Abstract

ABSTRACTA research on acute toxicity of effervescent tablet from Piper betle leaf extractum on wistaralbino male rats was done. This research aimed to determine acute toxicity potential ofeffervescent tablet from piper betle leaf extractum, evaluated clinical symptom that perhaps occurdue to the giving of effervescent tablet, single dosage orally on 24 hours to 15 days observation.The research uses male rats, which are divided into 5 groups. Group I was negative control withaquadest. Group II was given sample with 0,38 g/kgBW dosage. Then, succesively group III with1,03 g/kgBW dosage, group IV with 2,79 g/kgBW dosage and group V was given test sample withhighest dosage, that was 7,50 g/kgBW dosage. On the basis of data analysis result having beenperformed both quantitively and qualitatively, it could be said that in general, the giving ofeffervescent tablet from Piper betle leaf extractum single dosage orally on male rats from 0,38g/kgBW dosage to highest level (7,50 g/kgBW) or approximately 19,74 times of therapy dosage,didn’t cause toxic effect. It didn’t cause mortality on research animal. So we can determine thequasi LD50, that was bigger than 7,50 g/kgBW, according to Loomis criteria (1978), that acutetoxicity potential of effervescent tablet from piper betle extractum was practically not toxic category.Key words: acute toxicity, effervescent tablet, Piper betle

Abstract

ABSTRACTIt is well know that adipose tissue mass increase greatly in obesity. In Indonesia obesityprevalence tends to increase. It raised the risk of morbidity and total cost attributable to obesityrelated disease. The adipose tissue is energy storage, fatty acids being released when fuel isrequired. It is now adipose tissue expresses and secretes factors with impotant endicrine functions.These factor include adiponectin, adipsin, leptin, protein of the renin-angiotensin system,plasminogen activator-1, other cytokines and resistin. A better understanding of the endocrinefunction of adipose tissue will give evidence of links between increased productios of someadipocyte factors and the metabolic and cardiovascular complications of obesity.Key words: adipose tissue, obesity, endocrine organ

Abstract

ABSTRACTA research on the isolation and identification of xanthone from the fruit rind of mangosteen(Garcinia mangostana L.) was carried out. Extraction was done using maceration with methanol –water solvent and fractionation with n-hexane and ethyl acetate solvent. Column chromatographywith silica gel as the stationary phase and n-hexane – ethyl acetate (7 : 3) as the mobile phase wasused for the isolation. From the ethyl acetate fraction was obtained GM II-1 isolate (Rf 0,61 ; nhexane– ethyl acetate 7 : 3 as mobile phase) which showed a colour change from reddish brown toyellow under UV light 366 nm after spraying with AlCl3. Identification of the isolate with UVspectrophotometry gave maximum absorptions at wavelengths 241,5, 257,5, 317, 364,5 nm. GC -MS indicated that GM II-1 isolate had a molecular weight of 259, the isolate was presumed to be axanthone.Keywords: xanthone, Garcinia mangostana L., mangosteen, isolation, identification

Abstract

ABSTRACTZeorin or 6,22-Dihydroxyhopane is a compound isolated from Aegle marmelos Correacollected in Yogyakarta Indonesia. The molecular structure was confirmed in Universiti PutraMalaysia. This compound was obtained from petroleum ether extract of the leaves of Aeglemarmelos Correa. In present study, we investigated the effects of zeorin on the -hexoaminidaseenzyme release from mast cell culture. The experiment was performed by using rat basophilicleukemia (RBL-2H3) cell line, a tumor analog of mast cells. DNP24-BSA and thapsigargin were usedas immunologic and non-immunologic inducers for -hexoaminidase enzyme release from mastcells, respectively. The release of -hexoaminidase enzyme was determined by using colorimetricmethods with an enzyme substrate, p-nitrofenil-2-Acetamido-2-deoksi--D-gluko-piranosida, and amicroplate reader at 405 nm. In this study, treatment of 20 ng/mL DNP24-BSA and 0.5 Mthapsigargin could stimulate the release of -hexoaminidase enzyme from RBL-2H3 cells by25.421.62 % and 33.163.72 %, respectively. Zeorin showed potent inhibitory effects on the -hexoaminidase enzyme release, when the release induced by DNP24-BSA. In contrast, zeorin showweak inhibitory effects, when the -hexoaminidase enzyme release from RBL-2H3 cells induced bya Ca2+stimulant, thapsigargin. The IC50 values of zeorin’s effects on DNP24-BSA and thapsigarginexperiments were 33,71 M and >100 M, respectively. Based on the results, the inhibitory effect ofzeorin on the -hexoaminidase enzyme release from RBL-2H3 cells involving mechanisms relatedto the interaction of IgE on the mast cell surface or intracellular signal transductions involved inmast cell degranulation.Key words : Aegle marmelos Correa, zeorin, sel mast, -hexoaminidase enzyme

Abstract

ABSTRACTA study on formulation of cream in a base containing Virgin Coconut Oil (VCO) withPiroxicam (1%) as a model has been undertaken. VCO concentration in the cream-base weremade 0 %, 31%, 36% and 41% respectively to determine the influence of VCO concentration on thepenetration profile of the drug. The profil of penetration was determined by using Franz diffusioncell (vertical type) with the mice’s skin and pH 8 phosphate buffer as membrane and mediumrespectively. Concentration of piroxicam released was determined spectrophotometically atwavelength 353.2 nm. Results indicated that the penetration profile of piroxicam from formula 1, 2and 3 followed zero order kinetic with the slope (k) of 0.0171 (r = 0.9913); 0.0217 (r = 0.9869) and0.0217 (r = 0.9939) respectively, while that from formula 4 followed Higuchi equation with the slope(k) of 0,0570 mg/sec2 / 1(r = 0.9853). Abstract

ABSTRACTIndonesia has a lot of traditional medicines, which people used long time ago, one of theseis mindi (Melia azedarach L.). It have been known that ethanolic extract of mindi leaves hasanalgesic effect greater than paracetamol at dose 6.44 mg/kgBW and 12.89 mg/kgBW. Thisresearch conducted to find out the effectiveness ethanolic fraction of ethanolic extract as ananalgesic. The research was carried out in completely random one way design, used 35 micewhich have been fasted for 24 hours. The mice were divided into 7 groups i.e. negative control,positive control and 5 testing groups (in different doses). Fifteen minutes after injected with thefractions of mindi, the mice were given acetic acid 0.5% intraperitoneally injection. The cumulativewrithe reflects were calculated in every 5 minutes for one hour and were counted the protectionpercentage. Results showed that the ethanol fractions doses 12,88-103,04 mg/kgBW of mindileaves have the analgesic effect in mice and gave protection percentage 30.93-71.61%. Thechromatograms of TLC indicated that the fraction contain flavonoids and phenolic compounds.Key words: ethanol fraction of mindi, analgesic effect, TLC profile
