Abstract

The Sumatran tiger (Panthera tigris sumatrae) is one of several endemic Indonesian wild cat groups which population is critically endangered. A program to increase the population size had been conducted in captivity, especially in the zoo. In order to monitor the captive population and for the means of management in captivity, a logbook data recording system had been developed for individual animals. A compilation data from the Tiger International Stud Book from 1942 to 2000 was analyzed. The extraction data consisted of the reproduction performance of the animals, such as calving pattern, sex ratio, litter size etc. The results showed that mortality of cubs at ≤ 5 months old reached 59%, between 5 and 24 months old was 9.3% and above 24 months was 31.7%. Cubs were born all year round with concentration in July for Europe and North America regions. The mean of first reproductive age was at 4.6 years old (± 2.28), with the mean of the oldest reproductive age was at 8.3 years (± 3.63). Mean litter size was 2.21 cubs from dame born in captivity and 2.45 cubs from dame capture from the wild. Sex ratio of male to female was 53.8:46.2. The average lifespan of adult wild captive tiger was 5108.9 day (± 2365.4) day, while for adult (≥ 24 months of age) captive tiger was 4417.4 day (± 1972.7).© 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: Sumateran tiger, Panthera tigris sumatrae, reproduction, stud book.

Abstract

This research was conducted in the nature reserve on beach forest of Tangkoko Batu Angus. Study sites were located at 0 until 150 m above sea level. Data being collected were the type of feed, nest sites and habitat utilization. During the observation, 18 animals species were identified being consumed and consist of 77.7% insects, 16. 6% reptil and 5.5% bird. Abstract

The aim of this research was to expose the relation between tumor necrosis factor alpha (TNF-α) in the peritoneal fluid and in the serum of endometriosis patients. It was conducted at Dr. Muwardi Hospital Surakarta. Twenty patients undergoing laparoscopic operation because of endometriosis indication (Endometriosis Group), 20 women (aged 23 to 40) who undergo interval sterilization by means of laparoscopic technique. During laparoscopic operation, peritoneal fluid is taken to examine TNF-α by ELISA technique. At the same time, the serum isalso taken to examine TNF-α by the same technique. The research result indicated that by independent sample t-test, the TNF-α concentration in the Endometriosis Group is quite different from the control group (P=0.00). The simple linear regression test shows a strong positive one-way correlative relation between TNF-α concentration in the peritoneal fluid and TNF-α concentration in the serum in the Endometriosis Group. The research result indicated that the TNF-α concentration in the serum can be used as a reflection of endometriosis. Abstract

The hydrogen sulfide is one of compound which very often found in the shrimp pond caused by anaerobic decomposition or as a natural condition of the sea water which have volcano activity. This research was obtaining information of the differences of genetic expression between black tiger shrimp which could resist to H2S and the one which could not survive in this H2S. This research also trying to obtain information the genetic variety which could resist to H2S. The genetic variety of black tiger shrimp which could resist to H2S has been analysis with allozyme electrophoresis technique, using specific tissue meat and buffer CAPM (Citric Acid Aminopolimorpholine) pH 6.From the three enzymes analyzed it could be detected that IDH enzyme (Isocitrate dehydrogenase) has locus polymorphic, whereas enzyme α-GDP (α-glycerophosphate dehydrogenase) and LDH (lactate dehydrogenase) with monomorphic locus. The average heterozygosity for the group which could resist to H2S is 0.070, whereas the group which could not survive in the H2S is 0.042. The control group has heterozygosity 0.041. The group which could resist to H2S with higher heterozygosity will have bigger chance to survive and have better adaptation ability in environmental changes. Abstract

Indonesia as one of the megabiodiversity country has high biodiversity of plants, animals, and microbes. One of the microbe’s biodiversity is mould inhabiting plant rhizosphere. Research and exploration on mould inhabiting plant rhizosphere in Gunung Mutis Natural Reserve, NTT has never been conducted before. The objective of the study was to isolate and identify mould inhabiting several horticulture and reforestation plants rhizosphere in Gunung Mutis Natural Reserve, NTT. The mould isolation method was based on direct inoculation. Theresult showed that eight moulds genera with several species variants had been isolated. Abstract

Indonesian tropical climate have an opportunity for fungi growth as Aspergillus flavus Link which can produce aflatoxin within foodstuffs, include condiment of fermented shrimp. Aflatoxin B1 is the dangerous agent having roles as carcinogenic, mutagenic and teratogenic. The aims of this research were known kinds of moulds and detection of aflatoxin B1 on commercial condiments fermented of shrimp. Two brands of commercial condiments fermented of shrimp were taken from traditional markets and supermarkets in Surakarta. Isolation was done by made suspension of sample in aquadets. Suspension on appropriate dilutions was grown on CDA (Czapek’s Dox Agar) media withsurface spread. The grown colonies were separated and grown on PDA (Potato Dextrose Agar) slant media. Furthermore, isolates were cultured on CDA and MEA (Malt Extract Agar) media. The grown colonies were microscopes and microscopes examined and identified. Existence of aflatoxin B1 was known by Commercial RIDA Screen ELISA Kit that could detect qualitatively and quantitatively with detection sensitive < 1.7 ppb. Moulds that could be isolated from condiments fermented of shrimp were: Aspergillus flavus Link, Aspergillus niger vanTieghem, Aspergillus wentii Wehmer, Aspergillus PU1 or Aspergillus PU2 and Penicillium citrinum Thom. There was aflatoxin B1 contaminated to 2 brands of commercial condiments fermented of shrimp that were examined. Traditional markets’ commercial condiments fermented of shrimp contained higher aflatoxin B1 than supermarkets’. Abstract

The research of the physical and mechanical characteristic of rattans stem of 10 species of the genus Calamus from Bancea Nature Reserve and Lore Lindu National Park in Central Sulawesi had been conducted. The aims of this research were to describe phylogenetic relationship of those species based on physical and mechanical characteristic with cladistic approach (cladogram). The research had been used descriptive method, i.e. specific gravity, parallel attracting firmness of fiber, firmness stress parallels of flex firmness, and fiber/static curve. It was reconciled with standard of ASTM D no 143-52 with a few which has modified. The data was analyzed by ANOVA. The resultindicated that the mechanical and physical characteristic of Calamus rattan can be made as distinguishing evidences. Abstract

Sago Malintang Nature Reserve is one of conservation area in West Sumatra which have some potential plants. Ferns is one group which can be found ini this area, which several of them are attractive and useful. The aims of the research were to inventory ferns in Sago Malintang Nature Reserve and to know the acclimatization process of these species in Bogor Botanical Garden. The method used in this research were explorative method at the place and observation of the growth of living collections in Bogor Botanical Garden. The result showed that there were approximately 17 species of ferns in Sago Malintang Nature Reserve. Bogor Botanical Garden collected 11 species of them. The botanical information of each species is presented in this paper. Abstract

Endophytic microbes are microbes which live symbiotically inside the plant tissue. The potential of those microbes were studied for many purposes such as industrial and agriculture. The purpose of this research was to study the potential of endophytic microbes for bioactive compound production as plants protection agent. From 238 bacteria isolates tested, it were identified that 44 bacteria were against the Xanthomonas campestris, 49 isolates against Pseudomonas solanacearum, 28 isolates against Colletotricum gloeosporioides, 18 isolatesagainst Fusarium oxysporum, 19 isolates against Xanthomonas campestris and Pseudomonas solanacearum, 6 isolates against X. campestris and C. gloeosporioides, four isolates against X. campestris and F. oxysporum, two isolates against P. solanacearum and F. oxysporum, seven isolates against C. gloeosporioides and F. oxysporum, five isolates againt X. campestris, P. solanacearum and C. gloeosporioides. Fermentation process was conducted to collect the bioactive compound against pathogen. Isolates HL.50B.106 couldproduce bioactive compound against C. gloeosporioides, and F. oxysporum, isolates HL.13B.21 against X. campestris and P.solanacearum, isolates Hl.12B.19 against X. campestris, P. solanacearum and C. gloeosporioides. Thin layer chromatography analysis showed specific spot, different from standard (media). Spot was purple in color after sprayed with CeSO4. Rf value of HL.50B.106 extract were 0.51 and 0.53 (fraction 1 and 2), 0.62 and 0.64 (fraction 3 and 4).© 2006 Jurusan Biologi FMIPA UNS SurakartaKey words: endophytic bacteria, pathogen microbial against, TLC

Abstract

Fertility of Paraphalaenopsis serpentilingua (J.J.Sm.) A.D. Hawkes was investigated through experimental pollination. Several methods of pollen transfer were tested. In general, outcrossing demonstrate a better result compare to intercoss and selfing. This is indicated by seed morphological appearance and positive respond of germination test. However in the situation where flowers are limited by chance, viable seeds from selfing and intercross pollination can still be expected. The seed capsules reach maturity at 4-5 months after pollination.
