Abstract

Purwonegoro Fish Market, Banjarnegara district is the largest fish market in Central Java southern and western parts. This fish market serves as a center for buying and selling fish and fish seed traffic in a particular place to the location of the fish farmers. It will possibly spread a particular type of disease from one area to another. Trichodina sp. is found in the seed of gouramy fish hatchery center area with the pathogenicity level reached 80%. Trichodina sp. has a big role in the cultivation of gouramy production by reducing the physical condition of the fish so dangerous and cause death of gouramy in the seed phase. The results showed that as many as 150 gouramy fish fry were examined, 56 gouramy fish fry were infected Trichodina sp. All of Trichodina sp. are found as many as 573 individual from the gouramy seeds infected. Total intensity of Trichodina sp. from gouramy seeds amounted to 10.97 individual. The measurement results morphometric characters Trichodina sp. show the variation of morphometric with body diameter from 47.5 to 112.5 μm; denticle ring diameter from 22.5 to 47.5 μm; denticle diameter of 12.5 to 42.5 μm, adhesive disc diameter 37.5 to 65 μm; membrane width 2.5-7.5 μm and the number of denticle 13-28. Trichodina sp. allegedly found there are three types of T. heterodentata, T. nigra, and T. acuta.

Abstract

Filariasis limfatik merupakan penyakit disebabkan oleh cacing filaria Wuchereria brancofti, Brugia malayi, dan Brugia timori yang menyerang saluran dan kelenjar getah bening. Cx. quinquefasciatus merupakan vektor filariasis bancrofti di daerah urban. Kecamatan Cisayong endemis filariasis dengan Microfilaria rate >1%. Eliminasi  filariasis di  Indonesia ditetapkan dua pilar kegiatan yaitu pemberian obat massal pencegahan filariasis (POMP filariasis) di daerah endemis dan pengendalian vektor. Pengendalian vektor dan potensi vektor dalam penularan filariasis dapat diketahui melalui berbagai hal diantaranya dengan mengetahui spesies Culex, infection rate, parity rate dan kepadatan larva Culex.Tujuan penelitian yaitu mengetahui spesies nyamuk Culex, infection rate, parity rate, dan kepadatan larva nyamuk Culex. Metode yang digunakan dalam penelitian ini yaitu metode survei dengan teknik cluster sampling. Kelompok (Cluster) dalam penelitian ini yaitu RT berjumlah 39 RT yang ada di Desa Cisayong. Penangkapan sampel nyamuk dilakukan di 9 kelompok (RT) yang terpilih. Pengambilan sampel dilakukan pada malam  hari pukul 18.00-00.00 WIB. Hasil penelitan menujukkan bahwa spesies Culex yang ditangkap di Desa Cisayog diantaranya Cx. quinquefasciatus, Cx. hutchinsoni , Cx. sitien , Cx. vishnui , Cx. pseudovishnui , Cx. tritaeniorhynchus dan Cx. gelidus, infection rate 0%, sementara itu, parity rate Cx. quinquefasciatus 66%, Cx. hutchinsoni 17%, Cx. sitien 68%, Cx. vishnui 55%, Cx. pseudovishnui 21%, Cx. tritaeniorhynchus 53%, dan Cx. gelidus 54%. Kepadatan larva Culex sp. yaitu 3,38/cidukan.

Abstract

Begonia hirtella leaf are commonly used by people in mountainous areas to treat itchy due to insect bites and skin infection. This study aims to (1) determine proper solvent extract of B. hirtella leaf on antibacterial activity of S. aureus and E. coli and on antifungal activity of C. albicans (2) determine the lowest concentration of leaf extract of B. hirtella that indicate the formation of inhibition zone (3) know the content of bioactive compounds contained in each leaf extract of B. hirtella. The method used in this study is experimental with completely randomized design (CRD). The treatments were type of solvents (n-hexane, ethyl acetate, ethanol and water) and a serie of the ethyl acetate extract concentration of 500 ppm, 450 ppm, 400 ppm, 350 ppm, 300 ppm and 250 ppm against S. aureus, E. coli and C. albicans. Data were analyzed using analysis of variance (Anova) and the significant differences between the treatments were analyzed by Duncan test at 95% confidence level. The result showed that each solvent extracts affect microbes growth with a highly significant difference p <0.05. Solvent extracts that have the best inhibitory zone is ethyl acetate against S. aureus with an average diameter of inhibitory zone 13.75 ± 1.26 mm. Increasing concentrations of ethyl acetate extract of 250 ppm to 500 ppm increase inhibition zone against microbes. The lowest concentration that show inhibition zone was 300 ppm. The formation inhibition of zone on microbes growth happened due to their compounds in the extract. Extract of n-hexane contains stigmasterol, ethyl acetate contains neophytadiene, while the ethanol extract contains ethyl palmitate

Abstract

Amoxicillin is one of the antibiotics that commonly used on treatment of H. pylori infection. pbp1A gene mutation in H. pylori is a dominant cause of amoxicillin resistance. This study was aimed to evaluate the presence of H. pylori in patients with dyspepsia by using non-invasive method i.e. H. pylori stool antigen (HPSA) and invasive method i.e. pbp1A gene amplification, and also evaluate the amoxicillin resistance of H. pylori by assessing the pbp1A gene mutations. The samples were  26 faeces and 26 gastric biopsies of patients with dyspepsia from the Internal Disease of Prof. Dr. Margono Soekardjo Hospital in Purwokerto. DNA amplification performed by using polymerase chain reaction (PCR) to detect the presence of amoxicillin resistance encoding gene i.e. penicillin binding protein (pbp1A). Sequencing of the DNA sample was performed at the First Base Malaysian Company, to analyze the existence of a point mutation. DNA sequencing analysis of 12 samples showed the presence of a mutations in pbp1A gene from 2 samples, in the third motive of pbp i.e. amino acid changes, Alanine 599 substituted to Threonin and Threonin 592 to Alanine. Those mutations become a dominant risk factor for resistance of H. pylori, toward the bacterial peptidoglycan synthesis. In this research, it was known that the detection of H. pylori infection by using PCR remains more accurate and specific method. The presence of H. pylori mutant strains in this study may becomes the risk factors of resistance to amoxicillin treatment.

Abstract

Laporan ini memverifikasi keberhasilan protokol tiga tahap ginogenesis tawes melalui inaktivasi genom jantan dengan iradiasi ultra violet (UV), fertilisasinya, dilanjutkan diploidisasi zigot dengan kejut panas (400 C) selama 60 detik. Eksperimen Rancangan Acak Lengkap, tujuh perlakuan yaitu, kontrol positif (fertilisasi normal); kontrol negatif1 telur difertilisasi dengan milt encer yang di UV 1983,348 J/m2; kontrol negatif2 dosis iradiasinya 3966,96 J/m2; ginogenesis1 kontrol negatif1 lalu dikejut panas pada 10 menit pasca fertilisasi; ginogenesis2 ginogenesis1 beda waktu kejut pada 15 menit pasca fertilisasi; ginogenesis3 kontrol negatif2 lalu dikejut pada 10 menit pasca fertilisasi; dan ginogenesis4 ginogenesis3 beda waktu kejut pada 15 menit pasca fertilisasi. Materi gamet segar diperoleh dari induk yang diinduksi GnRH analog dan domperindon dosis 1,5 ml/kg bobot induk intramuskuler, 6-10 jam sebelumnya. Milt segar diencerkan 100 x dalam larutan Ringer. Data dianalisis dengan uji Anova, menggunakan program SPSS. Hasil penelitian menunjukkan bahwa data fertilitas, data penetasan dan data kelangsungan hidup yang terbukti homogen (p>0,05), menunjukkan bahwa ketujuh perlakuan secara statistik fertilitas, penetasan, dan kelangsungan hidupnya tidak nyata (p>0,05) antar pelakuan. Data persentase morfologi larva abnormal tawes menunjukkan bahwa perlakuan yang dicobakan memberikan pengaruh yang sangat nyata (P<0,01). Secara keseluruhan dapat disimpulan bahwa, keempat perlakuan mitoginogenesis yang diujikan efektif menghasilkan larva mitoginogenesis ikan tawes walaupun efektifitasnya masih tergolong rendah. Efektivitas perlakuan dibuktikan dari signifikansi data morfologi larva abnormal dariklompok kontrol negatif membuktikan efektivitas dosis inaktivasi genetis jantan dan morfologi larva normal hasil perlakuan ginogenesis membuktikan efektivitas diploidisasi kejut panas tidak berbeda dari morfologi kontrol positif.

Abstract

INH mempunyai peranan dalam menghambat sintesis asam mikolat yang merupakan komponen pembentuk dinding sel M. tuberculosis. Mengetahui pengaruh pemaparan obat INH terhadap karakteristik M. tuberculosis secara morfologi sel. Metode penelitian eksperimen dengan menggunakan Rancangan percobaan acak Lengkap dengan variasi waktu pemaparan 24 jam, 48 jam, 72 jam dan 96 jam pada konsentrasi INH 0,5 μg/ml dengan jumlah ulangan 6. Hasil penelitian menunjukkan ada pengaruh pada masing-masing perlakuan INH 0,5 μg/ml terhadap karakteristik M. tuberculosis, pada pemaparan 24 jam diperoleh persentase pertumbuhan 18,25%,  ukuran rerata sel 1,64 x 0,35 µm, persentase kerusakan sel 28,1%. Pada pemaparan 48 jam diperoleh persentase pertumbuhan sel  7,9%,  ukuran rerata sel 1,64 x 0,36 µm, persentase kerusakan sel 41%, pada pemaparan 72 jam diperoleh persentase pertumbuhan sel 7,0%,  ukuran rerata sel 1,64 x 0,32 µm, persentase kerusakan sel 53,4%, pada pemaparan 96 jam diperoleh persentase pertumbuhan sel 1,6%, ukuran rerata sel 1,61 x 0,29 µm,  persentase kerusakan sel 57%. Perubahan morfologi sel yang terpapar INH 0,5 μg/ml pada permukaan sel terjadi kerutan dan penyusutan panjang sel setelah regenerasi, permukaan sel mulai kasar dan sedikit ada kerusakan, sel mengalami lisis dan sebagian sel berbentuk oval , sel mengalami perubahan bentuk menjadi kokus. karakteristik M. tuberculosis yang terpapar INH 0,5 µg/ml mengalami perubahan morfologi sel, menjadi lebih pendek dan perubahan bentuk dari batang menjadi kokus, namun perubahan morfologi ini tidak terjadi secara permanen pada generasi berikutnya.Kata kunci       : M. tuberculosis, INH, Morfologi,

Abstract

Karsinoma nasofaring (KNF) merupakan tumor epitel yang terletak di nasofaring dan merupakan penyakit genetik multifaktor yang endemik. Penyebab utama KNF adalah infeksi oleh Virus Epstein-Barr (EBV). Keberadaan EBV pada penderita KNF dapat diketahui dengan ditemukannya DNA EBV dalam spesimen biopsi jaringan penderita KNF. Genom EBV dan ekspresi sebagian dari produk gen laten virus secara konsisten terdeteksi hampir di setiap sel dalam kanker ini, salah satunya adalah gen Latent Membrant Protein (LMP). Aktivitas mRNA EBV lebih mencerminkan patogenesis KNF yang  sesungguhnya dari pada diagnosis serologi dan pengukuran DNA EBV di sirkulasi. Penelitian dilakukan di Laboratorium PA RSUD Margono Seokarjo, Purwokerto/Lab PA Fakultas Kedokteran dan laboratorium genetika/molekuler Universitas Jenderal Soedirman Purwokerto. Analisis Ekspresi mRNA LMP2A EBV dengan teknik one step RT-PCR dan produk RT-PCR (amplikon cDNA) divisualisasi dengan elektroforesis gel agarosa 1%. Hasil ekspresi mRNA LMP2A EBV adalah 27,3% (6 dari 22 sampel). Kesimpulan, metode one step RT-PCR dapat digunakan untuk menganalisis ekspresi mRNA LMP2A EBV dari sampel biopsi jaringan KNF dalam blok paraffin dan hasil positivitas ekspresi mRNA LMP2A EBV sedang, sehingga berpotensi digunakan sebagai petanda biologi molekul diagnosis KNF.
