Abstract

Eagleton G.E. 2016. Review: Persistent Pioneers; Borassus L. and Corypha L. in Malesia. Biodiversitas 17: 716-732. This review traces advances in taxonomic and ethnobotanic understanding of the genera Corypha L. and Borassus L. gained from research since the time of publication of “Harvest of the Palm; Ecological Change in Eastern Indonesia’’ by James J. Fox in 1977. It posits testable hypotheses arising from the literature: firstly, that both genera were present in the furthest parts of island Southeast Asia prior to a definitive Indianized cultural expansion in the first millennium CE.; secondly, that two of their species ̶ “lontar” Borassus flabellifer L. and “gewang” Corypha utan Lam. ̶ were significant components of pre-agricultural economies of the archipelago, but that their full economic exploitation benefited from later cultural stimuli from the Indian subcontinent. To test these hypotheses, lines of research with potential benefits for local economies in semi-arid Indonesia are proposed.

Abstract

Iskandar J, Iskandar BS 2016. Ethnoastronomy-The Baduy agricultural calendar and prediction of environmental perturbations. Biodiversitas 17: 694-703. In the past, the village farmers of Java and other islands owned extensive the traditional ecological knowledge (TEK) on climate or pranata mangsa. It had culturally practiced as guidance to various agricultural activities, such as planting rice which is considered and fixed with dynamic climate conditions. Nowadays, however, the pranata mangsa has eroded and neglected by the majority irrigated rice (sawah) farmers. Unlike the sawah farmers, the Baduy people have culturally maintained the pranata mangsa (called by Baduy as pananggalan) for annual practicing the swidden farming (ngahuma). This paper discusses the way in which cultural practices of Baduy swidden farming based on traditional calendar. Method used in this study qualitative which is based on ethnoecology or ethnoastronomy approach. The result of study shows that the Baduy rice farming cycle isfixed annually with reference to an agricultural calendar. It has slightly affected by the various environmental perturbations, because the Baduy people have developed some strategies, such as by organizing the traditional calendar and applying the traditional agroforestry that productions can be used for both subsistence and commercial purposes.

Abstract

Amorphophallus muelleri Blume (Araceae) is valued for its glucomanan content for use in food industry (healthy diet food), paper industry, pharmacy and cosmetics. The species is triploid (2n=3x=39) and the seed is developed apomictically. The present research is aimed to identify genetic variability of six population of A. muelleri from Java (consisted of 50 accessions) using random amplified polymorphic DNA (RAPD). The six populations of the species are: East Java: (1) Silo-Jember, (2) Saradan-Madiun, (3) IPB (cultivated, from Saradan- Madiun), (4) Panti-Jember, (5) Probolinggo; and Central Java: (6) Cilacap. The results showed that five RAPD primers generated 42 scorable bands of which 29 (69.05%) were polymorphic. Size of the bands varied from 300bp to 1.5kbp. The 50 accessions of A. muelleri were divided into two main clusters, some of them were grouped based on their populations, and some others were not. The range of individual genetic dissimilarity was from 0.02 to 0.36. The results showed that among six populations investigated, Saradan population showed the highest levels of genetic variation with mean values of na = 1.500+0.5061, ne = 1.3174+0.3841, PLP = 50% and He = 0, 0.1832+0.2054, whereas Silo-Jember population showed the lowest levels of genetic variation with mean values na = 1.2619+0.4450, ne =1.1890+0.3507, PLP = 26.19% and He = 0.1048+0.1887. Efforts to conserve, domesticate, cultivate and improve genetically should be based on the genetic properties of each population and individual within population, especially Saradan population which has the highest levels of genetic variation, need more attention for its conservation.Key words: Amorphophallus muelleri, RAPD, genetic variability.

Abstract

Producion of nata de coco is growing fast, in line with increasing product of biocellulose demand. It is requrire a pure inoculum to reach the best biocellulose product. The aim of this reseach is to evaluate the most appropriate carrier material of gel inoculum for nata de coco. The four carrier material are Carboxy Methyl Cellulose (CMC), Agar, Sagu starch and biocellulose pap and its inoculated by Acetobacter sp. RMG-2 and A. xylinum. After inoculation, then put in the plastic bag (50 g/bag) and stored in 4° C. The texture of gel, population of cell andbiocellulose production were observer in 7 days. The result shown that all matterial were suitable to used as carrier for gel inoculum. Both CMC and biocellulose pap have good texture as standard qualities. Population of A. xylinum was 1.28x109 cfu/mL (in CMC carrier), 1.6x106 cfu/mL (in biocellulose pap carrier) after 15 weeks. The weight of biocellulose production was 500 g/L and 740 g/L medium respectively. While the population of Acetobacter sp. RMG-2 on CMC carrier was 1.79x108 cfu/mL and 7.75x107 cfu/mL on cellulose carrier with the weight of biocellulose production 630 g/L and 775 g/L medium respectively. Thus, the carrier material (CMC and Biocellulose pap) are able to keep bacteria without loss their capability to produce cellulose.Key words: Acetobacter sp. Abstract

The objective of the research was to investigate the Indonesian species of Rennellia Korth. based on their morphological characters. The investigation on morphological characters was based on herbarium specimens at the Herbarium Bogoriense and Herbarium Andalas University. Additional specimens also were collected during the course of the field work. There are four species recognized, i.e., Rennellia elliptica Korth., Rennellia elongata (King & Gamble) Ridl., Rennellia morindiformis (Korth.) Ridl. and Rennellia speciosa (Wall. ex Kurz) Hook.f. An identification key to the species was produced and detailed description of the taxa was also provided. The natural history data including habitat, distribution, uses and vernacular names were also presented.Key words: Rennellia Korth., Indonesia, morphological characters

Abstract

Putra DE, Sumadi, Kanazawa T, Hartatik T. 2016. Short Communication: Identification of Growth Hormone genepolymorphism for beef cattle in Pesisir Selatan District, West Sumatra, Indonesia. Biodiversitas 17: 711-715. This study aimed to determine gene polymorphism of growth hormone of domestic cattle in Pesisir Selatan District of West Sumatra. The research was conducted at Laboratory of Animal Genetics and Breeding Faculty of Animal Science, UGM from August 2013 to January 2014. Blood samples were collected from 66 individuals consist of 15 Pesisir cattle, 15 SimPes cattle, 15 SimPO cattle, 15 Bali cattle and 6 PO cattle. DNA was extracted from each blood samples after SDS-proteinase K digestion, and used for PCR-amplification for a region of growth hormone gene (211 bp), and then the PCR products were analyzed for restriction fragment length polymorphism (RFLP) using AluI restriction enzyme. The results showed that GH gene of Pesisir, PO and Bali cattle are monomorphic, which frequencies of L allele was1.00 and V allele was 0.000 while these LL genotype was 1.000. Frequency of L and V alleles in SimPO and SimPes cattle were 0.634, 0.366 and 0.700, 0.300, respectively. SimPO and SimPes cattle were polymorphic, LL and LV of SimPO cattle was 0.733 and 0.267 as well as SimPes cattle which LL and LV was 0.600 and 0.400, respectively. The correlation between genotype and the performance (body weight and body size) was not significant. The present study indicates that polymorphism of growth hormone gene in AluI site could not yet be used as a molecular marker for body weight and body size of beef cattle.

Abstract

Attempt to rehabilitate degraded natural forests in Indonesia is recently carried out by applying selective cutting and line planting (TPTJ) silvicultural system. One of the most important aspects of TPTJ silvicultural system is the procurement methods of large number of planting stocks. Shorea johorensis Foxw was investigated in this regards as one of promising Shorea species for rehabilitating degraded forests due to its fast growing character. The species is usually propagated by three different propagation techniques, namely up-rooted seedlings, seeds and cuttings. Genetic consequences due to different propagation methods in this species are poorly known and need to be studied in order to determine genetic variation and differentiation. Material from five origins (populations), namely: (i) up-rooted seedlings, (ii) seeds, (iii) cuttings, (iv) young plantation and (v) natural forest were randomly taken in the field and subsequently assessed by RAPD using threepreviously tested random primers of OPO-11, OPO-13 and OPO-16. Results showed that natural tree populations showed the highest levels of genetic variation with mean values na = 1.2593, ne = 1.2070, PLP = 25.93% and He = 0.1109. Cutting populations showed the lowest levels of genetic variation with mean values na = 1.1111, ne = 1.0773, PLP = 11.11% and He = 0.0445. Meanwhile, according to the propagation techniques, up-rooted seedling population revealed the highest levels of genetic variation with mean values na = 1.2222, ne =1.1613, PLP = 22.22% and He = 0.0886. Particular methods of plant propagation in this company, especially cutting method, reduced significant genetic variation of S. johorensis.Key words: Shorea johorensis Foxw, RAPD, genetic variation, silvicultural system

Abstract

The aims of this research were to: (i) study the effects of gibberellic acid (GA3) to the growth of Boehmeria nivea L. Gaudich, and (ii) study the effects of gibberellic acid (GA3) to the cellulose and lignin content of B. nivea. GA3 could delivered starch hydrolysis that could support α-amylase formation that would increase glucose. Increasing glucose concentration caused the rising of osmotic pressure inside the cell so that the cells expand. The initial synthetic path of cellulose was glucose, as well as lignin. If GA3 could increased the glucose amount inplant, then the cellulose and lignin amount also increased. This experiment use Completely Random Design with one factor, they are concentration gibberellic acid that consist of 6 treatment concentration are GA3 0 ppm, GA3 50 ppm, GA3 100 ppm, GA3 150 ppm, GA3 200 ppm, and GA3 250 ppm. The treatment to the rhizome had been done before it was planted. Some parameters like growth and anatomy parameter are measured. The results showed that treatment GA3 affect to the improvement of bud stem diameter, length of bud stem, thewet weight, dry weight, length of phloem bundle, phloem number, and lignin content but they does not affect to the change of bud number, leaf number, and cellulose content. The treatment of GA3 on GA3 200 ppm show the maximum result to the improvement of bud stem diameter, length of bud stem, the wet weight, dry weight, length of phloem bundle, phloem number, and cellulose content The treatment of GA3 on GA3 250 ppm show maximum result to the improvement of bud number, leaf number, and lignin content.Key words: gibberellic acid, Boehmeria nivea L. Abstract

The aim of this research was to study the physiological characters of kimpul [Xanthosoma sagittifolium (L.) Schott] in various light intensity (shading) and water availability. The physiological characters are growth, photosynthetic apparatus, tissue nitrogen and polyphenol content. Information about physiological character of tannia is important in order to their cropping development. The research done in randomizecomplete block design with 2 factors and 3 replicates. The first factor was light intensity (shading) in 3 levels (0%, 55%, and 75%). The second factor was water availability in 4 different fields capacities (40%, 60%, 80% and 100%). The treatments were be done in 6 weeks. There were 8 parameters measured (plant height, dry plant weight, total chlorophyll content, stomata index, nitrogen and polyphenol content. The data were analyzed by analysis of varians, followed by DMRT in 5% confident level. The result showed that the plant height significantly affected by the combinations of treatment (shading and water availability). The treatment also influenced the total chlorophyll content and leaf nitrogen content. The treatment didn’t give significantly effect on stomata index, and pholyphenol content.Key words: Xanthosoma sagittifolium (L.) Abstract

A study was conducted on artificial pollination, both self and cross pollinations, of Amorphophallus decus-silvae Back. & v.A.v.R. planted in a house yard in Bogor. It was observed that no fruit set was resulted from self pollination. On the other hand cross pollination successfully produced numerous fruits. It was suggested that phenomena was due to protogenic character rather than self incompatibility mechanism of the species. The stigmas mature 1-2 days earlier than the anthers (pollen shedding). Such a case was also reported earlier by otherauthors in A. titanium Becc.Key words: acung, Amorphophallus decus-silvae Back. & v.A.v.R., pollination, fertilization, fruit, seed.
