Abstract

The protein profiles of Pseudomonas pseudomallei exotoxins which derived from P. pseudomallei strains of different virulence and origin (J53, AN, C2, XI003 and 1328 strains), have been studied by electrophoretic analysis using SDS-PAGE (sodium dodecyl sulphate - polyacrylamide gel electrophoresis).Each of these P. pseudomallei strains is propagated in mucin broth and then incubated statically at 32 ° for 7 days. The exotoxin of each strain has been prepared from that broth culture by centrifugation and followed by ultrafiltration.The result indicated that all of the strains, regardless of its different virulence and origin, showed one protein band common to all, i.e. at a molecular weight of approximately 35 K.

Abstract

Biakan sel fibroblas embrio ayam yang dibuat dari embrio berumur 9 sampai 10 hari, yang bebas patogen tertentu digunakan untuk mengisolasi virus IBD dari beberapa tempatdi pulau Jawa, Medan, Lampung, Bali dan Maros. Virus diisolasi dari bursa Fabricius ayam penderita IBD. Sedangkan sera hiperimun dibuat dari cavia yang disuntik dengandosis 104 sampai 106 TCID50 masing-masing isolat virus. Uji netralisasi virus dilakukan dengan cara virus konstan dan antibodi bervariasi. Dari hasil uji netralisasi dan uji keterkaitan antigen dari 25 isolat virus yang telah dimurnikan terhadap ke 25 serum kebalnya dapat ditetapkan 3 isolat virus IBD yang tidak mempunyai keterkaitan antarasatu dengan lainnya. Ketiga isolat itu merupakan subtipe yang berbeda dan dapat digunakan sebagai bakal virus bibit untuk membuat vaksin. Diharapkan bahwa vaksin yang dibuat dari ketiga virus Gumboro tadi, tidak hanya mampu mencegah penyakit karena infeksi virus IBD yang 25 isolat itu, tetapi juga akibat virus IBD lainnya yang ada di Indonesia. Vaksin tersebut akan merupakan kombinasi dari 3 isolat virus 19.13 dan 11 atau 19.5 dan 11.

Abstract

Piglet neonatal diarrhoea occured in all the piggeries studied in Indonesia. Field studies demonstrated the prevalence of diarrhoea in all piggeries were at range from 13.4% to 43.7% with and average 24.7%. The majority of piglets with diarrhoea were in the first 2 weeks of life. Mortality of piglets were at rates between 12.2% to 31.6% with an average 17.9%. The distribution of piglet mortality preceded by diarrhoea recorded in an intensive study in a piggery G for a seven week period showed a high positive correlation with diarrhoea (r2 = 0.79; P

Abstract

The aim of this research is to observe the antiproliferation activity of temulawak (Curcuma xanthorrhiza Roxb.) ethanol (70%) extract on MCA-B1 and MCM-B2 cell lines in vitro. Cells were cultivated in tissue culture plate 24 wells with vinblastin (positive control) and 5 dose level extracts in 5 replicates. The concentrations of extract were 0 ppm (negative control), 15 ppm (P1), 30 ppm (P2), 45 ppm (P3), 60 ppm (P4), and 75 ppm (P5). After 3 days incubated at 37 derejad C, cells were harvested and total cells were counted using a haemocytometer Neubauer. The results showed that 70% temulawak extract had antiproliferation activity to MCA-B1 and MCM-B2 cell lines. The best result was occured on the 75 ppm doses, with the ant-proliferation activity reached for 70,0% in MCA-B1 cell line and 75.4% in MCM-B2 celll line. Overall, the results indicated that temulawak may have a possible therapeutic potential againts cancer.Keywords: tumor, temulawak, antiproliferation, MCA-B1, MCM-B2, in vitro.

Abstract

The objective of this research is to examine the presence of antibody of avian influenza (AI) H5 and detection of genetic material the subtype of AI virus (H5N1) in waterfowls in Lampung Province. Samples consisted of 673 serum and 673 cloacal swabs, and collected from water fowls Epidemiologycally, samples collection were performed by using multistage sampling method. Examination of AI antibodies by Haemagglutination Inhibition test (HI test) demonstrated that H5 antibodies were found at all district in Lampung Province. High frequency of antibodies were noted at district ofo Tulang Bawang (69.41%). The average of antibody titres of AI were low (under 24) and the lowest antibody titre was found at district of Tulang Bawang (20.92). Evaluation of genetic material H5N1 individually by using Reverse Transcriptase – Polymerase Chain Reaction (RT-PCR) was not found among water fowls. The results showed H5 and unknown N subtype (H5Nx); the second subtype was unknown H and N1 (HxN1). Abstract

The purpose of the research is to determine the influence of injection and inhalation anesthesia on oxygen saturation during enterotomy of Indonesian local cats (Felis domestica). Twelve local cats were divided into two groups. The first group (PI) received injection anesthesia with xylazin and ketamin, while the second group (P2) with the inhalation enesthetic isoflurane. Observations were made when the cat anesthetized with the observation interval every 15 minutes until the operation completed. The statistical test showed that oxygen saturation values in the provision of injection anesthesia and the inhalation anesthesia during enterotomy was not significantly different (P>0.05), however a decline in the value of injection anesthesia saturation was occured at minute 45 to 60. Both types of anesthesia caused significant decrease in the value of physiological parameters such as respiratory rate, heart rate and body temperature. Correlation test results showed that respiratory rate and body temperature were affecting the oxygen saturation during enterotomy. Oxygen saturation level during the inhalation anesthesia provide a relatively stable value when compared with the injection anesthesia, this is due to the supply of dissolved oxygen in inhalation anesthesia system.key words: anestesia, enterotomy, cats, oxygen, saturation.

Abstract

Mortality, growth and conception development after placentation are affected by capacity and placenta function which are influenced by hormonal and growth factor to succed pregnancy till birth. Growth hormone is protein or polypeptide hormone. It influences protein metabolism, increases amino acid up take, carbohydrate metabolism, gluconeogenesis, fat mobilizatin, minerals metabolism and the development of cartilage that will lead to growth. The purpose of this research is to observe the effectiveness of somatotropin on the growth of fetus. Forty pregnant rat were divided to 4 groups, they were K (not given somatotropin), A (given somatotropin 9 mg/kg BW at 12 to 20 days of pregnancy), B (given somatotropin  9 mg/kg BW at 2 to 11 days of pregnancy), C (given somatotropin  9 mg/kg BW  at 2 to 20 days of pregnancy). Parameters observed were ovulation rate, implantation rate, development/growth of uterus and placenta, size of fetus, and pups growth. The result showed that somatotropin could increase implantation succed, development of uterus and placenta that are followed by good performance of reproduction during pregnancy and also birth body weight pups.Keywords: Somatotropin, Pregnancy, Pups.

Abstract

The purpose of the research was to determine meiotic status of cat oocytes maturated in vitro at various duration times. Cat ovaries were collected from sexually mature queens after routine ovariohysterectomy. Oocytes were collected from those ovaries by slicing method and only good oocytes were maturated in maturation medium and incubated at 38,5oC in 5% CO2 humidified incubator in various duration. The durations were at 18-20 hours, 22-24 hours and 28-30 hours. At the end of maturation, the oocytes were fixed and stained with acetic orcein. The percentage of oocytes that reached metaphase II for 18-20 hours, 22-24 hours and 28-30 hours maturation were 60% ± 0.04 ; 68% ± 0.31 and 77% ± 0.2, respectively. There were no significant differences (P>0.05) on the percentage of oocytes in reaching the stage of germinal vesicle, anaphase-telophase and methapase II. Abstract

Detection of Edwardsiella tarda antigen on the 20 heads of experimentally infected-catfish tissue (Pangasius pangasius) by immunohistochemistry was done. Clinical sign of the infected fish were weak response, swelling, haemorrhages in the anal, abdomen and tail fin regions, ulcer in the above of linea lateralis. Gross findings of the internal organs were swelling, pale and bad smell. Histopathologically, there were necrotic area with  infiltration of inflamatory cells, melanomacrophages in the muscle, liver, kidney and spleen and some eosinophilic materials in the kidney was also found.  Immunohistochemical detection using specific antibody to E. tarda showed positive reaction in the internal organs.  Immunohistochemistry assay is one of the suitable methods for detection and diagnose of E. tarda infrction in fish.Keywords: Catfish, Edwardsiella tarda, immunohistochemistry

Abstract

Determinan of reproduction status was carried out 16 lactation dairy cows at Faculty of veterinary Medicine and Faculty of Animal Science - IPB bt measurement of milk progesteron level by enzyme Immunoassay.This result could concluded that : 1 cow post oestrus, 3 cows dubius pregnant and 5 cows pregnant.Confirmation by rectal palpation was done one week later and shown that: 4 of 5 diagnosed pregnant, 1 cows need recheck.The accuracy of milk progesteron assay by enzyme immunoassay (EIA) in this case was 80% for positive pregnant.-------------------------------------------------------------------------Telah dilakukan penentuan status reproduksi terhadap 16 ekor sapi perah yang berada di Fakultas Kedokteran Hewan dan Fakultas Peternakan - IPB, melalui pengukuran kadar estrogen susu dengan menggunakan Enzyme immunoassay.Dari hasil Penelitian tersebut dapat ditentukan 3 bentuk status reproduksi yaitu berahi, lewat berahi dan bunting, di mana 7 ekor sapi dubius bunting dan 5 ekor bunting.peneguhan diagnosa dilakukan dengan palpasi rektal seminggu kemudian dan menunjukan 4 dari 5 ekor dinyatakan bunting.Ketepatan diagnosa uji progesteron susu dengan  Enzyme Immunoassay (EIA) untuk kasus ini adalah 80% untuk positif bunting.
