Abstract

The use of local probiotics in the culture of aquatic organisms is increasing with the demand for more environmental-friendly aquaculture practices. The local bacterium isolate considered as a probiotic was added into the water of whiteleg shrimp (Litopenaeus vannamei) culture in a field trial. Four rectangular plastic ponds (ca. 20 m x 30 m per pond) were used for 100 days experimentation for six consecutive crops in two years experiment. Survival, harvest size, feed conversion ratio (FCR) and Vibrio bacterial count was compared with those of shrimp receiving and none of local isolate. Identification based on 16S rRNA gene sequence shown those isolate was Bacillus pumilus strain DURCK14 with 99% homology. Water shrimp pond added a local isolate had significantly higher survival at about 10.0% to 11.7% than shrimp without added the isolate (p<0.05), and better FCR, but no significant different in shrimp harvest size. Vibrio bacterial was undetected by total plate count. Moreover, it shown better projected yields on an annual basis (three crops per year).

Abstract

The purpose of these observations was to identify the stages of the embryo development of ikan palmas (Polypterus senegalus senegalus) and determine the length of duration of each stage. Broodstocks were cultured in aquaria 6 cm x 72 cm x 50 cm in size. The broodstock were stocked at ratio of 1:1 and fed ad libitum with earthworm, small feed fish (ting sea fish) and golden snail. The Palmas broodstocks here naturally spawned in artificial nests made of split plastic raffia in resembling the aquatic plant found in the natural habitat of ikan palmas. After 21 days of culture period, the broodstock began to lay eggs in gradually for 20 days. The average diameter of the eggs was 25 μm. The embryo developed in 24 hours after fertilization and hatched out three days after the embryo had developed.

Abstract

Mariculture is an important component of Indonesian fisheries and aquaculture production, directly contributing an estimated US$ 320 million in 2008. Because most mariculture production is focussed on producing for export markets, mariculture production is an important source of foreign earnings for the Indonesian economy. This paper reviews the current status and prospects for continuing development of mariculture in Indonesia. Currently the major mariculture commodity in Indonesia is seaweed for carrageenan production. Seaweed production accounts for 98% of total Indonesian mariculture production and 84% of value. The other major commodity groups are marine finfish and pearl oysters. Commodities being developed for mariculture in Indonesia include abalone and spiny lobsters. Prospects for continued development of mariculture in Indonesia appear positive. Indonesia has several advantages for mariculture development, including many potential mariculture sites, a stable tropical climate, and does not suffer from cyclonic storms. The Government of Indonesia is planning to increase aquaculture production substantially over the next four years, including mariculture production. Globally, demand for seafood products is expanding due to increasing population and increased per capita consumption of fish products. Constraints to the continued development of mariculture in Indonesia include: limited seed supply, particularly of species which cannot be economically produced in hatcheries, such as spiny lobsters; need to develop more efficient production systems for some marine finfish; the need to improve environmental sustainability by improving feeds and reducing environmental impacts; and market issues relating to environmental sustainability.

Abstract

Genetic diversity is an important aspect of a selective breeding program to produce fish broodstock carrying superior traits such as fast-growing, disease resistant, and other traits. We have carried out a breeding program to produce a fast-growing striped catfish (Pangasianodon hypophthalmus) since 2010. The aim of this study was to evaluate the genetic variation of the first (G-1) and second (G-2) generations of fast-growing striped catfish using microsatellite analysis. The G-1 and G-2 populations were selected individually from populations. DNA samples were collected from 40 ind. fish of each population and analyzed using five microsatellite loci (Pg1, Pg2, Pg3, Pg13, and Pg14). The results showed that the number of alleles per loci in the G-1 and G2 populations ranged from 4 to 7 alleles, with an average of five for each generation. The average of observed heterozygosity of the G-1 population (0.420) was lower than the G-2 population (0.495). Inbreeding level showed that the G-1 population was more inbred than the G-2 population. The study also found that both striped catfish populations had relatively low genetic variation. This result suggests that monitoring of genetic variation and better scheme of good spawning were needed on the next selection program to produce the intended fast-growing striped catfish.

Abstract

Organic and inorganic pollutants such as N, P, and heavy metals are a serious problem in water bodies (lake, reservoir, river, and stream) and have deleterious effects to pond productivity and human health. These pollutants produced from anthropogenic activities (i.e. industrial, agricultural, and settlement) are released into the water bodies and causing poor water quality. Constructed wetland (CW) is one of the technologies that have the capability to solve such problems. The purpose of this experiment was to examine the efficiency of constructed wetland in terms of reducing pollutants from the stream as a water resource for aquaculture area and increasing fish production. Two kinds of treatments were set up: one fish pond culture with constructed wetland and the other one without constructed wetland (control). Each treatment consisted of three replications. The result showed that water quality in the fish pond with CW was better than the control. Constructed wetland were able to reduce ammonia (NH3-N), nitrite (NO2-N), nitrate (NO3-N), TN, phosphate (PO4-P), TP, Cu, Pb, and As concentrations to 15.00%-88.27%, 9.52%-72.73%, 11.11%-57.14%, 20%-66.67%, 24.44%-80.77%, 12.04%-77.95%, 50.00%-100%, 56.25%-100%, and 46.83%-100%, respectively. Nile tilapia cultured in pond using CW was higher than the control in terms of total length, body weight, and survival rate. The t test revealed that nile tilapia (Oreochromis niloticus) cultured in pond with CW and the control has significant difference (P<0.01) in terms of total length, body weight, and survival rate. This is the evidence that constructed wetland technology is very useful on improving water quality and increasing pond productivity of nile tilapia cultured in the freshwaterpond.

Abstract

Spiny lobster (Panulirus homarus and Panulirus ornatus) are important commodities for Indonesia. The aquaculture of lobster is susceptible for several diseases like parasite, fungi, bacteria, and virus. Among those diseases, milky haemolymph disease (MHD) is often seen as a symptom to mass mortality occurred at lobster farms in Gerupuk Bay of Lombok. The purpose of this study was to determine the lobster diseases on cage culture in Gerupuk Bay of Lombok, West Nusa Tenggara. The study was undertaken from January to March 2015. Diseases status was determined by application of molecular plat-form, polymerase chain reaction (PCR) with designation of specific primer for MHD (254F/R), 254F: 5’-CGA-GGA-CCA-GAG-ATG-GAC-CTT-3’ and 254R: 5’-GCT-CAT-TGT-CAC-CGC-CAT-TGT-3’ with PCR size product of 254 bp. and for cloned the pathogen was used TA-cloning Invitrogen for the DNA plasmid as positive control for other analysis. Several tissue samples i.e hepatopancreas, haemolymph, part of muscle hepatopancreas P. homarus and P. ornatus were taken from cage culture farms at Gerupuk Bay then preserved on 90% ethanol for further analysis by PCR and then the amplificated DNA were cloned into pCR®2.1 plasmid and transformed into competent E. coli. The result showed that almost all lobster samples from Gerupuk Bay were positive infected by MHD, as the results of PCR amplification whereas the band appeared at 254bp. Also MHD plasmid has been successfully cloned and will be used for further examination. Histopathologically in hepatopancreas infection have seen necrosis that contain numerous of rickettsia-like bacteria.

Abstract

Seaweed is an important aquaculture commodity that could contribute on climate change mitigation, related to its ability on absorbing CO2, as one of the green house gases, through photosynthesis. This study aimed to analyze seaweed potencies on carbon sequestration in the context of climate change mitigation while still resulting optimum production as primary purpose and to analyze the carrying capacity of Gerupuk Bay in order to manage sustainability of seaweed aquaculture. Seaweed, (Kappaphycus alvarezii) was cultivated with long-line system in Gerupuk Bay, West Nusa Tenggara, during five months for three cultivation cycles. Samplings were conducted at days-15, 30, and 45 with CO2 absorption rates as main parameters. Water carrying capacity was calculated to determine the ability of Gerupuk Bay waters for supporting development of sustainable seaweed aquaculture. The results showed that absorption rates of CO2 by seaweed (K. alvarezii) were different at each sampling days of cultivation periods; the highest value was at 10-20 days of cultivation. CO2 absorption analysis resulted based on sampling days of cultivation period could be appl ied to formulate the strategies for management of sustainable seaweed aquaculture, with optimal production and positively contributed to the environment. However, waters carrying capacity should also be considered as major aspect in the application of seaweed cultivation management, thus it can run continuously without causing conflicts with other interests.

Abstract

The objective of this experiment was to find out the appropriate dosage for producing homogametic female of giant freshwater prawn (Macrobrachium rosenbergii de Man) by feminization. This experiment was carried out at Research Institute for Freshwater Fish Breeding and Aquaculture, Sukamandi. The giant freshwater prawn at PL-5 stage was treated orally by estradiol-17ß. Estradiol-17ß was given orally. The dosage levels are 0, 30, 50, and 70 mg/kg of feed. The hormonal feed was given for 30 days. Prawns were reared in aquaria for 30 days and hapas for 60 days. The parameters observed are sex ratio, survival rate, total length, and body weight. The results showed that estradiol-17ß increased production of female giant freshwater prawn. The best dosage was 70 mg/kg of feed that could produce 65.33% ± 5.64% of female. Survival rate and growth of giant freshwater prawn were not affected by estradiol-17ß (P<0.05).

Abstract

Big size betta (Giant) have a high economic value compared to normal size betta, and over expression of growth hormone gene can produce giant fish.  As an initial step of giant transgenic betta productions, this study was conducted in order to obtain DNA plasmid concentration which provide higher hatching and survival rate of betta larvae.  Construction of PhGH pCcBA gene contains growth hormone gene of Siamese catfish (PhGH) and it is controlled by the CCBA promoter. Betta imbellis broodstocks were spawned naturally, and embryos were collected 1-2 minutes after spawning time. One hundred embryos were dipped in 2 mL of transfectan X-treme gene which containp CcBA-PhGH construction genes (50 µg/mL), on room temperature for about 30 minutes. Treatments on this study were different transfectant : DNA plasmid ratiosnamely:A (0,75 µL: 0,25 µL); B (0,75 µL : 0,50 µL); C (0,75 µL: 0,75 µL), D as Control 1(without transfectant, 0,25 µL DNA); E.as Control 2(0,75 µL transfectant, without DNA), and Fas control 3 (without transfectant and without DNA). Every treatments was repeated three times.  Transfection embryos were hatched on a container (1L Volume). Study results showed that hatching rate and larvae survival rate  (4 days after hatching) on treatment A were the same with the control, but slightly higher than B and C treatments. PCR analysis with DNA template showing that PhGH gene were found on embryos and larvae (pooled sample) of treatment A, B and C. Furthermore, RT-PCR analysis showing the existence of mRNA PhGH expression on embryos and larvae (pooled sample). Therefore, embryo transfection with transfectant ratio 0,75 µL and  DNA 0,25 µLshowing the best results.

Abstract

The use of organic mineral (OM) has been recently introduced in aquaculture both as feed supplement and water quality improvement. A feeding experiment was conducted to evaluate a response dose of OM on growth, survival, and mineral content in whole the body and carapace of vannamei shrimp (Penaeus vannamei). Three diets were supplemented with different levels of organic mineral at 1 (OM1), 2 (OM2) and 4 (OM4) g/100 g diet. Positive control was a diet without OM inclusion but supplemented with commercial mineral mixture at level of 4 g/100 g diet. Juvenile vannamei shrimp with average initial body weight of 3.5±0.1 g were stocked into 12 tanks with a capacity of 200 L. After 75 days feeding trial, highly significant weight gains was observed in shrimp fed OM at all levels compared to the positive control. However, no significant differences were found among dietary OM groups. The growth response was clearly shown by the same values of SGRs in the three OM supplemented groups (1.1%/d) and only differed significantly from positive control. Increasing of dietary OM significantly improved survival rate of shrimp where the highest was observed in group fed OM1 and the lowest was in control diet. Effect of dietary OM on whole body Ca and P were quite similar while whole body Ca and P content of OM1 group was slightly high and tended to decrease in two groups with higher level dietary OM. However, no significant differences among the treatment groups. A clear response of supplementing OM in diet was detected on whole body Zn content. Increase of dietary OM resulted in an increase of Zn content in whole body. The effect was clearly shown when diet contained 2% and 4% OM. Carapace Ca content was highly significant when diet contained 2% OM. Similar to whole body Zn content, there was also a linear trend of response dose of dietary OM on carapace Zn content which the highest was found in dietary OM4. Based on growth, survival rate, and Zn content in whole body and carapace, dietary OM at 1 g/100 g diet can replace mineral mixture as mineral source in diet of vannamei shrimp.
