Abstract

The purpose of this research was to investigate the catch composition and catch per unit effort of shrimp captured using ciker net of fishermen in Tegalkamulyan Cilacap district. The sample was collected using ciker net between the first week of February and April 2008. The samples were separated between shrimp and fishes then measured total biomass weight. Individual shrimp was identifi ed, measured their weight and total length. The daily shrimp catch data was grouped based on lunar circulation into new moon, first quarter, full moon and third quarter. The t-test and anova was performed to the means catch between shrimp and fish, and shrimp and group of catch. The results showed that the catch during new moon was significantly higher than full moon, but not significantly different with the first and third moon. The average catch per trip of ciker net composted of shrimp (9.27 kg) which was not significantly different with fish weight (6.8 kg). The shrimp catch composed of fi ve species, namely: Penaeus monodon, Penaeus merguiensis, Metapenaeus ensis, Penaeus longipes and Parapenaopsis sculptilis. The highest average catch of shrimp was P. merguiensis 5.76 kg (62.2%) then followed by M. ensis 2.03 kg (21.9%), while the rest was other shrimp. Based on individual length and weight, P. monodon, P. merguiensis and M. ensis were categorized into big size shrimps, while others were small size. The sex ratio of P. monodon and P. merguiensis was 1:1, while others were not equally to 1:1.

Abstract

The aims of this experiment were to study the effect of depth to the growth and survival rate of pearl oyster (Pinctada maxima) during nursery period. The experiment was conducted in ten weeks from November 2008 to January 2009 at the Pegametan Bay using spat collector. The spat collectors were wrapped using polyethylene net, then hanged and placed in the floating cage. The growth and survival rate of juveniles were measured and counted weekly. The experiment used Completely Randomized Design (CRD) with four treatments and four replications. The treatments were 4 depths that were 0 m (near surface), 5 m, 10 m and 15 m. The results showed that the density of plankton in each depth was significantly different, which lead to the differencies of growth among treatment. The high growth and survival rate were obtained at the depth of 5 m. During ten week in nursery, the shell could growth from 3.0 mm to 36.5 mm in length, and from 2.45 mm to 30.1 mm in width. The survival rate was ranged from 2.68% to 25.15%.

Abstract

The purposes of this experiment were to establish loop-mediated isothermal amplification of DNA ( LAMP) method and its application to observe the presence and duration of koi herpes virus ( KHV) in freshwater fishes after infection. The infection was carried out using 4-6 cm length of java barb (Barbodes gonionotus), grass carp (Ctenoparingodon idella), gold fish (komet) (Carassius auratus), tambaqui (Colossoma macropomum) and koi (Cyprinus carpio koi) fishes. Fishes were infected intraperitoneally with KHV inoculums and 2 fishes were sampled everyday. DNA was extracted from gill and used for diagnosis with LAMP assays. The result of this experiment showed LAMP assay can be established and gave 100 times more sensitive than conventional PCR assay. Koi was confirmed as a host of KHV. Java barb, tambaqui, gold fish and grass carp can serve as vector for KHV and the presence of KHV was detected in java barb and tambaqui for 4 days, goldfish and grass carp for 5 days after infection.

Abstract

This research was aimed to isolate proteolytic bacteria that have high ability to hydrolize protein. The sources of bacterial samples were collected from mangrove sediment in Indramayu and Cilacap Regency. Collwel SWYE Modification Agar medium and Skim Milk Agar Medium, which contain pepton and casein, were used for isolating the bacteria. Forty isolates having proteolitic activity were obtained. Three isolates, i.e. Ps.23.1, Ps.27.3 and Ps.23.2, having the highest activity were selected. The selected isolates were able to grow at pH range of 5 to 8. They were also able to grow at NaCl concentration of 0.5-3.5%. Intramuscular injection test on white shrimp (Lithopenaeus vannamei) shows that Ps.23.1 and Ps.23.2 were not pathogenic to shrimp. On the other hand, Ps.27.3 was indicated to cause disease on shrimp. These results indicated that isolates Ps.23.1 and Ps.23.2 potentially can be used as inoculant for bioremedation of brackish water pond. Isolate Ps.23.1 was characterized to be an aerobic, nonmotile, short rod, gram negative bacteria having catalase and oxidase activities. It produced yellowish, circulair, entire colonies. Isolate Ps.23.2 was characterized to be an aerobic, motile, rod, spore forming, gram positive bacteria having catalase and oxidase activities.

Abstract

In aquaculture, efficiency of feed is very important to decrease cost production. Besides, excess of feed consumptions caused disturbance of water quality which affected on productivity and survival rate. Fasting day interval was one of best chosen method to make efficiency and effectively on feeding schedule. The research was aimed to know the effect on fasting day interval against its growth and survival rate. Experimental design used was completely randomized design with 4 treatments, 3 replication. The treatments were: P1 (1 day given feed, 1 day fasted), P2 (2 day given feed, 1 day fasted), P3 (3 day given feed, 1 day fasted), P4 (given feed everyday/control). Red nile used gave length 7–10 cm and weight 10 - 19 g were placed on cement pond 1 m2 of sized. Fish density was 20 fishes/m2 with fed consumption about 3% daily of total biomass weight. Result showed that treatment using fasting day interval on red nile fish did not affect on both compensatory growth and its survival rate. Better growth occurred at P4 which fed intake consumption everyday without fasted. While P1 revealed same result (p>0,05) on survival rate with P4.

Abstract

Production of high quality of shrimp broodstock needs proper domestication and improvement of their genetic quality. MCBAD Jepara has conducted research to evaluate genetic heterogeneity of Penaeus monodon broodstock F1 using Restriction Fragment Length Polymorphism (RFLP) and Random Amplified Polymorphic DNA (RAPD) analysis. For RFLP analysis, amplification product of 16SrDNA of mitochondrial DNA was digested with restriction enzymes. According to the RFLP analysis, heterogeneity value of P. monodon F1 broodtock population was 0.0422; male F1 population was 0.0613 and female F1 population was 0.1252. The primer used in RAPD analysis was OPA 2. According to the RAPD analysis, heterogeneity value of P. monodon F1 broodstock population was 0.0417; male F1 population was 0.0653 and female F1 population was 0.1104. The results from these researches have shown that either RFLP or RAPD can be used as a family specific marker for Penaeus monodon.

Abstract

The purpose of this study was to know the distribution of nutrients and their effect to the distribution of phytoplankton in the eastern area of Belitung water. Sampling was done on October 2006. Water samples were collected from the surface and at depth 10 m among 10 stations. For chemical and physical properties, samples were collected from both of surface and at depth 10 m while for phytoplankton content, samples were collected from surface water. Samples were analyzed for phosphate, nitrate, nitrite, ammonia, silicate, pH, dissolved oxygen, temperature, salinity and concentration of plankton. The result showed that the concentration of phosphate, nitrate, nitrite and silicate were tended to decrease followed to the distance of station from the shore line. Dissolved oxygen and phytoplankton were tended to decrease as the depth. There was positive correlation between salinity and concentration of plankton. It led to conclusion that the water was still suitable for living biota.

Abstract

The objectives of this research were to identify population character of balashark fish in Musi River base on morphometric character and to determine which were the main predictor as morphometric characters.This research was conducted in 2007 in Musi River South Sumatra Province. Samples were selected base on purposive sampling method, at sum 45 specimens for morphometric observation. Measurements were made on 22 morphometric characters on the left side of body. Morphometric data were subject to PCA (Principal Component Analysis) and Discriminate Analysis using Statistic version 6.0 softwareThe result showed base on morphometric character analysis of balashark population in Musi River, there were three different subpopulations as reference of unit management of balashark fishes in Musi River. As the main character predictor among the groups (subpopulation/group 1, 2 and 3) are IOW (Inter Orbital Width/22), DFL (Dorsal Fin Length/7) and PDL (Peduncle Length/4). First group (Group 1) has longest similiarty distance (most difference group) compared with others subpopulation (Group 2 and Group 3).

Abstract

The purpose of this research was to identify the condition and diversity of hard corals in Nusalaut Island. The research was conducted in five locations of Nusalaut Island, Central Maluku District, namely Nalahia, Ameth, Titawai, Abubu and Akoon. Sampling by LIT (Line Intercept Transect) method was applied in December 2007. During the research, 123 species consisting 15 families were found. Hard coral was classified into midle to very good catagory, and the percentage of coverage was 26.96 to 80.08 %.  Akoon island was good location and the diversity index was found 1.2 and  evenness index was 0.69 respectively.  In general, Nusalaut Island  was dominated by Porites lutea with value 10.64% and Acropora formosa with value was 8.01% from  all components.

Abstract

The Aims of this research were to know physical and chemical characteristic and plankton abundance of water in Lake Sentani. The research was done on September and November 2006 with stratified survey method. Samples were collected from four stations, namely Yope Bay, Butali Bay, Asei Besar and Kampung Harapan, at 0, 2, 4 and 8 m depth. Parameters were analyzed as temperature, transparency, dissolved oxygen, alkalinity, free CO2, N-NO2, N-NO3, P-PO4 and total organic matter. Water quality in Yope Bay, Butali Bay, Asei Besar and Kampung Harapan were show the same character. The averages of dissolved oxygen, total alkalinity, transparency were tend to high but free carbondioxide was low. The highest average of the phytoplankton abundance was found in Kampung Harapan about 104.423 ind/L and the highest average of zooplankton abundance was found in Asei Besar about 1.383 ind/L. Phytoplankton from Chlorophyceae dan zooplankton Copepoda were always found at every sampling stations.
