Abstract

Background: One of the most common difficulties found in leprosy management is how to manage leprosy reaction, even though MDT has proven to be effective for the patients. Reversal reaction is associated with cell mediated immunity (CMI), consists of T-helper (Th) 1 and Th2 activities. Moreover, subset of CD4+ Th17 and CD25+ FOXP3 Treg were known to have an important role in leprosy’s natural history. Prior study showed a decreasing number of CD25+ FOXP3 Treg and increasing number of CD4+ Th17 in type II reversal reaction (ENL). However, the distribution of Th17 and Treg in type I reversal reaction (RR) has not yet been identified.Objective: To compare the distribution of CD4+ Th17 and CD25+ FOXP3 Treg between RR and ENL patient groups.Method: A total of 50 samples, consisted of 27 samples of reversal reaction (RR) and 23 samples of ENL, were collected. Observation of CD4+ RORg-T Th17 and CD25+ FOXP3 Treg were conducted with immunohistochemistry staining technique using anti FOX-P3 and anti RORg-T. Expression of CD4+ ROR-g Th17 and CD25+ FOXP3 Treg in percentage were analyzed using T-test.Result: There is a significant difference in mean CD4+ ROR-g Th17 and IL17 cell distribution for RR patient group (14.96% and 10.72%) compared with ENL (9.12% and 4.28%). No significant difference were found between mean CD25+ FOXP3 Treg and TGF-β cell distribution in RR patient group (6.12% and 5.44%) compared with ENL group (6.16% and 5.96%).Conclusion: There is a significant increment od CD+RORg-T Th17 and IL17 in RR patients group compared with ENL patients group. however, the distribution of CD25+ FOXP3+ Treg and TGF beta in RR has no significant difference campared with ENL.

Abstract

Background: Breast cancer is on the first or second most malignancy in many places around the world. The incidence of breast cancer was increasing, also in Indonesia. Prevention and treatment of breast cancer have not been satisfactory, since patients and medical practitioners are searching for other therapeutic and preventive agents. Salvia miltiorrhizae Bunge roots is one of the therapeutics agent used in traditional medicine have been studied in foreign country.Objective: To prove the p53 and Bcl2 expression in C3H mice mammary adenocarcinoma using multilevel doses of the Salvia miltiorrhizae Bunge roots extract. Methods :The study is a randomized post-test only controlled group design. A total of 18 adenocarcinoma mice, divided into 3 groups. The first group was administered with 60mg/KgBW/day and the second group was given by 180mg/KgBW/day, for 21 days. All mice were terminated on day 22nd. The p53 and Bcl2 expressions were analyzed using Allred score, statistically analized by Kruskal Wallis followed by Post-Hoc Mann Whitney U test.Results :Immunohistochemistry staining shows the p53 expressions have been increased between groups, but Bcl2 were decreased. The Kruskal-Wallis and Post-Hoc Mann Whitney U test are significantly difference between groups.Conclusion :All of the p53 and Bcl2 expressions between groups are significantly differences.

Abstract

Background: In endometriosis there is a decrease in endometrial receptivity. Kebar and Metformin grasses are expected to improve the condition of endometrial receptivityObjective: To find out whether there is any effect of giving Kebar grass extract and metformin on endometrial receptivity Methods: The study was designed by using a simple experimental randomization. It was conducted in Obstetrics Gynecology Department - Faculty of Medicine Diponegoro University and Faculty of Veterinary Medicine Airlangga University. Sample calculation was employed. The study was done within five groups including control group with normal mice, group of endometriosis mouse models given placebo, group of endometriosis mouse models given Kebar grass extract, group of endometriosis mouse models given metformin, and group of endometriosis mouse models given the combination of Kebar grass – metformin extract. The examination materials of ανβ3 integrin and Leukemia Inhibitory Factors were taken from the uteri of the mice and measured by the Rammele Scale Index (Immuno Reactive Score), while the Superoxide Dismutase examination using ELISA was derived from mice serum. These examinations were performed by two veterinariansResults: The expression of ανβ3 integrin was significantly higher in the group of mouse models given Kebar grass extract as well as the combination of Kebar and metformin grass extracts (p <0.05) rather than in the group of endometriosis mouse models with no treatment, whereas LIF expression was significantly higher in the group given Kebar grass extract, metformin and combination of both extracts (p <0.05). Serum SOD levels remained the same.Conclusion: The expressions of ανβ3 Integrin and LIF are higher due to the provision of Kebar grass extract as well as the combination of Kebar grass and metformin extracts

Abstract

Background: Oligonol® (OLG) is a functional food product and ingredient for cosmetics derived from a lychee fruit polyphenol. It has been reported to act on the skin as an anti-inflammatory and prevent UVB-induced skin damage.Aim: In this study, with the aim of exploring new functionalities of OLG on the scalp, we investigated the effect of OLG on human dermal papilla cells by comparing with adenosine and minoxidil at the genetic level.Method: OLG, adenosine, and minoxidil were applied to human dermal papilla cell lines for 24 h, after which VEGF, FGF-7, WNT5a, and WNT10a mRNA expressions were measured by real-time PCR analysis. Additionally, using DNA microarrays, we investigated the effect on 205 inflammation-related genes.Result: Consequently, in human dermal papilla cell lines, FGF-7 and WNT10a mRNA expression were observed in 100 µg/mL OLG-supplemented cells. The results of the DNA microarray analysis showed that 10 genes were suppressed by OLG.Conclusions: OLG may be expected to affect function of human dermal papilla cell by regulating the expression of genes related to cell proliferation and inflammation.

Abstract

Background: Leprosy, a chronic infectious disease presents a broad clinical spectrum that is correlated with the immunological response of the patient, mainly related to Th1/Th2 cells. IL-10 is a major cytokine produced by Th2 cells inhibits imunostimulatory cytokine produced by Th1 cells. Suppressive effects of IL-10 in monocytes and cytokine synthesis by Th1 cells presumably because IL-10 has a general suppressive effect on immune function. Nigella sativa has a potent potentiating effect on cellular immunity through suppression of Th2 cells and IL-10, resulting in potentiation of Th1 cells. Method: The study design is a randomized pretest and posttest controlled design involving 40 subjects of MB leprosy patients. Serum levels of IL-10 were measured by ELISA. Result: The mean decrease in serum levels of IL-10 (IL-10 delta) in the treatment group (average fell 3.12 pg/ml) is greater than the control group (average rose 0.21 pg/ml), where the difference is statistically significant (p = 0.029). Nigella sativa giving significant correlation with a decrease in IL-10 compared to the control group (p=0.044, OR: 10.23). Conclusion: supplementation of Nigella sativa 3 x 1000 mg for 2 months in patients with MB leprosy can reduce levels of IL-10, thus increasing the cellular immune response in patients with MB leprosy.

Abstract

Introduction: Prostate is a male organ which might enlarge mostly, either benign or malignant. Hyperglycemia is one of the factor that increase the risk of  benign prostate hyperplasia. There is lack of studies which assessed the relationship between benign prostate hyperplasia and isolated hyperglycemia. The aim of this study was to evaluate the association between hyperglycemia and prostate volume in patients with benign prostate enlargement in dr. Kariadi Hospital Semarang.  Method: We conducted a retrospective analysis of clinical data which obtained from 640 men between 2010 and 2012 who admitted to the hospital with diagnosis of benign prostate enlargement. By their medical records, these patients were evaluated of their plasma glucose level and prostate volume by trans rectal ultrasound. The  presence  of  hyperglycemia  was  determined  based  on  the  American  Diabetes Association (ADA) and the European Association for the Study of Diabetes (EASD). Patients have already been diagnosed with controlled diabetes mellitus by an internist. We allocated the subjects into two groups: patients with hyperglycemia and non-hyperglycemia. Logistic regression analysis was used to assess whether hyperglycemia was associated with the increased risk of benign prostate enlargement.Results: Significant difference of prostate volume found between groups. Prostate volume was  significantly greater in hyperglycemia group compared with non-hyperglycemia ones in all sub-groups based on age (in decades). Odds Ratio (OR) in patients with hyperglycemia was 2.25 (95% CI: 1.23-4.11). By non-parametric Spearman test it obtained  Group 1 (P1) p = 0.000 and r = 0.669, group 2 (P2) it obtained p = 0.000 and r = 0.672, group 3 (P3) it obtained p = 0.000 and r = 0.415 which implied strong positive associationConclusion: Hyperglycemia and prostate volume were significantly associated in patients with benign prostate enlargement. Hyperglycemia became a significant risk factor  for  prostate  enlargement  in  patients  with  benign  prostate  enlargement  in dr. Kariadi Hospital Semarang.

Abstract

Background : Rusip consumption can change the microbial composition of the digestive system. Microbial changes due to rusip consumption that is measured by lactic acid bacteria count has not been conducted.Objective : The purpose of the study was to analyze the effect of rusip consumption to the number of lactic acid bacteria in the gastrointestinal tract.Methods : Experimental study with post test only control group design using twelve Rattus novergicus Sprague dawley strains aged 2-2.5 months which were divided into 2 groups. Rats adapted for a week, then they were given rusip 5.1 mg/g body weight for 14 days. Cecal content were taken on the 22th day to be analysed for the lactic acid bacteria count.Results : The number of lactic acid bacteria (LAB) in the group given rusip was lower compared to the control group, but not significantly different (p=0.180). Weight gain was greater in the treatment group compared to the control group. The given group tends to have more weight in cecal content and lower pH compared to the control group.Conclusion : Rusip consumption does not increase the number of lactic acid bacteria in gastrointestinal tract.

Abstract

Background: Preeclampsia and eclampsia arestill major problemsin the world. Maternal mortality of severe preeclampsia at the puerperal period is likely to be greater because of the cardiovascular diseases (CVD). HsCRP (high sensitivity C-Reactive Protein) is a usefull prediction for CVD among non preeclamptic patients. Before using hsCRP as a marker for that prediction of cardiovascular event from preeclamptic patient, we should do research to know difference of hsCRP between preeclampsia and normal pregnancy.Objective: To determine the differences levels of hsCRP among severe preeclampsia and normal pregnancyMethod : A cross sectional study was conducted to severe preeclampsia. The inclusion criteria were subjects in puerperal period (2- 6 weeks) with a history of severe preeclampsia and normotensive.Exclusion criteria were puerperal infection, chronic hypertension, metabolic syndrome, caesarean section delivery and refuse to join this research. All subjects were examined the levels of hsCRP maternal serum when blood pressure £140 / 90.Result: Subjects were consist of 26 severe preeclampsia (53%) and 23 normal pregnancy (46.9%). Level of hsCRP in severe preeclampsia was 4.73 +3.57while in normotensive 2.42 +4.14 (p <0.05). Severe preeclampsia group will increase the risk of hsCRP rise as much as 2.5 times compared to the normotensive group.Conclusion:Level of hsCRP in patients with preeclampsia post partum were higher than patients with normal pregnancies.

Abstract

Background: Glaucoma patients often come with intraocular pressure (IOP) that reach target pressure, but still have glaucoma progression due to IOP fluctuation. Water drinking test (WDT) can be used as a method to predict IOP fluctuation.Purpose: To compare prostaglandin analogue (PGA) and selective laser trabeculoplasty (SLT) capability to maintain IOP fluctuation on primary open angle glaucoma (POAG).Method: This clinical experimental research was conducted at Dr. Kariadi Hospital. Subjects were POAG patients selected with consecutive sampling. Intraocular pressure was measured before and after WDT. After WDT, IOP was measured every 15 minutes until 1 hour. Peak IOP and IOP fluctuation data were collected then analyzed with t-test.Results: Forty two eyes from 30 POAG patients were analyzed. Twenty six eyes in PGA group and 16 eyes in SLT group. Most of the subjects were male with mild glaucoma degree. WDT increased IOP significantly on both groups. Mean IOP pre WDT was 14.58 ± 2.580 mmHg and 16.94 ± 2.645 mmHg in PGA and SLT groups. Mean peak IOP post WDT was 22.73 ± 4.114 mmHg and 25.75 ± 5.859 mmHg in PGA and SLT groups. Mean IOP fluctuation was 8.15 ± 4.202 mmHg and 8.81 ± 5.344 mmHg in PGA and SLT groups. Peak IOP and IOP fluctuation result analysis on both groups were not significantly different.Conclusion: Prostaglandin analogue and SLT had equal capability to maintain IOP fluctuation but still higher than normal fluctuation (>6 mmHg) that affects glaucoma progression.

Abstract

Background: High fat diet is a diet containing large amounts of fat consistently, the increase dietary fat and cholesterol which have a key role in growing health problems. Extra virgin olive oil associated with prevention of LDL oxidation, beneficial changes in lipid ratios and low risk for CHD.Objective: to determine the anti-inflammatory effect  of extra virgin olive oil extract to  levels of COX-2 and LOX-1 in the blood in rats induced by high fat dietMethods: This research is an experimental study that used randomized posttest only control group design. 30 Wistar rats which were divided into five groups:  group of control (-) which received  normal diet and group of control (+) which received  high fat diet without  EVOO treatment and three high fat diet groups treated by EVOO 1 mL/kg/day, 2 mL/kg/day and 3 mL/kg/day orally for 2 months. The blood was collected from eyes rats and serum separation by centrifuge. COX-2, LOX-1 concentration was measured by the enzyme linked immunosorbent assay (ELISA).Results: The result of this study showed higher COX-2 concentrations in groups treated with EVOO than control group. COX-2 serum levels of negative control where significantly lower than those of rats treated with 2 ml/kg/day (p = 0.047) and 3 ml/kg/day EVOO (p = 0.014). The COX-2 serum levels of group received 1 ml/kg/day were significantly lower than those of rats received 3 ml/kg/day EVOO (p = 0.027). And showed  not significantly deferent among all groups (p = 0.570).Conclusions: The conclusion of this study has showed that extra virgin olive oil extract might have minor anti inflammatory and antioxidant effect in rats.
