Abstract

Calpain-1 gene (CAPN1) produces an calpain enzyme controlling structure of meat protein and tenderness. The aims of this study were to identify Single Nucleotide Polymorphism (SNP) in exon 5 and 6 of CAPN1 gene and its associate with carcass and meat characteristic traits in bali cattle. A total of 48 bali cattles from BPTU-HMT Bali Cattle, Bali Province were used in the research. SNP in exon 5 and 6 of CAPN1 gene were identify with direct sequencing using MEGA 5 program. Analysis of polymorphism was conducted by PopGen 1.32 software to identify frequencies of genotype, allele and Hardy-Weinberg equilibrium. The association of CAPN1 gene genotype with carcass and meat characteristic traits was analyzed using Generalized Linear Model (GLM) procedure of SAS. Sequencing analysis at exon 5-6 of CAPN1 gene in Bali cattle resulted in eight polymorphic SNPs. They are c.3669T>C, c.3854G>A, c.3881T>C, c.3899C>T, c.3908C>G, c.4002C>A, c.4021G>T and c.4037A>C. The SNPs c.3669T>C, c.3854G>A and c.3899C>T were significantly (P<0.05) associated with rump thickness (RT), rump fat thickness (RFT) and marbling score (MS), while SNP c.4037A>C was not significantly associated with carcass and meat characteristic traits. The SNPs were significantly associated with carcass and meat characteristic traits namely c.3669T>C, c.3854G>A and c.3899C>T. Those SNPs may be used as candidate marker for Marker Assisted Selection (MAS) in bali cattle.

Abstract

The objective of this research was to identify diversity of exon 5 UTMP gene fragment in Bali cattle using direct sequencing. The total 60 blood samples of Bali Cattle derived from BPTU Bali in Bali siland (20 heads), BPTU Serading in Sumbawa island (20 heads) and Village Breeding Center in Barru District South Sulawesi (20 heads) were used to evaluate their genetic diversity at exon 5 UTMP gene. The forward and reverse data sequences were analyzed using Bioedit program and alignment analysis was carried out using MEGA5 program. Meanwhile haplotype analysis was performed by DnaSPv5 program. The result showed that partial sequences in exon 5 UTMP gene had 16 haplotypes with the highest number of haplotypes ware found in VBC Barru district South Sulawesi (8 haplotypes). Moreover, the highest average of haplotype (h) and nucleotide (p) diversity were found in VBC Barru district South Sulawesi were 0.7949 and 0.0016, respectively. In addition, minisatellite insersion was found in exon 5 UTMP gene fragment on Bali cattle which are consist of 5'-CCA GTC ATG AAG AAG GCA GAG GTC GTC GTG CCG GCG AAA-3'. According to our results, haplotype and minisatellite variation in exon 5 UTMP gene fragment can be used as a candidate genetic marker specific for reproductive trait in the Bali cattle and for its strategy breeding program in the future.

Abstract

A descriptive study using 30 female local Lombok ducks age ready to lay was carried out to know the local Lombok egg duck production and its quality raised intensively in Centre Lombok for making a salted egg. The observation was done in three months. Variable observed were eggs production (monthly production, egg weight, egg index), shell quality (texture, thickness, color), and internal egg quality (yolk index, yolk color, yolk and albumen weight ratio, egg spot). Data were analyzed by using simple statistic to get the mean and the deviation standard. The observation results were: monthly egg production in the first three months were 58,15 ± 10,98%; 60,25 ± 10,60%; 50,00 ± 15,73%; average egg weight and its index were 61,42 ± 2,73 gram and 0,74 ± 0,10. The shell surface was smooth and the thickness was 0,40 ± 0,09 mm. The shell color was greenish dark blue. The yolk index was 0,47, the yolk color varied between 10 and 14, the yolk albumen ratio was 1.04, and there was not an egg spot. The quality of the local Lombok egg ducks was good for making a salted egg.

Abstract

Penelitian mengenai “Korelasi Bobot Badan dengan Ukuran Tubuh sapi Bali jantan muda di Balai Pembibitan Ternak dan Hijauan Makanan Ternak Serading Sumbawa” telah dilaksanakan pada bulan April 2021. Penelitian ini bertujuan untuk mengetahui korelasi bobot badan dengan ukuran tubuh Sapi Bali jantan muda di Balai Pembibitan Ternak dan Hijauan Makanan Ternak (BPT-HMT) Serading. Hipotesis dari penelitian ini adalah ukuran tubuh memiliki hubungan yang positif dengan bobot badan sapi Bali jantan muda. Materi yang digunakan yaitu data bobot badan dan ukuran tubuh (Panjang badan, Lingkar dada, dan Tinggi pundak) sapi Bali jantan muda sebamyak 34 ekor. Korelasi dan regresi bobot badan dengan ukuran tubuh sapi Bali jantan muda dianalisis menggunakan analisis korelasi dan regresi berganda dengan bantuan software SPSS versi 26. Hasil penelitian menunjukkan adanya hubungan yang positif dan sangat kuat antar bobot badan dengan ukuran-ukuran tubuh secara simultan dengan koefisien korelasi sebesar 0.96 dan nilai koefisien determinasi sebesar 0.919 dengan persamaan regresi berganda yaitu Y = -307.102 + 1.044X1 + 2.125X2 + 0.777X3. Koefisien korelasi antara bobot badan dengan Panjang badan, Lingkar dada, dan Tinggi pundak berturut-turut sebesar 0.76 ; 0.93 ; 0.79 dan persamaan regresi yaitu Y = - 185.761+3.562X ; Y = -239.362+3.008X dan Y = -238.188+3.716X.

Abstract

Growth Hormone Receptor (GHR) gene is a member of the super familycytokine/hematopoietin receptor. GHR plays a role in mediating the biological activity of growth hormone on target cells that lead to the affection of the nature ofgrowth in beef cattle carcasses and the nature of milk production in dairy cows. This study was designed to identify the genetic diversity of GHR genes exon 10 in Bali cattle. Genotype variation of gene candidates was identified using PCR-RFLP technique. The results showed that mutation at position of 755bp exon 10 of GHR gene as determined by Maskur (2012) can be identified using MBO II restriction enzymes at position of 125bp or at position of 282bp base nucleotidescaused by transition mutation of C/T and substitution of amino acid threonine to isoleucine.Genotype identification of GHR gene exon 10 resulted in three individual genotypes in Bali cattle population namely CC, TT and CT with frequency of 0.1931; 0.5455 and 0.2614respectively. The result of X² test and heterozygosity observationas well as heterozygosity expectation indicate that the genotypedistribution on Bali cattle population in this study were not in Hardy-Weinberg equilibrium (Hardy-Weinberg Equilibrium/ HWE). The value of GHR|MBO II gene Polymorphic Informative Content was 0.342051, might be used for genetic identifier, and was quite informative as gene identifier for linkage analysis in the population.

Abstract

The study was aimed to determine the effect of tapioca flour addition atvarious levels on the quality of chicken sausage. The research was design based on a Completely Randomized Design (CRD) with 4 treatments and 3 replications. The treatment of tapioca flour addition level were: 0%, 10%, 20% and 30%. Data ofsausage quality were analyzed using Analysis of Variance and continued with Duncan's Multiple Range Test. The results showed that the addition of tapioca flour in the process of chicken sausage making were significantly affect (P <0.05) moisture and protein content, while the fat and ash content were not significantly effected (P> 0.05) by the treatment. The addition of 10% tapioca flour obtained the highest nutritional value of protein (17.67 ± 0.93) % as compared to the other levels.

Abstract

Penelitian ini bertujuan untuk mengetahui karakteristik morfometrikSapi Bali di Kabupaten Sumbawa. Materi yang digunakan yaitu Sapi Bali yang terdiri dari 15 ekor pedet, 48 ekor muda, dan 61 ekordewasa.Pengambilan sampel dilakukan secara Purposive sampling dengan mengamati cirri-ciri morfometrik Sapi Bali. Hasil penelitian menunjukkan bahwa ukuran-ukuran tubuh sapi Bali betina dan jantan pada umur pedet memiliki tinggi badan, panjang badan, dan tinggi pinggul yang berbeda (P<0,05). Pada umur muda hanya parameter tinggi badan yang tidak berbeda (P>0,05) dan pada umur dewasa hanya parameter panjang badan yang berbeda (P<0,05).Lingkar dada memiliki koefisien korelasi yang paling tinggi terhadap bobot badan diikiuti panjang badan dan tinggi pinggul dengan nilai secara berurutan 0,88; 0,82; dan 0,80. Persamaan BB= -207,89 + 1,49PB + 2,5LD – 0,74TPdan derajat determinasi (R2) yaitu 0,80diperoleh melalui analisis regresi. Lingkar dada dan panjang badan secara signifikan (P<0,05) mempengaruhi bobot badan sedangkan tinggi pinggul tidak (P>0,05) dan secara simultan berpengaruh terhadap bobot badan (P<0,05).

Abstract

Penampungan semen pada kerbau seringkali menghasilkan semen yang tidak menunjukkan adanya pergerakan massa spermatozoa. Semen seperti ini disebut semen statik, sedangkan semen yang menunjukkan adanya pergerakan massa spermatozoa disebut semen motil. Penelitian dilaksanakan untuk mengevaluasi kualitas semen dan spermatozoa pada semen motil dan semen statik kerbau lumpur. Sumber semen diperoleh dari 3 ekor kerbau lumpur dengan kisaran umur 2.5 sampai 3,0 tahun dan berat badan 441 sampai 477 kg yang dipelihara di Teaching dan Experimental Farm, Fakultas Peternakan Universitas Mataram. Sebanyak sembilan puluh delapan ejakulat terdiri atas 52 ejakulat semen motil dan 46 ejakulat semen statik digunakan pada penelitian ini untuk mengevaluasi kualitas semen dan spermatozoa pada ketiga sumber semen tersebut. Kejadian ejakulat statik adalah sekitar 47 persen yang bervariasi dari pejantan ke pejantan. Penelitian menunjukkan bahwa perbedaan di antara kerbau jantan didapati tidak signifikan untuk semua karakteristik semen statik, kecuali volume semen. Ketika data karakteristik semen dari ketiga kerbau jantan disatukan, ejakulat statik menunjukkan pH, total spermatozoa abnormal dan ekor bengkok yang lebih tinggi (p<0,05) dibandingkan dengan ejakulat motil. Sebaliknya, ejakulat statik menunjukkan penurunan volume ejakulat, konsentrasi sperma dan spermatozoa hidup yang signifikan (p<0,05). Diperlukan penelitian lebih lanjut tentang kesesuaian semen motil kerbau lumpur yang dipreservasi dalam keadaan cair dan beku.

Abstract

Penelitian ini bertujuan untuk mengetahui karakateristik morfometrik dari sapi persilangan Sumbawa x Bali (Sumbal) di Kabupaten Sumbawa. Materi yang digunakan yaitu Sapi Sumbal yang terdiri dari 14 ekor pedet, 20 ekor muda dan 27 ekor dewasa. Pengambilan sampel dilakukan secara purposive sampling/ dengan mengamati ciri-ciri morfometrik Sapi Sumbal. Hasil penelitian menunjukkan bahwa ukuran-ukuran tubuh Sapi Sumbal jantan tidak berbeda (P>0,05)dengan betina pada kelompok umur pedet dan muda. Sedangkan pada kelompok umur dewasa, jantan lebih rendah dari betina (P<0,05) pada parameter tinggi pinggul. Lingkar dada memiliki koefisien korelasi yang paling tinggi terhadap bobot badan diikuti tinggi badan dan panjang badan dengan nilai secara berurutan 0,96; 0,83; dan 0,80. Persamaan BB = -173,104 - 1,837TB + 0,052PB + 4,493LD dan derajat determinasi (R2) yaitu 0,939 diperoleh melalui analisis regresi. Lingkar dada dan tinggi badan secara signifikan (P<0,05) mempengaruhi bobot badan sedangkan panjang badan tidak (P>0,05)dan secara simultan berpengaruh terhadap bobot badan (P<0,05).

Abstract

The purpose of this research was to identify polymorphism of leptin gene in Bali cattle and Kacang goat by PCR-RFLP technique. Material used was blood samples collected from 50 head of Bali cattle and 50 head of Kacang goat. Identification procedure was consisted of three stages namely extraction of DNA genom, PCR amplication of leptin gene (552bp on intron 2-exon 3) and RFLP method using enzyme restriction (BsaA1). Primer sequences used were forward primer: 5'GTCTGGAGGCA AAGGGCAGAGT-3'and reverse primer: 5'CCACCACCTCTGTGGAGTAG-3'. Data analyzed were frequency of allele, genotype frequency, heterozygosity value, and value of Polymorphic Informative Content (PIC). The results of this study indicate that three genotypes (AA, AG and GG) were found on Bali cattle whilst only two genotypes (AA andAG) were on Kacang goat. The values of heterozygosity observation (Ho) and expectation heterozygosity (He) on Bali cattle were 0.08 and 0.48 respectively. Kacang goat showed the value of 0,14 (Ho) and 0.14 (He). The PIC value of Leptin-BsaA1gene on Bali cattle was 0.43 (moderate high), and on Kacang goat considered as low (0.13). These results showed than Leptin-BsaA1 in Bali cattle and Kacang goat may be used as a genetic marker for selection.
