Abstract

The objectIves of the research were to study the effect of flooding and draining on the population and life cycle of root-knot nematode of rice (Meloidogyne graminicola). To study the effect of flooding and draining on the population of M. graminicola two experiments were done within two period of planting of rice IR 64. These experiments were arranged in completely randomize design with four replications. For studying the effect of flooding and draining on the life cycle of M. graminicola, the experiment was done within 30 days. The research results showed: 1) continuously flooding inhibit the penetration and population development of M. graminicola on rice IR 64, but the life cycle of M. graminicola was not affected; and 2) seventy two hours period of draining enhanced the development of M. graminicola compared to period of draining shorter than 72 hours.

Abstract

Many insects have been reported as pests of banana, among which the corm borer (Cosmopolites sordidus Germar) and pseudo-stem borer (Odoiphorus longicollis [Oliver]) are considered as the most important species. We found a beetle species during 2003 pest survey of banana in Rejang Lebong, Bengkulu, that had not been reported before in any publication. The adult feeds on and damages whorl of banana, and therefore we named it as "banana whorl beetle". The adult feeds on whorl causing irreguler elongated holes, forming a pattern of more or less parallel with leaf midrib when the leaf has fully opened. The holes often fuse and form larger ones. The larvae bores and tunnels into leaf midrib and pseudostem, very often from the base of leaf midribs, and develop to pupae inside the tunnel. Feeding activity of larvae and adult on banana cause serious growth retardation and failure to flowering. The beetle is sluggish, stays inside whorl or at the base of leaf midribs. It is bright/light brown in color, rather flattened, elongated, 17-19 mm length and 4.5-5.5 mm wide. The antennae are filiform, half of the body in length, consist of 11 segments. The elytra cover the whole abdomen, with parallel lines along them. Tarsi consist of four segment. The larvae have a bristle like structure along side the lateral and anal forceps at the tip of abdomen. Thoracic legs are absent. This beetle belongs to the family Chrysomelidae and subfamily Hispinae. The distribution of this insect is restricted to the area >400 m asl. and serious damages often occur above >700 m asl.

Abstract

This research aimed to know the intensity of fungal contamination in maize grain cattle-feed during storage. Five kilogram of grain sample were collected from Klaten, Sleman, and Muntilan, then stored at CV. Ragil Jaya’s warehouse for two months. Every two weeks the water content were measured and the grain were tested using PDA and blotter methods. Incubation during isolation process were conducted for seven days at 12 hour darkness and 12 hour light. Results showed that dominant fungal contamination from Klaten, Sleman, and Muntilan wasAspergillus sp. As2 isolate with contamination intensity as much as 89% (blotter), 73% (PDA), and 44% (blotter). The results also showed that factors which influenced the intensity of fungal contamination in cattle-feed maize is the grain condition before storage such as broken grain, dirt, and insect; and not caused by the planting location.  INTISARIPenelitian ini bertujuan untuk mengetahui intensitas cemaran jamur dominan pada biji jagung yang digunakan sebagai pakan ternak selama penyimpanan. Biji jagung pakan ternak dari Klaten, Sleman, dan Muntilan sebanyak 5 kg disimpan di gudang CV. Ragil Jaya, Magelang selama 2 bulan. Pengukuran kadar air dilakukan setiap 2 minggu dan kemudian diuji dengan metode PDA dan blotter untuk mengetahui cemaran jamur pada biji jagung. Hasil penelitian ini menunjukkan bahwa dari sampel biji jagung pakan ternak yang berasal dari Klaten, Sleman, dan Muntilan, jamur cemaran yang mendominasi, yaitu Aspergillus sp. isolat As2 dengan intensitas cemaran jamur tertinggi di daerah Klaten 89% (blotter), Sleman 73% (PDA), dan Muntilan 44% (blotter). Hasil ini menunjukkan bahwa hal yang mempengaruhi intensitas cemaran jamur pada jagung pakan ternak adalah kondisi awal bahan yang disimpan yaitu ada tidaknya kerusakan, kotoran, dan serangga; bukan lokasi penanaman jagung.

Abstract

An experiment concerning to the production of B. bassiana starter had been conducted to facilitate its use to the farmers. The production of starter was conducted by using maize rice mill as the growing medium of B. bassiana. To examine the effectiveness of the starter, it tested to H. hampei in vitro. The result showed that isolate of B. bassiana from Garut applied at 50 g/ml was the most effective to cause mortality of H. hampei. The percentage H. hampei mortality caused by this trial attained to 76 %.

Abstract

Cocoa pod borer (CPB), Conopomorpha cramerella Snellen (Lepidoptera: Gracillariidae) is a major pest of cocoa. Detection of the pest infestation using sex pheromone traps in the early growth and development of cocoa pods is important for an early warning system programme. In order to prevent the pest infestation the young pods were wrapped with plastic bags. A research to study the CPB incidence was conducted at cocoa plantations in Banjarharjo and Banjaroya villages, District of Kalibawang; Hargotirto and Hargowilis villages, District of Kokap; and Pagerharjo village, District of Samigaluh, Yogyakarta. The experiments design used RCBD with four treatments (sex pheromone trap, combination of sex pheromone trap and pod wrapping, pod wrapping, and control) and five replications. As many as 6 units/ha pheromone traps were installed with a distance of 40 m in between. Results showed that one month prior to the trap installation in the experimental plots there were ripen cocoa pods as many as 9-13%, which were mostly infested by CPB. During the time period of introducting research on August to Desember 2016 there was not rambutan fruits as the CPB host, hence the CPB resource was from infested cocoa pods. The CPB moth trapped as many as 0−7 (1.13 ± 0.14) moths/6 traps/12 observations. The seed damage due to CPB larvae in the pheromone trap treatments (23.98%) was relatively similar with the control (20.25%). Seed damage rate in combination treatment of pheromone trap and pod wrapping (0.59%) was relatively the same with the pod wrapping (0.20%). The pheromone trap was more usefull for monitoring tool rather than for control, meanwhile pod wrapping was an effective control measure of CPB. IntisariPenggerek Buah Kakao (PBK), Conopomorpha cramerella Snellen (Lepidoptera: Gracillariidae) merupakan salah satu hama utama kakao. Deteksi serangan hama PBK dengan perangkap feromon seks pada awal pertumbuhan dan perkembangan buah kakao penting dilakukan sebagai sistem peringatan dini. Dalam upaya pencegahan serangan hama tersebut dilakukan pembrongsongan buah muda. Penelitian dilakukan di kebun kakao rakyat di Desa Banjarharjo dan Banjaroya, Kecamatan Kalibawang, Desa Hargotirto dan Hargowilis, Kecamatan Kokap, serta kebun PT Pagilaran di Desa Pagerharjo, Kecamatan Samigaluh, Yogyakarta. Percobaan dengan empat perlakuan (perangkap feromon, kombinasi perangkap feromon dan brongsong, brongsong, kontrol) dan lima ulangan menggunakan rancangan RCBD. Perangkap feromon sebanyak 6 unit/ha dipasang dengan jarak antar perangkap sekitar 40 m. Hasil percobaan menunjukkan bahwa satu bulan sebelum pemasangan perangkap terdapat buah tua sebanyak 9−13 % dan umumnya terserang PBK. Selama kajian berlangsung tidak terdapat buah rambutan sebagai inang PBK sehingga sumber PBK diduga hanya dari buah kakao yang terserang. Ngengat PBK yang tertangkap yaitu sebanyak 0−7 (1,13±0,14) ekor/6 perangkap/12 pengamatan. Intensitas kerusakan biji akibat ulat PBK pada perlakuan perangkap feromon (23,98%) relatif sama dengan kontrol (20,25%). Kerusakan biji kakao pada perlakuan kombinasi perangkap feromon dan pembrongsongan (0,59%) relatif sama dengan pembrongsongan saja (0,20%). Perangkap feromon lebih berfungsi sebagai alat monitoring daripada alat pengendalian PBK, sedangkan pembrongsongan buah merupakan teknologi pengendalian PBK yang efektif.

Abstract

Corn borer is one of the important pests on corn plant in Gorontalo Province, especially in three districts which are the center of maize planting development. Corn borer and its natural enemy complex in Gorontalo Province has not been clearly studied. The purpose of this study is to determine the complexity of the natural enemies of maize stem borer by calculating the diversity and composition. The survey method was used, in which the older larvae, pupae, and corn borer natural enemies were collected from the field in the three districts (Gorontalo, Boalemo, and Pohuwato). Observation plots in each district area of 2,500 m2, were divided into 3 sub plots with the size of 3×3 m, each plot is consisted of 45 plants at the spacing of 75×25 cm. The data were analysed descriptively and organized into tables. Shannon Index was used to count the diversity and composition of the pests and their natural enemies. The results showed that the corn borer species was Ostrinia furnacalis, while the dominant natural enemy was the tachinid fly. The analysis of Shannon diversity index for natural enemies of three districts of Gorontalo, Boalemo, and Pohuwato were 0.63, 0.61, and 0.97 respectively.

Abstract

Cucurbits are grown throughout the Java Island as dry season crops. Plants having mosaic, mottling, chlorosis and leaf distortion symptoms were frequently found in most of the cucurbit fields during the survey which conducted in Central Java including Sleman, Kulon Progo, and Klaten during July–September 2000 and 2001. Using double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA); Cucumber mosaic virus (CMV), Cucumber green mottle mosaic virus (CGMMV) and Kyuri green mottle mosaic virus (KGMMV) were found infecting cucurbits. CMV was widespread, infecting 48.9% of the samples tested followed by CGMMV (12.8%) and KGMMV (6.4%), while others samples (31.9%) were not tested, double infections were common with 8.5 % of the samples being infected with two viruses (CGMMV and KGMMV) and 34% with three viruses (CMV, CGMMV, and KGMMV). Severe mosaic and mottle symptoms were associated most often with single infection of CGMMV and KGMMV respectively. In addition, these are the first detections of CGMMV and KGMMV infecting cucurbit plants in Indonesia.

Abstract

The development of the pink fungus stages showed that among the five stages, cobweb stage is the most important, since this stage may form the other four stages. I. Cobweb stage, develops following the germination of either basidiospores or conidia both on exposed and shaded side of branches. II. Pseudonodular stage, develops from symphogenous aggregation of cobweb stage mycelia on the surface of lenticel or cracks on shaded bark. III. Teleomorph, develops in two ways: a. Teleomorph incrustation develops from cobweb stage mycelia through four layers: basal layer, intermediate layer, subhymenial layer, and hymenial layer; b. Teleomorph pustule, develops from pseudonodular stage, through four layers, mirror to teleomorph incrustation. IV. Nodular stage, develops from meristogenous aggregation of cobweb stage mycelia on exposed bark. V. Anamorph, develops in two ways: a. From nodular stage; b. From cobweb stage mycelia on exposed bark.

Abstract

There were ten processed wastes (molase, vinase, sludge, ciu, brem, tofu, milk, cacao, fish, and beer) used in these experiments. The experiments were conducted in two separated arrangements: no choice test and choice test. The no choice test was carried by testing the attractiveness of B. carambolae on each waste was compared with water. Each run consisted of four replication, for each replication consisted of 20 females and 20 males. The choice test was conducted by placing all wastes in a cage at the same time. Each run consisted of 15 replication, for each replication consisted of 100 females dan 100 males. The no choice test results showed that B. carambolae tend to visit waste more than water. The choice test results showed that B. carambolae tend to be more frequent visits to beer and cocoa waste. The content of protein inside the waste seemed to play role in attracting B. carambolae to come with.

Abstract

There were some commonly found mosaic diseases on soybean in Indonesia that were suspected to be caused by viruses. The main virus diseases of soybean plant are Soybean mosaic virus (SMV) and Soybean stunt virus (SSV). The occurrence of mosaic diseases are very harmful because they might reduce soybean grain yield. The aim of this research is to determine the disease incidence and distribution of mosaic disease in the field that caused by SMV and SSV. The surveys were conducted in some regions of the area of soybean production center in Indonesia including: Special Region of Yogyakarta (DIY), Central Java, East Java, West Nusatenggara (NTB), and South Sulawesi. Detection of the viruses were done by serological test using I-ELISA and by molecular analysis using PCR. The field surveys showed that the mosaic symptoms always found in all the field location. The disease incidence and the disease severity varied in any locations. The disease symptoms showed as mild mosaic, yellow mosaic, mosaic with blister, and mosaic with leaves malformation by curling and stunting. The I-ELISA showed that all the samples collected were double infected by SMV and SSV. Both of the viruses were widespread all over the soybean production center in Indonesia. By PCR using CI-SMV primer, the DNA of virus could be amplified to about 638 bp which indicated SMV infection, but using cp-SSV primer, the DNA could be amplified to about 657 bp that indicated of SSV infection.  Pada pertanaman kedelai di Indonesia banyak ditemukan gejala mosaik yang diduga disebabkan virus. Virus penyebab penyakit mosaik pada tanaman kedelai antara lain Soyben mosaic virus (SMV) dan Soybean stunt virus (SSV). Keberadaan penyakit mosaik pada pertanaman kedelai sangat merugikan karena berpotensi dapat menurunkan angka hasil kedelai. Penelitian ini bertujuan untuk mengetahui tingkat kejadian dan sebaran penyakit mosaik yang disebabkan SMV dan SSV di berbagai sentra produksi kedelai di Indonesia. Survei dilakukan di DIY, Jawa Tengah, Jawa Timur, NTB, dan Sulawesi Selatan. Deteksi keberadaan virus dilakukan secara serologi dengan teknik I-ELISA (Indirect-Enzyme-linked Immunosorbent Assay) dan secara molekuler dengan teknik PCR (Polymerase Chain Reaction ). Berdasarkan pengamatan di lapangan gejala mosaik selalu ditemukan pada setiap lokasi pengamatan. Kejadian dan tingkat keparahan penyakit yang ditemukan bervariasi. Gejala yang ditemukan adalah mosaik ringan, mosaik kuning, mosaik dengan tonjolan warna hijau tua (blister), dan mosaik dengan malformasi berupa daun keriting dan tanaman kerdil. Berdasarkan uji I-ELISA semua sampel daun bergejala mosaik terinfeksi ganda oleh SMV dan SSV. Kedua jenis virus tersebut sudah tersebar luas di semua sentra pertanaman kedelai di Indonesia. Deteksi PCR menggunakan primer spesifik CI-SMV dari beberapa lokasi dapat mengamplifikasi pita DNA berukuran sekitar 638 bp yang mengindikasikan terinfeksi SMV, sedangkan menggunakan primer universal cp-CMV dapat mengamplikasi pita DNA berukuran sekitar 657 bp yang mengindikasikan terinfeksi SSV.
