Squalen Bulletin of Marine and Fisheries Postharvest and Biotechnology

Squalen publishes original and innovative research to provide readers with the latest research, knowledge, emerging technologies, postharvest, processing and preservation, food safety and environment, biotechnology and bio-discovery of marine and fisheries. The key focus of the research should be on marine and fishery and the manuscript should include a fundamental discussion of the research findings and their significance. Manuscripts that simply report data without providing a detailed interpretation of the results are unlikely to be accepted for publication in the journal.

Abstract

This research aims to screen and to identify marine bacteria able to degrade Polyaromatic Hydrocarbons (PAHs). 7 bacterial isolates were screened for the ability to degrade 6 PAHs (phenanthrene, naphthalene, dibenzothiophene, fluorene, phenothiazine, and pyrene). This screening step was conducted by sublimation method, in which the color change or clear zone appearance was used as an qualitative indicator to show the capability of the selected isolate to degrade PAHs. Two bacterial isolates, designated as LBF-1-0102 and LBF1-0103, showed capability in degradating the PAHs tested. In particular, the isolate LBF-1-102 was potential in degrading phenanthrene, naphthalene, dibenzothiphene, fluorene, phenothiazine, and pyrene.The isolate LBF-1-0103  showed the ability in degrading 2 (two) PAHs, i.e. dibenzothiophene and  fluorene. Based on partial 16S rDNA analysis, LBF-1-0102 and LBF-1-0103 were identified as Pseudomonas balerica and Brachybacterium sp., respectively.

Abstract

Dietary fiber is one of the most essential food component, mainly used to maintain health andbalance of digestive system. Dietary fiber plays an important role in health, especially in reducingblood cholesterol, improving glucose absorbtion for diabetic patients, preventing colon cancer,and reducing weight. Several studies showed that seaweed is a potential source of dietary fiberwith more advantages compare to that of land-based crops. Contrary to its production, seaweedconsumption in Indonesia is relatively low. To date, dietary fiber intake for the majority of Indonesiansis fulfiled from land-based crops.

Abstract

Marine algae is known to contain a wide variety of biomedical compounds having pharmaceutical applications. The aim of this research was to evaluate cytotoxic activity and apoptosis induction of Turbinaria decurrens extract on T47D cell lines.  Cytotoxic activity test was conducted by using MTT assay whereas detection of apoptosis was evaluated by DNA fragmentations and flow cytometry analysis. The MTT test showed that crude extract had medium cytotoxic activity to T47D, HepG2, and C28 cell lines with IC50 value of 172, againts 360 and 330 µg ml-1, respectively. After solvent partition of crude extract, the cytotoxic activity of n-hexane and ethyl acetate fractions T47D cell increased, the cytotoxic activity of n. hexane and ethyl acetate fractions T47D cell increased with  IC50  value of with IC50  43.1 and 51.9 µg ml-1, respectively, whereas IC50 value of methanol fraction was 383.0 µg ml-1. Analysis of DNA fragmentation of T47D cell showed that  both n-hexane and ethyl acetate fractions could not fragment DNA as a features of apoptosis. However, flow cytometry analysis by using annexin-V and propidium iodide staining revealed that n-hexane and ethyl acetate fractions could induce apoptosis in T47D cell. This research indicated that Turbinaria decurrens had potency to induce apoptosis in T47D cells.

Abstract

Previously designed endpoint PCR has been adapted for use with real time PCR to detect the presence of Salmonella typhimurium and Salmonella enteritidis  in fish. Optimization of a standard curve in the presence of herring sperm DNA as background matrix indicated that the real time PCR highly efficient with the Pearson coefficient of determination (R2) value = 0.99937 and slope (M) value = -3.44. An enrichment method (overnight culture) significantly increased (p0.05) the sensitivity of real time PCR. Comparison of real time PCR and the conventional isolation method based on biochemical tests has been conducted. In terms of their sensitivity, real time PCR and the conventional methods are not significantly different in the level of confidence 95%. Both real time PCR with enrichment method and conventional biochemical method can detect the presence of Salmonella spp. in spiked sample. However the direct extraction method was only detecting the presence of Salmonella in higher concentration. While the sensitivity both conventional and real time PCR are similar, the real time PCR has an advantage to detect the pathogen qualitatively and quantitatively depending on processing method.

Abstract

Emerging pollutants (EPs) or contaminants of emerging concern (CECs) have become global awareness since few decades ago, including in Indonesia. Intensive usage of industrial compounds has led to the massive emission to the environment and therefore their potential adverse effects may endanger aquatic organisms and human health. Based on the available literatures, organotins and flame retardants are two main groups of EPs from industrial emission identified in Indonesian seafood, as well as in those worldwide. However, concentration of both EPs group detected in Indonesian seafood is relatively low than that in developed countries and the majority of South East Asian countries. Toxicological studies revealed that EPs emitted from industrial activities have to be concern, as most of the EPs attributed to endocrine disrupting chemicals. Nevertheless, study on the exposure assessment of EPs in Indonesia is very limited. Even though, the concentrations of EPs in Indonesian seafood produce exposure below tolerable daily intake (TDI) to the local consumer, the long term exposures have to be aware due to their possible continues emission and elevating concentration such as shown by flame retardants. Governmental regulation, monitoring programs, and laboratory approach are among issues to be concern in addressing EPs in Indonesia, especially from industrial emission.

Abstract

Two species of Caulerpa, locally known as “Pedesan” and “Latuh” have been traditionally consumed by coastal communities at Binuangeun, Banten. This study aimed to identify “Pedesan” and “Latuh” using the DNA barcoding method and to evaluate their nutrient and heavy metal contents. Fatty acids were determined by Gas Chromatography Flame Ionization Detector (GC FID), amino acids using Ultra Performance Liquid Chromatography (UPLC), and minerals using Inductively Coupled Plasma Optical Emission Spectrometry (ICP OES). Based on the tufA gene sequences, the “Pedesan” was identified as Caulerpa racemosa var. macrophysa and “Latuh” as Caulerpa chemnitzia. Thirteen fatty acids were detected in C. racemosa var. macrophysa and twelve fatty acids in C. chemnitzia. Of the total fatty acid content, C. racemosa var. macrophysa contained 41.0% unsaturated fatty acids, dominated by linolenic acid and eicosapentaenoic acid. Meanwhile, C. chemnitzia contained 47.5%, dominated by oleic acid. Both seaweeds contained fatty acids with the w6/w3 ratio lower than 10, which could prevent heart disease risk based on World Health Organization (WHO) recommendation. The primary amino acids content in C. racemosa var. macrophysa were glutamic acid, alanine, serine and aspartic acid, while those in C. chemnitzia were glutamic acid, serine, aspartic acid, and glycine. The high content of glutamic acid in both samples indicated their potential use as food flavor enhancer. The Na/K ratio of C. racemosa var. macrophysa (40.31) and C. chemnitzia (27.48) were higher than those recommended by WHO. Heavy metals were not detected in either “Pedesan” nor “Latuh”, indicating that they are safe for consumption.

Abstract

Extraction and partial characterization of lectin from Indonesian Padina australis and Padina minor had been carried out. The crude extract of the P. australis and P. minor were examined for hemagglutination activity (HA) using native and trypsin-treated of rabbit and human A, B, O type erythrocytes. Both extracts agglutinated all of the trypsin-treated erythrocytes tested in the HA assay. Strong HA was detected in the crude extract of P. minor with trypsin-treated of human type A and O erythrocytes. However, the sugar-binding specificity study through the quantitative hemagglutination inhibition (HI) assay showed that P. minor extract could not specifically recognize the glycans tested. Apparently, the HA of the P. minor was more due to its co-extracted polyphenols content than its lectin content. On the other hand, the HI assay showed that asialo transferrin human (aTf) and asialo porcine thyroglobulin (aPTG) were the most powerful in inhibiting the HA of P. australis. Those indicated that P. australis protein extract was able to specifically recognized aTf and aPTG. The stability of P. australis and P. minor HA over various temperatures, pH ranges, and divalent cations studies showed that the P. minor HA was stable on a wide range of pH and temperature; not affected by the presence of EDTA, but decreased by Ca2+ and Mg2+ additions showed that P. minor protein extract  was not a metallic protein. The HA of P. australis decreased at 60 oC and was inactivated at 90 oC; increased at strong acidic (pH 3 4) and strong basic (pH 9 10) and dependent by the presence of either EDTA or Ca2+ and Mg2+ divalent cation.
