Abstract

One of the pathogen infecting peanut in Indonesia is Peanut stripe virus (PStV,), causing stripe and blotch symptoms on infected peanut leaves. The objectives of this research were to evaluate effects of PStV infection on yield of 10 introgression lines of peanut derived from crossing of Arachis cardenasii and A. hypogaea, and to determine the tolerance of these lines against PStV infection. Peanut plants were grown in polybag containing 10 kg of potting soils and were grown under glasshouse conditions. The experimental unit consisted of four plants grown separately in four containers and for each treatment was replicated four times. Peanut plants were inoculated mechanically with Bogor isolate of PStV at 15 days after planting (dap) and harvested at 95-100 dap. Results of the experiment indicated peanut cv. Gajah belonged to moderate tolerance while Kelinci was sensitive against PStV infection. Introgression line NC-CS11, CS30 and WS4 were grouped as tolerance while NC-CS51, WSl, and WS3 were moderate tolerance. The tolerance lines showed mild mosaic symptoms, did not show reduction of plant height and peanut yield upon inoculation with PStV. Introgresion line NC-CS15, CS20,CS22, and CS50 were sensitive against PStV infection, showed moderate to severe mosaic/blotch symptoms, reduction of plant height and peanut yield due to PStV infection. Among the tolerance and moderate tolerance lines, only NC-CS30 showed higher yield as compared to peanut cv. Gajah or Kelinci. Therefore, this line may be developed further as commercial peanut cultivar or be use as donor germplasm for PStV tolerance mechanisms in peanut breeding.

Abstract

The objectives of this research were to determine the species composition, distribution pattern and virus transmission properties of N. virescens. Sweep net sampling of the leafhopper and others tungro vectors population were conducted from endemic areas in Central Java, East Java and Bali and also non endemic areas from Central Java and East Java. The species composition and distribution pattern of the leafhopper were studied in seedbeds and early stage of rice crop (30 days after transplanting). Inoculation test using Cisadane seedlings was conducted to determine the efficiency of virus transmission by N. virescens colonies. Four tungro virus vectors were found in all areas, namely N. virescens, N. nigropictus, N. malayanus, and Recilia sp. The domination of N. virescens occurred either in the endemic area or the non endemic areas. However, the domination of N. virescens did not correlate with the occurrence of the disease in the endemic area. The distribution pattern of N. virescens was regular. The number of N. virescens active transmitter in the endemic area was higher than that in the non-endemic areas. In addition, the female of N. virescens was more efficient in transmitting the tungro virus than that of the male.

Abstract

The existence of Clavibacter michiganensis subsp. michiganensis (Cmm), the pathogen causing bacterial canker, on 22 lots of tomato seeds distributed commercially in Indonesia was evaluated. Isolation of suspected Cmm colonies were conducted by agar dilution plating on semiselective SCM medium followed by confirmation of colony identity on YDC medium. Identity of suspected Cmm colony was confirmed using IF test, pathogenicity test on tomato seedlings, hypersensitivity test on leaf of Mirabilis jalapa and Nicotiana tabacum, ELISA, and Cmm specific DNA amplification by PCR. After seed extraction and evaluation of the extract on semi-selective SCM medium and confirmastion by IF test, at least six tomato seed lots were contaminated with Cmm. After more confirmation using pathogenicity and hypersensitivity test, ELISA, and PCR amplification, at least three seed lots were confirmed possitively to carry Cmm.

Abstract

This study was designed to investigate the paraquat toxicity toward root nodulation by Rhizobium on Macroptilium atropurpureum as an indicator plant. The legume was grown in Thornton medium treated with several concentrations of paraquat and inoculated with R.japonicum 143 (Rj-143) or Rhizobium sp. C-1.1. These bacteria represent cross-inoculation of soybean and cover-crops legumes, respectively. Nodule formation and Rhizobium population were measured periodically. At the end of planting time, nitrogenase activity of the nodules was analysis based on ARA (Acethylene Reduction Analysis) method. The results showed that nodules in plants inoculated with Rhizobium without addition paraquat, were formed within four weeks. There was no nodulation when paraquat was added. Paraquat was toxic to the plant, causing chlorosis, stunting, drying of the plant tissues, and death. The symptoms were detected at the second week after planting time. Paraquat also decreased Rhizobium population from 10^6 to 10^2 or 10^1 CFU/mL at 40 and 100 pp, respectively. These results depicted that paraquat disturbed the plant before nodulation, and at the same time Rhizobium populatin decreased until below minimal population required for nodulation. Therefore, the process of nodulation was disturbed, and in some treatments there was nodulation. It was concluded that paraquat was toxic to both plant and the Rhizobium, which cause nodulation failure.

Abstract

The barn owl, Tyto alba javanica (Gmel.); is one of the important predator of rats. For controlling the rat population, the predator must retain in the rice ecosystem in which it is supported by continuous availability of its prey and suitable nest. The barn owl does not make its own nest, hence, the suaitable nest are necessary vailable in the ecosystem. The rat population fluctuatesand in the certain time it is difficult to find, hence, alternative preys have to be available. The nest type and prey species of the barn owl in the rice-field ecosystem had been survayed in Kendal District, Central Java Province, from October 2003 until March 2004. The area represented the rice-field ecosystem type with various habitats. The prey species were described from analysis of 2.261 pellets that were collected from 106 nests. The nests were found and distributed in 72 sub-sub district and in 17 subdistricts. Hairs, pulmes, skulls, teeth, obtained from the pellets were identified following the Hafidzi & Naim method (2003). The nest types consisted of building plafon (76,41%), trees (20,75%) and nest box (2,84%). Rats were the predominant prey-species (86,90%); consisted of Rattus argentiventer 77,08%, Rattus norvegicus 9,86%, Rattus rattus diardii 0,58%, Bandicota indica 0,01% and Rattus tiomanicus 0,005%),followed by aves (5,49%), insects (3,98%), bats (2,07%), and others (1,59%) i.e. squirrels, gecko, house-lizard, and frogs.

Abstract

Research was conducted in Jogjakarta from April to October 2004, to know the existence and description of Dacus spp, in Indonesia especially in Jogjakarta Special Province: The result of the exploration shows that there are two species of Dacus spp. attracted to cue lure traps. The species are Dacus (Callantra) longicornis and Dacus (Callantra) petioliforma with characteristics as follows: notopleuron and mesopleural stripe are brownish on D. longicornis and yellow on D. petioliforma; mesonotum uniformly red-brown with midline dark marking and a small yellowish spot present on D. petioliforma and absent on D. longicomis; apical scutellum is brown fulvous on D. longicomis and yellow on D. petioliforma, on both of them are broad black fulvous on basal band.
