Abstract

This study was conducted to determine the effect of Citrus hystrix as fat protector on unsaturated fatty acids, cholesterol and chemical composition of lamb meat. The research design applied was completely randomized design with 3 treatments and 5 replications. Fifteen local male lambs aged 9-12 months weighing 14-17 kg, were divided into 3 groups of different diet treatments (P0 : sheeps were only given a basal diet; P1 : sheeps are given basal diet and 3% cooking oil; P2 : sheeps are given basal diet, 3% cooking oil and protected by 3% Citrus hystrix powder). The data were analyzed by analysis of variance, the differences among treatments were tested by Duncan’s New Multiple Range Test. Abstract

By mixing with pork, beef adulteration is frequently found in the traditional  market that very disturbing Moeslem community in Indonesia. This study was conducted to detect pork contamination in fresh and cooked beef using genetic marker mitochondrial DNA cytochrome b (mt-DNA Cyt b) by duplex-PCR. A total of twelve samples was used in this study consisting six fresh meat samples and six cooked meat samples, respectively. Those beef and pork were bought from animal slaughterhouse and a supermarket in Surakarta. Cooked samples were prepared by boiling the meats in hot water at 100oC for 30 minutes. We designed pork contamination in beef in the level of 0, 1, 5, 10, 25%, respectively. The DNA genome was extracted and polymerase chain reaction (PCR) was performed using species specific primer to isolate mt-DNA Cyt b gene from the samples. The results showed that the DNA genome was successfully extracted from pork, beef, and contaminated meat samples. In addition, visualization of duplex-PCR on 1.5% agarose gel was able to detect pork contamination in both fresh and cooked beef up to very small proportion (1%). The existence of pork in beef was indicated with the presence of specific 398 bp DNA band. It can be concluded, duplex-PCR of mt-DNA Cyt b gene was very sensitive in detection of pork contamination in fresh and cooked beef.

Abstract

The use of vinegar as a natural preservative to inactivate microbial growth in meat was investigated. Vinegar was prepared from banana peel and coconut water, and the efficacy against Escherichia coli O157:H7 was tested. Chicken meat were treated with vinegar solution (equals to 1% acetic acid), acetic acid solutions (1%), and lactic acid solutions (2%), control (distilled water). Samples were soaked with acid solution treatment for 1 minute (1.125:1 w/v; chicken meat: solution treatment). Treated samples were inoculated with E. coli O157:H7 on the surface of the chicken meat, then left for 20 minutes for absorption of bacteria into the meat. Observation was done at 0, 3, 6, 9, and 12 days at low temperature storage, and at 0, 6, 12, 18, 24 hours at room temperature storage. The results showed that banana peel vinegar and coconut water vinegar were effective as natural preservative. In conclusion, the vinegar were potentially inhibited E. coli O157:H7 growth at chicken meat until 12 hours at room temperature storage and 9 days at cold temperatures.

Abstract

The objective of this research was to identify diversity of exon 5 UTMP gene fragment in Bali cattle using direct sequencing. The total 60 blood samples of Bali Cattle derived from BPTU Bali in Bali siland (20 heads), BPTU Serading in Sumbawa island (20 heads) and Village Breeding Center in Barru District South Sulawesi (20 heads) were used to evaluate their genetic diversity at exon 5 UTMP gene. The forward and reverse data sequences were analyzed using Bioedit program and alignment analysis was carried out using MEGA5 program. Meanwhile haplotype analysis was performed by DnaSPv5 program. The result showed that partial sequences in exon 5 UTMP gene had 16 haplotypes with the highest number of haplotypes ware found in VBC Barru district South Sulawesi (8 haplotypes). Moreover, the highest average of haplotype (h) and nucleotide (p) diversity were found in VBC Barru district South Sulawesi were 0.7949 and 0.0016, respectively. In addition, minisatellite insersion was found in exon 5 UTMP gene fragment on Bali cattle which are consist of 5'-CCA GTC ATG AAG AAG GCA GAG GTC GTC GTG CCG GCG AAA-3'. According to our results, haplotype and minisatellite variation in exon 5 UTMP gene fragment can be used as a candidate genetic marker specific for reproductive trait in the Bali cattle and for its strategy breeding program in the future.

Abstract

The objective of the study was to determine the effects of feeding Lactobacillus species (Lactobacillus sp.) and inulin derived from dahlia tuber powder on antioxidant activity, calcium mass, and protein mass of crossbred kampong chicken meat. A total of  168 birds of 21 days old crossbred kampong chickens were randomly allocated into 6 treatments with four replications per treatment. The present experiment was assigned in  a completely randomized design with 2 x 3 factorial scheme. The first factor was levels of dahlia tuber powder, namely 0.8% (A1) and 1.2% (A2), and the second factor was levels of Lactobacillus sp., namely none (B0), 1.2 mL (108 cfu/mL/B1) and 2.4 mL (108 cfu/mL/B2). Abstract

A study was conducted to identify the variations of the myostatin and also to evaluate the existence of genetic marker for “double muscling” (11-bp deletion in the third exon of the myostatin gene) in the first generation of Belgian Blue cattle in Indonesia using the Single Strand Conformation Polymorphism (SSCP) and the sequencing analysis. A total of 8 DNA samples belonged to Karya Anugerah Rumpin (KAR) Farm were used in the Single Strand Conformation Polymorphism (SSCP) and the sequencing analysis. There were 3 allele types of myostatin gene based on the SSCP analysis. The Belgian Blue sire has type A allele. The Simmental, Wagyu, SO x BX, Charolais, and the PO cattle have the type B allele, while the Belgian Blue x FH and the Belgian Blue x SO have the type C allele (heterozygous). There are 11-bp deletion in the third exon myostatin gene for the Belgian Blue sire based on the sequencing analysis. The myostatin gene in the Belgian Blue F1 generation individual was heterozygous. This study provides scientific evidence that the 11-bp deletion in the third exon of myostatin gene in the Belgian Blue sire was inherited to its F1 generation (male and female).

Abstract

Two adult male sheeps fitted with rumen cannula were used in two experiments to study the effects of synchronization of carbohydrate and nitrogen supply in sugarcane bagasse based total mixed ration (TMR) on in situ nutrient degradation. The first experiment was aimed to create three TMR with different synchronization index. Ingredient feedstuffs of TMR were evaluated for its in situ organic matter (OM) and nitrogen (N) degradation kinetics. On the basis of the OM and N degradation kinetics of feedstuffs, three sugarcane bagasse based TMR were formulated with synchronization indexes of 0.37; 0.50; and 0.63; respectively. The TMR had similar levels of crude protein (CP), total digestible nutrients, and neutral detergent fiber (NDF). In the second experiment, the three TMR with different synchronization index were evaluated for in situ degradability characteristics of OM, CP, NDF, and sulfur. The in situ degradation of OM in TMR were decreased (P<0.05) with the increasing of synchronization index. The higher synchronization index in TMR increased (P<0.05) CP degradation of CP. The NDF degradation decreased slightly by the alteration of synchronization index in TMR. The higher synchronization index in TMR reduced (P<0.05) in situ sulfur degradation, and this may not support to effects of synchronization of carbohydrate and nitrogen supply.
