dan Triticum Starch ABSTRACT Pharmaceutical-grade dextrose monohydrate, one of raw materials used as active pharmaceutical ingredients (API) and additives, can be made from starch. There are five types of local Indonesian commercial starch that are potentially used, namely tapioca (Manihot esculenta), sago (Metroxylon sagu), corn (Zea mays), rice (Oryza sativa), and wheat (Triticum) starch. This study aimed to compare these five starches as raw materials for preparing pharmaceutical-grade dextrose monohydrate which was expected to meet the requirements of the Indonesian Pharmacopoeia (5th Edition) and the United States Pharmacopeia (USP). The starch was converted into dextrose monohydrate through liquefaction hydrolysis, saccharification hydrolysis, activated carbon purification and filtration, ion exchange purification, evaporation, crystallization and drying.  High Performance Liquid Chromatogram (HPLC) and the Luff-Schoorl methods were used for dextrose equivalent value (DE) analysis. The results showed that only three of the starch types produced pharmaceutical-grade dextrose monohydrate, namely (DE) sago starch (107.23% and 100.77%), corn starch (97.86% and 96.19%), and tapioca starch (85.18% and 99.20%).Keywords: dextrose equivalent, dextrose monohydrate, hydrolysis, pharmaceutical grade, starchABSTRAKDekstrosa monohidrat kualitas farmasi, salah satu bahan baku yang digunakan sebagai active pharmaceutical ingredient (API) dan bahan tambahan, dapat dibuat dari bahan pati-patian. Penelitian ini bertujuan membandingkan lima jenis pati tersebut sebagai bahan baku pembuatan dekstrosa monohidrat kualitas farmasi yang diharapkan mampu memenuhi standar persyaratan dari Farmakope Indonesia Edisi V dan United States Pharmacopeia (USP). Pati diubah menjadi dekstrosa monohidrat melalui hidrolisis likuifikasi, hidrolisis sakarifikasi, pemurnian karbon aktif dan filtrasi, pemurnian ion exchange, evaporasi, kristalisasi dan pengeringan. Metode High Performance Liquid Chromatogram (HPLC) dan Luff-Schoorl digunakan untuk analisis dextrose equivalent (DE). Abstract

Spleen Histology and Hematology of Mice Infected by Escherichia coli after Oral Administration of Humic Acid from Borneo PeatABSTRACTHumic acid compounds have an immunostimulatory effect. The aim of this research was to determine the effect of humic acid on the spleen of mice infected with Escherichia coli. The study used a completely randomized design with six treatments and five replicates. The treatments were normal control, negative control, positive control of isoprinosine, humic acid dose of 62.5; 125; and 250 mg/kg body weight (BW). The results showed that E. coli infection caused diarrhea symptoms and significant weight loss. There were significant differences (P<0.05) on hematocrit value and a total leukocyte count of humic acid, in which isoprinosine treatment was higher than those of negative control and normal control. There was no significant difference in the spleen weight of the mice subjected to the different treatments, but through histologic observations a significant difference (P<0.05) was found in the histologic size of the spleen. Humic acid treatment of 250 and 125 mg/kg BW resulted in the widest white pulp (495.8 ± 58.2 µm) and the highest leukocytes count (6725 ± 1018 cell/mL), respectively. On the red pulp serving as negative control numerous clusters of lymphocyte cells were found.Keywords: Escherichia coli, humic acid, peat soil, spleen, white pulp ABSTRAKSenyawa asam humat mempunyai potensi imunostimulan. Penelitian ini bertujuan mengetahui pengaruh pemberian asam humat terhadap organ limpa mencit yang diinfeksi bakteri Escherichia coli. Penelitian ini menggunakan rancangan acak lengkap dengan enam perlakuan dan lima ulangan. Perlakuan tersebut yakni kontrol normal, kontrol negatif, kontrol positif isoprinosin, asam humat dosis 62,5; 125; dan 250 mg/kg berat badan (BB). Hasil penelitian menunjukkan bahwa infeksi bakteri E. coli pada mencit menyebabkan mencit mengalami gejala diare dan penurunan berat badan yang signifikan. Perbedaan signifikan (P<0,05) pada nilai hematokrit dan jumlah leukosit total perlakuan asam humat dan isoprinosin lebih tinggi dibandingkan kontrol negatif dan kontrol normal. Tidak terdapat perbedaan nyata (P>0,05) pada berat limpa mencit antar perlakuan, melalui pengamatan histologi ditemukan perbedaan ukuran histologi limpa mencit. Perlakuan asam humat 250 mg/kg BB mempunyai ukuran pulpa putih (495,8 ± 58,2µm) dan perlakuan asam humat 125 mg/kg BB mempunyai nilai leukosit tertinggi (6725 ± 1018 sel/mL). Pada pulpa merah perlakuan kontrol negatif ditemukan banyak sel limfosit yang menggerombol.Kata Kunci: asam humat, Escherichia coli, limpa, pulpa putih, tanah gambut

Abstract

Expression of Mycobacterium tuberculosis Ag85B Antigen in Mammalian Cell CultureTuberculosis (TB) continues to be a major health problem worldwide, affecting millions of people each year. The only vaccine approved for the prevention of TB is Bacillus Calmette-Guérin (BCG). However, one of the limitations of BCG is that its preventive effect against pulmonary TB varies from person to person. Therefore, there arises a need for a new TB vaccine to replace BCG. This study aims to obtain the Ag85B recombinant protein which has characteristics similar to the native Ag85B antigen from Mycobacterium tuberculosis. In this study, we cloned and expressed recombinant Ag85B in mammalian cell culture. In the initial step, we cloned synthetic Ag85B into mammalian expression vector pFLAG-CMV4 and expressed the gene in CHO-K1 cells. Interestingly, a specific band around 30 kDa was observed in the culture media of transfected cells by Western blot analysis. The results from our research showed the potency of mammalian expression system to produce recombinant protein Ag85B for new TB vaccine candidate.Keywords: Ag85B, mammalian cells, tuberculosis, vaccine, expression ABSTRAKTuberkulosis (TB) terus menjadi salah satu masalah kesehatan dunia yang mempengaruhi jutaan manusia setiap tahun. Satu-satunya vaksin untuk TB yang ada adalah Bacillus Calmette-Guérin (BCG). Namun demikian, vaksin BCG ini memiliki kelemahan berupa terjadi efek preventif yang bervariasi dari satu individu terhadap individu lainnya.  Oleh sebab itu diperlukan pengembangan vaksin TB yang dapat menggantikan vaksin BCG yang sudah ada. Penelitian ini bertujuan memperoleh protein rekombinan Ag85B yang memiliki karakteristik mirip dengan antigen Ag85B native dari Mycobacterium tuberculosis. Pada penelitian ini, telah dilakukan kegiatan pengklonaan dan ekspresi gen Ag85B pada galur sel mamalia.  Pada tahap awal dilakukan pengklonaan gen sintesis Ag85B ke dalam plasmid pada sel mamalia pFLAG-CMV4 dan diekspresikan gennya pada sel CHO-K1. Hasil analisis Western blot menunjukan tersekresinya gen target berukuran 30 kDa pada media kultur dari sel mamalia yang ditransfeksi. Hasil dari penelitian ini menunjukkan potensi dari sistem ekpresi untuk protein rekombinan Ag85B pada galur sel mamalia sebagai kandidat vaksin TB yang baru.

Abstract

Eurycoma longifolia (pasak bumi) is known as a medicinal plant that contains biologically active compounds. Studies on endophytic bacteria associated with pasak bumi and their biocontrol activities have not been widely reported. The objective of this study is to isolate potential endophytic bacteria associated with E. longifolia possessing biocontrol activity against plant pathogenic fungi, Sclerotium rolfsii, Fusarium solani, F. oxysporum, and Colletotrichum gloeosporioides. Endophytic bacteria were isolated from the roots of E. longifolia using the plant piece method and identified based on 16S rRNA genes analysis. Antagonist test of bacterial isolates was conducted by dual confrontation method. The mechanisms of fungal growth inhibition were evaluated based on their ability to produce hydrolytic enzymes, antibiotic, and volatile organic compounds. Two isolates were obtained and identified as Stenotrophomonas maltophilia (Apb1) and Serratia marcescens (Apb2). Apb2 was able to inhibit the growth of four tested fungi and showed protease, chitinase as well as cellulase activities. The crude extract and volatile organic compound produced by Apb2 were active against F. solani growth.Keywords: biocontrol, endophytic, Eurycoma longifolia, fungi, inhibition mechanism ABSTRAKEurycoma longifolia (pasak bumi) dikenal sebagai tanaman obat yang mengandung beberapa senyawa aktif secara biologis. Penelitian mengenai bakteri endofit yang berasosiasi dengan tanaman pasak bumi berikut aktivitas biokontrolnya belum banyak dilaporkan. Penelitian ini bertujuan mengisolasi bakteri endofit potensial dari tanaman E. longifolia yang memiliki aktivitas biokontrol terhadap empat strain uji jamur patogen tanaman, yaitu Sclerotium rolfsii, Fusarium solani, F. oxysporum, dan Colletotrichum gloeosporioides. Bakteri endofit diisolasi dari akar E. longifola menggunakan metode plant piece dan diidentifikasi berdasarkan analisis gen 16S rRNA. Uji antagonis isolat bakteri dilakukan dengan metode konfrontasi ganda. Mekanisme penghambatan jamur patogen tanaman dievaluasi berdasarkan kemampuannya dalam memproduksi enzim hidrolisis, senyawa antibiotik, dan senyawa organik volatil. Dua isolat bakteri endofit berhasil diperoleh dan teridentifikasi sebagai Stenotrophomonas maltophilia  (Apb1) dan Serratia marcescens (Apb2). Isolat Apb2 mampu menghambat pertumbuhan keempat jamur yang diuji dan menunjukkan aktivitas protease, kitinase dan selulase. Ekstrak kasar dan senyawa organik volatil yang dihasilkan oleh isolat Apb2 aktif menghambat pertumbuhan F. solani.

Abstract

Effect of Fermentation on Antioxidant Activities and Betacyanin Content of Functional Beverages from Dragon Fruit and BeetrootKombucha is a traditional beverage prepared by fermenting polyphenol-rich tea using a consortium of bacteria and yeast (SCOBY). Dragon fruit and beetroot contain a significantly high amount of polyphenols and betacyanin as antioxidants which are beneficial in reducing the risk of cardiovascular disease, cancer, and the body's natural degeneration related to the early-aging process. This study aims to determine changes in antioxidant activity and levels of betacyanin from fermented dragon fruit and beetroot as functional drinks during different fermentation times. The results showed that there was a correlation between fermentation time and antioxidant activity in its ability to counteract free radicals, ferric reducing antioxidant power (FRAP), and the levels of betacyanin produced. Longer fermentation time caused an increase in free radical inhibition and reduction of iron ions but reduced the levels of betacyanin. The optimum value of free radical inhibitory activity (DPPH) was obtained in the 12-day fermented dragon fruit drink with an inhibitory value of 80.76%, ability to reduce iron ions by 197.94 µg ascorbic acid mL-1, and betacyanin level of 0.055 mg L-1Keywords: antioxidants, dragon fruit, betacyanin, beetroot, functional beverages ABSTRAKKombucha merupakan minuman tradisional hasil olahan fermentasi teh yang kaya polifenol dengan konsorsium bakteri dan yeast (SCOBY). Buah naga dan umbi bit memiliki polifenol dan senyawa betasianin yang cukup tinggi sebagai antioksidan yang bermanfaat dalam menurunkan risiko penyakit kardiovaskular, penyakit kanker dan degenerasi alami tubuh terkait proses penuaan dini. Penelitian ini bertujuan untuk mengetahui perubahan aktivitas antioksidan dan kadar betasianin dari fermentasi buah naga maupun umbi bit sebagai minuman fungsional selama waktu fermentasi yang berbeda. Hasil penelitian menunjukkan bahwa adanya korelasi antara waktu fermentasi terhadap aktivitas antioksidan dalam kemampuannya menangkal radikal bebas, kemampuan mereduksi ion besi (FRAP), dan kadar betasianin yang dihasilkan. Semakin lama waktu fermentasi menyebabkan peningkatan penghambatan radikal bebas dan reduksi ion besi, namun menurunkan kadar betasianin. Nilai aktivitas penghambatan radikal bebas (DPPH) optimum diperoleh dari minuman fermentasi buah naga selama waktu fermentasi 12 hari dengan nilai penghambatan sebesar 80,76%, kemampuannya dalam mereduksi ion besi sebesar 197,94 µg asam askorbat mL–1, dan kadar betasianin sebesar 0,055 mg L–1
