Abstract

The purpose of this study was to evaluate the effect of water in oil in water (w/o/w) emulsion containing 17a-methyltestosterone on gonad development of climbing perch. The results showed administration of 200 ug/kg 17o-methyltestosterone in w/o/w emulsion enhanced gonad development of climbing perch. In this treatment, testosterone concentration increased but estradiol-17p concentration decreased. Both hormones ranged 10,55-15,13 ng/dl and 0.96-5.66 ng/dl, respectively. This treatment also accelerated the development of egg diameter which were eggs e" 0.20 mm became 71.00% after 60 days, and development of egg nucleus position into germinal vesicle breakdown (GVBD) in day 60 was about 17.50%.

Abstract

This study was to identify the pathogenicity of local isolates of Salmonella enterica serotipe enteritidis (S. enteritidis/SE) in experimental chickens and mice. Thirty six local isolates of S. enteritidis isolated from chickens, eggs and human was inoculated 8 intra peritoneally (ip) to each 5 chickens and mice per group. Each group received 0.2 ml of broth culture containing 2 x 10Â CFU of S. enteritidis. Chickens and mice were observed daily at 1, 2, 3, 4, 5, 6 and 7 days after inoculation for clinical signs and mortality. The result showed that the overall mortality rate of chicken and mice were 78.4% and 63.9% respectively. The highest mortality rate was in the first day after inoculation, 51.1% for chickens and 40.0% for mice, however the lowest mortality rate was in the sixth day (0.0%) after inoculation in chickens and in the seventh day (0.6%) in mice. The bacteria of S. enteritidis can be isolated from various organs (liver, heart and spleen) and caeca tonsil of every dead chickens and mice. The study showed that local isolates of S. enteritidis isolated from chickens, eggs, and human were very pathogen for experimental chickens and mice.

Abstract

Green catfish is a commodity that is not only potential to be cultivated in the pond and in the floating net cages, but also easy to adjust to the artificial feed. The objective of this research is to evaluate the effect of different feed types (sinking and floating)on the growth performance of green catfish(Hemibagrus nemurus. One thousand and six hundred fingerlings averaging 2.50 g of 3 individual body weight were stocked in each of earthern ponds (10 x 8 x 1.2 m ). They were fed daily for four months with the same protein content (31%) and lipid (6%) diets. The feed was given in pelleted form at 8-4% of the total body weight. The result of this study showed that no significant difference (P>0.05) between different feed types. The sinking feed gave specific growth rate (2.35%), feed conversion ratio (2.49), protein retention (18.92%) and protein efficiency ratio 1.13 while the floating type gave the values 2.49%, 2.37, 20.48% and 1.21, respectively. Lipid retention of sinking feed (34.84%) is lower than that of floating type(48.39%).The survival rates ranged between 76.56-84.54%.Feed type sinking with protein content of 31% can be used in the rearing of fish in earthern pond.

Abstract

Pollution of heavy metals mercury (Hg) is a serious problem especially in gold mining areas. This study aims to isolate the bacteria in the gold mining of Pongkor (West Java) and testing the bacterial resistance to mercury (Hg). The results showed that of the 5isolates tested, qualitatively only 2 isolates (P3 and P5) that resistant to HgCI2, each at a concentration of 30 and 40 ppm.Furthermore, up to 70 ppm concentration of HgCl2, P5 bacteria can still grow well even require a longer time lag phase in period.

Abstract

The experiment was deal with propagat cell line from encephalon and retina of grouper Chromileptes altivelis. The cell was isolate propagation of method described following by Chen et al.(1982) from carp and crussian carp fin. The cell line from enchepalon and retina of grouper grew very fast and heavy in medium L-1S.The cell line were then used for viral replication giving, isolated virus with high pathogenicity indicated by mortal effect in short time after administration. The preservation of cell line is in 10% glycerol under -80°C for one year. Thawing will be done every 3 months to ensure the viability of the cell.

Abstract

The research was subjected to identify 26 endophytic and phyllosphere bacteria isolated from 4 rice varieties using sequencing method of 16S rDNA.The sequences were then aligned with reference sequence from Gen-Bank data library by BLAST program from NCBI to find the most related identity of the isolates analysed. The sequencing analysis revealed that some isolates have a high identity to Staphylococcus and Serratia (5 isolates each), Bacillus (4 isolates), Microbacteria (3 isolates), Pseudomonas (2isolates) and Klebsiella, Acidovorak, Bulkholderiaceae, Agrobacterium and Shewanella (1 isolate each). No specific isolate dominated in both endophytic and phyllosphere group, nor host-specific isolate found in the four rice varieties.

Abstract

Fruit growth and development occurred in plant for producing the offspring. Pollination and fertilization will form embryo and seed, which produce and supply plant growth regulators (PGR) for fruit growing. The role of seed can be exchanged with supply of POR exogenously and produce seedless fruit. Seedless fruit could be produced from triploid plant by crossing between tetraploid x diploid plants, in vitro culture of endosperm/irradiated pollen and genetic engineering or treated diploid plant by using PGR, pollen stress, chemical agent/ antibiotic and environment/ plant control. The benefit of triploid plant is quicker growth and produce of seedless fruits directly, whereas diploid plant could produce seedless fruits after fruit induction with special treatment. The other advantages of seedless fruit on certain plants are increased yield, fruit size/ weight, carotene and total sugar contents, decreased harvest period, yield fluctuation and blossom-end rot. Seedless fruit is preferential of consumer and give a higher price than the seeded fruit. Recently, seedless fruits of tomato, grape, citrus, cucumber and watermelon have been produced in commercial industries
