Abstract

Pranoto, Rosariastuti R, Prihandoko A. 2018. Short Communication: The utilization and effectiveness test of andisol soil-bioball-Agrobacterium sp. toward heavy metal chrome removal. Biodiversitas 19: 1955-1959. This study was aimed to determine chrome metal adsorption and bioremediation ability using andisol soil, bioball, and Agrobacterium sp. The andisol soil characterization was performed by Fourier Transform Infrared Analyzer (FTIR), X-Ray Diffraction (XRD), and Surface Area Analyzer (SAA) while the cell quantity of Agrobacterium sp. was calculated by hemocytometer method. Determination of adsorption and bioremediation effectiveness was done using several parameters such as effect of pH solution, ratio variation of andisol soil-bio ball-Agrobacterium sp, and contact time. The pH variation was ranged from 1-6, while the composition variation of andisol-bioball-Agrobacterium sp. were 2: 0: 0, 1, 5: 1: 5, 1: 2: 10, 0, 5: 3: 15, and 0: 4: 20 (gr: item: mL), and contact time of 30,60,90,120, and 150 minutes. The result of this research shows that the optimum adsorption and bioremediation of Cr metal at pH 5, while the optimum ratio of andisol soil-bioball-Agrobacterium sp was 1, 5: 1: 5 (gr: item: mL), with the contact time for 120 minutes with Cr decrease percentage of 71.3%. The adsorption isotherm followed Langmuir and Freundlich isotherm.

Leunca (Solanum americanum Mill.): The uses as vegetable in two villages in Upper Citarum Area, Bandung, West Java, Indonesia. Biodiversitas 19: 1941-1954. Leunca is known as botanical name as Solanum americanum Mill, Family of Solanaceae. In recent years, academic interest has been increasing. After so long studied as weeds, today leunca has also studied because of its important meaning as crop that has high nutritional and economic value in relation to food resilience of developing countries, as because of its chemical substances with its medicinal properties. Leunca was recorded in colonial period by botanists or agricultural scientists’ report as local vegetable in rural of West Java also in modern time by anthropologist or ecologists who studying rural population. In the recent time in Indonesia, leunca studies almost all have been focused on its pharmacological, agronomic, and economic aspects. The aspect that is related to Sundanese sociocultural system was almost neglected. This paper presents the finding of research on ethnobotany of leunca includes landraces, agronomical and utilization, traditional institutional aspect, and culinary culture food habits of leunca in rural Sundanese people. Method used in this study mixed-method of qualitative and quantitative was applied in this study, while some techniques including observation and semi-structured interviews were carried in the field research. The result of study showed that based on informants it has revealed that 7 kinds of plant that are named as leunca, however, only 3 kinds of leunca that are grown in their village. Among 7 kinds of leunca, leunca biasa (S. americanum) has been predominantly consumed both fruits and leaves. There is various food dishes are consumed fresh or cooked. Various dishes of leunca biasa have been culturally integrated everyday life of people and culturally as part of people identity of Sundanese people (urang Sunda). Other kinds of leunca, including leaves of leunca manuk (variety of S. americanum) have been consumed leunca as cooked vegetable, and its leaves have been used as traditional medicine of pet chicken disease.

Abstract

Medina CE, Díaz DR, Malaga BA, Medina YK, López E. 2018. Short Communication: Second record of Eumops chiribaya (Chiroptera, Molossidae) in Peru. Biodiversitas 19: 1979-1984. The bat Eumops chiribaya was recently described from southwestern Peru, based on unique specimen collected in the coastal desert of Moquegua Department. Herein, we document the second record for the country based on a specimen collected in Arequipa Department. This specimen is described in detail and compared with the holotype of E. chiribaya. This record represents a significant extension of the geographic range for this species, both in latitude and elevation, and it show the high diversity of bats living in the coastal valleys of southwestern Peru. Our record adds one genus and species to the Arequipa Department, totaling 17 bat species in the Department.

Predicting impacts of future climate change on the distribution of the widespread selaginellas (Selaginella ciliaris and S. plana) in Southeast Asia. Biodiversitas 19: 1960-1977. The current global climate is moving towards dangerous and unprecedented conditions that have been seen as a potentially devastating threat to the environment and all living things.  Selaginella is a fern-allies that needs water as a medium for fertilization, hence its distribution is presumed to be affected by climate change. In Southeast Asia (SEA), there are two widely distributed selaginellas, namely Selaginella ciliaris and S. plana. S. ciliaris is a small herb (up to 4 cm), annual, abundant during the rainy season, and found in the middle-high plains, whereas S. plana is a stout large herb (up to 80 cm), perennial, and mainly found in the lowlands. The purpose of this study was to determine the potential niche distribution of S. ciliaris and S. plana under current climatic conditions, and to predict its future distribution under the impacts of climate change. We used Maxent software along with bioclimatic, edaphic, and UV radiation variables to model the potential niche distribution of those two selaginellas under current and future predictions climate conditions. We generated future predictions under four detailed bioclimatic scenarios (i.e., RCP 2.6, RCP 4.5, RCP 6.0, and RCP 8.5) over three times intervals (2030, 2050, 2080). The results showed that future climatic conditions in the SEA had been predicted to significantly disrupt the distribution of suitable habitat of S. ciliaris and S. plana, and alter their geographic distribution patterns. Although some areas were predicted to become suitable habitat in the early period of future climate change, the overall projections show adverse effects of future climate conditions on the suitable habitat distribution of S. ciliaris and S. plana, as estimated losses of suitable habitat will be higher than the gains.

Priyono DS, Solihin DD, Farajallah A, Arini DID. 2018. Anoa, dwarf buffalo from Sulawesi, Indonesia: Identification based on DNA barcode. Biodiversitas 19: 1985-1992. Anoa is an endangered endemic species in Sulawesi. The controversial issue of anoa conservation until now is the taxonomic status of lowland and mountain anoa. This study aims to test the ability of DNA barcoding techniques to identify the taxonomy between mountain anoa and lowland anoa. A 681bp fragment of cytochrome oxidase subunit 1 (COI) gene was obtained and used to solve the molecular taxonomic problem and to resolve the phylogenetic relationships of the two types of anoa. Our results showed that the DNA barcode is useful in assigning the taxonomic position of anoa. In the phylogenetic tree, we found that the two types of anoa were in separate clades. We also found that based on the Kimura-2 parameter (K2P), the genetic distance between the two types of anoa showed higher values (3.4%) than the threshold of the separating species level. We, therefore, proposed that the binomial nomenclature for both types of lowland and mountain anoa are respectively Bubalus depressicornis and Bubalus quarlesi. We suggest that the use of DNA barcode techniques in anoa taxonomic studies and their implementation will be useful in conservation management

Koi Herpesvirus (KHV) has caused considerable losses in the cultivation of the Cyprinidae family worldwide. In addition to infecting Cyprinidae, KHV can also infect other freshwater fish. This study reports histopathology and ultrastructural analysis of KHV that infect Nile tilapia. Although there are no specific clinical symptoms, pathognomonic indicating Nile tilapia fish has been infected with KHV have been found, i.e. through the formation of intranuclear inclusion body and cell swelling which essentially experience chromatin margination. In addition, histopathology results indicate changes or damage to other tilapia tissue, i.e. necrosis, hypertrophy, edema, hemorrhage, inflammation, congestion, degeneration, and apoptosis. These results indicate that KHV has been distributed and replicated in tilapia organs. Based on the Transmission Electron Microscopy (TEM) analysis, KHV virions have been detected in the cytoplasm or outside the cells on a size of 150-170 nm.

Abstract. Yemata G, Desta B, Fetene M. 2019. In vitro antibacterial activity of traditionally used medicinal plants against Xanthomonas campestris pv. musacearum in Ethiopia. Biodiversitas 20: 555-561. In Ethiopia, traditional medicinal plants have long been used to treat human and livestock ailments. Nevertheless, studies about the use of these plant extracts to control crop diseases are scarce. Therefore, the aim of the present study was to evaluate the antibacterial activity of traditionally used medicinal plants against Xanthomonas campestris pv. musacearum (Xcm). The bioactive chemicals from leaf samples were drawn using methanol by maceration method. Total phenolic content of the extracts was determined by Folin Ciocalteau reagent. The antibacterial activity of leaf extracts was evaluated by disc diffusion method. The phytochemical analysis revealed the presence of alkaloids, flavonoids, phenolic compounds, terpenoids, tannins and saponins. Extracts with higher total phenolic content had greater antibacterial activity. The extract of each species showed antibacterial activity against Xcm on a dose dependent manner. Significant differences were recorded between species and test concentrations. The extract of Bersama abyssinica exhibited the strongest antibacterial activity at 200 mg/mL followed by Ricinus communis, Eucalyptus citriodora and Acokanthera schimperi. At lower test concentrations, extracts of E. citriodora and R. communis revealed higher antibacterial activity. Moreover, these species had lower bacteriostatic and bactericidal concentrations. The results showed the potential potency of E. citriodora and R. communis leaf extracts in controlling enset bacterial wilt. However, further studies on the identification of chemical compounds and in vivo evaluation of the extracts are recommended.

Biodiversitas 19: 670-675. Agroforestry is an ecologically and environmentally sustainable land use that offers great promise to carbon (C) sequestration. Forests play a significant role in reducing greenhouse gas emissions through maintaining current carbon stores and by increasing the rate of carbon sequestration. Vegetation carbon stocks are necessary to be quantified to evaluate the carbon sequestration potential in the ecosystem. Reasonable methods for estimating tree biomass and carbon storage on forest land are increasingly crucial given concerns of global climate change. This study aimed to evaluate C sequestration potential by agroforestry in North Sumatra Indonesia. This study was conducted at the Agroforestry system in Aek Nauli, Simalungun District, North Sumatra. Data collection for primary data was done through a field survey. The present study was carried out to determine above ground tree biomass of Toona sureni (Blume) Merr and Coffea arabica. Data retrieval of T. sureni and C. arabica was done by non-destructive sampling by measuring the diameter at breast height (dbh). The results showed that the potential of average above-ground biomass and carbon storage of T. sureni and C. arabica was 6.25 t ha-1 and 2.88 C t ha-1, respectively. Total aboveground biomass of Toona sureni and C. arabica in the study area was 93.75 ton, while total of carbon storage was 43.16 ton

Riyanto R, Widodo I, Abbas B. 2018. Morphology, growth and genetic variations of sago palm (Metroxylon sagu) seedlings derived from seeds. Biodiversitas 19: 682-688. Propagations of Sago palm using seeds will result in large variations of seedlings due to segregation resulting in the genetic diversity. The objectives of this study are to observe the morphology, growth and genetic variations of sago palm seedlings derived from seeds resulted from natural pollination. Morphological diversities of sago palm seedlings showed in high variations with the similarity coefficient ranging from 10% to 69%. Growth patterns of sago palm seedlings were divided into three categories, i.e., slow, medium, and fast growth. Genetic characters of sago palm seedlings showed that sago palm seedlings were varied with coefficients ranging from 23.6-74.5% and seedlings samples were separated into three groups of 49% the differentiation level. Based on the morphological, growth, and genetic characteristics of sago palm seedlings derived from seeds, it is predicted that sago palms in the natural condition may occur because of cross-pollination.

Ekyastuti W, Ekamawanti HA. 2018. Short Communication: The role of microbial rhizosphere in enhancing plant growth of Jatropha curcas in soil contaminated mercury. Biodiversitas 19:  701-705. Soil in the area of ex-gold mining, has the chemical-physical constraints to the growth of plants. These chemical-physical constraints are low organic matter, poor of nutrient, acid pH, very low CEC, soil texture dominated by sand, and mercury contamination. This area needs to be rehabilitated. Previous research has found that Jatropha curcas as a plant tolerant to mercury. On the other hand, some types of microbial rhizosphere such as arbuscular mycorrhizal fungi (AMF) and mercury reducing bacteria (MRB) also have an ability to reduce mercury. The purpose of this study was to determine the role of microbial components of AMF and MRB in enhancing the growth of J. curcas in tailings contaminated mercury. The study was conducted in two places, in the greenhouse and in the tailing area of ex-gold mining, using factorial completely randomized design. Results showed that interactions between AMF and MRB were simultaneously able to enhance the growth of J. curcas not only in the greenhouse, but also in the field (tailing area). In the greenhouse (nursery), several isolates of Bacillus sp, Bacillus sp + Glomus SS11 and Bacillus sp + Glomus SS18 in the forms of inoculum were very effective in enhancing the seedling growth of J. curcas. However, results were apparently changed after those seedlings were planted in the field (tailings of ex-gold mining). The combination of Bacillus sp. + Glomus SS18 was the best treatment to enhance the growth of J. curcas of all used treatments. This result proves that the role of microbial rhizosphere, especially AMF and MRB, could effectively enhance the growth of J. curcas in tailings contaminated with mercury.
