Abstract

Low productivity of shallot in Indonesia can be improved through development of high yielding variety. The availability of genetic information related to the character being improved is inevitable for effective breeding program. In this study, seven shallot genotypes were evaluated for their combining ability for yield improvement using half diallel cross.  Heterosis effect and heritability estimate was also investigated for yield-related characters.  The results showed that there were significant differences in general combining ability (GCA) and specific combining ability (SCA) among the evaluated genotypes. Tiron and Timor had the greatest GCA.  The greatest SCA and heterosis value was found in crosses of Kuning/Tiron, Timor/Bima Juna, Tiron/Timor and Kuning/Sibolangit. Heterosis effect varied from low to high. Broad sense heritability estimate for all characters was high, but narrow sense heritability was low for most charaters.  The dominant gene action observed on all yield-related characters suggests that the evaluated genotypes are potential to be used in breeding for high yielding hybrid varieties.   Keywords: GCA, SCA, heterosis, heritability, shallot

Abstract

Gamma irradiation to induce mutation in plant has been used intensively since several decades ago.  On maize, 275 Gy gamma irradiation have been known to increase genetic variability indicated by their morphological variation.  Identification on genetic changes by molecular technique is important to answer whether there is mutation happening on DNA level of the plants.   The objective of this research was to identify RAPD marker polymorphism on gamma irradiation mutants compared to their parents.  The initial step of the research was to select random primers could positively amplify the maize DNA.  The result showed that selection on 60 random primers yielded 15 primers that positively amplified the maize DNA.  Amplification on both mutants and their parents by those 15 selected primers indicated that only 5 primers yielding polymorphism between mutants and their parents.  Polymorphisms on mutant G1, G3 and G6 were detected on one locus, meanwhile on mutant G7, G8 and G9 were on two loci.

Abstract

EnglishThe government of Situbondo Regency has not decided the location of  the agropolitan development. In this regard, a study on livestock-based regional sustainability development is necessary before the implementation of the agropolitan program  This study aims to analyze the sustainability index and the status of Situbondo area through five dimensions of sustainability. The study used a Multidimensional Scaling analysis (MDS) method called Rap-BANGKAPET and the results are shown in the form of sustainability index and status. The Leverage and Monte Carlo analyses were used to determine the attributes that sensitively affect the index and the status of sustainability and its impact. Sustainability analysis indicate that the ecological dimension is less sustainable (46.50%), economic dimention is sufficiently sustainable (69.53%), social and cultural dimensions is also sufficiently sustainable (55.14%), infrastructure and technology dimensions are at low level of sustainability (45.48%), and similarly at low level of sustainability for legal and institutional dimensions (47.46%). Of the 73 attributes, 24 of these attributes require direct treatment immediately because of its sensitive effect on sustainability index and status (significant at 95% of confidence level). The study suggests a progressive-optimistic scenario to improve the sustainability of the future status (long term) with overall improvement of sensitive attributes. IndonesianPemerintah Daerah Tingkat II Kabupaten Situbondo sampai saat ini masih belum menetapkan wilayahnya untuk pengembangan kawasan agropolitan. Untuk itu sebelum program agropolitan dilaksanakan perlu dikaji terlebih dahulu tingkat keberlanjutan wilayah berbasis peternakan di Kabupaten Situbondo. Penelitian ini bertujuan untuk menganalisis indeks dan status keberlanjutan wilayah berbasis peternakan di Kabupaten Situbondo ditinjau dari lima dimensi keberlanjutan, yaitu: dimensi ekonomi, sosial budaya, infrastruktur dan teknologi, serta hukum dan kelembagaan. Penelitian meggunakan metode analisis Multidimensional Scaling (MDS) yang disebut RAP-BANGKAPET dan hasilnya dinyatakan dalam bentuk indeks dan status keberlanjutan. Untuk mengetahui atribut yang sensitif berpengaruh terhadap indeks dan status keberlanjutan dan pengaruh galat dilakukan analisis Leverage dan Monte Carlo.   Hasil analisis keberlanjutan menunjukkan bahwa dimensi ekologi berada pada status kurang berkelanjutan (46,50%), dimensi ekonomi cukup berkelanjutan (69,53%), dimensi sosial budaya cukup berkelanjutan (55,14%), dimensi infrastruktur dan teknologi kurang berkelanjutan (45,48%), serta dimensi hukum dan kelembagaan kurang berkelanjutan (47,46%). Dari 73 atribut yang dianalisis, 24 atribut yang perlu segera ditangani karena sensitif berpengaruh terhadap peningkatan indeks dan status keberlanjutan dengan tingkat galat (error) yang sangat kecil pada taraf kepercayaan 95 persen. Dalam rangka meningkatkan status keberlanjutan ke depan (jangka panjang), skenario yang perlu dilakukan adalah skenario progresif-optimistik dengan melakukan perbaikan secara menyeluruh terhadap semua atribut yang sensitif dalam peningkatan status kawasan.

EnglishThe Government of Bengkayang Regency has decided that the border area is one of the agropolitan region development programs. Hence, a study on the sustainability level of border area as the agropolitan region development is required.  The first objective of this study was to analyze the index and sustainability status of the border area, Bengkayang Regency, based on five sustainable dimensions. The analysis used Multi Dimensional Scaling (MDS) Method, called Rap-BENGKAWAN and the results were stated in the index and sustainability status. The second objective was to analyze the attributes that affect sensitively on index and sustainability status and the effect of error using Laverage and Monte Carlo Analysis. The results of the study revealed that ecological dimension was in the status of less sustainable (40.37%), economical dimension was sufficient sustainable (66.54%), socio-culture dimension was sufficient sustainable (67.06%), dimension of infrastructure and technology was not sustainable (24.49%) and dimension of law and institutional was sufficient sustainable (60,10).  Out of the 47 attributes analyzed, there were 22 attributes need to be handled immediately as they affect sensitively on the increase of index and sustainability status with a negligible error in the level of 95% confidence limit. Abstract

Extending the planting area to a less-fertile soil, such as Ultisols, become inevitable choice to increase the national chili pepper production in Indonesia as this types of soil is the largest part of dry land of Indonesia, about 29.7% or around 51 million ha.  However, efforts to increase its productivity by providing sufficient fertilizers application is most frequently inefficient because in the conventional fertilizer applications, broadcasted fertilizer on the soil bed, are not readily available for the plant as their solubility is limited under the plastic mulch and still in the form of granular until the end of planting season.  Fertigation, therefore, maybe the answer for this problem.  The objective of this study was to determine the best NPK fertigation method on four chili pepper hybrids.  The experiment was conducted in a randomized complete block design with three replication, and the treatments were arranged in a split plot design.  As the main plots were NPK fertigation intensity, i.e. 100%, 75%, 50%, 25% and 0% fertigation with the remnant fertilizers were circularly broadcasted surrounding the plant.  As the subplots were chili pepper hybrids, i.e. ‘Maxima’, H39, H14, and ‘UNIB CHR17’.  The results showed that there was no interaction effect of fertigation intensity indicated that all hybrid genotypes showed similar pattern of responses.  The H39 hybrid had better vegetative growth compared to other hybrids in term of plant height, dichotomous height, stem diameter, number of branches, plant fresh-weight.  Whereas the hybrid cultivar ‘Maxima’ showed the highest yield followed by H14 and cultivar ‘UNIB CHR17’.  The 100% fertigation was the best method of NPK fertilizer application for chili pepper in Ultisols.keywowrd: fertigation, chili pepper, fertilizer application, acidic soil

Abstract

Tomat (Lycopersicum esculantum) merupakan jenis sayuran yang terus berkembang menjadi komoditas penting di dunia termasuk Indonesia. Permintaan yang terus meningkat secara kuantitas dan kualitas menuntut ketersediaan varietas unggul tomat. Penelitian ini bertujuan mendapatkan informasi keragaman genetik beberapa karakter pertumbuhan dan karakter produksi 30 genotipe tomat lokal. Penelitian dilakukan pada bulan Mei sampai Agustus 2013 di Kebun Percobaan Leuwikopo IPB, menggunakan 30 genotipe tomat lokal hasil eksplorasi dari berbagai provinsi di Indonesia, yaitu Aceh, Riau, Jawa Timur, Nusa Tenggara Timur,Nusa Tenggara Barat, Sulawesi Selatan, Maluku, dan Papua. Penelitian disusun berdasarkan rancangan kelompok lengkap teracak dengan tiga ulangan. Tiap unit percobaan terdiri dari 20 tanaman. Penanaman di lapangan dilakukan dalam bedeng berukuran 1 m x 6 m dengan jarak tanam 50 cm x 60 cm. Berdasarkan hasil analisis data diketahui bahwa genotipe berpengaruh sangat nyata terhadap karakter tinggi tanaman, jumlah daun, persentase tanaman hidup, jumlah buah total, bobot buah per tanaman, dan persentase pecah buah. Karakter-karakter yang diuji memiliki keragaman genetik yang luas sehingga efektif dilakukan seleksi. Seleksi terhadap karakter bobot buah dapat dilakukan pada generasi awal karena memiliki nilai heritabilitas yang tinggi. Genotipe Aceh5, KEF9,LOM4, MER2, dan PAPUA memiliki potensi hasil yang tinggi. Genotipe KEF12, KEF6, dan MAK1 toleran terhadap pecah buah.

Abstract

Perakitan varietas unggul cabai merah toleran Al (Aluminium) merupakan solusi terbaik untuk mengatasi persoalan rendahnya produkvitas cabai di lahan kering masam. Identifikasi tetua toleran dan marka SSR (Simple Sequence Repeats) sangat bermanfaat dalam meningkatkan efektifitas program pemuliaan tanaman untuk merakit varietas toleran terhadap cekaman aluminium. Seleksi primer SSR adalah bagian awal yang penting dalam pengembangan marka molekuler tersebut. Penelitian bertujuan untuk menentukan kelompok genotipe toleran alumunium dan menentukan primer prospektif dalam pengembangan marka SSR untuk sifat toleran terhadap Al pada tanaman cabai. Evaluasi tingkat toleransi 27 genotipe cabai terhadap cekaman aluminium dilakukan menggunakan hidroponik sistem sumbu dengan larutan hara AB mix ditambah 500 ppm AlCl3.6H2O. Tahap selanjutnya adalah menguji 20 primer SSR dalam mengamplifikasi DNA genom cabai toleran dan peka yang mewakili. Hasil analisis cluster sifat toleransi terhadap 27 genotipe cabai menghasilkan 7 kelompok tingkat toleransi terhadap aluminium. Individu PBC396 dan PBC518 adalah genotipe yang sangat toleran terhadap cekaman aluminium yang unik sehingga keduanya sangat baik digunakan sebagai tetua dalam pengembangan cabai toleran aluminium. Primer SSR yang potensial menghasilkan marka polimorfik untuk sifat toleran terhadap cekaman aluminium pada cabai adalah TMS7 dan CAMS358. Primer tersebut selanjutnya harus diuji pada mapping population untuk menentukan pita yang dapat digunakan sebagai penanda toleransi terhadap aluminium. Abstract

Bamboo is naturel mixture because it can survive in many habitats and belongs to the order of angiosperms monocots. There are around 1200 – 1500 species of bamboo in the world that grow in hilly areas but do not grow in alkaline, desert and rice fields. Bamboo shoots can be processed into food and even herbal and traditional medicines. Bamboo shoots are a sourece of fiber that can be used as a nutraceutical. Bamboo shoots contain as much as 2.23 – 4.20 gfiber in 100 wet weights in the form of flour, thin slices and capsules. Dendrocalamus asper bamboo shoot flour, Bambusa tuldoides and Bambusa vulgaris with air content < 10 g /100, protein, lipid and ash content< 3 g /100 g and can obtain mineral content > 60 g /100g. Bamboo shoots are a good source of dietary fiber. This review supports the increasing benefits of bamboo shoots in the supports the increasing benefit in the health world to prevent an increase in blood sugar, as antihypertention, and antihyperuresemia. Bamboo shoots can be processed into fermented products containing Lactid Acid Bacteria (BAL) which are beneficial for intestinal microflora which are recommended as probiotics.
