Abstract

Virus was isolated from infected koi Cyprinus carpio koi with KHV and confirmed by polymerase chain reaction (PCR) and transmission electron microscope (TEM). Fish that were naturally living near koi pond such as catfish Clarias batrachus, nila Oreochromis niloticus, koki Carrasius auratus, and komet Macropinna microstoma did not infected after the cohabitation test. Based on the results KHV was the agent responsible for mortality of million cultured koi in East Java Province Indonesia. Absolute mortality was occurred in koi between 48-72 h post infection, while other cyprinids family was not caused in mortality. Other fresh water fish such as catfish Clarias batrachus, nila Oreochromis niloticus, koki Carrasius auratus, and komet Macropinna microstoma was not infected. Virus-like particles as found in gill ofinfected fish with KHV.

Abstract

The experiment was deal with propagat cell line from encephalon and retina of grouper Chromileptes altivelis.The cell was isolate propagation of method described following by Chen et al.(1982) from carp and crussian carp fin. The cell line from enchepalon and retina of grouper grew very fast and heavy in medium L-1S.The cell line were then used for viral replication giving,isolated virus with high pathogenicity indicated by mortal effect in short time after administration. The preservation of cell line is in 10% glycerol under -80°C for one year. Thawing will be done every 3 months to ensure the viability of the cell.

Abstract

Indonesia shrimp products are often rejected by foreign countries due to high antibiotic content, whereas a few years ago Japan and European countries are set 0.05 ppb permitted to consume by the people, but now the restriction will be smaller is 0.01 ppb. Some farmer was stopped to use antibiotics but the residues still found in fish flesh. Possible origins antibiotics are environment because are antibiotic residues still exist in the mud, water organism which lived in pond. Samples originating from shrimp, mud, water and other biota will be examined according to the method of Thomson (1966) of the Pfizer Corporation and the Association of Official Analytical Chemist (Horwitz, 1980). The purpose of this study was to determine the origin of antibiotic present in the cultivation or farming system that does not use antibiotics in shrimp production.: a. Antibiotics obtained from ponds in the Tuban area is 1.8 ppm (mud), 0.8 ppm (water) and 2.1 (biota). b. a. Antibiotics obtained from ponds in Gresik were 0.6 ppm (mud), 0.3 ppm (water) and 1.2 (biota). c. Antibiotics obtained from ponds in the area of Situbondo are 0.3 ppm (mud), 0.1 ppm (water) and 0.6 (biota). d. a. Antibiotics obtained from ponds in the area are 0.3 ppm Probolinggo (mud), 0.2 ppm (water) and 0.6 ppm (biota).

Abstract

The vibriosis in hatchery and farm easily recognized by the luminous the bacteria present in the water and shrimp itself. The Vibrio sp. used in this experiment were isolated from shrimp and rearing water and the LD50 of V.parahaemolyticus is 10 2'6 and V alginolyticus is 102.5 CFU/fish. The best results obtained from 10 mg/ml mixed of both V.parahaemolyticus and V alginolyticus. The protection of OMP vaccinated fishes was relatively low, the reason could be the OMP contained the low antigenic substance. The SR of fish vaccinated FKC and ICC is 65-70 percent 70-75 percent. Even though it was not proved that FKC and ICC contents more antigenic substance, the FKC and ICC have the higher protein than OMP in measurement of protein and electrophoresis.

Abstract

The rod-shape virus particles were found in the hyperthrophied nucleus and cytoplasm of diseased white shrimp Penaeus merguiensis naturally infected by White spot syndrome (WSBV). In natural infection cumulative mortality of shrimp were very high, 95% of population were dead in 3-7 days, and 5% of shrimp population survived. The disease was outbreak in intensive culture farms used the closed circulation sea water system. Others crustacean such as crabs and bentatos were not infected by WSBV at time of sampling. The virus particle were rod-shape ranging from 78 ± 10 nm in diameter and 280 ± 10 nm in length. The pathogenic bacteria mainly dominated by genus Vibrio sp were isolated from shrimp.

Abstract

Penyebab kematian lou han Cichlasoma synspilum adalah Aeromonas hydrophila dan Escherichia coli, dengan ciri adanya pendarahan dan luka pada sirip dada. E. coli kemungkinan besar tidak menyebabkan kematian ikan sebab sampai sekarang belum pernah ada laporan bahwa bakteri tersebut menyebabkan sakit pada ikan di mana pun. Gejala klinis utama yang terlihat dari luar adalah pendarahan di sirip dada dan menimbulkan luka yang cukup dalam. Cairan ascites menyebabkan membengkaknya perut, pembengkakan liver, dan kosongnya usus dari ikan yang sakit. Gambaran histopatologi menunjukkan nekrosis dan adanya vacuole pada hepatocyte. Sedangkan pada usus adalah rusaknya mucosal ephitelium, lamina propia stratum compactum, stratum granulosum dan muscularis. Ginjal mengalami kerusakan berat, hampir semua organela ginjal berantakan. Ikan ini dipelihara di aquarium sehingga menurunnya kualitas air atau kontaminasi lewat pakan dan penanganan yang buruk diduga sebagai portal of entry dari penyakit.

Abstract

The extra cellular product (ECP) was rapidly induced leucocyte number compared to Edwardseilla tarda NUF251 in eel blood. The reason could be live cells need time to multiplied in eel body whereas the ECP composed of high protein therefore induce the leucocyte production in short time. The number of leucocyte in eel blood were not different between inactivated ECP and control eels. Although the leucocyte number increased gradually in live cells injected eel but have not correlation to increasing phagocytosis index (PI) in vivo phagocytosis. The PI and phagocytic rate (PR) of in vivo phagocytosis relatively constant from 24-96 h post injection.

Abstract

Tuna is one of the main export commodities from Indonesia. Indian Ocean has the potential of large pelagic fish resources among them include tuna amounting to 386.260 tonnes per year with a production of 188.280 tons per year and the utilization rate of 48.74%. The purpose of this field practice is to acquire additional knowledge, experience and skills in the process of canning tuna with high temperatures in the PT. Aneka Tuna Indonesia, Pasuruan. This field practice was held in PT. Aneka Tuna Indonesia, Jl. Surabaya-Malang Km. 38 Desa Gempol, Kecamatan Gempol, Kabupaten Pasuruan, East Java at January 18th until February 19th, 2015. the practice used descriptive method and took data with primary and secondary data. The data retrieved from observation, interview, active participation, and literature review. Products canned tuna was originally derived from the receipt of raw materials tuna, then do the grouping size of each type of fish, included cold storage, wash or do thawing, cutting or cutting, then cooked or cooking, done showering or fish cooling, after that is done pre cleaning and cleaning, do metal detecting, and subsequent filling of meat, charging medium, closing cans or seaming, sterilization, incubated (stored in warehouses), performed labeling and packaging, and finally collected into containers or stuffing.
