Abstract

The study investigated the optimal dose of molasses for the cultivation of commercial probiotics containing Bacillus amyloliquefaciens and Bacillus subtilis. A two-month laboratory experiment was conducted at the Faculty of Fisheries and Marine Sciences, Jenderal Soedirman University, using a Completely Randomized Design (CRD) with four treatments: K (0 ppm), A (5 ppm), B (10 ppm), and C (15 ppm), each with three replications. The study measured optical density (OD) and bacterial growth over 48 hours. The results showed that Treatment C (15 ppm) produced the highest OD value (0.014), indicating substantial bacterial growth. However, it exhibited the slowest growth phase. In contrast, Treatment A (5 ppm) demonstrated the fastest growth phase and achieved the highest bacterial density throughout the study. This suggests that 5 ppm of molasses is ideal for rapid bacterial proliferation. Treatment K (0 ppm) resulted in the lowest OD (0.0042) and led to bacterial death by the end of the experiment, highlighting the necessity of molasses for bacterial survival and growth. In conclusion, a 5 ppm molasses dose (Treatment A) proved to be the most effective for promoting optimal bacterial growth and density, while higher doses resulted in slower growth, and no molasses inhibited bacterial survival

Abstract

Vibrio harveyi can cause mass death in vaname shrimp. The β-Glucan content in Saccharomyces cerevisiae can stimulate the immune system non- specifically. Meanwhile, the endophytic bacteria Sonneratia alba has antibacterial activity. This study aims to determine the effectiveness of the probiotic fee formulation of β Glucan S. cerevisiae using a combination of antibacterial S. alba leaves as an effort to inhibit the growth of V. harveyi in vaname shrimp (L. vannamei). The research method used is an experimental probiotic formulation from a combination of β Glucan S. cerevisiae and endophytic bacteria S. alba through shrimp feed with 6 treatments and 4 replications, The parameters used are Total Haemocyte Count, Differential Haemocyte Count and Survival Rate. This study was conducted in vivo tests for 10 days of probiotic feed maintenance and 7 days of infection with a total research period of four months at the Faculty of Health, Medicine, and Natural Sciences, Airlangga. The results of the research show that the combination of ingredients can inhibit the growth of V. harveyi through the immune response and survival of vannamei shrimp. The best treatment was P3 with a Total Haemocyte Count (THC) of 7,54x106 cells/ml and Differential Haemocyte Cout (DHC) (60,56% granular, 16,68% semi-granular, 22,76% hyaline) with 85% survival. The provision of probiotics had a real effect and became the optimum formulation to inhibit the growth of V. harveyi.

Abstract

Indonesia is the third largest global shrimp exporter after Ecuador and India. Shrimp production increased by 21% from 1.21 million tons to 1.48 million tons in 2021-2022, but decreased to 1.097 million tons in 2023. This is thought to be due to AHPND (Acute Hepatopancreatic Necrosis Disease). Our research aims to determine the biofabrication, characterization and effectiveness of gold nanoparticle biofabrication mediated by Chaetoceros calcitrans as an antibacterial against AHPND in white shrimp. The method used is experimental. The findings were the antibacterial potential of gold nanoparticle biofabrication mediated by C. calcitrans as an AHPND drug in vaname shrimp. Based on the research results, biofabrication was successfully formed with UV-vis characterization at a wavelength of 533 nm, in the FTIR test the AU3+ group was successfully reduced by the bioactive compound C. calcitrans. The results of the SEM and TEM tests showed that nanoparticles were dominated by a spherical shape, the particle size according to the PSA test was 8.25 nm and the XRD test answered that Np-Au crystallization had formed with the appearance of a high intensity peak.

Abstract

Whiteleg shrimp (Litopenaeus vannamei) cultivation can be carried out in areas far from the sea with the RAS (Recirculating Aquaculture Systems) system using artificial seawater (ASW) added with bittern as a mineral source. This study aimed to determine the effect of artificial seawater formulation using bittern minerals on growth, survival, glucose levels, and hepatopancreas histology in whiteleg shrimp. This study used a completely randomized design (CRD) with 5 treatments and 4 replications including (P0) seawater; (P1) 18.56 grams of crossover salt + 8.4 ml/liter of bittern; (P2) 14.93 grams of crossover salt + 16.86 ml/liter of bittern; (P3) 11.53 grams of crossover salt + 25.13 ml of bittern/liter; (P4) 23 grams of crossover salt/liter. This study began with the crystallisation and characterisation of bittern, continued with mineralisation tests, and prepared artificial seawater using a combination of salt and bittern solutions. Cultivation was carried out for 20 days, and then survival, growth in length and weight, water quality, blood glucose, and histology of hepatopancreas tissue were observed. The formulation of artificial seawater treated with P1 was the best, with a survival rate of 78% and growth no different from control seawater. Therefore, artificial seawater has the potential to be used for the cultivation of whiteleg shrimp.

Abstract

Product otak-otak ikan tenggiri is a promising fishery diversification product but is prone to spoilage. Edible coatings can help slow down spoilage. The purpose of this study was to determine the effect of different edible coating formulations on the characteristics of edible coating and to determine the protective potential of edible coatings against bacterial growth and organoleptic product otak-otak ikan tenggiri during storage. This research is an experimental study that is divided into two stages, the first stage is the characteristics of edible coatings using a completely randomized design (CRD) with testing including antimicrobial activity, water absorption, and viscosity with 5 treatments and 4 replicates, and the second stage is coating applications on the length of storage of product otak-otak ikan tenggiri testing including TPC and organoleptic using two-factor CRD 3×6 treatments and 3 replicates. Results indicated a significant impact (P < 0.05) of coating formulations on these characteristics. The best formulation, modified tapioca-WPI with CSNP (P4), showed antibacterial activity with a 9.16 mm inhibition zone, 3.46% water absorption, and 15.33 cP viscosity. This formulation maintained microbial stability and organoleptic quality for up to 3 days at room temperature and 12 days under cold temperature.

Abstract

Pempek is a very popular traditional Indonesian food because it has a delicious taste. However, the shelf life of pempek is only about 16 hours at room temperature. This study aims to develop and evaluate edible coatings based on cellulose and chitosan nanoparticles (CSNPs) to extend the shelf life of pempek from Tenggiri fish (Scomberomorus commerson). This study is experimental research divided into two stages. The first stage involves characterizing the edible coating using a Completely Randomized Design (CRD), with tests including antimicrobial activity, water absorption, and viscosity, conducted with 5 treatments and 4 repetitions. The second stage involves applying the coating to pempek and evaluating its shelf life, focusing on TPC and organoleptic qualities using a two-factor CRD with 3×6 treatments and 3 repetitions. The results showed a significant effect (P<0.05) of various formulations on the characteristics of edible coatings. The results showed that the variation of cellulose derivatives and CSNP in the preparation of edible coating significantly affected the coating characteristics (P<0.05). The combined formulation of CMC and CSNP (P5) was selected as the pempek coating, with antibacterial activity of 9.31 mm, water absorption of 1.23%, and solution viscosity of 33.50 cPa.s. In the second stage, the treatment of pempek coating with CMC and CSNP (C2) effectively maintained the quality of pempek until day 3 at room temperature and day 12 at refrigerator temperature.

Abstract

The widespread use of antibiotics in aquaculture has been associated with the development of multidrug resistance in the environment and in humans. This is also exacerbated by the application of high stocking densities in fish farming systems, including catfish (Clarias sp). This study aims to see the potential of Sargassum aquifolium extract, which is a brown macroalgae, as an antibacterial agent for fish disease, Aeromonas hydrophila. Previous studies have revealed the phytochemical profile of S. aquifolium extract, but its effectiveness as an antibacterial agent has not been studied. The ethanol extract concentrations used were 20%, 40%, 60%, 80%, and 100% with two repetitions. The MIC test method was carried out to determine the lowest concentration of S. aquifolium ethanol extract that could inhibit the growth of Aeromonas hydrophila bacteria, while the MBC test was carried out to determine the bactericidal properties against A. hydrophila bacteria. SEM analysis was carried out to determine the morphological damage to A. hydrophila bacterial cells that had been treated with S. aquifolium ethanol extract. Based on the results, it was revealed that S. aquifolium extract could inhibit the growth of A. hydrophila highest with a very strong category at a concentration of 100%. The MIC value of S. auifolium extract was shown at a concentration of 100% but did not cause bacterial cell death based on the MBC value and SEM observations. These findings indicate that S. aquifolium extract has the potential to inhibit the growth of A. hydrophila. Increasing in vivo parameters such as the immune system, production performance, and survival in catfish is still needed to strengthen these results.

Abstract

This study investigated the interaction between salinity and container colour on survival, growth, and feed conversion ratio in barramundi (Lates calcarifer). This study used a Randomised Block Design (RCD), with juvenile sea bass (2-3 g weight, 5-6 cm length) stocked into a 70 litre aquarium. The study was conducted with a combination of three salinity levels (0, 15, and 30 ppt) and three container colours (blue, black, and orange). Each treatment consisted of 35 juveniles fed 40% crude protein pellets (0.7-1 mm) at 7% of biomass, three times a day for 28 days. There was a significant interaction between salinity and container colour on L. Calcarifers growth. Blue containers supported better weight gain at all salinity levels. Meanwhile, orange containers reduced the negative impact of high salinity on growth. Although, this interaction was not significant for length growth or feed conversion.

Abstract

The demand for grouper fish continues to increase, with the Ministry of Fisheries' annual production target of 9,000 tons. However, intensive cultivation systems pose risks due to poor water quality, making grouper fish susceptible to disease and health problems. This study investigated the impact of Trichodina sp. infection on the blood glucose levels of hybrid grouper fish. The study was conducted in May 2024 by observing 30 fish and measuring blood glucose. The results showed that 20 fish showed glucose levels above the normal range (> 57.6 mg/dL), ranging from 61 to 114 mg/dL. In contrast, nine fish had glucose levels in the normal range (28.8 - 57.5 mg/dL), and one fish had low glucose levels of 26.7 mg/dL. High glucose levels in most fish indicate stress or metabolic disorders, possibly related to Trichodina sp. infestation. This parasitic infection can compromise the fish's immune system, making the fish more susceptible to disease. The results emphasize the need for effective parasite management and optimal water quality in intensive culture systems. To maintain grouper health and productivity, it is important to monitor water quality, control ectoparasites, and conduct further research on the long-term effects of these infections on blood glucose and overall health.

Abstract

The introduction of non-native species into new environments poses significant ecological risks, including the potential for hybridization with native species, leading to genetic pollution and the creation of hybrid species. This study investigates the natural occurrence of hybridization between koi (Cyprinus carpio var. koi), an ornamental species, and the native mahseer (Tor dourorensis) in an artificial pond in Padang, West Sumatra, Indonesia. The coexistence of these species over five years resulted in the emergence of hybrid offspring, which exhibited a blend of morphological traits from both parent species. Meristic and morphometric analyses revealed that the hybrids closely resembled mahseer in certain characteristics, particularly dorsal and caudal fin rays, while also displaying koi-like traits such as body color and fin shapes. The hybridization of these two species raises concerns about the genetic integrity of native mahseer populations and the potential ecological consequences. Hybrids may possess competitive advantages that could further threaten already vulnerable mahseer populations. The findings emphasize the urgent need for effective management strategies and regulations to prevent the release of non-native species into sensitive ecosystems. Public awareness campaigns and regulatory measures should be strengthened to mitigate the risks of hybridization and protect biodiversity in freshwater environments. This study serves as a critical reminder of the far-reaching consequences of introducing non-native species, particularly in regions with delicate ecological balances.
