Abstract

Intraspecific crossing of giant gouramy was done to get better hybrid population. This study was conducted to evaluate the hybrid vigour or heterotic value of the hybrids line, especially for growth rate character. Besides that, we want to know the effect of hybridization on performance of phenotypic variability. Two days post hatching age larvae of four populations i.e. Bastar and Blue-saphire strain, F-1 hybrid and F-1 hybrid reciprocal, were used in this experiment. All fish were reared in aquaria for 30 days period in indoor hatchery. The fish were fed with live food such as Moina and Daphnia for the first ten days, followed by Tubifex for the second ten days and finally, commercial feed were given until the end of the study. The collected data were total length, standard length, and body weight of each population. The results showed that the phenotypic variability of the hybrid population was higher than that of inbred population. Hybrid population from female of Bastar and male of Blue-saphire has better growth than both of its parents. Heterotic values of total length, standard length, and body weight of the hybrid population was 9.5%, 2.9%, and 15.6% respectively. Maternal effect was detected in hybrid population.

Abstract

Giant gourami (Osphronemus gouramy Lac.) is one of freshwater fish having high economic value and very popular in Indonesian food restaurant. This fish have been cultured throughout Indonesia. Since two years ago, cultured fish in ponds infected by unknown pathogenic agent and may cause diseased fish and mass mortality. Mass mortality of pond cultured giant gourami occurred in West Java, Central Java, and Bali-Indonesia. The symptoms of diseased fish are the body color becomes darker, resting on the water surface, and internal organs of fish characterized by enlargement of spleen. In the present study diseased fish samples were diagnose by Polymerase Chain Reaction (PCR) techniques using specific primers of red sea bream iridovirus (RSIV) and histopathologically analysis to determine the causative agent of mortality. Based on PCR analysis, show that all fish sampled were positive infected by megalocytivirus and infected fish mostly died. This is a new and first time reported virus infection in cultured giant gourami in Indonesia and it is proposed to be named as giant gourami iridovirus (GGIV).

Abstract

Red snapper, Lutjanus sebae is favored in mariculture activities because it has a relatively good market and price. Technology for big scale seed production of this species has been developed and is now adequate to supply seed for grow-out activities. However, the availability of artifical diets for L. sebae is still a major constraint for grow-out production. Data on optimum dietary protein and lipid requirements for this fish as a basic information in feed development is not available yet. The objective of the present study was to find out dietary protein and lipid requirements for juvenile of L. sebae. A 70-day feeding experiment was conducted in 24 fiberglass tanks, 200 L volume. Each tank was equipped with a flow-through water system. Twenty five hatchery-produced juveniles of L. sebae (43.1 g BW) were randomly selected and stocked in each tank. The fish were fed with the experimental diets twice everyday at a level of 3% of biomass for the first 4 weeks, and then 2% of biomass afterward. Twelve experimental diets were prepared in form of dry pellet containing casein and fish meal as the main protein sources. Experimental diet had 4 levels of crude protein (32%, 37%, 42%, and 47%) and each protein level consisted of 3 levels of lipid (7%, 12%, and 17%). The experiment employed factorial method with completely random design using 12 combination treatments and 2 replications for each treatment. Result of the experiment showed that there was no significant effect of dietary protein and lipid on growth, feed consumption, and feed efficiency of tested fish. Growth and feed efficiency of fish fed on diet containing 42% and 47% crude protein were significantly higher than that of fish fed on diet containing 32% and 37% crude protein. High lipid content in the diet (17%) resulted in poor growth and poor feed efficiency. This data indicates that Lutjanus sebae has limited ability to utilize dietary lipid as an energy source. Result of the present study recommends that dietary protein and lipid requirement for good growth of L. sebae should be 42% and 12%, respectively.

Abstract

The aims of this study were to identify and to determine life cycle of marine leech isolated from cultured hybrid grouper “cantik” (Epinephelus fuscoguttatus fx E. polyphekadion m) in the northern Bali waters of Indonesia under laboratory conditions. Observation of the life cycle of the marine leech was done using petri-dishes (9 cm in diameter) arranged into two groups. In group-1, a petri-dish was filled with sterile seawater (with water exchange of 50%-60% every two days) and in group-2, a petri-dish was filled with continuous running water. DNA sequence was aligned with the sequences from GenBank by BLAST program. Results of similarity index with GenBank sequence exhibited that the nucleic acid of the marine leech isolated from the hybrid grouper “cantik” showed high similarity (99%) with Zeylanicobdella arugamensis. One adult leech could produce 1-63 eggs. The eggs were developed into morula, blastula, and gastrula within five days. The early phase of the embryo with daily water exchange treatment started on day-6 and hatched into larvae on day-10. The eggs incubated with continuous running water had hatched faster (eight days). However, not all eggs hatched at the same time. Some of the eggs hatched 1-3 days after the first one. Hatching rate of eggs varied from 2.70% to 100%. The newly hatched Z. arugamensis larva has transparent color and length of 1.0-1.5 mm. On day-6, Z. arugamensis larvae were already seen attaching to the body of the fish. The size of the Z. arugamensis larvae ranged between 3-11 mm on day-9. In that stage, they were able to produce eggs. Therefore, we argue that Z. arugamensis only requires 17 to 22 days to develop into the adult stage.

Abstract

Vertebral development is one of the main indicators of organism growth. The aim of this study was to know the vertebral development of cobia Rachycentron canadum in larval stage (20 day post hatch). Vertebral assay was done with double staining methods. The result showed that cobia larvae from 0 dph up to 5 dph did not have cartilage. On 5 dph up to 10 dph had pre cartilage phase composed by calcium and on 10 dph up to 18 dph were cartilage phase and marked with blue color by alcian blue. Vertebral was formed perfectly as bones on 18 dph marked with red color by alizarin red. On 20 dph, cartilage had been fully transformed to bones, and the segment of vertebral was clearly formed. Measurement showed that length of cobia vertebrae was 20.20±3.90 mm, vertebrae segment was 0.91±0.11 mm and number of vertebral segments were between 25-26 segments.

Abstract

In previous study, the fast growth transgenic founder of African catfish was produced harboring a growth hormone (GH) gene construct containing a stripped catfish growth hormone (PhGH) cDNA. This study was conducted to investigate transgene (PhGH) transmission and expression in F-1 transgenic African catfish. The transgenic founders (female) were crossed with non-transgenic (male) to produce heterozygous F-1 progeny. PhGH gene was detected in the embryo, larvae, and seed of the transgenic F-1 using PCR method. Expression levels of transgene in embryo and larvae were analyzed using real-time quantitative PCR (qPCR) method. The transgene was detected in embryo, larvae and seed of F-1 transgenic African catfish. Founder could transmit PhGH gene to transgenic F-1 lines in ranged 36% to 48%. Expression level of Phgh gene in embryo was higher than that of the larvae; whereas in the embryo was 1.5 x 105 - 5.2 x 105 copies or 0.49-9.82 fold, while in the larvae was 1.1 x 105 - 2.5 x 105copies or 0.19-5.80 fold.

Abstract

Cultured mud crabs (Scylla spp.) are commonly fed with ‘trash’ fish. Insufficient supply, high cost and variable quality of ‘trash’ fish has lead to a need to develop costeffective and environmentally friendly formulated diets. This study was conducted to determine quality of selected feed ingredients as protein sources in mud crab diets based on their nutrient composition and digestibility coefficients for dry matter (ADMD), crude protein, lipid and energy. The digestibility coefficients for ADMD ranged from 82.46% to 89.20%. Animal-based feedstuffs such as shrimp head, tiny shrimp and squid liver meal had higher ADMD values than fish meal. Of the plant-based feedstuffs, soy bean meal had the highest ADMD values (89.20%) and corn gluten had the lowest (82.46%). Corn gluten had the lowest protein digestibility (78.81%) and soy bean meal had the highest (96.05%). The lowest energy digestibility (71.13%) was obtained in corn gluten meal. Soy bean meal had a higher energy digestibility value (98.48%) than fish meal (85.95%). All animal meal sources had similar energy digestibility values (85.86%—92.09%).

Abstract

The Cyca-DAB1*05 allele of major histocompatibility complex class II genes is recently suggested to have a link with the European common carp strain resistant to koi herpesvirus (KHV). In this study, a set of specific primers for Cyca-DAB1*05 was designed and applied as a marker to identify broodstocks of majalaya common carp strain subsequently used as a candidate resistant to KHV infection. From a total of 23 broodstock subjected to PCR analysis, two female and male fish, both having (P) and no Cyca-DAB1*05 (N), were selected and then diallelly mated. Disease resistance of progenies from 10 crosses was determined by a survival analysis in pond rearing and a laboratory challenge-test using cohabitation method. The results have revealed that the average survivals of PxP progenies for pond rearing and KHV challenge test were 86% and 100% higher (P < 0.05) respectively compared to that of NxN fish. Survival rate of PxN/NxP progenies was significantly lower (P < 0.05) than that of PxP fish. Furthermore, PCR analysis showed that almost 91% progenies of PxP crosses seemed to have a KHV resistant gene marker. Thus, this study suggests that the marker is associated with the KHV resistance in majalaya common carp strain, and farming of PxP progenies can be useful to increase common carp production.

Abstract

A breeding program for giant freshwater prawn has been developed in Indonesia to supply good quality prawn seed to industry. To achieve the goal of the program, optimum conditions of both environment and nutrition must be provided. Calcium is the main compound of the prawn carapace, influencing moulting processes, especially during the premating moult. The effect of calcium supplementation in the prawn feed on moulting process and egg quality of giant freshwater prawn, Macrobrachium rosenbergii, was investigated. Experimental units consisted of 300 L plastic tanks stocked with 4 adult prawns comprising 3 female and 1 male. A standard prawn feed containing 30.39% crude protein; 0.82% Ca and 0.47% P was used as the experimental diet. A complete randomized design was‘applied in the study with 5 dosage levels of dicalcium phosphate i.e. 0%, 2%, 4%, 6%, and 8% with 4 replicates. Prawns were reared over 45 days. There were significant differences (P<0.05) in premoult, moult, and egg hatching rate, while no significant differences in intermoult or fecundity were observed (P>0.05). Egg hatching rate increased liniearly from 26.5%±9.9% to 50.8%±10.3% as calcium dosage increased from 0% to 8%. The number of eggs per spawn was not significantly different (P>0.05), ranging from 40,096 to 46,131 for females weighing 30.19 to 32.94 g. The results of this study suggest that giant freshwater prawns require dietary calcium supplementation to support moulting process and egg quality when reared in soft water.

Abstract

Acid sulfate soils (ASS) contain sufficient pyrite which, when oxidised following excavation for brackishwater aquaculture ponds, will generate acid and mobilise toxic metals. Production in affected ponds can be low due to poor growth of shrimp and fish, mass mortalities of stock and low plankton blooms. The resultant low soil pH can also cause poor klekap production due to the retention of phosphorus associated with elevated concentrations of Fe and Al in the pond soils. A series of experiments was conducted to determine the effects of different soil amelioration techniques and dosage of phosphorus (P) on soil and klekap production under laboratory conditions. The treatments consisted of two factors. The first factor tested was different techniques for ASS improvement (non-improvement, improvement through liming and improvement through remediation involving forced oxidation of pyrite, flooding and flushing of oxidation products). The second factor tested was phosphorus dosages, that is, with phosphorus and without phosphorus-based fertilizer. Each treatment had three replications. The experiment showed that liming and remediation had the same effect on several soil variables; they raised the soi pH (pHF, pHFOX, pHKCl) and decreased SPOS, Fe and Al. Remediation of ASS decreased retention of P and increased available-P of soil, whereas liming did not show a significant effect on retention of P and available-P in the doses used for this experiment. The interaction between the different soil improvement techniques and phosphorus fertilising showed a significant effect on klekap production with the highest klekap production of 23.21 mg/cm2 found in remediated soil and with a phosphorus fertiliser dosage of 75 kg/ha.
