Abstract

Multiplication ofpepper clone variety Panniyur in vitroThe study was conducted to determine effect of Benzyl Adenine (BA) on shoot multiplication and regeneration capacity of callus fom vaiety Panniyur. This esearch was undertaken at the Laboratory of Plant Genetic Resources and Beeding from Apil to October 1998. Nodal segment was used as explant source, taken from plantlet already gown on Murashige and Skoog (MS) medium. The expeiment was divided into two steps, shoot multiplication and regeneration of callus. The BA concentration used for shoot multiplication was 0.3, 1.0, 1 .5, 2.0. 2.5,3.0. and 5.0 mg/1 while for callus regeneration was 0.3. 0 5, and 0.7 mg/1. The expeiment was designed as completely randomized with 10 and 6 replications, respectively. Results showed thai the treatment with BA 2.5 mg/1 appeared to be the best medium for shoot multiplication indicated by the highest number of shoot, number of leaves and shoot height as much as 10.3, 5.6. and 4.40 cm in eight weeks, respectively. Culture performance from such culture was also the best with green leaves, fesh and vigorous. Addition of BA 0.7 mg/1 onto regeneration medium also the best treatment for shoot egeneration fom callus with average number of shoot 8.67. From such treatment was also obtained the highest growth rate with 4.35 cm shoot height and culture performance showing green leaves, average size and vigorous.

Abstract

Tanaman ylang-ylang (Canangium odoratum f. genumea), merupakan tanaman penghasil minyak atsiri yang nilainya cukup tinggi. Evaluasi potensi produksi bunga bertujuan untuk mendapatkan individu- individu yang produksi bunganya linggi untuk diteliti lebih lanjut dalam mendapatkan pohon unggul sebagai sumber benih Penelilian dilakukan di Sukamulya lahun 2000-2002 pada areal petanaman 1 ha (± 200 pohon) Petanaman berumur 12-15 tahun Evaluasi dilakukan dalam 2 tahap. Pada lahap petama, evaluasi dilakukan secara visual. Pohon-pohon yang dipilih adalah pohon dengan penampilan baik dengan ciri-ciri morfologis khas ylang-ylang, yaitu berbatang lurus, percabangan terkulai, permukaan batang licin dan terdapat bekas cabang (scars). Dari sekitar 200 pohon (erpilih 15 pohon yang berpenampilan paling baik. Pada lahap kedua, ke 1 5 pohon tersebut diamati petumbuhan dan produksi bunganya selama tiga tahun berturut-turut Hasilnya menunjukkan bahwa petumbuhan yang meliputi lingkar batang dan jumlah cabang tidak berbeda antar pohon. Sebaliknya produksi bunga sangat beragam antar pohon. Rata-rata produksi selama liga tahun berkisar antara 652-12 551 g/ph/tahun. Berdasarkan produktivitas dan fluktuasi produksi antar musim telah tcrpilih lima pohon yang produksi bunganya paling tinggi, yaitu No. 2/143, 8/06, 12/64, 14'113, dan 15/16 dengan produksi masing-masing 7 177, 8 352; 7 177; 12 551, dan 12 398 g/ph.Kata kunci : Canangium odoratum f. genumea, cvaluasi, potensi produksi ABSTRACT Evaluation of the potency offlower yield of Ylang-ylangYlang-ylang (Canangium odoratum f genumea) is the essential oil producing crop, which has high economic value. An evaluation of yield potential was aimed al selecting high yielding individuals which arc futher selected lo produce outstanding individuals as seed source. The evaluation was conducted in Sukamulya (Sukabumi) from 2000-2002 on the area of I ha ((+ 200 trees) Ihe trees was 1 2-15 years old. The evaluation was performed in 2 stages In Ihe irst stage the evaluation was conducted visually and based on Ihe specific morphological characters of ylang-ylang, which has erect trunk, plain surface, drooping branches and showing scars on Ihe surface of the trunk. From about 200 trees, 15 trees were selected as showing ihe best performance. In the second stage, the 15 selected trees were evaluated for their growth performance, including trunk circumference, number of branches and yield of fresh flowers for 3 years. Result showed that the trunk circumferences and the number of branches were nol different among the trees On the other hand, the yield of flowers was greatly variable, cither between the trees or between seasons. The average yield of flower in 3 year observation ranged from 652 to 12 551 g-'trcc/ycar. Based on the productivity the highest yielding trees were No. 2/143, 8/06, 12/64, 14/113, and 15/16 with the yield of flowers 7 177, 8 352; 7 177; 12 551, and 12 398 g'tree/ycar respectively.Key words : Canangium odoratum f. genumea. evaluation, yield potency

Abstract

Pseudocercospora liebenbergii the cause of leaf spot disease on Rauwolfta serpentinaThe experiments wee conducted at Reseach Institute for Spice and Medicinal Crops, rom Ocober 1995 o March 19%. The objective of the experiments wee to identify the causal agent of leaf spot disease on Rauwofia serpentina and some ecobiological aspects. The expeiments were divided into two steps. The first step was observation on the disease intensity and identification of the causal agent. The second step was testing the effect of temperature and light on the mycelia growth and the germination of conidia, and the survival of conidia at diferent ranges of elative humidity. The results showed that the pathogenic of leaf spot disease is Pseudocercospora liebenbergii, newly eported pathogenic fungus in Indonesia. The fungus infected leaf only. The disease intensity ranged between 0-85%/plant with the average 22.23%. The first symptoms i.e. light spots wee observed 2-3 weeks after the inoculation. Stroma wee found on the upper and botom of the oldest spots The optimum temperatue for fungal growth ranged between 20-28°C. Continuous light was able o induce the growth of aerial mycelia but failed against the fungal rate. The germination of conidia occured at temperature between 20-32°C with an optimum of 24°C. Conidia on the surface of the oldest leaf spots were still viable ater the leaves wee incubated for 48 hours at 55% elative humidity.

Abstract

Pepper is an introduced species and has always been propagated vegetatively, so it has narrow genetic base. High genetic diversity is necessary to produce new varieties, especially for breeding of resistance to foot rot disease. Increasing genetic diversity can be done through gamma ray irradiation. This research aims to evaluate response of black pepper Ciinten variety at seed and radicle emergence phases to gamma ray irradiation. The research was conducted in PAIR BATAN and greenhous e IMACRI from June 2014 to April 2015. The plant material was Ciinten variety at the seed and radicle emergence phases. The experimental design used was completely randomized design (CRD) with one factor which is dose of irradiation with seven levels (0, 25, 50, 75, 100, 125, 150) Gy. Each treatment consisted of three replications, each replication consisted of 60 seeds. Both phases showed significant differences in perfomances between dose in plant height, leaf length, number of leave, number of internode. Radiosensitivity of pepper on radicle emergence phase was higher than the seed phase indicated by LD50 (Lethal Dose 50). LD50 at seed phase was 68.15 Gy, whereas LD50 of the radicle emergence phase was 30 Gy. The higher irradiation dose that given to both treatment phases caused reduction in plant height, leaf length, while the number of leaves and nodes decreasing. Irradiation dose 25 dan 50 Gy in seed phase and 25 Gy in radicle emergence phase significantly increase genetic diversity base on quantitative, qualitative characters, anatomy and leaf resistence to P. capsici infection. Eighteen putative mutants resistant to infection P. capsici. Eighteen mutant putative Ciinten pepper varieties were expected to be high yielding varieties with more research in the greenhouse and in the field to determine the production and quality potential.

Abstract

Response of four new coconut hybrids to bud rot and premature nutfull diseasesThe objective of Ihe experiment was to find out Ihe resistance of four new coconut hybrids GRA x DMT. GKB x DMT, GKN x DTE. GKB x DTE against bud rol and premature nutfall diseases due lo Phytophthora palmivora. The field observation on bud rot was conducted in coconut trial plots at Kima Atas experimental garden. Minahasa District, North Sulawesi Province for 4 years (1995-1998). The field trial was arranged in a randomized block design with 6 treatments (4 coconut hybrids. 2 control) and 60 replicates The experiment under laboratory condilion was conducted to know the artificial infection development of P palmivora on coconut nut The experiment was arranged in a completely randomized design with 6 treatments (4 coconut hybrids, 2 control) and 10 replicates. The result of the field observation showed that percentage of coconut infection by bud rot per cultivar were PB-121 (23.33%) as the highest and then followed by GKN x DTE (8.33%), KHINA-1 (5.0%) and GKB x DMT (3.33%) while cultivars of GKB x DTE and GRA x DMT unlil 4 years observation were not infected by bud rot In laboratory trial, Ihe growth rate of pathogen was very high on KHINA-1 (12.62 mm2/day). The growth rate of pathogen in GKB x DIE and GRA x DMT were slowest (8 9 mm'/day) while those in PB- 121, GKN x Dll and GKB x DMT were moderate (9-10.2 mm2/day).

Abstract

Tujuan utama pemuliaan kapas di Indonesia adalah meningkatkan produktivitas dan kualitas serat dalam upaya meningkatkan pendapatan petani dan memperbaiki mutu benang tcnun seta kualitas tekstil yang harus bersaing di pasar internasional. Scjumlah enam persilangan telah dilakukan antara dua varietas dai India. I.RA 5166 dan SRT-1 dengan dua varietas dai Amerika Serikat, Dcltapine 55 dan Deltapinc Acala 90 dan satu vaietas dai Australia, Siokra. Seleksi individu, seleksi galur dan seleksi individu dalam galur dilaksanakan pada generasi F2 sampai F5 berdasarkan jumlah buah, tingkat kerusakan daun terhadap Sundapteryx biguttula. dan mutu serat; semua proses di atas dilakukan pada kondisi lahan tadah hujan, dan tanpa penggunaan insektisida terhadap tanaman; dari proses di atas diperoleh 12 galur harapan. Sejumlah 13 percobaan dilakukan antara tahun 1993 sampai dengan 2001 untuk mengamati kcragaan galur-galur baru tersebut; pengujian dilakukan di Jawa Timur dan Sulawesi Selatan, menggunakan teknik-teknik penelitian standar. Dengan proscdur ini dapat diidcntifikasi beberapa galur yang menunjuk¬ kan perbaikan serenlak hasil dan kualitas serat kapas. Beberapa penelitian juga dilakukan untuk mcngcvaluasi tanggap galur-galur tersebut pada tumpangsari dengan kedelai dan kacang hijau di Jawa Timur. Dua galur, 88003/16/2 dan 92016/6 (sudah dilepas dengan nama vaietas Kanesia 8 dan Kanesia 9 pada bulan Juni 2003), menunjukkan produktivitas dan kualitas serai yang lebih linggi. Rata-rata, kedua vaietas menghasilkan 1.85 ton dan 191 ton kapas berbiji per hektar atau 8-12% lebih tinggi dai hasil vaietas Kanesia 7 yang sudah dilepas sebelumnya. Persentase serat 35.2%, kekuatan serat berkisar antara 22.6-24.7 gram tex'1, serat lebih panjang dan berkisar 29.2-30.3 mm sedangkan angka mikroncr lebih rendah yang menyatakan bahwa serat lebih halus. Semua perbaikan di atas menunjukkan perbaikan mutu serat. Kanesia 8 dan Kanesia 9 juga menunjukkan peningkatan ketahanan terhadap Sundapteryx biguttula dan komplcks hama kapas. Kanesia 8 dan Kanesia 9 kurang kompctitif dalam tumpang sari dengan kedelai jika dibandingkan dengan Kanesia 7. Pada tumpang sari dengan kacang hijau Kanesia 8 juga mengalami kehilangan hasil yang tinggi, sedangkan Kanesia 9 menunjukkan toleransi yang tinggi dalam kompctisi dengan kacang hijau. Pelepasan Kanesia 8 dan Kanesia 9 akan memberikan pilihan varietas yang lebih banyak bagi petani dan perusahaan pemintalan untuk menyesuaikan dengan produk akhirnya.Kata kunci : Gossypium hirsutum, prosedur pemuliaan, produktivitas, kualitas serat, Sundapteryx biguttula, tumpangsari ABSTRACT Genetic improvement on two new cotton varieties, Kanesia 8 and Kanesia 9The main objective of cotton breeding in Indonesia is to improve productivity and fiber quality which is aimed to increase farmers' income and to make beter yam and textile quality that has to compete in international market Six crosses were made between two Indian varieties, LRA 5166 and SRT-1 with two USA vaieties, Deltapine 55 and Deltapinc Acala 90 and one Australian variety, Siokra. Individual plants, lines and individual within lines were selected on F2-F5 generations based on boll- counts, leaf-damage by jassids and fiber traits, those were conducted under rainfed and insecticide-ree condition; twelve promising lines were produced from this process. A total of 13 trials were carried out to observe performance of these new lines during 1993 to 2001; those were located in East Java and South Sulawesi using the standardized experimental techniques. By these procedures make it possible to identify several breeding lines showing simultaneous improvement in yield and fiber quality. Several tests were also made to evaluate response of those lines under intercropping with soybean and mungbean, which were located in East Java. Two breeding lines, 88003/16/2 and 92016/6 (those have been released as Kanesia 8 and Kanesia 9 in 2003), showed higher productivity and fiber quality. In average, these new vaieties produced 1.85 and 1.91 ton ha'1 seed cotton respectively or 8 to 12% higher than those on Kanesia 7, the previously released vaiety. Lint turn-out was 35.2% fiber-strength was varied from 22.6 to 24.7 gram tex'1 , fiber lengths ranged from 29.2 to 30.3 mm with lower micronaire-valucs indicating better fiber-ineness. All of those improvements represented a trend toward a higher quality iber. Kanesia 8 and Kanesia 9 also showed a slight improvement in resistance to jasssids and insect pest-complex. Kanesia 8 and Kanesia 9 performed lower competitive ability under intercropping with soybean in comparison with Kanesia 7. Under intercropping with mungbean Kanesia 8 also suffered high yield loss, wherein Kanesia 9 showed good tolerance to mungbean. The release of Kanesia 8 and Kanesia 9 is expected to give a broader choice for the cotton growers and spinning-mills to match with their inal product.Key words: Coton (Gossypium hirsutum), breeding procedure, productivity, liber quality, Sundapteryx bigullul. inter¬ cropping.

Abstract

Thee wee a number of factors causing diffeences in coconut growth and soil properties under ield condition. To cope with the existing vaiation fanning system in relation to soil properties, six coconut farming systems with four species of perenial crops as inter- crop(s) namely banana, papaya, coffee and pineapple wee conducted understudy. The objectives of the study were to study soil chemical and physical properties and nutrient element content in the coconut leave under various coconut cropping systems. Six coconut farming system as used for the study namely (I) coconut (control), (2) coconut + pineapple, (3) coconut + coffee, (4) coconut + papaya + pineapple, (5) coconut + banana + coffee, and (6) coconut + banana ♦ papaya + coffee + pineapple. Total area for each system is 0.5 ha. Data on leaf analysis parameters, soil chemical poperties and soil physical parameters, wee statistically analyzed in randomized complete block design (RCBD) with three replications. The organic matter, pH and cation exchange capacity of the soils did not differ significantly with cropping pattern, although intensively cropped farm tended to have higher organic matter and cation exchange capacity values. Nitrogen, phosphorus and potassium wee also significantly higher in intercropped farms but calcium and magnesium did not vary signiicantly. While moistue content, water holding capacity, bulk density of the soil did not show significantly difference with cropping patterns. Leaf nitrogen and calcium incease while potassium deceased with intensity of cropping. Phosphorus and magnesium showed no diinite trend. Leaf nitrogen, phosphorus and calcium were all positively correlated with soil nitrogen but soil phosphorus was weakly correlated with all nutrient element in the leaf. The exchangeable potassium in the topsoil was positively and insigniicantly with nitrogen, potassium and calcium but negatively insignificantly with phosphorus and magnesium concentration in coconut leaf. While the exchangeable calcium in the subsoil was significantly and positively correlated with nitrogen in the leaf but negatively corelated with potassium.

Abstract

ABSTRAKInformasi jarak genetik dan hubungan kekerabatan sangatdiperlukan dalam merakit varietas unggul. Semakin jauh jarak genetikantar tetua maka peluang dihasilkan kultivar baru dengan variabilitasgenetik yang luas akan menjadi semakin besar. Sebaliknya, persilanganantar tetua berkerabat dekat akan menghasilkan variabilitas yang sempit.Penelitian ini dilaksanakan pada bulan Mei 2007 di Kabupaten MinahasaUtara, Minahasa Selatan dan Bolaang Mongondow, Sulawesi Utara. Disetiap kabupaten ditetapkan tiga desa contoh pada ketinggian yangberbeda. Tujuan penelitian ini adalah untuk mengetahui tingkatkekerabatan dan jarak genetik sembilan aksesi plasma nutfah kelapa asalSulawesi Utara yang akan digunakan sebagai materi pemuliaan dalamperakitan kelapa unggul. Untuk mengetahui kekerabatan antara sembilanaksesi kelapa tersebut diukur jarak genetiknya dengan menggunakanperhitungan nilai D 2 statistik dari Mahalanobis didasarkan pada delapankarakter komponen buah, yaitu panjang buah, lebar buah, berat buah utuh,berat buah tanpa sabut, berat buah tanpa air, berat daging buah, tebaldaging buah, dan berat tempurung. Hasil penelitian menunjukkan bahwakesembilan aksesi kelapa tersebut membentuk lima kelompok dan jarakgenetik terbesar terdapat antara kelompok II (Dalam Lansot, DalamMongkonai, Dalam Dua Saudara) dan IV (Dalam Kaleosan, Dalam Kema)dengan nilai D 2 = 2.196,57. Sumbangan terbesar terjadinya jarak genetiktersebut diperoleh dari karakter tebal daging buah.Kata kunci : Cocos nucifera L., kekerabatan, genetik, kelapa dalamABSTRACTGenetic relationship among nine coconut accessions fromNorth SulawesiThe research was conducted in May 2007 at North Minahasa, SouthMinahasa, and Bolaang Mongondow Regions, North Sulawesi Province.From each region, three villages with different elevation were determined.The objective of the research was to find out genetic relationship amongnine coconut germplasm accessions for breeding material in composinghigh yielding coconut. The genetic relationships were estimated using D 2Mahalanobis Statistics based on eight characters of fruit component, suchas length of fruit, width of fruit, fruit weight, unhusked fruit weight,weight of fruit without water, weight of endosperm, thickness of kernel,and weight of shell. The result showed that the nine accessions weredivided into five groups and the widest genetic distance had been foundbetween group II (Lansot Tall, Mongkonai Tall, Dua Saudara Tall) and IV(Kaleosan Tall, Kema Tall) with the D 2  value of 2,196.57. The highestcontribution to the genetic relationship was thick of kernel (50%contribution). Lansot Tall and Kaleosan Tall can be used as parents forprepotent coconut.Key words : Cocos nucifera L., genetic relationship, tall coconut

Abstract

Percobaan lapang telah dilakukan di Kebun Percobaan Pekuwon, Bojonegoro, 1992, untuk mcmpelajai sifat-sifat agronomis dan kimiawi tembakau pada berbagai waktu tanam sorgum pada sistem tumpangsai tembakau + sorgum. Percobaan disusun dalam rancangan acak kelompok dengan enam ulangan. Perlakuan terdiri dai 5 taraf waktu tanam sorgum yaitu 4 dan 2 minggu sebelum tanam tembakau, bersamaan dengan waktu tanam tembakau, 2 dan 4 minggu setelah tanam tembakau. Ukuran petak 10.8 m x 12.0 m. dengan 240 tanaman tembakau per petak dan 720 tanaman sorgum per petak. Analisis N, P, K, nikotin, dan gula beturut- turut dengan Kyeldhal, Spektrofotometi, Flamefotometi, Titrasi dengan NaOH dan Luff-Schroll. Hasil percobaan menunjukkan bahwa dengan mempcrcepat waktu tanam sorgum dari 4 minggu setelah tanam tembakau menjadi 4 minggu sebelum tanam tembakau sangat menurunkan pertumbuhan, hasil dan mutu. Scbaliknya perlakuan tersebut meningkat¬ kan kadar N-total, P, dan K, dan hasil sorgum tumpangsai, serta tidak berpengaruh pada kadar nikotin, gula, nisbah/nikotin, dan N/nikotin tembakau. Pada keadaan kering yang dialami oleh percobaan ini walaupun hasil tembakau rendah namun mutu hasil masih dalam kisaran yang baik dan persaingan dikuasai oleh tanaman sorgum.Kata kunci: Nicotiana tabacum, sorgum bicolor, tumpangsai, waktu tanam ABSTRACTAgronomics and chemicals properties of tobacco under different planting dates ofsorghum in tobacco -Horghum intercropping systemThe ield expeiment was conducted at Pekuwon Expeimental Station, Bojonegoro, in 1992, to study the agronomic and chemical propeties of tobacco grown under diferent planting dates of sorghum in tobacco+sorghum intercropping system. The expeiment was arranged in randomized block design, with 6 replications. The treatment consisted of 5 levels of sorghum planting, 2 and 4 weeks ater tobacco planting. Plot size was 10.8 m x 12.0 m, with 240 and 720 plants of tobacco and sorghum respectively. The methods for analyses N, P, K, nicotine and sugar analyses were Kyeldhal, Spectrophotometry, Flame photometry, Titration with NaOH, and Luf-Schroll, respectively. The growth, yield, and quality of tobacco were decreased sharply, but the N, P, K contents of the leaves were increased by accelerating planting date of sorghum from 4 weeks ater to 4 weeks before tobacco planting. The content of nicotine, sugar, sugar/nicotine. N/nicotine of the leaves were not afected by this treatment. In dry condition, although the yield of tobacco was low, but the quality was in good category, and the competition in tobacco ♦ sorghum intercropping system was dominated by sorghum.Key words : Nicotiana tabacum, sorghum bicolor, intercropping, planting date
