Abstract

ABSTRAK. Penelitian ini bertujuan mengetahui pengaruh pemberian PGF2 dalam pengencer semen komersial (Andromed) terhadap peningkatan motilitas spermatozoa kambing Nubian. Penelitian ini menggunakan sampel semen yang dikoleksi dari 3 ekor kambing Nubian berumur 2-3 tahun menggunakan vagina buatan dan dievaluasi kualitasnya secara makroskopis dan mikroskopis. Setelah dievaluasi, sampel semen ditambahkan pengencer Andromed lalu dibagi atas 3 kelompok perlakuan, yaitu: P1; P2; dan P3 yang masing-masing ditambahkan NaCl fisiologis; 37,5 g PGF2; dan 75 g PGF2. Seluruh sampel disimpan dalam refrigerator selama 4 jam dan dilakukan pemeriksaan motilitas spermatozoa. Data yang diperoleh dianalisis dengan sidik ragam pola satu arah (ANOVA) dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan, bahwa motilitas spermatozoa (%) kambing Nubian pada P1; P2; dan P3 masing-masing adalah 26,335,5; 62,03,5; dan61,810,13 (P0,05). Disimpulkan bahwa penambahan PGF2 pada pengencer Andromed dapat meningkatkan motilitas spermatozoa kambing Nubian.(The improvement of sperm motility in Nubian goat after PGF2 administration in andromed semen diluents)ABSTRACT. The study aims to determine the administration effect of PGF2 in a commercial semen diluents (Andromed) on improvement of Nubian goat sperm motility. This study used semen samples that collected from three Nubian goats aged 2-3 using artificial vagina and their quality evaluated macroscopically and microscopically. After evaluated, semen samples were added with Andromed diluents then divided into three groups (P1, P2, and P3) where each group was then added with 0,9% physiologic NaCl, 37.5 g PGF2, and 75 g PGF2, respectively and stored in a refrigerator for 4 hours and subsequently spermatozoa motility was examined. The data obtained were analyzed by one-way analysis of variance (ANOVA) and followed by Duncan test. The results showed that the spermatozoa motility (%) of Nubian goats at P1, P2, and P3 were 26.335.5, 62.03.5, and 61.810.13, respectively. Based on the statistical tests showed that the administration of PGF2 at P2 and P3 had a significant effect (P0,05) on the motility of spermatozoa of Nubian goats, but the motility decreased in P1. The conclusion of this study is the addition of PGF2 to Andromed diluents can increase the motility of spermatozoa of Nubian goats.

Abstract

ABSTRAK. Tingkat peradangan ambing merupakan salah satu faktor yang dapat mempengaruhi produksi dan kualitas susu sapi. Penelitian ini bertujuan mengetahui hubungan antara produksi dan kualitas susu yang dihasilkan sapi mastitis. Materi yang digunakan adalah 30 ekor sapi perah Friesian Holstein (FH) dengan periode laktasi III-V dan bulan laktasi 3-4. Tingkat peradangan ambing diuji menggunakan California mastitis test (X1) dan jumlah sel somatik (X2) dengan metode Breed dihubungkan dengan produksi (Y1) atau kualitas susu (lemak : Y2, protein = Y3 dan laktosa = Y4). Analisis yang digunakan adalah analisis regresi linier berganda. Hasil penelitian menunjukkan sebanyak 30% kuartir ambing dinyatakan sehat sedangkan 70% kuartir ambing terinfeksi mastitis subklinis dengan tingkat peradangan yang berbeda. Terdapat hubungan antara tingkat peradangan ambing dengan produksi susu berpola linier negatif nyata (P 0,05) dengan persamaan Y1 = 16,126 - 3,064X1 - 0,001X2 dan koefisien korelasi kuat (r = 0,740). Hubungan antara tingkat peradangan ambing dengan kualitas susu (lemak, protein, laktosa) berpola linier negatif nyata (P 0,05) dengan persamaan Y2 = 3,481 - 0,157X1 - 0,000X2 (r = 0,739), Y3 = 3,048 - 0,124X1 - 0,000X2 (r = 0,653) dan Y4 = 4,605 - 0,106X1 - 0,001X2 (r = 0,623). Kesimpulan dari penelitian ini adalah terdapat hubungan antara tingkat peradangan ambing dengan produksi dan kualitas susu dimana semakin tinggi tingkat peradangan ambing maka produksi dan kualitas susu akan mengalami penurunan.(Evaluation of milk production and quality in mastitis cows)ABSTRACT. The level of udder inflammation is one of the factors that can affect the production and quality of cow's milk. This study was aimed to determine the relationship between milk production and quality produced by mastitis cows. The material used was 30 Friesian Holstein (FH) dairy cows with a lactation period of III-V and a lactation month of 3-4. Inflammation degree of the mammary gland was determined by the California mastitis test (X1) and somatic cell count (X2) with a breed method relationship between milk production (Y1) or milk quality (fat: Y2, protein: Y3, and lactose: Y4) were determined by the multiple linear regression. The results showed that 30% of the udder quarter were healthy while 70% of the udder quarter were infected with mastitis with different levels of inflammation. There was a strong negative correlation (P 0,05; r = 0.740) between milk production, CMT and SCC score with Y1 = 16.126 - 3.064X1 - 0.001X2. Moreover, the correlation between milk production and milk quality (fat, protein, lactose) also showed a strong negative linear correlation (P 0,05) with the equation Y2 = 3.481 - 0.157X1 - 0.000X2 (r = 0.739), Y3 = 3.048 - 0.124X1 - 0.000X2 (r = 0.653) and Y4 = 4.605 - 0.106X1 - 0.001X2 (r = 0.623). In conclusion, the increased mammary inflammation level will result the decrease in milk production and milk quality.

Abstract

ABSTRAK. Penelitian ini bertujuan untuk mengkaji pengaruh perbedaan lama pemeraman dan aras starter mikroba pencerna serat dari rumen kerbau terhadap komponen serat dan perubahan struktur jaringan sabut kelapa fermentasi. Percobaan menggunakan rancangan acak lengkap pola faktorial 3x3 dan 4 ulangan. Fermentasi sabut kelapa menggunakan mikroba pencerna serat dari rumen kerbau sebagai starter dengan perlakuan aras starter (0, 2,5 dan 5%) dan lama peram (0, 7 dan 14 hari). Parameter yang diamati adalah komponen serat dan perubahan struktur jaringan. Data dianalisis menggunakan analisis ragam, dilanjutkan uji jarak berganda Duncan. Hasil penelitian menunjukkan tidak terdapat pengaruh interaksi (P0,05) antara perbedaan aras starter dan lama peram terhadap kadar neutral detergent fiber (NDF), acid detergent fiber (ADF), hemiselulosa, selulosa dan lignin. Kesimpulan penelitian ini adalah penurunan kadar komponen serat serta peningkatan kerusakan jaringan seiring dengan peningkatan aras starter dan lama pemeraman. Hal ini disebabkan oleh penurunan kadar selulosa dan lignin sedangkan kadar hemiselulosa tidak dipengaruhi. Hal ini didukung oleh pengamatan terhadap perubahan struktur jaringan.(Fiber components of fermented coconut husk using fibre degrading microbes from buffalo rumen)ABSTRACT. This study aimed to examine the effect of different fermentation periods and levels of fiber-degrading microbial starter from buffalo rumen fluid on fiber component and tissue structure alteration of coconut husk. The experimental design used was factorial randomized complete 3x3 and 4 replications. The coconut husk fermentation was using fiber-degrading microbial obtained from buffalo rumen fluid as a starter with different treatments of starter levels (0, 2.5 and 5%) and fermentation periods (0, 7 and 14 days). Parameters observed were fiber component and tissue structure alteration. The data were analyzed by analysis of variance and continued by Duncans Multiple Range Test. The results of this research showed that there was no interaction effect (P0.05) between starter level and fermentation period on NDF, ADF, hemicellulose, cellulose and lignin contents. The conclusion of this research was decreased fiber component content and damage to tissue structure of coconut husk along with increased starter level and fermentation period. This is caused by decreased levels of cellulose and lignin while hemicellulose levels were not affected. This is supported by observations of changes in tissue structure alteration.
