Abstract

Blast disease caused by Pyricularia oryzae is one of the limiting factors for rice production world wide. The use of resistant varieties for managing blast disease is considered as the most eco-friendly approaches. However, their resistances may be broken down within a few years due to the appearance of new virulent blast races in the field. The objective of the present study was to identify the quantitative trait locus (QTL) conferring resistance to blast disease using 126 recombinant inbred (RI) lines originated from a crossing of a durably resistant upland rice genotype (Laka) and a highly susceptible rice accession cultivar (Kencana Bali). The RI population was developed through a single seed descent method from 1997 to 2004. Resistance of the RI lines was evaluated for blast in an endemic area of Sukabumi, West Java, in 2005. Disease intensity of the blast was examined following the standard evaluation system developed by the International Rice Research Institute (IRRI). At the same year the RI lines were analyzed with 134 DNA markers. Results of the study showed that one major QTL was found to be associated with blast resistance, and this QTL was located near RM2136 marker on the long arm of chromosome 11. This QTL explained 87% of the phenotypic variation with 37% additive effect. The map position of this QTL differed from that of a partial resistant gene, Pi34, identified previously on chromosome 11 in the Japanese durably resistant variety, Chubu 32. The QTL, however, was almost at the same position as that of the multiple allele-resistant gene, Pik. Therefore, an allelic test should be conducted to clarify the allelic relationship between QTL identified in this study and the Pik. The RI lines are the permanent segregating population that could be very useful for analysing phenotypic variations of important agronomic traits possibly owned by the RI lines. The major QTL identified in this study could be used as a genetic resource in improvement of rice varieties for blast resistance in Indonesia

Abstract

Arrowroot (Maranta arundinacea L) is a multi-functional plant used as a source of medicinal, carbohydrate (especially the green leaf type) and also as ornamental plant (the streaked white leaf type). A leaf blight disease is recently found on the streaked white type in Bogor. Preliminary observation indicated that the disease was associated with bacterial infection. The cause of the disease has not been studied. This study was aimed to identify the cause of bacterial leaf blight disease. Experiments were conducted in the laboratory of Research Institute for Spice and Medicinal Crops in Bogor. Suspected bacteria were isolated from diseased leaves. The results showed that the bacterium produced white to brownish colonies on rich agar media containing peptone or cassamino acid. 3-5 mm in diameter, circular, and did not yield fluorescent pigment on King’s B medium. The bacterium formed rod cells, Gram negative, accumulated poly β hydroxybutyrate, utilized glucose under aerobic condition, not hydrolyse arginine and starch, positive catalase, insensitive to tetrazolium salt (0.1%), and grew at 35oC. The bacterium neither producted xanthomonadin pigment nor reduced nitrate to nitrite. The pathogen was tolerant to penicillin and oxolinic acid, but sensitive to streptomycin and oxytetracycline at high concentration (1.000 ppm). These characteristics met to those of Pseudomonas cepacia. Pathogenicity test on detached leaves showed that the typical symptom of blight was similar to that of natural infection on arrowroot. This is the first report on occurrence of P cepacia on arrowroot plant.

Abstract

Development of somatic embryos of sago (Metroxylon sagu Rottb.) on agar-solidified medium are highly varied producing heterogeneous seedlings. Understanding of this phenomenon may help in improving the cultural procedures and conditions of sagosomatic embryogenesis to obtain uniform seedlings in a large scale. This experiment was conducted at the laboratory for plant cell culture and micropropagation, Indonesian Biotechnology Research Institute for Estate Crops from January to March 2006 to examine morphological changes i.e. color and development stages of sago during their somatic embryo development on an agar-solidified medium. Twenty single globular somatic embryos of sago with specific color (yellowish, greenish, and reddish) were cultured in a Petri dish supplemented with a solid medium. The medium was a micronutrients-modified MS (MMS) with half strength of macronutrients containing 0.01 mg l-1 ABA, 2 mg l-1 kinetin, 20 g l-1 sucrose, 0.5 g l-1 activated charcoal, and 2 g l-1 gelrite. Parameter observed was the percentage of embryo’s number based on color and developmental stage. The result showed that at the end of 6-week culture passage, most originally greenish (80.8%) and reddish (95.8%) embryos remained unchanged in their colors, whereas almost half of the originally yellowish embryos turned to greenish and only 30%remained yellowish. At the same time, single globular embryos have changed gradually into the next developmental stages, although not all of the embryos were germinated. The initial color of embryo affected the rate of the developmental stage changes. Yellowish and greenish globular embryos developed more rapidly into cotyledon or germinant stages at 58% and 55% respectively, in 6 weeks than the reddish ones (41%). Therefore, the yellowish and greenish embryos are the best sources of material for in vitro mass propagation and synthetic seed production of sago.

Abstract

Planting Al tolerant crops is an economically justifiable approach in crop production on acid soils. Experiments were conducted to study the mechanisms of Al tolerance among species and varieties of tropical rice, maize, and soybean with previously known levels of Al tolerance. These varieties were hydroponically cultured in 0, 5, 10, and 30 mg l-1 Al with complete nutrient solution at pH 4. The results show that root/shoot ratio of dry weight at 10 mg l-1 Al treatment was an important parameter to indicate differential Al tolerance in maize. Oxalic acid exudation from roots cannot always explain the Al tolerance. Total organic acid concentration in roots at 10 mg l-1 Al treatment indicated a difference of Al tolerance in soybean and lowland rice. Aluminum translocation from roots to shoots was lower in  tolerant varieties than in sensitive varieties of soybean. Increased Al concentration in shoots with increased Al level in the solution was larger in soybean and maize than in lowland or upland rice. Among varieties of  soybean, the Al concentration in shoots increased drastically in Wilis (Al-sensitive variety) with increase Al level, while in Kitamusume (Al-tolerant variety) it did not.

Abstract

Amylase enzyme has a great significance for industrial usages in  Indonesia. However, this enzyme is still imported. The use of bacteria in biotechnological process of industrial products such as enzyme production has stimulated the exploration of extracellular amylase producing  bacteria. This study aimed to identify and analyze the potential use of amylolytic bacterial enzymes for hydrolyzing cassava starch. Two bacterial isolates, i.e. MII-10 and DKW-8 originated from Indonesia soil were identified based on their morphological, physiological and biochemical properties according to the standard protocol. The isolates were then  cultivated on fermentation medium and their growth pattern and  enzymatic assays were observed. The acetone-precipitated crude enzyme harvested based on predetermined cultivation time was used for  enzymatic hydrolysis product characterization on cassava starch using thin layer chromatography (TLC). The results showed that the mesophilicbacteria isolates (MII-10 and DKW-8) were belonged to Bacillus licheniformis. The maximum bacterial cell growth and enzyme activity were reached at 48 hours after incubation. The MII-10 isolate was found more stable than DKW-8 in producing amylase enzyme. Amylase produced by the MII-10 and DKW- 8 isolates was identified to be an endo-a-amylase as confirmed by oligosaccharides and dextrin of the random hydrolysisproducts. Relatively high dextrose equivalence (DE) value of a-amylase of MII-10 (DE of 9.96) suggests that the enzyme is prospective for  saccharification of starchy material in glucose syrup industry.

Abstract

Late blight resistance gene (RB gene) isolated from Solanum bulbocastanum, is a broad resistance gene against all races of Phytophthora infestans. The gene was transformed into Katah-din event SP904 and SP951 using Agrobacterium tumefaciens and these transgenic plants have been crossed with susceptible potato cultivars Atlantic and Granola. Populations of the crosses have been molecularly characterized for the integration of the RB transgene. The study aimed to evaluate the resistance of the populations of crosses between transgenic Katahdin RB  and susceptible non-transgenic parents (Atlantic and Granola) to late blight in a confined field trial at Pasir Sarongge, Cianjur, West Java. A total of 84 clones originated from four popula-tions were evaluated for resistance to late blight. These included 22 clones of Atlantic x transgenic Katahdin SP904, 16 clones of Atlantic x transgenic Katahdin SP951, 19 clones of Granola x transgenic Katahdin SP904, and 27 clones of Granola x transgenic Katahdin SP951. Observations of the late blight infection were conducted when late blight symptoms were detected, i.e. at 56, 60, 63, 70, and 77 days after planting (DAP). The result showed there were high variations in the resistance level of all the 84 clones tested. Clones of crosses between susceptible parents (Atlantic or Granola) and resistant parents (transgenic Katahdin SP904 or Katahdin SP951) showed a similar pattern based on the area under disease progress curve (AUDPC) value, i.e. 377.2 greater than the AUDPC of the resistant parents (180.1), but smaller than that of the susceptible parents (670.7). Observation at 77 DAP resulted four resistant potato clones having resistance score of 7.0-7.6, higher than the transgenic parents Katahdin SP904 (4.6) and Katahdin SP951 (6.8), i.e. clone B8 (Atlantic x transgenic Katahdin SP951) with resistance score of 7.6 and clones B26 (Atlantic x transgenic Katahdin SP951), C183 (Granola x transgenic Katahdin SP904), and D89 (Granola x transgenic Katahdin SP951) with resistance score of 7. These four transgenic potato resistant clones need to be further developed as promising potato clones to late blight.

Abstract

Resistance traits to brown planthopper on rice varieties are controlled by dominant and recessive genes called Bph/bph. Bph17 is one of dominant genes that control rice resistance to brown planthopper.  Marker of Bph17 allele can be used as a tool of marker assisted selection (MAS) in breeding activity. Association of Bph17 allele and resistance to brown planthopper in Indonesian landraces and new-improved varieties of rice is not clearly known. The study aimed to determine the association of Bph17 allele in landraces and new-improved varieties of rice resistant to brown planthopper. Twenty-one rice genotypes were used in the study, consisting of 13 landraces, 5 improved varieties, 3 popular varieties and a check variety Rathu Heenati. Two simple sequence repeat markers linked to Bph17 allele were used, i.e. RM8213 and RM5953. The results showed that association of Bph17 allele in landraces and new-improved varieties of rice resistant to brown planthopper resistance was very low (r = -0.019 and -0.023, respectively). The presence of Bph17 allele did not constantly express resistance to brown planthopper. The study suggests that Bph17 allele cannot be used as a tool of MAS for evaluating resistance of landraces and new-improved varieties of rice to brown planthopper. Further research is needed to obtain a specific gene marker that can be used as a tool of MAS and applicable for Indonesian differential rice varieties.

Abstract

Twenty seven species of Durio have been identified in Sabah and Sarawak, Malaysia, but their relationships have not been studied. This study was conducted to analyse phylogenetic relationships amongst 10 Durio species in Malaysia using PCR-RFLP on two chloroplast DNA genes, i.e. ndhC-trnV and rbcL. DNAs were extracted from young leaves of 11 accessions from 10 Durio species collected from the Tenom Agriculture Research Station, Sabah, and University Agriculture Park, Universiti Putra Malaysia. Two pairs of oligonucleotide primers, N1-N2 and rbcL1-rbcL2, were used to flank the target regions ndhC-trnV and rbcL. Eight restriction enzymes, HindIII, BsuRI, PstI, TaqI, MspI, SmaI, BshNI, and EcoR130I, were used to digest the amplicons. Based on the results of PCR-RFLP on ndhC-trnV gene, the 10 Durio species were grouped into five distinct clusters, and the accessions generally showed high variations. However, based on the results of PCR-RFLP on the rbcL gene, the species were grouped into three distinct clusters, and generally showed low variations. This means that ndhC-trnV gene is more reliable for phylogenetic analysis in lower taxonomic level of Durio species or for diversity analysis, while rbcL gene is reliable marker for phylogenetic analysis at higher taxonomic level. PCR-RFLP on the ndhC-trnV and rbcL genes could therefore be considered as useful markers to phylogenetic analysis amongst Durio species. These finding might be used for further molecular marker assisted in Durio breeding program.

Abstract

Rice bran oil has a potential in lowering blood cholesterol. The oil content extracted from rice bran isa influenced by several factors such as raw material and processing method. This research was conducted to study the effect of heating on the physicochemical characteristics of rice bran oil. The study was carried out at the Food Technology and Nutrition Laboratory and Biotechnological Science Laboratory of the Inter University Center, Bogor Agricultural University, from February to MAy 1997. Bran used was from IR64 rice variety obtained from a local milling rice in Tambak Dahan, Binong-Subang, West Java. Rice bran was subjected to different period of heating (0, 15 and 30 minutes) at 121oC by using an autoclave, then the oil was extraxted by using hexane solvent followed by standard processing at room temperature (28-30oC) and 40oC for two hours. The parameters observed were oil content and its quality such as saponification, iodine, acid, peroxide, tiobarbituric acid, and triglyceric acid values as well as oil color. The results showed that oil extracted from rice bran was high, ranging from 9.65 to 11.02%. Heating (sterilizing) of the rice bran for 15 and 30 minutes at 120oC proir to extraction increased the oil content. The Quality of rice bran oil extracted met the standard of AOAC.
THE FINANCIAL FEASIBILITY OF RICE DRYERS: A Case Study in Subang District, West Java Dewa Ketut Sadra Swastika

Abstract

Drying is a critical activity in rice postharvest handling. Any delay in drying will result in quality and quantity losses. Farmers and traders prefer to sell wet grain rather than dried grain of unhulled rice due to less attractive incentive for them to do drying. The study was conducted in Subang District, West Java, during March-June 2011 using survey method to analyze the financial feasibility of rice dryers. The respondents interviewed were 15 rice millers who have either sun-drying floor or mechanical dryer, or both. The main collected data were the investment cost, operational cost, maintenance cost, and the revenue of the dryers. The data were analyzed by using shortterm and long-term financial analyses to assess the profitability and feasibility of some drying technologies. The results showed that there were at least three drying technologies in the study area, namely sun-drying floor, flatbed dryer using gas fuel (gas dryer), and flatbed dryer using rice husk fuel (husk dryer). Under the mean capacity, the short-run MBCR of technological change from sun dryer to gas dryer was 1.29, while from sun dryer to husk dryer was 1.45. The long-run analysis showed that the IRRs were 44.44% for sun dryer, 233.47% for gas dryer, and 260.49% for husk dryer. These results showed that among the three drying technologies, husk dryer is the most profitable, most feasible, and most prospective to be adopted. Therefore, this type of dryer should be intensively promoted to the farmers’ group associations, rice traders, and rice millers. The government support in terms of rice dryers for farmers’ group associations should be focused on husk dryer.
