Abstract

This research was conducted to examine the chemical properties, lactic acid bacteria (LAB) populationand angiotensin converting enzyme (ACE) inhibitory activity of fermented beef meat with the addition of bamboo shoots. This research used factorial completely randomized design.Factor A was the preparation method of bamboo shoots: chopped, ground, and extracted. Factor B was the ratio of meat and bamboo shoots (B) which was1:0.75,1:1, 1:1,25ratios. The chemical properties observed were total protein, soluble protein, hydrolysisdegree, pHandtitratableacidity value. The results showed that bamboo shoot preparation methods by ground and extracted were able to reduce total protein, and pH value. The ratio of meat and bamboo shoot (1:0.75 and 1:1) wereable to decreasedtotal protein but increased total titratable acidity. The interaction of the factors A and factor B only occurredin increasing hydrolysisdegree. Both of two factors (A and B) couldnot significantly effect to the number of LAB.The ACE inhibitory activity ranged from 36.5 to 79.6%. It can be concluded that the preparation methods ofbamboo shoots by ground and 1: 0.75 and 1: 1ofbeef : bamboo shoot ratio couldincreasehydrolysis degree and had highest ACE inhibitory activity.

Abstract

The diversity of leptin genes in cattle has an impact to the physiological function of the leptin hormone as anti-obesity. This study was aimed to determine polymorphisms of the Leptin gene based on Single Nucleotide Polymorphism on exon 3rd and their effect to body weight, of Padjadjaran sheep. DNA samples were obtained from the blood of 26 Padjadjaran rams 1 years. Data of leptin gene diversity were analyzed from the results of PCR product sequencing by Sanger Methode of 696 bp leptin exon 3. Point mutations (SNPs) on nucleotide sequence were analyzed using Bioedit and MEGA 7 Program. Growth parameters which were measured i.e. monthly body weight gain until one year of age. The association of leptin gene polymorphism with body weight was find out by unpaired T test. The results showed there were SNPs in Leptin gene in exon 3 of Padjadjaran sheep, namely g.170 G> A and g. 332.A> G. Both of these mutations are non-synonymous mutations because there were altered amino acids from Arginine to Glutamine. Genotyping was done based on the two mutations above and got two alleles G and A, each of which has an allele frequency of 0.75 and 0.25, with the frequency of each genotype, AG and GG was 0.5. There was not association between SNPs on leptin gene with body weight from birth to 12 months of age.

Abstract

Central Java is the region that produces the second largest duck egg production in Indonesia or reaches 13.55% of the total national duck egg production. Therefore, duck farming in Central Java requires an economic evaluation to identify the success of the business. The economic analysis in the duck farm industry will update the latest statistics on production costs, outputs and profitability. This study aims to assess the analysis of laying duck business, which includes: marginal production costs, total net income and input-output ratio of duck egg production in various regions in Central Java, Indonesia. The study was conducted using a survey method using multi-stage sampling involving 120 duck farmers from the regency of Tegal, Brebes, and Pemalang. Each district was taken 40 farms as respondents by purposive random sampling with criteria for the number of breeders who had a minimum duck population of 100 ducks and a production period of 8-12 months. The analytical tools used include descriptive statistics and farm budgetary analysis. This study showed that duck breeders incur large costs to obtain production inputs, especially from feed aspects. Although three regencies reported profit based on the estimated input-output ratios, it is only a marginal profit that amounted to1: 1.382, 1:1.658, and 1:1.433 in Tegal, Brebes, and Pemalang, respectively. The conclusion of the study is the performance of duck farms in Central Java are still profitable even in the smallholder duck farm industry, with the highest profits obtained by farmers in Brebes Regency.

Abstract

The objective of the current study was to investigate the effect of addition an amino acid or its combination with EDTA on DNA integrity and morphometry sperm heads of freeze-dried bovine spermatozoa. Spermatozoa were freeze-dried in medium with the addition of an amino acid glycine, cysteine, glutamine, or its combination with EDTA. Sperm head morphometry was identified at 400X magnification using Axio Vision(Zeiss Company, Germany), while for membrane plasma integrity (MPI) determined by calculation of the percentage of spermatozoa having intact plasma membrane by osmotic resistance test method and DNA integrity analysis using acridine orange staining. Sperm head had declined in size after the freeze-drying process, MPI of FD spermatozoa gradually increased significantly when FD solution was added with an amino acid solution (glycine, cysteine) and its combination with EDTA. DNA integrity of all freeze-dried spermatozoa treatments was remaining intact, no significantly different (P>0.01) among treatments. The present study concluded that the addition of an amino acid (glycine, cysteine) or its combination with EDTA could be reduced morphometric sperm heads and plasma membrane damage of freeze-dried bovine spermatozoa, however, DNA integrity of bovine sperm nucleus remaining intact after the freeze-drying process without addition both amino acids and EDTA.

Abstract

This study aimed to analyze the impact of electronic marketing utilization and to determine the economic determinants to increase the income of small dairy farmers in Boyolali, Central Java, Indonesia. A survey of 80 dairy farmers from the Cepogo District, Boyolali was determined by simple random sampling. The results showed that the average milk production was 8.3 liters/head/day, the revenue of IDR 3,824,000 per month, production costs of IDR 2,105,000 per month, and income of IDR 1,719,000 per month. Furthermore, by using electronics marketing, farmers could sell 40% of their products to coffee shops and generate revenue of IDR 5,411,500, production costs of IDR 2,705,000 and income of IDR 2,706,500. The utilization of the electronic marketing could increase R/C from 2.1 to 2.43. Economic factors that influence income are market prices, transportation and communication costs, marketing costs and distance traveled. Based on the results of the study it can be concluded that the use of the electronic market can expand the market, increase milk prices and ultimately will increase income

Abstract

The study was conducted to investigatethe effect of L-Carnitine supplementation in Tris-eggyolk (TEY) diluents to optimize the quality of Pasundan bull liquid semen. Semen samples were collected from three Pasundan bulls (6 – 7 years old)usinganartificial vagina. Semen samples were evaluated macroscopically and microscopically, semen having ≥70% progressive motility, ≥500×106ml-1 sperm concentration and ≤ 20% sperm abnormalities were divided into 5 equal part. Each part  wasdiluted with TEYand supplemented with0 mM(control), 1 mM, 2 mM, 3 mM and 4 mM L-Carnitine. Liquidsemen was storedat5°C.Sperm motility was evaluated every 12 h  objectively using Computer Asissted Sperm Analysis (CASA) until progressive motilityreached 40%. The best diluent was demonstratedby progressive motility (PM)by TEY supplemented with 1 mM L-Carnitine(43.08±0.49%) that remained up to 108 h compared tothecontrol group (43.63±0.70%) that remained up to 72 h (P<0.05). In conclusion, 1 mM L-Carnitine in TEY was the best concentration for preservation of Pasundan bull semen stored in the liquid form.

Abstract

This study was aimed to analysis the effects of roughage sources and oil sources in a total mixed ration (TMR) on voluntary feed intake, digestibility and blood chemistry. The study was conducted using four crossbred Thai Native X American Brahman cattle, aged 1.5 years old and with a live weight of 177±34 kg, which were trialled by using a 2x2 factorial in 4×4 Latin square design. All cattle received TMR diets supplemented with bagasse and palm oil, bagasse and sunflower oil, rice straw and palm oil, rice straw and sunflower oil. Each period of feeding lasted for 21 days. During the experimental periods, all cattle were fed ad libitum and with a water supply. The results revealed that roughage sources and oil sources did not affect voluntary feed intake, digestibility and blood chemistry. It was concluded from this experiment that using bagasse or rice straw as the roughage sources with palm oil and sunflower oil mixed in the TMR were suitable for use in beef cattle diets, especially using bagasse with palm oil because of the lowest price (0.28 USD/kg) when compared to the other treatments.

Abstract

This study aimed to identify the variation of 5’UTR Myostatin (MSTN) gene and its association to body weight in Boer, Kacang, and Boerka goats. DNA samples were obtained from 149 heads of the goats from the Indonesian Goat Research Center, Sungei Putih, North Sumatera. Polymorphism identification was conducted by direct sequencing and PCR-RFLP with DraI as the restriction enzyme for indel g.1256/TTTTA. Analysis of variance for body weight was carried out using the General Linear Model (GLM). The 5’UTR MSTN gene|DraI was monomorphic in Kacang but polymorphic in Boer and Boerka. Genotype frequencies for Boer 0.40(AA), 0.43(AB), 0.17(BB); Kacang 1.00(AA); Boerka 0.53(AA), 0.37(AB), 0.10(BB). The allele frequencies for Boer 0.62(A), 0.38(B); Kacang 1.00(A); Boerka 0.72(A), 0.28(B), respectively. AB was the most frequent genotype among Boer, but AA was the most frequent in Kacang and Boerka. Indel g.1256/TTTTA has a significant effect (P<0.05) only on birth weight (BW) of Boer, but no significant effect on other bodyweight parameters both in Boer and Boerka.AA genotype has the highest BW (P<0.05) than AB, but it’s not significantly different from BB. Indel g.1256/TTTTA could be used as a genetic marker for the birth weight of Boer but not in Boerka goats.
