Abstract

As  of  nearly  the  end  of  2007,  National  Tuberculosis  Program  have  not  achieved    MDG’S  indicator  timely especially  in  South  Sumatera.  This  was  caused  by  Local  and  National  Tuberculosis  Control  Program administered  merely  by  governments  with  minimal  private  sector  and  community  participation.  A  Network  of Pulmonary Tuberculosis Eradication Program was established in Ilir Barat II District of South Sumatera. This network  is  directed  by  four  components  of  TB  control:  cadres,  Physician  Practitioner,  Head  of  subdistrict, district local authority, and district tuberculosis program. The Network aims at accelerating TB Contol Program since first October 2006 until first October 2007. The final goal is to have “District Free TB”. The”Network” was founded  by  decree  of  the  Mayor  of  Ilir  Barat  II  Distric  .  The  result  of  one  year  implementation  indicated significant increased of the Indicator of TB program, more than the MDG’starget.  If the network  is applied at Palembang  city  and  all  South  Sumatera,  those  areas  will  reach  100%  conversion  rate    by  the  end  of    2008 and 100% case detection rate by the end of 2010.  Keywords:  Tuberculosis  Control  Program,  Networking  Model,  Health  Cadre,  Private  Physician,  Local  Government, Millenium Development Goal’s

Abstract

The aim of this research was to investigate the event of loss of heterozygosity of retinoid acid receptor (RAR β2) gene on the chromosom 3p24 in the pathogenesis ofbreast cancer. The breast cancer is the most malignant tumor that leads the second cause of death among the female. The recent concept of the theory of pathogenesis breast cancer is molecular genetic approaches such as apoptosis gene, oncogen, suppressorgene, and growth promoting factor gene. However, the diagnostic and therapy of breast cancer are still not satisfied. The lost or decreased in the expressionof RAR  β2gene on 3p24 chromosome via loss of heterozygosity (LOH) mechanism are the role of pathogenesis of breast cancer. Design of the research is by experimental and observational. Human female breastcancer and blood samples were gained from public hospital RSUD dr. Saiful Anwar, Malang. DNA was extracted from lymphocyte and tissue using Macherey-Nagel system kit. Loss of heterozygosity (LOH) incident in 3p24 chromosome was detected by PCR and Chromogenic in situ Hybridization (CISH) method.

Abstract

Mast cell are derived from haematopoietic progenitors in the bonemarrow. Developing mast cell subsequently migrate to peripheral tissues, such as the skin, mucosa and air way, where they terminate their differentiation under  the  influence  of  factors  in  the  tissue  micro-enviroment.  This  study  aimed  to  know  distribution  and strucutre of mast cell on rat mesentery. The mesentery was stained with a 1 % safranine solution or toluidine blue  on  the  a  pepared  slide.  The  prepared  slides  were  immediately  viewed  by  a  observer  through  a microscope.  The  result  showed  that  mast  cells  stained  with  safranine  or  toluidine  blue  were  closely associated with postcapillary in the rat mesentery. Abstract

Hypoxia exposed trophoblast cells will cause apoptotic process, that correlate with up-regulation of p53 and Bax protein expression; and down-regulation of anti-apoptotic Bcl-2 protein. Apoptotic process and up-regulation of p52, and down-regulation of anti-apoptotic Bcl-2; also find both on trophoblastic samplefrom preeclampsia/ eclampsia complicated pregnancy, and  normal pregnancy. The aim of this study is, to prove that the amount of apoptotic trophoblast cells on trophoblast tissue sample from preeclampsia/ ecclampsia complicated pregnancy, were higher then those of normal pregnancy. This research was laboratory cross sectional study, that observed immunohystochemically, p53 and BCl-2 protein expression; also the DNA-fragmented technically (TUNEL) to detect apoptotic  cells on trophoblastic samples from preeclampsia/ eclampsia complicated pregnancy, compare by the normal pregnancy. On this study, we found that the amount of trophoblast cells that expressed of p53 protein (22.70 ± 2.90), were higher then those of normal pregnancy. (8.2 ± 4.99). The different was statistically significant. (t test; p  ≤0.000). We also found that an apoptotic process on trophoblast cells of preeclampsia/ eclampsia placenta (15.3 ± 3,56) were higher  then those of normal pregnancy (4.7 ± 1.89). The different was statistically significant. (t test; p ≤0.000).

Abstract

Coronary  Heart  Diseases  (CHD)  are  a  serious  health  problem  that  commonly  occur  not  only  in  modern countries, but aslo  in developing countries like  Indonesia. The high cholesterol level in  blood is  a risk factor that  may  cause  CHD  by  atherosclerosis  mecanism.  dietary  fiber  may  be  used  to  prevent  atherosclerosis. Cornmeal and cornmeal-soy are products of corn, that rich in fiber. This research was aimed to study about the  effect  of  cornmeal  and  cornmeal-soy  on  serum  cholesterol  level  of  6-8  mounths  male  wistar  rats.  The subjects  are  25  male  wistar  rats.  These  mice  will  be  provided  into  five  groups.  (1)  gruop  that  given atherogenic  diet  only,  (2)  gruop  that  given  atherogenic  diet  with  cornmeal  3.4  g/day,  (3)  gruop  that  given atherogenic diet with cornmeal 6.8 g/day, (4) gruop that given atherogenic diet with cornmeal-soy 3.6 g/day, (5)  gruop  that  given  atherogenic  diet  with  cornmeal-soy  7.2  g/day.  Data  collecting  include  first  weight,  last weight,  weight  gain,  nutrient  intake,  and  serum  lipid.  Statistic  analysis  that  be  used  is  ANOVA  one  way. Results showed that  wistar rats which given atherogenic diet with cornmeal and cornmeal-soy  had trend to increase  cholesterol  and  LDL  level.  According  to  statistic  analysis,  cholesterol  and  LDL  level  among  groups were not significant (p > 0.05). However wistar rats which given atherogenic with cornmeal and cornmeal-soy in  high  doses,

Abstract

Cerebral and lung damage during malaria infection is believed to be caused by free radicals activitiesthat are produced during immunology process. The free radicals react with lipid component of cellular membrane which generates malondialdehyde (MDA) as its end-product. The aim of the research was to determine whether combination of artemisinin and NAC was moreeffective in decreasing cerebral and lung MDA level compared to artemisinin mono-therapy . The researchwas post-test-control-only design using 5 groups consisted of group A (negative control group), group B mice which infected with P.berghei without therapy (positive control group), group C mice which infected with P.berghei and received artemisinin mono-therapy (0.04 mg/g BW for 7 days), group D mice which infected with P.berghei and received artemisinin in combination with NAC (1 mg/g BW for 7 days) and group E mice which infected with P.berghei and received artemisinin in combination with NAC (1 mg/g BW for 3 days and tapered into ½ mg/g BW for 4 days). On the 3rd, 5th,and 7thday, 3 mice from each group were scarified and assayed for MDA level. On the 3rd day, a decreasing trend of cerebral and lung MDA level wasobserved on all treatment groups. On the 5thday, a decreasing trend of cerebral and lung MDA level wasobserved in group that received artemisinin and NAC whereas group’s that received artemisinin mono-therapy increased. Cerebral and lung MDA level of groupthat received artemisinin mono-therapy was significantly different with group that received combination of artemisinin and NAC in constant dose (p = 0.014) and with group that received combination artemisinin  and NAC in tapering dose (p = 0.004).

Abstract

Long term ethanol administration may cause morphological changes in the hippocampus, followed by deficit in  the  hippocampal  function  including  the  impairment  of  memory.  The  aim  of  this  study  is  to  investigate  the changes of spatial working memory of rats after chronic ethanol administration at adolescent age. Twenty five adolescent  (30  days  of  age)  male  rats  (Rattus  novergicus)  divided  randomly  into  5  groups  :  control  without treatment (C1), control with treatment (C2), treatment 1 (T1), treatment 2 (T2), treatment 3 (T3) group. Each group consist of 5 rats. C2 group was given physiologic saline, T1, T2, and T3 group were given ethanol with doses  1,  2,  and  3  g/kg/day  for  30  days  by  intraperitoneal  injection.  After  chronic  ethanol  administration  the spatial  working  memory  of  5  groups  were  tested  by  using  8  arm  radial  maze  for  12  days.  Spatial  working memory measured by total number of entered arm maze and error A type. The results of this study showed that  chronic  ethanol  administration  at  adolescent  age  caused  decreasing  of  spatial  working  memory  of  the rats. The total number of entered arms maze and error A type for treatment group (especially P3 ) is higher than  control  groups  (p<0,05).  It  is  concluded  that  in  rats,  chronic  ethanol  administration  at  adolescent  age caused decreasing spatial working memory.   Key words : Ethanol, Spatial working memory, Adolescent

Abstract

The incidence of hypertension in menopausal and post menopausal women related to plasma estrogen leveland sympathetic nervous activity. Estrogen may have a specific effect to modulate adrenergic vasoconstriction by modulating adrenergic receptors. Blood vessels contractility is regulated mainly by the sympathetic nervous system, in aorta mediated particularly by  α1Dadrenergic receptor.We hypothesize that 17βestradiol decreases vascular smooth muscle contractility by decreasing  α1Dadrenergic receptor density. To prove the above mechanism in the present study we performed several interrelated assays, i.e bioassay using isolated organ and protein blotting. An isolated rat aortic ring without endothelium (2-3 month,150-200 gram) was incubated in 17β-estradiol (10-5M, 10-4M,and 10-3M ) for two hours prior to stimulation using phenylephrine as α1Dadrenergic receptor agonist. The response of the rat aortic ring to the stimulation of α1Dadrenergic receptor agonist was measured as the subsequent change on aortic contractility using bioassay, and the change of  α1Dadrenergic receptor density using Western and dotblot.
