Abstract

Penelitian ini bertujuan untuk mencari kondisi yang tepat dalam percepatan pembungaan tanaman Phalaenopsis amabilis transgenik yang telah disisipi gen penentu waktu pembungaan Ubipro::PaFT. Metode penelitian ini menggunakan tanaman transgenik pembawa gen Ubipro::PaFT umur 18 bulan setelah penanaman. Tanaman ditumbuhkan pada inkubator dengan pencahayaan menggunakan lampu LED putih dan kombinasi LED putih biru, dengan fotoperiodisitas 8 jam terang 16 jam gelap, suhu 25ºC pada fase terang dan 20ºC pada fase gelap selama 20 minggu. Setelah 20 minggu pertumbuhan tanaman, dilakukan analisis profil protein dengan metode SDS-PAGE untuk mengetahui protein yang diproduksi pada setiap fase pertumbuhan yang diamati.Hasil penelitian menunjukkan kombinasi cahaya LED putih dan biru meningkatkan pembentukan daun sebesar 60%, panjang daun 70,58%, tetapi belum diperoleh kemunculan infloresen. Analisis profil protein menunjukkan terbentuknya protein dengan ukuran 108,57; 71,30; 56,16; 40,85; 26,79; 13,27; dan 13,12 kilodalton pada tanaman transgenik, tetapi tidak terdeteksi protein dengan ukuran 19,65 kDa yang sesuai dengan berat molekul protein PaFT, sementara protein dengan ukuran sekitar 56,16 kDa sesuai dengan berat molekul protein POH1(Phalaenopsis Orchid Homeobox1). Hal ini menunjukkan bahwa gen vegetatif POH1 mampu menghambat aktivasi gen PaFT pada tanaman P. amabilis transgenik umur 20 minggu, sehingga tanaman masih dalam fase juvenil dan belum mampu diinduksi untuk berbunga.

Abstract

The properness of concepts, principles, and biological contents of natural science textbooks is essential. The aim of this research was to compare the biology content of  natural science textbooks for VIII graders in second semester. The indicators used to determine the properness were the accuracy of Core Competence/CC and Basic Competence/BC, concept, principle, illustration, and word editing. The textbooks analyzed were those which published by Indonesian Ministry of Education and Culture (government) and private publisher. The sample of this qualitative descriptive research was three natural science textbooks which applied Curriculum 2013. The research procedure referred to Milies and Huberman. The results showed that the properness percentages of CC and BC of science textbook published by governmental and private publisher were 86.25% (excellent) and 74.28% (good) respectively. There were inaccuracy of 11 concepts, 1 principle, and 5 illustrations found in the textbooks published by governmental. Furthermore, there were 5 inaccurate concepts found in the textbook published by private publisher. Moreover, there were 37 typographical errors found in the Natural Science Textbooks published by government  and none found in the book published by private publisher. Therefore, the textbooks published by private publisher is better than those which by govermental publisher.

Abstract

Examination of Phalaenopsis amabilis orchid resistance to hygromycin antibiotic is an important step to doprior to Agrobacterium-mediated genetic transformation in this orchids using Hygromycin phosphotransferase(HPT) gene as a selection marker in the T-DNA that harboring a desired gene to be transfered. We exposedthe plant on hygromycin containing medium. The experiment was conducted using 6 weeks old P. amabilisprotocorms. These protocorms were subcultured onto NP medium supplemented with various concentrationof Hygromycin (0, 5, 10, 20, 1nd 40 mg/l). The number of survival protocorms were examined every week for4 weeks after subcultured (WAS). The resistancy of hygromycin was calculated as ratio of death protocormsper total protocorms). The result showed that 10 mg/l hygromycin with 1 weeks of application caused deathclose to LD 50. This data indicate that P. amabilis resistance to hygromycin treatment on the appropriateconcentration 10 mg/l, and this concentration can be used for other purposes in orchid system.

Abstract

Tujuan penelitian ini adalah mengetahui pengaruh kombinasi penambahan air kelapa dan vitamin B1 terhadap pertumbuhan anggrek Dendrobium nobile serta mengetahui konsentrasi vitamin B1 yang optimum untuk pertumbuhan anggrek pada media kultur in vitro berbasis pupuk. Penelitian ini merupakan penelitian eksperimen dengan menggunakan rancangan tersarang (nested design) dengan faktor I adalah perlakuan air kelapa (dengan air kelapa dan tanpa air kelapa) dan faktor II adalah variasi konsentrasi vitamin B1 (0 ml.L-1, 0,25 ml.L-1,0,5 ml.L-1, dan 1 ml.L-1). Sampel yang digunakan adalah bibit anggrek D. nobile umur 11 bulan dalam kultur in vitro yang mempunyai keseragaman karakter pertumbuhan. Data penelitian ini dianalisis menggunakan analisis ragam (Anova) metode rancangan tersarang dan uji Least Significant Difference (LSD). Hasil penelitian menunjukkan bahwa penambahan air kelapa memberikan pengaruh yang signifikan terhadap pertambahan diameter batang dan berat basah tanaman sedangkan penambahan vitamin B1 memberikan pengaruh yang signifikan terhadap pertambahan jumlah daun, jumlah akar, dan panjang akar anggrek. Konsentrasi vitamin B1 yang optimum untuk pertumbuhan anggrek adalah 0,5 ml.L-1 baik pada media yang mengandung ataupun tidak mengandung air kelapa.OPTIMATION FERTILIZER BASED IN VITRO CULTURE MEDIA COCONUT WATER AND B1 VITAMIN ADDITION FOR Dendrobium nobileThis study was aimed at determining the effect of the coconut water and vitamin B1 as a combination on the growth of Dendrobium nobile orchids and determining the optimum concentration of vitamin B1 for orchid growth in fertilizer-based in vitro culture media. This study was an experimental study using a nested design which factor I was the treatment of coconut water (with and without coconut water) and factor II was the variation in the concentration of vitamin B1 (0 ml.L-1, 0.25 ml. L-1, 0.5 ml.L-1, and 1 ml.L-1). The samples used were 11 months old D. nobile orchid seeds in in vitro culture which had simultaneous growth characters. The data were analyzed using Anova, nested design method and Least Significant Difference (LSD) test. The results show that the addition of coconut water has a significant effect on the increase in stem diameter and plant wet weight. The addition of vitamin B1 has a significant effect on the increase in the number of leaves, the number of roots, and the length of the orchids. The optimum concentration of vitamin B1 for orchid growth is 0.5 ml.L-1 both in media with or without coconut water.

Abstract

In Vitro Transformation of Flowering Gene through Agrobacterium tumefaciens on Vandatricolor Orchid. Research concerning of Agrobacterium-mediated transformation of Vanda tricolor orchid had been done during March to August 2014. Objectives of this research wasto investigate the effects of period of inoculation with Agrobacterium suspension and application of acetosyringone (AS) on the percentage of transforman candidates. The T-DNA harbored the PaFT gene under the control of promoter of Ubiquitin and the hpt gene, ahygromycine resistant gene as a selectable marker for transformant selection. This T-DNA was constructed in the PGAS vector and transferred to the plant genome mediated by A. tumefaciens. The period of inoculation with Agrobacterium and application ofAcetosyringone (AS) were trialed in this research. The result showed that a period of one hour inoculation with Agrobacterium suspension added with 25 ppm AS resulted in the highest number of transforman candidates. i.e. 7.04% compared to 4.10% (one hour inoculation without AS), 3.76% (two hours inoculation without AS), and 4.48% (two hours inoculation with 25 ppm AS).

Abstract

Bunga adalah faktor yang sangat penting dalam budidaya anggrek. Salah satu kendala yang sering dijumpai dalam budidaya anggrek adalah lama fase vegetatif yang dibutuhkan tanaman tersebut untuk dapat berbunga. Pada penelitian ini induksi pembungaan dilakukan pada tahap pertumbuhan awal tanaman secara in vitro. Tanaman umur enam bulan setelah tanam (bst) disubkultur pada medium New Phalaenopsis (NP) dengan konsentrasi Nitrogen (N) setengah dari NP (1/2NP), dengan atau tanpa pemberian Benzyl Adenine (BA), variasi konsentrasi KH2PO4 (1,5 mM dan 3 mM), serta dengan atau tanpa pemotongan akar. Kombinasi perlakuan dengan pemberian BA 22.2 µM, kandungan P tinggi (3 mM), dan pemotongan akar mampu mempercepat pembungaan in vitro anggrek Phalaenopsis amabilis (P. amabilis) asli Indonesia pada umur 18 bst.Flowering is very important in orchid cultivation. However, the long vegetative phase to be able to bloom of the plant becomes an important problem. The orchid needs three up to five years after sowing to bloom. In this study, flowering induction is done in the early growth stages of plants. At six months after sowing (mas), plants were sub-cultured on New Phalaenopsis (NP) medium witha half Nitrogen(N) concentration of NP (1/2NP), with or without Benzyl Adenine (BA), concentration variations of Phosphor/P (1,5 mM and 3 mM),  and with or without roots cutting. In vitro flowering of Indonesian Phalaenopsisamabilis (P. amabilis) can induced on medium that contain 22.2 µM BA and 3 mM P with roots cutting at 18 mas.

Abstract

Due to the high cost of the most often used basic media in tissue culture, it is vital to identify more affordable alternatives. This research aimed to determine the best alternative culture media for the protocorm and plantlet growth of Coelogyne pandurata Lindl. It employed a completely randomized design, eight treatments and four replications. The treatments referred to the different types media and consisted of M1 = New Phalaenopsis (NP) medium, M2 = a medium made from foliar fertilizer (FFM), M3 = NP + 2 cc L-1 AB mix solution (a media for hydroponics), M4 = FFM + 2 cc L-1 AB mix solution, M5 = NP + 50 cc L-1 of tomato extract, M6 = NP + 50 cc L-1 of bean sprout extract, M7 = FFM + 50 cc L-1 of tomato extract and M8 = FFM + 50 cc L-1 of bean sprout extract. The M4 medium exhibited the best results in terms of average leaves count (4.80), average shoot length (2.68 cm), average root length (4.35 cm), the average fresh weight per plantlet (214.5 mg) and dry weight of plantlets (73.1 mg). The average number of roots per plantlet was 4.25, acquired using the less expensive M8 treatment, which also produces a negligible number of leaves (4.50). In conclusion, the M4 medium is the most appropriate medium for growing protocorm and plantlet of C. pandurata. The experiment also found that the FFM basic medium combined with 50 cc L-1 of bean sprout extract can be used as another cheaper alternative for growing protocorms of C. pandurata.

Abstract

This research aimed to find out the effectiveness of experiential learning (EL) model with mind mapping to improve the science process skills (SPS) on fungi materials. It was quasi-experiment research by using pre-test post-test control group design. The population of the study was all of the tenth-grade students of Kalasan State 1 Senior High School amount of 127 students in the odd semester in 2018/2019 of the school year. Sample techniques were cluster random sampling. The sample consisted of 26 students (10 students male and 16 students female) as the experiment class by implementing EL model with mind mapping and 26 students (9 students male and 17 students female) as control class by using 5M (observing, asking, collecting, associating, communicating) learning model. The used instrument was ten essay questions. The data of the research were analyzed by using t-test (independent sample t-test). Based on the result of the normalized gain score, it showed that there was an improve of SPS about 0,85 in the experiment class higher than that in the control class about 0,70. Besides, it also showed a significant value (2-tailed) that was 0,000 (<0,05). It meant that there was a difference in science process skills between the experiment class and control class, so the learning process through EL learning model with mind mapping proved useful to improve the students’ science process skills.
