Abstract

Salmonella spp. is a bacterium that causes Salmonellosis and is a bacterium that can cause food-borne illness to humans. The presence of Salmonella spp. in fresh milkfish can affect quality aspects, quality deterioration in fishery products, and food safety. Microbiological testing is an important indicator in checking for contamination in food. The main factor causing contamination can be the presence of pathogenic bacterial contamination in food ingredients. The purpose of this study was to detect Salmonella spp. on fresh milkfish (Chanos chanos) sold at Fish Auction Places (FAP), Gresik Regency using the Total Plate Count (TPC) test indicator and Salmonella Shigella Agar (SSA) media culture. This study used a qualitative descriptive study which began with observing the location and taking samples of milkfish in the field, then proceeding with the isolation of Salmonella spp. from in-vitro samples of milkfish and analyzed qualitatively. The results showed that fresh milkfish samples were contaminated with microbes with the highest number of 6×105 CFU/g and were detected to contain Salmonella spp. on SSA media, so that milkfish sold at the Gadukan Lumpur Fish Auction Place in Gresik Regency cannot meet the Indonesian National Standard for fresh fish (2729-2021).

Abstract

Plantation areas have the potential as a source of biological agents for entomopathogenic bacteria. This study aims to determine the potential of entomopathogenic bacteria from various locations in plantation crop cultivation areas, and compare their potential in North Kalimantan Province. This study carried out soil sampling, bacterial isolation, and selection of entomopathogenic bacteria using insect baits. This type of research uses a survey method on the potential of entomopathogenic bacteria. The survey was conducted at three plantation crop cultivation locations, namely coffee cultivation areas in Malinau, cocoa in Sebatik, and oil palm in Sebuku. The potency is distinguished based on the number and macroscopic characteristics of the bacterial isolate colonies obtained from each location. Furthermore, selection was carried out through testing, namely comparing bacterial isolates with a dilution of 10-1, stock, and control. Analysis of survey results using descriptive analysis and potential testing using t-test. The results showed that there were 13 bacterial isolates, and the coffee cultivation area in Malinau had the highest bacterial isolates. The selection results found 7 (seven) bacterial isolates that had the potential to be entomopathogenic bacteria, and coffee cultivation locations in Malinau had a higher number of isolates that had the potential to be entomopathogenic compared to other locations. However, isolate ST6-4U2 at dilution 10-1 from soil in the oil palm plantation area in Sebuku has the highest virulence potential as an entomopathogenic bacteria.

Abstract

Skipjack tuna (Katsuwonus pelamis) is one of the dominant capture fisheries products in Maluku. This is supported by the geographical location of the Maluku Islands which are surrounded by the sea. Bacteria are often found in traditional processed fishery products such as smoking fish. This study aims to determine the total bacteria Salmonella spp. and differences in the abundance of Salmonella spp. on smoked skipjack (Katsuwonus pelamis) products sold in several markets in Ambon City. This research is an experimental research. The research was carried out by taking samples with a total of 5 samples from the people's market location, and bacterial calculations were carried out using a dilution technique. Bacteria that have been obtained from the dilution results, then isolated using the quadrant scratch method technique on SSA media (Salmonella Shigela Agar). Bacterial biochemical tests, including: indole test, Methyl-Red test, Voges Proskauer test, citrate test, and Triple Sugar Iron Agar (TSIA) test, and gram staining were performed to confirm/identify the isolated bacteria. The results showed that smoked skipjack sold at Mardika Market was identified as having the total number of Salmonella spp. respectively 6700 CFU/ml and 8400 CFU/ml. The total number of Salmonella spp. on smoked skipjack at Mardika market is different from the results of the calculation of the total bacteria of smoked skipjack sold in modern markets and small hative markets, which did not identify any Salmonella spp bacteria contamination.
