Biodiversitas 21: 2638-2644. The assessment of genetic diversity is a key step in evaluating the adaptation of populations to new environmental conditions, and thus in the selection of new cultivars. The present investigation aimed to assess genetic variability of bread wheat (Triticum aestivum L.) genotypes grown in Algeria. The 17 hexaploid genotypes of bread wheat were assessed using 16 molecular microsatellites of SSR (Simple Sequence Repeat). Among the 16 microsatellite markers tested, only 11 markers were the most polymorphic and reproducible. The Polymorphism Information Content (PIC) values per locus varied from 0.14 to 0.70 with an average of 0.48 and 0.49. Genetic similarity between genotypes varied from 0.27 and 0.92 with an average of 0.60. The highest genetic distance value of 0.92 has been scored between Kauz/Pastor/Fiscal and Wbll1*2/Brambling. The lowest value of 0.27 has been scored between Cham6 and Pastor/Wbll1. Genetic similarity was calculated by molecular derived data and used to produce a dendrogram. The genotypes were grouped in two clear clusters according to their origin and pedigree. The first cluster included wheat genotypes Ain Abid (local genotypes), Attila/2 Pastor, and 5119 (introduced genotypes). The second group revealed high polymorphism and was subdivided into four sub-groups. This high level of diversity revealed among the accessions of wheat, grown in Algeria could be used in breeding programs.

Protein profile, amino acids, and taurine composition of sea slug (Paromoionchis tumidus) from Sumenep sea waters, Madura, Indonesia. Biodiversitas 21: 2430-2436. Sea slug (Paromoionchis tumidus) is one of the marine gastropods and inhabits the intertidal zone. Sea slug has long been used by coastal communities as food and medicine for breast cancer. Sea slugs are rich in nutritional content, a protein with the composition of essential amino acids, unsaturated fatty acids, and minerals, which are important as food and medicine in the future. The objective of the study was to investigate the composition of proximate and amino acids, protein profile, and taurine content of sea slug from the sea waters of Sumenep Madura. In this work, the sea slug collected from Sumenep was analyzed for protein, amino acids, and taurine. We compared the composition of proximate and amino acids before and after dried (fresh and dried), and part of the body of a sea slug (whole, muscle, viscera, and mucus) for the protein profiling and taurine content. The results showed that protein content reached 9.32% (fresh) or 60.82% (dried). The molecular weight of the protein was ranging from 22.53 kDa (viscera) to 49.61 kDa (viscera). The total amino acids were found at 6.14% (fresh) and 53.87% (dried). Furthermore, taurine was present at range from 53.568 µg/mL (viscera) to 158.784 µg/mL (whole). This experiment imparted a consistent result with previous studies regarding the richness of protein content in sea slug. Thus there is a wide opportunity for using it as taurine rich products in functional food and medicine.

Slamet, Central Java, Indonesia. Biodiversitas 21: 2946-2954. Saurauia microphylla de Vriese has the potentials for medicinal uses, yet it is listed in the IUCN Red List under Vulnerable status. The natural population of this species is only found in highlands one of which is in Mount Slamet, Central Java, Indonesia. This study aims to determine the population structure, habitat preference and predicted spatial distribution of S. microphylla in the Mount Slamet area. The method used was purposive sampling by establishing 103 observation plots with size of 20x20 m2 for each plot. The research parameters recorded were abiotic variables and biotic associations. Data were analyzed using Principal Component Analysis (PCA) to determine the most influencing environmental factors on the presence of S. microphylla. Species association of S. microphylla was analyzed using the Jaccard index equation. Spatial distribution model was analyzed using Maxent (Maximum Entropy) software. The results showed that S. microphylla populations were found in highland forest areas with elevations of 1689-2265 m above sea level, slope of 3-40°, temperature of 16°-26°C, air humidity of 49.3-90%, soil pH 5.8-7, soil moisture 5-70% and canopy cover of 0-92%. Our analyses revealed that elevation is the most influential factor in the presence of S. microphylla. There are 20 tree species which found around S. microphylla habitat with 4 species have significant association with the species. The spatial distribution model showed that S. microphylla spread across all the slopes of Mount Slamet with the southwestern slope was predicted had the most extensive habitat suitability for the species and become smaller to the north, east, and south slopes.

Molecular identification of bacteria isolated from culture medium of rotifer fed on fishery waste diet. Biodiversitas 21: 2735-2740. The aim of this study was 16S-rRNA sequences based molecular identification of bacteria isolated from culture medium of rotifer fed with fishery waste diet (FWD). We cultured rotifer Brachionus rotundiformis in sterilized seawater (salinity 25 ppt) using FWD, following the procedure in Patent No. P00201609066. Bacteria from the culture were collected, homogenized, diluted 10 to 1000 fold, spread on agar plates and incubated at 370C for 24 to 48 hours. Representative colonies of the bacteria according to their morphologies were isolated for further characterization. Genomic DNA of the isolates were extracted, and the 16S rRNA gene of the isolates were amplified. Polymerase Chain Reaction (PCR) product of each isolate was sequenced and queried against the NCBI GenBank database. Six different isolates based on size, color, elevation, margin, and colony were observed during 24-48 hours incubation at 370C. The 16S rRNA genes of the six isolates were successfully amplified and produced DNA band at 1300-1500 bp, with quality value equal to or greater than 20 (QV20+) of each entire sequence around 941-1253 bases. Basic Local Alignment Search Tool (BLAST) queries in the NCBI GenBank and EzBioCloud database using the 16S-rRNA gene sequences showed that the six isolates belong to four different genera, i.e: Bacillus, Staphylococcus, Vibrio, and Alteromonas.

Perceptions on utilization, population, and factors that affecting local distribution of Mimusops zeyheri in the Vhembe Biosphere Reserve, South Africa. Biodiversitas 24: 6432-6439. Mimusops zeyheri Sond., was once an important plant species to local communities in Southern Africa but nowadays is considered as underutilized plant. Undoubtedly, the neglect of indigenous knowledge associated with M zeyheri could lead to its gradual disappearance. This study aimed to evaluate local people‘s perceptions about the contribution of M. zeyheri to their livelihoods, population status and factors affecting its local distributions in the Vhembe Biosphere Reserve, Limpopo Province, South Africa. Semi-structured research questionnaires were administered to 52 randomly selected respondents. The majority of respondents (67.31%) perceived the decline in the population of M. zeyheri within the studied sites. More than three-quarters of laypeople (78.6%), two-thirds of farmers (66.7%), and 57.1% of environmentalists stated that M. zeyheri possessed more economic benefits than other livelihood benefits. A total percentage of 44.20% of participants perceived fire as the main factor that negatively affects the distribution of M. zeyheri in the studied regions. Documenting information about people's perceptions regarding the contributions of M. zeyheri to local livelihoods and assessing factors that affect its local distribution is a fundamental step toward understanding its associated sociocultural values, socio-economic, and threats. Future studies should incorporate both ethnobotanical and ecological data-gathering methods and validations.

Abstract. Ramarumo LJ, Maroyi A. 2020. An inventory of useful threatened plant species in Vhembe Biosphere Reserve, Limpopo Province, South Africa. Biodiversitas 21: 2146-2158. Scientists and conservation managers are seeking to understand and monitor plant species that are likely to be on the verge of extinction risk. Monitoring of threatened plants’ extinction risk can be better achieved through insights about indigenous knowledge dynamics associated with those species. This study aimed to document detailed information about useful threatened plant species in the Vhembe Biosphere Reserve, Limpopo Province, South Africa. Data was collected through interviews with 203 participants and literally counting of individuals as per the IUCN’s Red List Criteria. A total of 13 useful and native threatened plants belonging to 12 families were recorded. The majority of the threatened plant species were being used for medicinal purposes only (46.0%) followed by the mixture of medicinal and ornamentals (23.0%). The frequently cited useful threatened species with UV > 0.024, RFC > 0.059 and FL > 5.911%, includes Asparagus sekukuniensis, Bowiea volubilis, Brackenridgea zanguebarica, Ocotea bullata, Rhynchosia vendae, Siphonochilus aethiopicus and Warburgia salutaris. About 47.0% of the recorded useful threatened plants were distributed in remote areas of the Thathe Von?o and its surroundings. Threatened plants with the population size < 100 adult individuals constitute an overall of 61.54% of all the recorded species. The current study provides substantial information about useful threatened plant species in the studied region. Detailed information about threatened plant species remains fundamental for making informed decisions that are important for managing species of conservation concern.

Metagenomic analysis and biodiversity of Lactic Acid Bacteria (LAB) on masin (fermented sauce) from Sumbawa, West Nusa Tenggara, Indonesia. Biodiversitas 21: 3287-3293. Masin is a spontaneously fermented sauce from Sumbawa, West Nusa Tenggara Indonesia that is made of shrimp paste, chili, turmeric flower, and herbs mixed with some spices. This study aims to isolate Lactic Acid Bacteria (LAB) and analyze the metagenomic and biodiversity of its bacteria. Genome isolation for metagenomic analysis was using ZymoBIOMICSTM DNA Miniprep Kit. Sequencing analysis to identify LAB strain form masin was using 16S rRNA universal primer. Metagenomic analysis showed that relative abundance bacteria in masin for order taxon was Lactobacillales, the family taxon was Enteroccocaceae, and the genus was Tetragenoccocus. Six different groups were obtained from the phylogenetic tree analysis of 40 isolates found in masin. The representatives of each group taken were isolates number 2, 17, 11, 34, 28,  and 5. Based on the results of sequencing analysis, the 6 isolates found in masin are Staphylococcus piscifermentans strain CIP103958 (Isolate Code 2), S. piscifermentans strain BULST54 (Isolate Code 17), S. piscifermentans strain SK03 (Isolate Code 11), S. piscifermentans strain ATCC 51136 (Isolate Code 34), S. piscifermentans strain PCM 2409 (Isolate Code 28) and S. piscifermentans strain PU-87 (Isolate Code 5). Through this research, it has been described a whole diversity of bacteria contained in masin, one of them is Lactic acid bacteria. The six LAB isolates that have been identified can be developed as starter candidates for masin production.

Abstract. Ao A, Changkija S, Tripathi SK. 2020. Species diversity, population structure, and regeneration status of trees in Fakim Wildlife Sanctuary, Nagaland, Northeast India. Biodiversitas 21: 2777-2785. The plant species composition and diversity represent the overall health of the forest ecosystems and provide useful insight on forest conservation, and the species regeneration is important in assessing the potential of the forests to serve the society on sustained basis. This present study aimed to assess the species diversity, population structure, and regeneration status of the trees in temperate forest of Fakim Wildlife Sanctuary, Nagaland, Northeast India. Tree species composition, population structure and regeneration status were analyzed by randomly establishing eight plots of 0.1 ha within the sanctuary. Density, basal area, abundance, evenness, and other diversity indices were calculated for mature trees with girth at breast height (GBH) ? 30 cm. A total of 60 tree species belonging to 40 genera and 27 families were recorded. The most dominant families recorded were Lauraceae and Rosaceae. The total tree density recorded from the forest was 432.5 individuals ha-1 with a basal area 42.8 m2 ha-1. Cinnamomum zeylanicum and Lithocarpus pachyphyllus had the highest density with 17.58 individuals ha-1 and Lithocarpus pachyphyllus had the highest basal area with 2.64 m2 ha-1. The species with the highest IVI was Betula alnoides (17.13). Girth class of 30-60 cm had the highest density and basal area with 280 individuals ha-1 and 20.7 m2 ha-1. The diversity indices such as Shannon-Wiener index, Simpson index, Pielou evenness index, and Margalef species richness index were 3.90, 0.93, 0.92, and 11.59, respectively in the wildlife sanctuary. The present study showed that 41% of tree species exhibited good regeneration status, 31% as fair regeneration, 8% as poor regeneration and 20% had no regeneration. The present data will be useful to understand the current status of tree species and will serve as baseline information to the forest department, policymakers and conservationists to develop management plans for the conservation of priority species in the area.

Traditional ecological knowledge of the bird traders on bird species bird naming, and bird market chain: A case study in bird market Pasty Yogyakarta, Indonesia. Biodiversitas 21: 2586-2602. The bird-keeping in Indonesian society has been popular for a long time. Because the demand for birds has increased in an urban area, plenty of bird species are predominantly traded in urban bird markets. Bird market is a place for selling and buying various birds. The rise of bird trade in the bird markets in many regions of Indonesia has provided social and economic benefits for many people. But in addition to providing many benefits, the plentiful bird trade in bird markets that are not well managed is very worrying about the extinction of various bird species in rural ecosystems. This paper aimed to elucidate the Traditional Ecological Knowledge (TEK) of bird traders on bird species traded, bird naming and folk classification of bird species, and market chain system based on a case study in bird market of Pasty, Yogyakarta. The method used in this study was a qualitative method with the ethnoornithological approach. The results of the study showed that it was documented 106 bird species representing 40 families traded in the Pasty bird market. The common local names and folk bird classification are culturally based on bird vocalization, special colors, morphological characteristics, special habitats, involvement in bird song contests, flying racing competitions, selling prices, and wilderness. Most wild and semi-bird species are traded in the Pasty bird market are predominantly supplied from a rural ecosystem.

Abstract. Hartini YS, Nugroho LH. 2020. Short Communication: Antibacterial effect of red betel (Piper crocatum) extract in combination with vancomycin against  Staphylococcus aureus. Biodiversitas 21: 3271-3274.  The occurrence of bacterial resistance to a single antimicrobial compound has prompted the search for antimicrobials in the form of compound combinations. The current study was performed to show the antibacterial effect of red betel (Piper crocatum Ruiz & Pav.) in combination with vancomycin against Staphylococcus aureus. The combination of the antibacterial compounds from plant with the antibacterial drug was expected to increase antibacterial activity resulted in the reduction of resistance. The interaction effect of extract and antibiotic combination was performed using the microdilution checkerboard method. The combination of red betel extract with vancomycin showed significantly (p < 0.05) greater inhibition growth of S. aureus compared to red betel or vancomycin alone. The addition of red betel extract to vancomycin reduced Minimum Inhibitory Concentration (MIC) vancomycin to be a-4 fold reduction against S. aureus. There was a synergistic effect of the red betel extract in combination with vancomycin against S. aureus. It is the potential result for future research on the infectious treatment of S. aureus resistant to vancomycin.
