The study on the life cycle of Neocaridina spp. is important because it plays a major role in the economy and trade between countries. However, there is a lack of sufficient studies on this particular topic. One of the factors that influence Neocaridina spp. life production is its life cycle and the associated water parameters. The objectives of this study were to conduct, observe, and record the life cycle of Neocaridina denticulata and N. palmata and the associated with water parameters (temperature, pH, ammonia, nitrite, and oxygen) in the aquarium. The results showed that there was no difference in the life cycle stages between the two shrimp species. Both species reached the first sexual maturity stage of the life cycle at 75 days, with the maximum body length of the mature shrimp ranging from 2.30-3.00 cm. The eggs became larvae in 15 days, larvae reached the juvenile stage after 60 days, and the juvenile became mature in 15 days. The number of eggs was influenced by the body size of the female shrimps; smaller females produced fewer eggs. Optimal water quality varies by species and must be monitored to ensure growth and survival. The temperature range in this study was 25-28°C and the pH was 6.5-8.0. The ammonia level was in the range between 1.21 and1.72 mg/L, the nitrate was between 0.06 and 7.91 mg/L, and the oxygen was around 5.5-7.9 mg/L. All of the water quality parameters in this study are within the acceptable range for Neocardina cultivation.

Abstract. Yulita KS, Atikah TD, Wardani W, Susila. 2020. Unraveling genetic variations of Dalbergia latifolia (Fabaceae) from Yogyakarta and Lombok Island, Indonesia. Biodiversitas 21: 833-841. Dalbergia latifolia Roxb. (Fabaceae) or sonokeling is a timber species extracted for its beautiful heartwood used mainly for musical instruments. D. latifolia was listed in Appendix 2 of CITES since 1997, and the demand for its timber is increasing. The species is considered vulnerable (VU A1cd ver 2.3) to extinction according to the IUCN Red List, while cultivations were mainly done from a root cutting. To anticipate the further loss of population size while maintaining sustainable use of the species, it is essential to evaluate the genetic diversity since the genetic diversity level could serve as an indicator of plant adaptability to its environment. This present study was the first to report the genetic diversity of five populations of D. latifolia from Yogyakarta and Lombok Island, West Nusa Tenggara, Indonesia. Ten combinations of Sequence Random Amplified Polymorphism (SRAP) primers were used to amplify 48 samples of D. latifolia from five populations in Yogyakarta and Lombok Island. The 137 SRAP loci were used for a cluster, and population genetic analysis, with mean NPL, was of 13.6/locus, PLP of 56.18%, H of 0.16, and PIC for all population was of 0.28. The lowest gene diversity (H = 0.11) was observed in population WL 1 of Lombok Island, while the highest (H = 0.20) was recorded for population KP of Yogyakarta. The Yogyakarta and Lombok populations were genetically structured at a medium level (Gst = 0.27), with low genetic distance (0.0310-0.1281), and a moderate level of variation observed among individual trees (ca. 0.5 coefficient similarity). The results of this present study were expected to provide necessary information on further research of D. latifolia from Indonesia to support the sustainable use of D. latifolia in Indonesia and the preservation of this species.

Short communication: The genotype of growth hormone gene that affects the birth weight and average daily gain in crossbred beef cattle. Biodiversitas 21: 941-945. Growth Hormone gene has been proposed to direct search for quantitative trait loci, and polymorphisms at these loci have been associated with several production traits in bovine. This research aimed to investigate the association of the genotype of partial growth hormone gene with birth weight and average daily gain in crossbred beef cattle. The materials of this research consist of 47 samples from four different breeds. Birth weight was measured just after calving delivery from the mother. The average daily gain was calculated from the period since birth weight to the second weight (90 days). Genotyping of the growth hormone gene was determined by the sequencing and PCR-RFLP method with the AluI restriction enzyme. The association between genotype and growth traits data was analyzed using analysis of variance (ANOVA) by RStudio version 1.1.383. The results showed the significantly different (P<0.001) in birth weight and average daily gain according to the different breed. Friesian Holstein breed showed the highest birth weight, followed by Belgian Blue Bull cross and others. The genotype valine/valine only occurs in Belgian blue bull cross and Wagyu bull cross cattle and showed moderate birth weight. Genotype affects birth weight significantly (P<0.001). The birth weight for genotype leucine/leucine, leucine/valine, and valine/valine was 34.82±18.13, 25.31±13.10, and 29.35±13.65, respectively. There was no significant difference in average daily genes according to the different genotypes. In conclusion, the growth hormone gene was probably one of the molecular genetics markers for excellent growth traits in different crossbred beef cattle.

Abstract. Tajuddin M, Santosa E, Sopandie D, Lontoh AP. 2020. Characteristics of growth, flowering and corm yield of iles-iles (Amorphophallus muelleri) genotypes in third growing period. Biodiversitas 21: 570-577. Iles-iles (Amorphophallus muelleri Blume) is an emerging crop for producing glucomannan, and it is predominantly cultivated in agroforestry systems. The study aimed to evaluate characteristics of growth, flowering, and production of genotypes in the third growing period in order to develop new clones. The experiment used 2-year-old corm of ten genotypes, i.e. BKB, CF, CR, LSP, SB, SBM, SG-BKK, SGH, SR and STS. The corms were planted in June 2018 to May 2019 in the field under 45% artificial shading net. Plant growth was evaluated on weekly basis from bud emergence until dormant. Flowering rate was ratio the number of corms with flower to number of planted corms in each replication. Corm dry mass was measured after harvest, which was conducted at dormant stage at 46 weeks after planting. Candidate clone was selected from the highest total score of five criteria i.e. flowering rate, corm shape, corm healthiness, marketable yield, and total yield. Results demonstrated that genotypes exhibited variation in growth, flowering rates, and yield. Based on emerging time, three genotypes (SB, SGH, SG-BKK) were classified as early emergence, five genotypes (CF, CR, LSP, SBM, STS) as a medium, and two genotypes (BKB, SR) as the late emergence. All genotypes produced inflorescence at rate 1 to 52%, and 3 genotypes (CF, CR, STS) had flowering rate >20%. Five genotypes (CR, LSP, SBM, SGH, STS) had an average corm weight > 1500 g. CR and SBM genotypes produced the largest average corm weight, i.e., 2,509.33 g and 2,129.25 g, respectively, while the BKB produced the smallest one (912.94 g). All genotypes had similar dry mass content, i.e., 18.73-20.70%. The scoring evaluation recommended CR, SBM, and STS genotypes as candidates of new clones with productivity > 30 ton/ha. It needs further evaluation of selected clones in the farmer production field.

Morphometric variations of Asian Common Palm Civet (Paradoxurus Hermaphroditus, Pallas 1777) from Bali Island, Indonesia as the basis of morphometrics diversity data. Biodiversitas 21: 1027-1034. Asian Common Palm Civet (Paradoxurus hermaphroditus, Pallas 1777) is one of the small carnivores that also lives in Bali island, Indonesia. Civets can process coffee beans by fermenting them in the digestive tract, so the coffee beans have a unique aroma. This coffee product, commonly known as Kopi Luwak, is the most expensive type of coffee in the world. The purposes of this study were to obtain the morphometric variations, genetic diversity, and genetic relationship of Bali civets. In this study, we involved 73 civets from four different areas (Bangli, Tabanan, Gianyar, and Denpasar) of Bali Islands, Indonesia. The quantitative character was the body size of animals, and the qualitative character was their hair color. The body size among populations was not significantly different (P>0.05). The complete canonical structure analysis showed that tail length (1.04) and body length (0.76) in canonical 1 and body height (0.96) and head width (0.94) in canonical 2 could be used as a group differentiator. The genetic distance among Bali civets was categorized as close except Bangli civets having far distance than others.

Abstract. Susanti R, Yuniastuti A. 2020. Short Communication: Prolactin and x-collagen genes polymorphism in Central Javanese local ducks, Indonesia. Biodiversitas 21: 605-610. Genetic conservation is an effective approach to preserve unique and protected species in Indonesia, including indigenous duck (Anas platyrhynchos Linnaeus, 1758). One of the most important things is an identification effort by studying genetic characteristics to support genetic resource conservation programs. This study aim was to analyze the genetic polymorphisms of Central Javanese local duck based on prolactin (PRL) and collagen-x (COLX) genes. A total of 35 local ducks in Central Java was collected as samples, the duck’s calamus feathers were taken for tissue DNA isolation. Polymorphism analysis of PRL and COLX genes were performed using single nucleotide polymorphism (SNP) markers and PCR-RFLP method. The amplicons were then digested with DraI restriction enzyme (AhaIII) (5'TTT/ AAA) for PRL gene, and HaeIII (BsuRI) enzyme (5'GG/ CC-3 ') for COLX gene. The genotype was determined based on RFLP PCR amplicon band results from each target gene. For each gene, data on genotype frequencies, allele frequencies, expected heterozygosities (HE), and Hardy-Weinberg equilibrium were analyzed using PopGene32 software. Based on the PRL gene, there were 25 ducks (71.43%) with BB genotype and 10 ducks (28.57%) with AB genotype. While based on the COLX gene, as many as 25 ducks (71.43%) had TT genotypes, 9 ducks (25.71%) had TC types and 1 duck (1.86%) had CC types. Both PRL and COLX genotypes were clustered the Central Javanese local ducks into 5 haplotypes (A-E). The high allele frequency has belonged to B allele (85.71%) of the PRL gene and T allele (84.25%) of the COLX gene that showed high egg production and quality. Allele B (PRL gene) and T allele (COLX gene) could be predicted as weight eggs’ alleles marker.

The royal palm (Roystonea regia (Kunth) F.Cook is one of the palm species that are many planted along the streets as part of urban landscapes. As biologically products, in nature they can self-cleaning with drop off their big leaves. The study aims to examine the morphological characteristics of ultrastructure and elemental content, crystallinity, as well as fibers derivation characteristics from the royal palm. The samples were taken out from leaf sheaths that divided into three sections along the width of i.e. outside, middle, and inside; in two conditions of leaf sheaths namely green and brownish-dry. The morphology structure and elemental content were observed by the Scanning Electron Microscopy (SEM) and the Energy Dispersive X-ray analysis (EDX), while the crystallinity was analyzed using X-ray diffraction (XRD), and fiber characteristics were measured to determine cell dimensions as well as to calculate Runkle ratio (RR), muhlsteph ratio (MR), coefficient of rigidity (CR), felting powder (FP), and flexibility ratio (FR). The diameter of palm fiber bundles was decreased from outside towards inside. The model of vascular tissue was rounded which located in the central region of the fiber bundles. The silicon content in the inside section was lower than those in the middle and outside sections. It was in line with the degree of crystallinity in the inside section which that higher than other sections. The highest degree of crystallinity was about 18 %. The fiber length of royal palm leaf sheath was in values around 6000 ?m. However, the royal palm leaf sheath had thick fiber walls and thin lumen, as well as other fiber derivation characteristics of muhlsteph ratio, the coefficient of rigidity and the flexibility ratio, were in low-quality values. For that reason, the study provides information that the royal palm sheath was recommended as a composite reinforcing material.

Characteristics of Bacillus thuringiensis isolates indigenous soil of South Sumatra (Indonesia)  and their pathogenicity against oil palm pests Oryctes rhinoceros (Coleoptera: Scarabaeidae). Biodiversitas 21: 1287-1294. Bacillus thuringiensis is a gram-positive, entomopathogenic bacterium that could be isolated from soil and be used to control various plant pests. Oryctes rhinoceros is an important pest in oil palm. Application of B. thuringiensis-based bioinsecticides is an alternative in controlling these pests. The purposes of this study were to isolate and identify B. thuringiensis bacteria from the soil of South Sumatra, production of B. thuringiensis-based bioinsecticides and to test their toxicity to O. rhinoceros larvae. The study was conducted in several cities/districts in the province of South Sumatra. Soil samples were taken from various habitats and B. thuringiensis isolates were grown on NGKG agar media. Among 76 soil samples (6 districts and 2 cities) B. thuringiensis colonies were obtained leading to 24 isolates of B. thuringiensis. Toxicity screening tests for armyworm Spodoptera litura were 55.79% (53 isolates) and their mortality to 25.26% O. rhinoceros larvae (24 isolates). From these isolates whose effectively killed O. rhinoceros larvae, 10 isolates were taken and propagated with Nutrient Broth (NB) and biourine enriched with 5% molasses. Number of spores produced was counted during 24, 48 and 72 hours. Furthermore, a bioassay test was carried out on O. rhinoceros larvae for 7 days. Isolate of KJ3P1 caused the highest mortality of O. rhinoceros larvae after 7 days of observation. SDS Page resulted in KJ3P1 and KJ3R5 isolates showing several bands whose content of various types of protein molecular weight. Isolation of B. thuringiensis in South Sumatra produced 2 isolates potentially to be active ingredients in production of bioinsecticides which were effective in killing O. rhinoceros larvae.

Bioconversion of isoflavones glycoside to aglycone during edamame (Glycine max) soygurt production using Streptococcus thermophillus FNCC40, Lactobacillus delbrueckii FNCC41, and L. plantarum FNCC26. Biodiversitas 21: 1358-1364. Due to its strong radical-scavenging and antioxidative activity, isoflavones in soybeans have received great attention for the development of functional foods. This study focused on bioconversion of isoflavones glycoside into its aglycone form of edamame green soymilk fermented with three lactic acid bacteria (LAB), i.e., S. thermophilus FNCC40, L. bulgaricus FNCC41, and L. plantarum FNCC26 to produce soygurt. Green soymilk was fermented with 6% (v/v) of LABs as a starter culture for 24 hours at 37ºC. Its antioxidative activity were measured using DPPH free radical scavenging activity method. Daidzein and genistein released during fermentation were fractionated using HPLC and detected further by LCMS to confirm the presence of these two substances. The results showed that the population density of starter culture in green soymilk reached 109 CFU/mL, and the pH decrease from 6.8 to 3.5. All LABs cultures used in the fermentation process were able to produce free aglycone, releasing more daidzein and genistein. Increasing daidzein and genistein content in soygurt results in increasing antioxidative activity. The highest antioxidative activity (IC50 = 41.01 mg/mL) was found in the soygurt fermented with S. thermophilus FNCC40. This finding indicates that S. thermophilus FNCC40, L. bulgaricus FNCC41, and L. plantarum FNCC26 are potential as an effective starter culture to produce a soygurt with good antioxidant activity.

Biodiversitas 21: 1074-1081. Studies on the effects of varying cutting intensity on the abundance and species richness of Diptera in tropical rainforest are limited, particularly in Southeast Asia region. The aim of the study was to assess the temporal effect of cutting intensity on Diptera community in tropical rainforest in the eastern Borneo rainforest, Indonesia, which was logged using the Indonesian Selective Cutting and Planting system. The field study was carried out in 2016. Responses of Diptera to the Indonesian Selective Cutting and Planting systems in the eastern Borneo rainforest, Indonesia were examined. We compared the abundance and morphospecies composition of Diptera before cutting and after cutting at three different treatments: low cutting intensity, medium cutting intensity and high cutting intensity. Diptera was collected using a malaise trap. Selective cutting of tropical rainforest altered biodiversity of Diptera. The abundance and morphospecies composition of Diptera were greater after cutting than before cutting at all cutting intensities. Our study showed that cutting intensity did not significantly affect the abundance and morphospecies composition of Diptera. Results of the study clearly indicated that the percentage of forest canopy cover could be a single predictor for abundance and morphospecies composition of Diptera in the natural rainforest of eastern Borneo, Indonesia.
