The indigenous and traditional foods of the Alas tribe have a long history and unique traditions that have been passed down for thousands of years. Unfortunately, food tradition is strongly intertwined with the health-related and therapeutic reasoning of food ingredients and preparation methods. However, changes in people's consumption patterns, which prefer fast food, lead to a decline in local knowledge of traditional foods, particularly among the younger generation. Therefore, the aim of this study was to document the traditional Alas food in Southeast Aceh District, Indonesia. The study was conducted in eight subdistricts of Southeast Aceh, Indonesia, namely Lawe Alas, Babul Rahmah, Tanoh Alas, Babussalam, Darul Hasanah, Lawe Bulan, Lawe Sumur, and Bambel subdistricts. The study enlisted 80 respondents (10 from each district) who were chosen at random. The interview was conducted using a questionnaire that covered the respondents' backgrounds, traditional food names, food composition, mode of preparation, and uses. A total of 16 traditional Alas foods were documented, including six types of traditional cuisine and 10 types of traditional snacks. All respondents recognized Manuk Labakh, Ikan Pacik Kule, Puket Megaukh, and Buah Khum Khum as traditional Alas foods. On the other hand, less than 25% of respondents were familiar with some other traditional Alas foods, such as Menekem. The Alas tribe has a great system in place for passing down traditional knowledge from generation to generation. A total of 24 plant species consisting of 21 genera and 14 families was used as traditional Alas food. The Alas tribe prepares a variety of foods from various plant species to maintain their health and contribute to conservation efforts.

Abstract. Growth response of Dendrocalamus asper on elevational variation and intra-clump spacing management. 2021. Title. Biodiversitas 22: 3801-3810. Dendrocalamus asper (Schult. Schult. F.) Backer Ex. K. Heyne is a well-known commodity classified as a non-timber forest product (NTFP) to substitute wood-based products in the future. While bamboo is widely distributed in various habitats, and it could impact growth performance and quality. Nevertheless, the development of bamboo research on the upstream level is quite rare, specifically for clumping bamboo species. Therefore, our study aimed to reveal the performance in elevational variation and to discover the intra-clump spacing and diameter relationship. The elevational variation was divided into three levels, which were lower, middle, and higher levels. Each elevation was established in 9 plots with parameters observed were culm diameter at breast height (DBH), culm height (H), and culm volume (V) of D. asper. The intra-clump spacing was used to assess the relationship between the clump density and diameter growth. The research is complemented with in-depth interviews to explore the traditional silvicultural practices of Dendrocalamus asper and spatial analysis to generate land surface temperature and soil moisture index. The result showed that there is no effect (P > 0.05) of elevational variation to D. asper growth and development, while the availability of intra-clump spacing showed a significant result (P < 0.05) on the culm DBH of D, asper. Furthermore, our result suggests a wider intra-clump spacing (0.4-0.6 m2) is more recommended than a narrow intra-clump spacing for optimal culm diameter growth. Additionally, bamboo plantation was still less managed and utilized. Therefore, improving the productivity of D. asper by maintaining plantation, i.e., fertilizing, managing spacing among clump bamboo, and harvesting to achieve sustainable development of the bamboo plantation is useful.

Tannins in mangrove plants in Segara Anakan Lagoon, Central Java, Indonesia. Biodiversitas 22: 3508-3516. Mangrove tannin is polyphenol compound and extractive matter in mangrove vegetation. Mangrove tannins have the potentials to support paint industry, animal feed, tanners and wood adhesives industry. This research is aimed to investigate the mangrove species in Segara Anakan, Central Java, Indonesia that produce tannins, and to analyze the distribution and contents of tannin in the part of mangrove plant. A total of 342 samples from 19 mangrove species was collected from 37 sampling points in Segara Anakan. UV-Visible Spectrophotometry was used to analyze the tannin content in the samples. The results showed that the tannins contained in bark and stems were higher (66.6%) than those in leaves (33.4%). Mangrove vegetation can be classified into five classes in terms of tannin percentage with Heritiera littoralis, Nypa fruticans, and Rhizophora mucronata had highest tannin percentage. The clustering analysis of mangrove tannin showed that Ceriops decandra-Ceriops tagal, Acacia auriculiformis-Sonneratia alba, Sonneratia caseolaris-Xylocarpus granatum, and Avicennia marina-Rhizophora apiculata had high similarity of tannin percentage. The distribution of tannins in mangrove species is as follow: 0.59-10.14 kg trees-1 (bark and stem of mangrove diameter > 10 cm) and 0.20-3.74  kg trees-1 (leaves of mangrove diameter > 10 cm) until 8.84-158.96 kg trees-1 (bark and stem of mangrove diameter > 40 cm) and 4.60-91.65 kg trees-1 (leaves of mangrove diameter > 40 cm). R. mucronata and R. apiculata had the highest total tannin content, ranged between 386.60-460.38 kg trees-1.

Application of wheat flour (Triticum aestivum) on spore density and sporulation efficiency of Bacillus megaterium isolated from Litopenaeus vannamei gastrointestinal tract. Biodiversitas 22: 3709-3715. Bacillus megaterium is frequently used in fish farming, such as white shrimp (Litopenaeus vannamei) pond, which can produce spores with high stability in its implementation. Currently, spore production still requires the availability of high-cost carbon sources. The objective of this research was to evaluate the effect of different wheat flour doses on spore density and sporulation efficiency of B. megaterium BM1. In flasks, 50 mL of each test medium was treated with different doses of wheat (10, 20, 30 and 40 g. L-1, respectively) and glucose was used as a control. Each treatment was inoculated with B. megaterium BM1 (2.6 x 108 CFU. mL-1) and incubated in a shaker incubator (120 rpm) at 37 °C for 120 hours. The results showed that the highest vegetative cell concentration (17 x 108 CFU. mL-1), growth rate (0.8 hour-1) and spore (14.7 x 108 spores. mL-1) were found in the wheat flour dose of 30 g. L-1. Furthermore, the highest sporulation efficiency was achieved at 20 g. L-1 of wheat (91.30%) and germination should be done at a dose less than 40 g. L-1. The size of the spores was 1.35-1.39 µm. Thus, 30 g. L-1 of wheat flour is a potential dose to produce spore for probiotic candidates.

Abstract. Agussalim, Umami N, Nurliyani, Agus A. 2021. The physicochemical composition of honey from Indonesian stingless bee (Tetragonula laeviceps). Biodiversitas 22: 3257-3263. The demand of honey has recently increased significantly, but this situation is widely used by irresponsible humans who made a fake honey. This present study aims to evaluate the quality based on the physicochemical composition of honey from Indonesian stingless bee Tetragonula laeviceps. Honey was obtained from three geographical origins in Indonesia, i.e. Sleman, Gunungkidul, and Lombok. The physicochemical composition of honey was analyzed such as moisture, ash, protein, pH, glucose/moisture ratio, and electrical conductivity, and amino acids. The results showed that the geographical origins had a highly significant effect (P<0.01) on moisture, pH, and significant effect (P<0.05) on ash, electrical conductivity, protein, and glucose/moisture ratio. Honey from Indonesian stingless bee T. laeviceps was found 17 amino acids were arginine, histidine, lysine, phenylalanine, isoleucine, leucine, methionine, valine, threonine, tyrosine, proline, glutamic acid, aspartic acid, serine, alanine, glycine, and cysteine. The dominant amino acids of honey from Sleman were glutamic acid, histidine, lysine, and arginine. Honey from Lombok were glutamic acid, aspartic acid, lysine, and histidine. Furthermore, honey from Gunungkidul were lysine, arginine, histidine, and phenylalanine. Honey from Sleman and Lombok have the best quality than honey from Gunungkidul based on the chemical composition and amino acids profile.

Vatica venulosa Blume is categorized as Critically Endangered A1c ver 2.3, according to the IUCN Red List. A study of the reproductive biology of V. venulosa Blume was carried out in August 2019-February 2020 at the Research Center for Plant Conservation and Botanic Gardens, Bogor, Indonesia. The research observed flower morphology, flower development, type of pollination, fruit and seed structures. V. venulosa is categorized as having a sub-annual flowering pattern. The flowering phenology of V. venulosa from flower bud emergence until fruit senescence takes about 6 months. The flower is hermaphroditic, with position of the pistil is higher/longer than the stamen; and pollen was released prior to stigma being receptive (protandrous). V. venulosa is a cross-pollinated plant, the flower visitors are insect nymphs of Thrips sp. (Thysanoptera: Thripidae), which are white and 1-1.5 mm in size. The percentage of blooming flowers was 32.3-37.9%, fruit set 10.9 %-12.6%. Seed physiological maturity  is reached at the age of 101±3-106±3 days after the flowers bloom when the wings are yellow green or green orange in color. V. venulosa seed is a non endospermous seed, and the cotyledons are composed mainly of small clumps of starch, being 42.5% carbohydrate.

Soil organic carbon (SOC) is one of the important components that can improve soil conditions and maintain agriculture sustainability. This study was conducted to determine the potential of soil organic carbon in grassland and bush forest in Aceh Besar District. We used purposive random sampling by taking soil samples at various depths with five samples for each land type. The parameters of soil bulk density, % carbon soil, the potential of carbon and carbon stock were analyzed. Soil organic carbon (SOC) analysis followed the Walkley and Black method using analytical guidelines from the Indonesian Soil Research Institute. The results showed that land type and soil depth affected bulk density, % carbon soil, the potential of carbon and carbon stock. Land type affected soil organic carbon in which grasslands had a higher soil carbon than bush forests. In total, total soil carbon stock in the bush forest of Aceh Besar District was higher than that in grassland due to the much larger extent of bush forest in the district. Land use management efforts to improve soil SOC need to be carried out and in collaboration with various parties. Thus, it is necessary to maintain the availability of soil carbon stock for sustainable land management, including agriculture.

Biodiversitas 22: 3401-3408. Leaves of betel vine (Piper betle Linn.) have been used as a traditional medicine in various regions in Indonesia. However, the genetic diversity of this plant has not been well recorded. Considering its diverse usage in traditional and folk medicine, it is essential to analyze and document the genetic diversity of Piper betle L. with the aim to collect the scientific data of betel vine genetics in Indonesia. This study aims at analyzing the population structure and genetic diversity of betel vine from the Singkil, Gayo Serbajadi, Baduy, Bandung, Hutan, Kalisusu, Kaledupa, and Mekongga accession groups of Indonesia using the sequence-related-amplified polymorphism (SRAP) marker technique. Genomic DNA was isolated from each accession and then used as a template for DNA amplification using PCR. As many as 16 SRAP primer combinations were screened for the genetic diversity analysis. Optimization of primer combinations resulted in 7 selected combinations based on their ability to generate clear amplification patterns and polymorphic bands. These were then employed for genetic diversity analysis. The genetic distance dendrogram showed the lowest similarity coefficient was 0.62 and that the betel vine grouping pattern was not based on genotype. The Singkil population had the highest genetic diversity and the Hutan population had the lowest. The mean value of Nei's genetic diversity index was 0.0985, while Shannon's information index was 0.01459 and the percentage of polymorphic loci was 25.81. This study concluded that the level of betel vine diversity is low.

Biodiversitas 22: 3685-3692. Edaphic mites play crucial roles in maintaining ecosystem services that are essential to human needs. However, the conversion of natural habitats followed by agricultural intensification may adversely affect edaphic mites. The objective of this study was to investigate the influence of different management systems on edaphic mite abundance, richness, and diversity in tropical rain forests in Indonesia. There were five forest management systems, which were as follows: secondary forest, production forest (pine monoculture), and three agroforestry systems (pine + coffee, mahogany + coffee, and mahogany + new cocoyam). We established a transect containing five research plots for each forest management system. Litter and soil from each plot were collected from December to March 2021. Temperature, relative humidity, and pH of litter and soil, as well as litter thickness, were measured. We found that edaphic mite abundance, richness, and diversity in the secondary forest were similar to managed forests. However, the aforementioned variables were significantly higher in “pine” systems than in “mahogany” systems. Our analysis evidenced positive correlations between litter thickness and edaphic mite abundance, richness, and diversity. Our findings may assist in selecting the appropriate forest management systems to rationalize the conversion of secondary forests to production forests and agroforestry.

Biodiversitas 22: 3556-3561. Environmental pollution by plastic from food packaging can be reduced by applying edible film as a biodegradable casing and safe for consumption. Utilization of Indonesia's biodiversity, especially purslane plants (Portulaca oleracea L.) at Boyolali, Central Java, Indonesia as an antimicrobial compound that can improve the quality of chitosan edible films. This study aims to determine the effect of purslane extract (Portulaca oleracea L.) as an antibacterial, antifungal, and antioxidant agent in edible films in inhibiting microbiological and oxidative damage to sausages. The film was made by mixing the extract (0%; 2.5%; 5%; 10%) into the chitosan solution and sorbitol, then applied to casing the sausage. The sausage was then stored at room temperature (27°C) and in refrigerator (4°C). Sausage quality testing was carried out on days 0; 3; 6; 9; 12; 15 during storage, including determination of Total Plate Count, Yeast Mold Count, and thiobarbituric acid test. The results of tensile strength, water vapor permeability, elongation, antioxidant, FTIR film, and morphology observed by using SEM of the films with and without the purslane extract addition were different. After 15 days of storage, Kruskal-Wallis analysis also revealed that the addition of purslane extract significantly inhibited the growth of microorganisms and oxidative damage to sausages.
