Abstract

A research was conducted with objectives to look for the best DNA extraction methods for whisker plants, to optimize random amplified polymorphism DNA-polymerase chain reaction (RAPD-PCR) by testing annealing temperatures and screening random primers, and to identify morphology of these plants in Bengkulu Province. DNA’s extraction followed the protocols of Geneaid and Epicentre kits were conducted for 11 clones of whisker plants. Optimalization of RAPD-PCR was tested by six levels of annealing temperatures and 16 random primers. Morphological identifications were done in 4 locations in Bengkulu Province, i.e. Bengkulu City, Kuro Tidur (North Bengkulu), Kepahiang, dan Air Dingin (Curup)  Results showed that Geneaid demostrated better results as compared with Epicentre.  RAPD-PCR performed optimum at 34.8 oC of annealing temperature; whereas, three random primers showed optimal amplification were I-01, N-01 and P-01, which nucleotide sequences were ACCTGGACAC, CTCACGTTGG, and GTAGCACTCC, respectively.  Meanhile,  no morphological variatons was found among the the samples, except that growths of whisker plants were taller and bigger in Kepahyang and Air Dingin Curup than in others.

Abstract

Bengkulu province area elevating from 0 m to over 1000 m above sea level possesses high rice germplasm diversity. To obtain salinity controlling gene(s) from those germplasm, it is required to determine a suitableprotocol. This research was objected to determine NaCl concentration to select Bengkulu rice landraces. The standard most salinity tolerant genotype used in this research was ’Kuning’, a local variety usually grown on tidal area of Seluma Regency for generations. The growing media was Yoshida nutrient culture supplemented with a series of NaCl concentration of 0, 2000, 4000, 6000, 8000 or 10000 ppm. Observation was done every 2 days to find out the response pattern of rice seedling growth on different NaCl concentration. NaCl stress level was determined at 90% seedling dead (LC90) at the eighth day. The results showed that the increase of plantheight was deceased at concentration of 6000 ppm. Based on the mathematical equation generated from the percent mortality data distribution, the LC90 was at 3910 ppm NaCl concentration.

Abstract

Jengkol pod exocarp can be used as anorganic fertilizer for rice cultivation. A research aimed to determine an appropriate formulation and application time of jengkol pod exocarp on paddy rice seedling growth. Thisstudy consisted of 2 factors. The first factor was formulation, namely liquid, powder or granules. The second factor was time of application, namely at planting, 1 or 2 weeks after planting. These treatments were arrangedin a Completely Randomized Design with 3 replications. The results showed that the application of jengkol pod exocarp in the form of granule improved root growth, N, P, K uptake, photosynthesis rate, but not dry weight. Thetime application of jengkol pod exocarp two weeks after planting increased N and P uptake, transpiration rate, photosynthesis rate and root dry weight.

Abstract

Planting wetland paddy by setting row space could be a method to increase rice production. Legowo system is row planting method where we are able to apply different row types on paddy field. The objective ofthis research was to identify the best row planting type of ‘Jajar Legowo’ on growth and yield of wetland paddy. Six row plantings, including 2:1 type (means 2 rows are splited by wide space/ legowo), 3:1 type, 4:1 type, 5:1type, 6:1 type, and 7:1 typ, were tested in a Randomized Complete Block Design with 3 replications Results of the experiment revealed that the highest growth and yield as showed by total number of tillers (28), number ofproductive tillers (25), dry-mill rice (66.16 g per plant), and yield difference of dry-mill rice (50.43% per plot) was found at row planting type 2:1. Therefore, row planting type 2:1 can be selected as the best row.

Abstract

The availability of high rate of transplanting succes is very important for early growth of snakefruit. An experiment aimed to identify both optimal concentration and frequency of foliar fertilizer application on growthof snakefruit during nursery. The experiment was conducted in Pondok Kelapa, Bengkulu Tengah, from August to October 2008. Concentration and frequency treatments were arranged in a Split Plot base on a Randomized Complete Block Design. Spraying frequency (weekly, two-weekly or three-weekly) were used as the main plot;whereas, concentration of liquid nutrient Hyponex Merah with composition of 25:5:20-N:P:K, consisted of 0, 2, 4 and 6 g L-1 water, was the sub-plot Results demonstrated that concentration of liquid nutrient significantlyaffected seedling growth with optimum concentration for plant height was 4.6 g L-1, the greatest leaf number was 39 g L-1, the largest leaf area was 4.1 g L-1, the greatest stomata number was 4.2 g L-1, the greatest chlorophyl content was 3.9 g L-1 and seedling dry weight was 4.6 g L-1. Both frequency and its combination with concentration of foliar fertilizer had no significant effects on the growth snake fruit seedlings
