Phytochemical screening and antioxidant activity of wild mushrooms growing in tropical regions. Biodiversitas 22: 4716-4721. This study aims to determine the presence of secondary metabolites which generally have bioactivity that is antioxidant with phytochemical tests and antioxidant tests in Auricularia auricula, Schyzophyllum commune, Microporus xanthopus, and Trametes versicolor fungi that grow in the wild. Phytochemical analysis was carried out by testing color changes and modifications, while antioxidant tests were carried out by using a UV-Vis spectrophotometer and a DPPH solution (1,1-diphenyl-2-picrylhydrazyl). This research found that edible wood fungus that grows, wild such as A. auricula, contains secondary metabolites, namely flavonoids, triterpenoids, saponins, and tannins, but does not contain steroids, alkaloids, carotenoids, or coumarin, while S. commune contains secondary metabolites, namely flavonoids, steroids, tannins, and coumarin, but does not contain triterpenoids, alkaloids, carotenoids, or saponins and non-edible wood fungus, such as M. xanthopus, contains secondary metabolites, namely, flavonoids, saponins, tannins, coumarin, and steroids, but does not contain triterpenoids, alkaloids, or carotenoids, while T. versicolor contains compounds, secondary metabolites, flavonoids, saponin, tannins, coumarin, and triterpenoids, but does not contain steroids, alkaloids, or carotenoids. The antioxidant test results are based on IC50 calculations; it is known that the IC50 value of A. auricula is 499.25 g/mL, while S. commune is 121.37 g/mL, M. xanthopus is 251.20 g/mL, and T. versicolor is 493.04 g/mL. Based on the research results, edible and inedible wood mushroom extracts contain several phytochemicals and show potential for antioxidant activity.

Abstract. Chumroenphat T, Saensouk S, Saensouk P. 2021. Chemical composition and antioxidant activity of three species of Cornukaempferia in Thailand. Biodiversitas 22: 4036-4044. Genus Cornukaempferia (Zingiberaceae) were used as medicinal plants in Thailand. Cornukaempferia has never been reported the chemical compounds and antioxidant activity before. This study was designed to identify the chemical composition and antioxidant activities in root, rhizome, stem, and leaves of Cornukaempferia aurantiflora, C. larsenii, and C. longipetiolata. DPPH scavenging activity and Ferric reducing antioxidant power (FRAP) assays assessed the chemical composition and antioxidant activity. The results showed that total phenolic content (TPC) and total flavonoid content (TFC) found different plant parts obtained in this study were in the order of root>rhizomes>leaves>?stems. The C. aurantiflora had a high TPC of 416.39 mg GAE/g dry weight, while individual phenolics, such as vanillic acid at 256.90 mg/g dry weight, showed high contents in the leaf part. For C. larsenii,  TFC and individual flavonoids, such as catechin, have high 62.52 and 56.47 mg/100g dry weight values in the roots, respectively. The antioxidant activities in different species of different plant parts (DPPH: root>leaves?> rhizomes?>?stems; FRAP: leaves?>root>?rhizomes?>?stems). The genus Cornukaempferia is a rich source of bioactive compounds and antioxidant activity. Therefore, the genus Cornukaempferia will be used as medicine and food to produce products with health benefits in the future.

Abstract. Yuningsih L, Hearmansyah, Ibrahim E, Marsi. 2021. Diversity, structure and composition of vegetation in post-coal mining reclamation area in Sumatra, Indonesia. Biodiversitas 22: 3392-3400. The majority of coal mines are operated through an open-pit system which has a significant impact on the environment. Therefore, reclamation and revegetation activities are required to recover ecological sustainability. This study aimed to analyze the diversity, structure, and composition of vegetation resulting from the post-mining revegetation process in a mining concession in Sumatra, Indonesia at various intervals of post-reclamation periods, namely 11 years, 10 years, 8 years, and 5 years. Data collection was conducted by sampling with a 2.5 percent intensity. The study discovered 23 species of naturally grow understory plants and 25 species of woody plants. The stand structure at the eleventh, tenth, and eighth years of revegetation resembled an inverted J curve. The Species Richness Index (R) was determined to be low, the Shannon diversity index (H’) was is low to moderate, whereas the evenness index was moderate to high. We concluded that the structure of vegetation in the post-mining reclamation region resembled that of a natural forest ecosystem, yet the biodiversity indicators remain low and below those of natural forest. We expected the result of this study can enrich the limited knowledge of the ecology of post-mining reclamation in the tropics.

Abstract. Khan K, Listyanto T, Soraya E. 2022. Moisture content, density, and allometric model for estimating above-ground biomass of Peronema canescens trees in the private forest. Biodiversitas 23: 1132-1139. The objectives of this study were to quantitatively measure the above-ground biomass (AGB) of Peronema canescens Jack species in a private forest in Indonesia using a destructive method and to develop an allometric model for the same. Six trees were sampled for above-ground biomass and their components (stem, branch, and leaves) were weighted and measured. Regression analysis was used for developing the allometric equation by using predictor variables of diameter at breast height (DBH), squared diameter at breast height together with height (DBH2H), and diameter and height separately (D & H). The average AGB of six sampled trees was 0.264 (26.38%) tons per tree. The fit allometric equation AGB = 0.254(DBH2H)0.886 was developed for estimating above-ground biomass of P canescens and it had 99.2% accuracy. The implications and recommendations were provided based on the results of the study.

Abstract. Geraldi A, Tay CC, Ni’matuzahroh, Fatimah, Hanafi WNW. 2021. Unraveling the bacterial diversity of Cangar Hot Spring, Indonesia by Next Generation Sequencing of 16S rRNA gene. Biodiversitas 22: 4060-4066. This study is the first attempt at using the Next Generation Sequencing (NGS) method with 16S rRNA to understand the bacterial community structure in an Indonesian hot spring. This study aims to unravel the bacterial diversity of the Cangar Hot Spring as one of the most explored natural hot springs in East Java, Indonesia. We found Proteobacteria and Bacteroidetes as the two most abundant phyla. We discovered the first occurrence of genera Cloacibacterium and Methylobacillus in the hot spring ecosystem, which was the most dominant genera at Cangar Hot Spring. We also found several potential bacteria for bioindustry and bioremediation, such as Acinetobacter junii and Pseudomonas alcaligenes. Besides that, we also observed opportunistic pathogens from genera Comamonas and Vogesella. This study result will provide valuable information for further bioprospecting of bacteria with commercial potential and the development of health and safety measures in the Cangar Hot Spring, among others. Hopefully, this report would encourage the use of NGS technology for studying other hot springs in Indonesia.

Abstract. Feliatra F, Mardalisa M, Efendi I, Adelina A, Feliatra VA. 2021. Biodiversity of Escherichia coli bacterial resistance to multidrug isolated on the Dumai coast of Indonesia. Biodiversitas 23: 10-16. Anthropogenic pollution around the Dumai coast of Indonesia disturbs marine microorganisms, including commensal bacteria such as fecal Escherichia coli. Escherichia coli have often been used as a sensitive indicator for the spread of Antibiotic Resistance Genes (ARGs) among pathogens. This study aimed to analyze the development of E. coli resistance, originating from the Dumai sea waters using six antibiotics against seven E. coli isolates (E1, E4, E6, E8, E11, E13, and E15) at the five stations. Furthermore, a sensitivity test was performed on Mueller Hinton Agar (MHA) solid media, based on the Kirby-Bauer disk diffusion method. The results showed 100% E. coli isolates resistant to narrow-spectrum antibiotics (Penicillin, Isoniazid, Streptomycin, and Erythromycin) at the five stations. Meanwhile, high resistance of E. coli against the broad-spectrum antibiotics was observed in station 4 (mangrove habitat) existed in Chloramphenicol, whilst no resistance was found in Ciprofloxacin. Escherichia coli isolates showed Multidrug Resistant (MDR) index value higher than 0.2 which suggests there is a high-risk of antibiotics pollution in Dumai seawater. Bioinformatics analysis of 16S rRNA gene sequences represents the identity of E. coli isolates at the species level (97.84% - 99.74%). This study revealed the presence of antibiotic pollution in Dumai seawater which may impact public health. Hence, the local community must be discipline about the use of antibiotics freely.

Abstract. Viersanova A, Purwanto H. 2021. Genome analysis of Lysinibacillus sphaericus isolate 6.2 pathogenic to Culex quinquefasciatus Say, 1823 (Diptera: Culicidae). Biodiversitas 22: 5211-5222. Lysinibacillus sphaericus is an entomopathogenic bacteria that is specific to vector mosquitoes, especially Culex spp., and Anopheles spp., so it has been widely used as a bioinsecticide. L. sphaericus has a wide variation of toxicity efficiencies, which have led to continuous exploration of new isolates with higher toxicity and a new toxin to deal with resistance problems. This study aimed to identify the genomic characteristics and toxin characteristics of isolate 6.2 based on whole genome analysis and analyze the identification of isolate 6.2. Isolate 6.2 was previously obtained from rhizosphere in Yogyakarta. To analyze the genome and toxins, the NGS technique was used and then the analysis was carried out using a couple of freely available bioinformatics tools. Molecular identification was carried out with the 16SrRNA gene and the relationship was analyzed by reconstructing the phylogenetic tree using Neighbours-Joining. The genomic analysis of isolate 6.2 showed good results with G+C content and genome size that matched the reference genome of L. sphaericus. The result of the 16SrRNA gene blasting showed that the closest related gene of isolate 6.2 is L. fusiformis (NR_042072.1). However, the reconstructed phylogenetic tree did not show the formation of clusters according to the species. Toxin analysis indicates that isolate 6.2 has Mtx, s-layer protein, hemolysin, and chitin-binding protein genes. All of which are known to be associated with the toxicity of L. sphaericus to binary toxin resistant population of Culex quinquefasciatus.

Bacterial abundance and community composition in green, brown and red water from intensive Catfish (Clarias sp.) culture ponds in Yogyakarta, Indonesia. Biodiversitas 22: 3677-3684. Catfish (Clarias sp.) is an important aquaculture commodity in Indonesia and cultured in an intensive system. Microorganisms play an important role in maintaining water quality of aquaculture system. The objective of this study was to determine the bacterial abundance and community composition of green, brown and red water collected from intensive catfish culture ponds in Yogyakarta using next-generation sequencing method. The water samples were collected from intensive catfish culture ponds with different colors, namely green, brown and red ponds located in Yogyakarta. The DNA from water samples was extracted using DNA extraction kit and used as template for 16S rRNA amplification. The V3-V4 hypervariable regions of the 16S rRNA gene were amplified apply for next-generation sequencing technology. This study could explore effectively the bacterial community in water samples. The bacterial communities in this catfish culture water showed higher bacterial richness compared to the other aquaculture system. The diversity of the green, brown and red catfish culture water ponds was similar with the number OTUs of the green, brown and red water samples, which were 1269; 1387 and 1323 OTUs respectively. The 694 OTUs (34.42%) were common core microbiomes in all catfish culture ponds, the 212 OTUs (10.51%) are present on green and brown water ponds, the 182 OTUs (9.02%) were on green and red water ponds, and the 183 OTUs (9.07%) were present on green and brown water ponds. However, the composition of the bacterial community was different. The most dominant phylum in green and brown water ponds was Proteobacteria with relative abundance in green water and brown water 71.6% and 47.0% respectively, whereas, the most dominant phylum in red water was Firmicutes (29.5%). The dominance of Firmicutes phylum in red water ponds may be caused by application of probiotic bacteria, the high organic content, and low oxygen concentration.

Vegetation structure, aboveground biomass, and carbon storage of wono¸ local forest management in Gunungkidul, Yogyakarta, Indonesia, across three geomorphological zones. Biodiversitas 22: 3207-3218. Wono is local community-based forest management in Gunungkidul District, Yogyakarta. This land use has the potential to reduce carbon dioxide emissions through their carbon sequestration capacity as well as to produce renewable energy sources through wood biomass for charcoal and wood pellet. Since Gunungkidul is unique in terms of geomorphological characteristics, study on the vegetation structure, biomass estimation, and carbon storage of wono across geomorphological zones are important. Therefore, this study describes the vegetation structure of wono in three geomorphological zones of Gunungkidul District, as well as estimates the aboveground living biomass (AGB) and aboveground living carbon storage (AGC). The quadratic sampling technique was used to collect data for vegetation analysis with the size of the plots were 20 m x 20 m, 10 m x 10 m, 5 m x 5 m, and 2 m x 2 m for trees, poles, saplings, and seedlings, respectively. A total of 32 plots were established, consisting of 18 plots in Nglanggeran Village, 12 plots in Dengok Village, and six plots in Girisekar Village, each village representing geomorphological zones of Batur Agung, Ledok Wonosari, and Pegunungan Seribu, respectively. The AGB was performed non-destructively and estimated using referenced allometric equations. Furthermore, the AGC was calculated using a conversion factor of 0.47 from the obtained AGB. The results showed that the identified species at wono in Batur Agung, Ledok Wonosari, and Pegunungan Seribu zones were 13, 7, and 8, respectively. Swietenia macrophylla had the highest important value index (IVI) of 185.22% in the Batur Agung zone, while Tectona grandis was the most important species in both the Ledok Wonosari and Pegunungan Seribu zones with IVI= 238.27% and 178.60%, respectively. The biodiversity in these three zones was very low in terms of species diversity (H' < 2) and species richness (R1 < 3.4). The estimated AGB and calculated AGC in the Batur Agung, Ledok Wonosari, and Pegunungan Seribu zones were 210.96 ton ha-1 and 99.15 ton C ha-1, 73.58 ton ha-1 and 34.58 ton C ha-1, and 57.92 ton ha-1 and 27.22 ton C ha-1, respectively.

Silba adipata (Diptera: Lonchaeidae) parasitoids on cayenne pepper (Capsicum frutescens) in Bali, Indonesia. Biodiversitas 22: 3929-3935. The black fig fly (Silba adipata) is newly emerging pests that attack cayenne pepper (Capsicum frutescens L.) plants in Bali Province, Indonesia. This study aimed to determine the parasitoid type, community structure, distribution, and parasitization rate of the parasitoids of S. adipata on cayenne pepper in Bali. The sampling method employed in this study involves purposive sampling techniques on 100-150 pieces of cayenne pepper infested with S. adipata at each location point, with the emerging parasitoids identified morphologically. Three types of parasitoid species were associated with S. adipata, which include Asobara japonica Belokobylskij, Fopius arisanus (Sonan), and Diachasmimorpha longicaudata (Ashmead). The parasitoid community structure associated with S. adipata had a low abundance index, low diversity index, and a moderate dominance index. Based on the distribution of the three parasitoids, D. longicaudata is distributed evenly in Bali, whereas F. arisanus was not found in Jembrana . Conversely, A. japonica was only found in Badung, Bangli, Gianyar, and Klungkung. As summary, D. laungicaudata had the highest parasitization rate compared to the F. arisanus and A. japonica. D. longicaudata is a potential parasitoid, which can be studied for the future biological control of S. adipata.
