Abstract

The Effect of Ethyl Methane Sulphonate (EMS) on Growth and Variations of Marigold (Tagetes sp.) The aims of this research are to determine the variation of marigold (Tagetes sp) derived from seed treated with EMS and to recommend the EMS concentrations that are able to induce varietion. Seeds of marigold cv Narai Orange were soaked in water for 6 hours, followed by soaking in EMS at concentration of 0%, 0.3%, 0.6% and 0.9% for 4 hours. This study employed Randomized Complete Blok Design with 10 replicates and each replicate consisted of 10 plants. Six plants were randomly chosen for measurements. The total number of samples observed were 240 plants. Observations were made on the percentage of the growth, plant height, number of leaves, number of branches, diameter and weight of flowers. Data obtained from the observations were analyzed using Analysis of Variance (ANOVA), followed by DMRT (Duncan’s Multiple Range Test) if there is a significant difference between treatments. The EMS treatment reduced all characters observed. The EMS concentration of 0.6% showed plant that had yellow flowers. The 0.9% EMS treatment resulted in one plant with chimera, 6 dwarf plants, 2 plants with thin stems, and 1 short plant with many branches. Untreated plants did not show any variation.

Abstract

This experiment was conducted in the village of Sibang Gede, District of Abiansemal, BadungRegency on November 2011 until February 2012. The research objective is to enhance the growth andyield of maize hybrid BISI-2 using rhizobacteria Pantoea agglomerans as PGPR in compostformulations. This study used a randomized block design (RBD) single factor with 5 treatments and 5replications. The treatments tested were P. agglomerans BS7a, P. agglomerans BS7b, P. agglomeransBS2a, and P. agglomerans BS5a. All treatments P. agglomerans formulated in the form of compostand given a dose of 50 g/hole. As a control dose of compost 50 g/hole without PGPR. The resultsshowed that the application of rhizobacteria P. agglomerans increased the maximum plant height, netassimilation rate, crop growth rate, chlorophyll content of leaves, fresh weight of roots per plant, ovendried weight of stover per plant, number of seeds per plant, cob length, cob diameter, seed weightmoisture content of 12% per plant and seed yield of 12% water content per hectare.

Abstract

Mathematical Model in Determining Effective Time for Growth Rate of Red Chili The growth rate showed a sigmoidal shaped pattern with initially slow growth rate and speeding up till a certain period before slowing down to approach a limit. So it is very important to define the plant growth model. The aim of the study is to estimate the effective time of plant growth by looking at the mathematical model. The statistical analysis showed that the growth ofCapsium annum Cult. Hot Beauty’s can be fit well by Janoscheck model and the effective growth time occurs as the plant is in the interval of 14 to 21 days after transplanting.

Abstract

The Quality Test on Soybean Seeds (Glycine max L. Merril) for Grobogan Variety that Produced by Using Applications of 10 Isolats PGPR. This research is aimed to know the ability of isolat Plant Growth Promoting Rhizobakteria (PGPR) producing soybean seed with quality and high seed quality. The complete random plans (RAL) are used in this research. Tens kinds of isolat rhizobakterias and 1 control (without rhizobakteria) are tested. Observation is done toward variable of sprouts’ energy, vigor (energy supplying), electrical conductivity, ratio of seed coat, and vigor of seeds growth (tall, number of leafs, dry weight for oven on ground, and dry weight for roots). The outcome shows of roots that 10 isolat rhizobakterias are able to improve the seed yield and soybean seed quality. 10 isolat rizobakterias produce soybean seeds with sprouts’ energy > 80% to fulfill seed quality standard “international seed testing association (ISTA)”. Having three isolats which are effective namely, R35 (it is isolated from cajanus cajan’s root), R3 (it is isolated from stylosanthes guianensis’s root), and R26 (it is isolated from solanum nigrum’s root). Three of those isolats produce high harvest namely R35 (10.4g), R3 (10.10g), R26 (10.86g). Those three isolats also produce seed with physical quality and high physiology.

Abstract

Adaptation Test of Promising Upland Rice Mutant Line On Dry Highland With Wet Climate. Upland rice productivity is still low, productivity can be improved by using high yielding varieties through the breeding programs, by the Agency for agricultural research (BATAN). Test adaptation is necessary to determine the superiority of promising lines. The objective of this research was to determine adaptability, performance of growth, and productivity of mutant lines of upland rice. Research used randomized complete block design (RAK) single factor with 10 lines of treatment the results of gene mutation and 2 check varieties with three replications. Those treatments were PMG 01/Psj, PMG 02/Psj, PMG 03/Psj, PMG 04/Psj, PMG 05/Psj, PMG 06/Psj, PMG 07/Psj, PMG 08/Psj, PMG 09/Psj, PMG 10/Psj, Limboto and Local. The research was conducted on August 2014 until February 2015 in Tiga Village, District Susut, Bangli regency at high dry land wet climate. Data were analyzed by analysis of variance (Annova) and LSD test at 5% level. The results showed that tested of promising mutant lines show significant differences were observed in all parameters. Treatment of lines PMG 05/Psj, PMG 06/Psj, and PMG 08/Psj gave a yield of 2.22; 2.66; and 3.18 tons GKG/ha or higher than Limboto and Local check varieties as big as 2.04 and 0.90 tones GKG/ha. The three lines showed the parameters of flowering dates, number of filled grain, empty grain, and weight of 1000 grain, harvest time, KAR leaf, and chlorophyll better than check varieties. Lines of PMG 08/Psj gave the results 36.49% higher than Limboto and 71.78% higher than local varieties.

Abstract

Study on Soil Nutrients in Rice Field to Determine Soil Fertility Status.Research of soilnutrients in rice field to determine soil fertilitystatus had been done on rice field in district Penebel,Tabanan regency.This study aimed to assess level of soil fertility status on rice field.The research wasconducted by survey and laboratory analysis. The study consists of several stages such as, the formationof the land unit based on the compilation maps of soil type, geology and slope.It’s obtained 11 landunits. Each unit tookone samples. The soil sample was analyzed for Cation Exchange Capacity andCation Saturated (NH4OAC extractional pH 7,0); N-Total (Kjeldhal); P-total and K-total (HCl 25% extraction); Salinity (EC), Organic matter(Wokly and Black)and pH (H2O)(1 : 2,5). The analysisproceseshave done at Soil laboratory, Faculty of Agriculture, Udayana University.The result oftheresearch show that status of soil fertilityon rice field at district Penebel, Tabanan regency are low1.478,551ha (29.16%), medium2.286,721 ha (63.83%), and high2.286,721 ha (6.01%).

Abstract

In Vitro Transformation of Flowering Gene through Agrobacterium tumefaciens on Vandatricolor Orchid. Research concerning of Agrobacterium-mediated transformation of Vanda tricolor orchid had been done during March to August 2014. Objectives of this research wasto investigate the effects of period of inoculation with Agrobacterium suspension and application of acetosyringone (AS) on the percentage of transforman candidates. The T-DNA harbored the PaFT gene under the control of promoter of Ubiquitin and the hpt gene, ahygromycine resistant gene as a selectable marker for transformant selection. This T-DNA was constructed in the PGAS vector and transferred to the plant genome mediated by A. tumefaciens. The period of inoculation with Agrobacterium and application ofAcetosyringone (AS) were trialed in this research. The result showed that a period of one hour inoculation with Agrobacterium suspension added with 25 ppm AS resulted in the highest number of transforman candidates. i.e. 7.04% compared to 4.10% (one hour inoculation without AS), 3.76% (two hours inoculation without AS), and 4.48% (two hours inoculation with 25 ppm AS).
