Cendekia Eksakta journal focuses upon aspects of exact sciences: engineering, agriculture, health and pharmacy . The Cendekia Eksakta journal is an research journal and invites contributions of original and novel fundamental research. Abstract

Ethanol extract of clove (Syzygium aromaticum) flower contains triterpenoid compounds, coumarins, flavone aglycones, flavonoids, tannins and saponins which have antibacterial activity against Streptococcus mutans. Fractionation was carried out to separate compounds based on their polarity. This study aims to determine the chemical compound groups and antibacterial activity of fractions of clove flower extract against S. mutans. Ethanol extract of clove flower was partitioned successively with n-hexane, ethyl acetate and water. The extracts and fractions obtained were subjected to phytochemical screening. Antibacterial activity tests were carried out on fractions of concentration 20; 25; 30; 35%, using positive control ciprofloxacin 5μg/disk and negative control Dimethyl Sulfoxide (DMSO) solvent with disc diffusion method. The result of the antibacterial activity test characterized by the formation of zone of inhibition ​​which were then analyzed statistically for the n-hexane fraction, meanwhile descriptive analysis for the ethyl acetate and water fractions. The results show that the phytochemical compounds from all fractions contained coumarin group, flavone aglycones, and flavonoids while triterpenoid, phenolic and saponin compounds were found respectively only in the n-hexane fraction, ethyl acetate fraction and water fraction. The n-hexane fraction has antibacterial activity with an average zone of inhibition of 15.63±0.38 mm at 35% concentration, with a radical zone type. The ethyl acetate and water fractions produce an irradical zone type. Statistical analysis showed differences in zone of inhibition ​​between concentration of n-hexane fraction against S. mutans, except for concentrations of 20 and 25% and 30 and 35%.

Abstract

Kombinasi parasetamol, guaifenesin, dan klorfeniramin maleat dan sering digunakan untuk meringankan gejala flu dan batuk pada anak-anak. Penetapan kadar ketiga senyawa dalam sediaan memerlukan metode  yang simultan dan valid, salah satunya yaitu metode analisis KCKT. Tujuan penelitian ini adalah validasi metode KCKT untuk analisis campuran parasetamol, guaifenesin dan klorfeniramin maleat serta mengaplikasikannya dalam sediaan sirup flu dan batuk. Pengembangan metode analisis dilakukan pada aspek panjang gelombang deteksi dan komposisi fase gerak. Penelitian ini menggunakan KCKT (Jasco) dengan detektor UV-Vis pada panjang gelombang 265 nm, fase diam C18 berukuran 125 mm x 4,6 mm dan fase gerak berupa campuran metanol: air: asetonitril (75:15:10 v/v/v) dengan laju alir 1 mL/menit. Uji validasi meliputi parameter linieritas, akurasi, presisi, selektivitas, dan sensitivitas. Hasil penelitian menunjukkan bahwa metode analisis dapat dikembangkan dan tervalidasi dengan memenuhi semua persyaratan parameter yang diujikan serta dapat diaplikasi pada sirup obat batuk. Uji  validasi pada parameter linieritas menunjukkan metode linier untuk analisis ketiga senyawa dengan nilai koefisien korelasi parasetamol, guaifenesin, dan klorfeniramin berturut-turut adalah 0,999; 0,998; dan 0,999 (r> 0,995). Uji presisi ketiga senyawa menghasilkan nilai %RSD ≤ 2%. Uji akurasi menghasilkan nilai perolehan kembali yang baik pada rentang 98-102%. Uji selektivitas menghasilkan pemisahan yang baik dengan nilai daya resolusi ≥ 1,5. Uji sensitivitas menghasilkan nilai LOD dan LOQ parasetamol sebesar 2,20 µg/mL dan 7,33 µg/mL, LOD dan LOQ guaifenesin sebesar 1,27 µg/mL dan 4,24 µg/mL, serta LOD dan LOQ klorfeniramin maleat sebesar 0,05 µg/mL dan 0,18 µg/mL. Hasil penetapan kadar parasetamol, guaifenesin, dan klorfeniramin maleat menggunakan metode yang telah dikembangkan berturut-turut adalah 100,36%, 100,25% dan 100,04%. Kandungan ketiga senyawa memenuhi persyaratan Farmakope Indonesia.
