Abstract

Menurunnya populasi ikan soro di alam akibat kerusakan lingkungan habitat dan penangkapan berlebih merupakan ancaman bagi kelestarian ikan ini. Pemijahan buatan telah dapat dilakukan dengan rangsangan hormon, namun hasil yang diperoleh masih perlu disempurnakan. Tujuan penelitian ini adalah untuk mendapatkan kombinasi jenis hormon yang tepat, dosis terbaik dan masa laten tercepat terhadap keberhasilan ovulasi dan pemijahan ikan soro. Perlakuan yang diberikan adalah Ovaprim 0,5 mL/kg Induk (O1), kombinasi Ovaprim + hCG (O2), Ovaprim+AI (O3), dan AI + Oxytocin (O4), Hasil yang diperoleh memperlihatkan bahwa perlakuan O4, (kombinasi antara AI dengan Oxytocin) memiliki waktu laten untuk merangsang ovulasi yang tercepat (17,5± 0,52 jam) dibandingkan perlakuan lainnya. Secara umum derajat pembuahan perlakuan O4 (AI + Oxytocin) lebih tinggi dibandingkan dengan perlakuan lain yaitu 96,60 ± 1,00%. Demikian halnya dengan derajat penetasan sebesar 81,05±1,77% dan tingkat kelangsungan hidup larva yaitu 98,88 ± 1,37 %. Hasil yang diperoleh memberikan indikasi bahwa O4 (AI + Oxytocin) merupakan kombinasi hormon terbaik yang dapat diinduksi untuk ovulasi dan pemijahan semi alami (tanpa "stripping") pada soro.

Abstract

The purpose of this study was to evaluate the effect of water in oil in water (w/o/w) emulsion containing 17a-methyltestosterone on gonad development of climbing perch. The results showed administration of 200 ug/kg 17o-methyltestosterone in w/o/w emulsion enhanced gonad development of climbing perch. In this treatment, testosterone concentration increased but estradiol-17p concentration decreased. Both hormones ranged 10,55-15,13 ng/dl and 0.96-5.66 ng/dl, respectively. This treatment also accelerated the development of egg diameter which were eggs e" 0.20 mm became 71.00% after 60 days, and development of egg nucleus position into germinal vesicle breakdown (GVBD) in day 60 was about 17.50%.

Abstract

The conversion of androgen to estrogen is catalyzed by an enzyme, cytochrome P-450 aromatase (P-450arom). The P-450arom cDNA fragment was cloned from brain and ovary of the primitive teleost, Japanese eel. The p-450 arom cDNA fragment (1.1 kb) from brain and ovary of the Japanese eel are identical. The deduced amino acid from P-450 arom cDNA in Japanese eel have conserved domains for common putative functional properties of P-450 super family. P-450arom mRNA was predominantly present in the brain and pituitary, weak in the ovary and none in the other tissues. Key Word : Aromatase. Fragmen cDNA P-450 arom telah diklon dari otak dan ovari ikan teleos primitif, sidat Jepang. Fragmen cDNA P-45 arom (1,1 kb) dari otak dan ovari sangat identik. Asam amino deduksi dari cDNA P-450arom memiliki daerah konservatif untuk sifat-sifat fungsional dari super famili P-450arom. Abstract

In oviparous animals, the qualities of offspring are determined by the materials deposited in the developing oocytes that are synthesized and produced by the hepatocytes under the control of estrogen.  This study was designed to evaluate the effect of turmeric powder supplementation on liver performance to support the reproduction performances of catfish (Pangasianodon hypophthalmus). The study used 60 catfish with average body weight of ± 2 kg. The experimental catfish were assigned into a completely randomized design with 4 treatments and each treatment consisted of 15 replications. The treatment was the dose of turmeric powder supplementation in the commercial feed i.e., 0, 240, 480, and 960 mg/100 g feed.  The experimental catfish were fed two times a day in the morning and in the evening as much as 3% of body weight. The results showed that turmeric powder supplementation at a dose of 240 mg/100 g feed could inhibit liver cell destruction in the catfish as was indicated by the increase in body weight, liver weight, and DNA concentrations of liver, as well as the decrease in the liver malondialdehyde, and serum glutamic piruvic transaminase, serum glutamic oxaloacetic transaminase concentrations. Turmeric powder supplementation can be used in oviparous animals and fish to increase liver function and capacity to synthesize vitellogenin to be deposited in the developing follicles during reproduction phase.

Abstract

Dried Ulva reticulata has been proven as a feeding stimulant for the collector sea urchins (Tripneustes gratilla). The purposes of this study were to determine the optimal dose of dried U. reticulata as a feeding stimulant into artificial feed and its effect on the gonad condition of the collector sea urchins, T. gratilla. Previously, starved sea urchins (ca. 66 mm diameter, n=7) was held in 80-l aquarium volume with flow through seawater system (6.25 l/hour inlet and outlet). Sea urchins were fed with diet containing 0, 10, 20, or 30% of dried U. reticulata (designated control, U-10, U-20, and U-30 respectively) ad libitum for 38 days. Each treatment was given 3 replicates. Survival rate, feed consumption, and gonad conditions (gonad index, biochemical composition of the gonad, gonad histology, and egg diameter) parameters were quantified and compared to sea urchins fed with the mixture of fresh macroalgae (Gracilaria lichenoides and Padina) and seagrass (Enhalus acoroides). The results showed that the addition of dried U. reticulata into artificial feed by 10% from feed weight (U-10) was the optimal dose recommended. Sea urchins treated U - 10 diet resulted in gonad index and egg diameter (5.02 % and 82.66 µm each), were higher than other diet treatments. However, this diet treatment was not able to homogenize the reproductive phase composition of T. gratilla. Keywords: Ulva reticulata, Feeding Stimulant, Tripneustes gratilla, Gonad Condition

Abstract

Eel is a consumable fish that has a unique taste and is rich in vitamins A, B1, B2, B6, C, D, protein albumin, DHA and EPA which is better known for omega-3, as well as its mineral content. This research aimed to determine the protein and steroid content and tissue structure in various parts of the body in eel fish (Anguilla bicolor bicolor). Eels are analyzed on three different body parts covering the front, central, and back. The protein profile was identified by the SDS-PAGE and Bradford methods, steroids by extraction method, tissue structure by the microscopic method. SDS-PAGE analyzed on eel fish meat has a protein molecular weight ranging from 9.825–199.196 kDa. Dissolved protein levels at the front, central, back are 0.307±0.02 mg/mL, 0.298±0.00 mg/mL, and 0.349±0.05 mg/mL. Water soluble and salt soluble proteins at the front, central, back are 0.469±0.01 mg/mL, 0.336±0.03 mg/mL, 0.437±0.00 mg/mL and 1.571±0.16 mg/mL, 1.312±0.11 mg/mL, 1.242±0.11 mg/mL. Eel fish contained steroids in ethyl acetate solvents. The structure of meat tissue and eel fish skin contained muscle fiber and stratum compactum layer.

Abstract

The experiment was conducted in order to determine the role of each supplement combination of Spirulina platensis and Curcuma longa in the diet as well as the induction of Oodevto to tinfoil barb maturation and rematuration process. Randomized block design (RBD) with two factors consists of twelve treatments and five replicates of fish individuals was used. The dietary treatment comprises of four type, i.e. control diet, 3% kg-1 fish weight of turmeric supplementation in the diet, 3% kg-1 fish weight of spirulina supplementation in the diet; and the combination of turmeric and spirulina with 6% kg-1 supplementation in the diet. Meanwhile, the hormonal factor consists of three levels, i.e. 0.0 ml kg-1 fish weight, 0.25ml kg-1 fish weight (FW) and 0.50 ml kg-1 fish weight. We used sixty female broodstock with initial weight ranged 150-400 g and at stage II of gonad maturation. Fish were fed twice a day at 07.00 am and 05.00 pm at satiation level during 14 weeks. During the rearing period, gonado somatic index, maturation level, maturation period, 17 β estradiol concentration, eggs diameter and fecundity were observed. The result shows that all diet types which was combined with 0.50 ml kg-1 fish weight hormonal induction could improve reproductive performance up to 220%. This result comparatively higher than 0 ml kg-1 and 0.25 ml kg-1 hormonal inductions. Supplementation of 3% turmeric meal in the diet could produce 100% of mature broodstock out of season spawning. Thus, the combination of supplement either spirulina or tumeric with 0.5 ml kg-1 fish weight hormone induction can improve the induction maturation up to 2.2 times faster on the out-of- season spawning. Abstrak Penelitian ini dilakukan dengan tujuan untuk mengevaluasi peran kombinasi Spirulina platensis dan Curcuma longa dalam pakan dengan induksi Oodev terhadap proses pematangan gonad dan pematangan gonad kembali ikan tengadak. Metode yang digunakan adalah rancangan acak kelompok (RAK) dua faktor yang terdiri atas 12 kombinasi perlakuan dan 5 ulangan. Ulangan yang digunakan berupa ulangan individu. Faktor pakan terdiri atas empat jenis pakan yaitu pakan kontrol, pakan ditambah tepung kunyit 3% kg-1 bobot ikan, pakan ditambah tepung spirulina 3% kg-1 bobot ikan, dan kombinasi kunyit dengan tepung spirulina 6% kg-1 bobot ikan. Faktor hormon terdiri atas tiga taraf yaitu: 0,0 mL kg-1 bobot ikan, 0,25 mL kg-1 bobot ikan, dan 0,50 mL kg-1 bobot ikan. Ikan uji yang digunakan sebanyak 60 ekor ikan betina dengan bobot 150-400 g dan sudah mencapai tingkat kematangan gonad II. Ikan diberi pakan dua kali sehari (pagi dan sore) secara at satiation selama 14 minggu. Selama masa pemeliharaan parameter yang diamati meliputi indeks kematangan gonad, tingkat kematangan gonad, waktu pematangan gonad, konsentrasi 17β estradiol, diameter telur, dan fekunditas. Hasil penelitian menunjukkan bahwa semua jenis pakan yang dikombinasi dengan induksi hormon 0,50 mL kg-1 bobot ikan dapat meningkatkan kinerja reproduksi lebih baik (220%) dibandingkan induksi hormon 0,25 dan 0,0 mL kg-1 bobot ikan. Penambahan tepung kunyit 3% dalam pakan dapat menghasilkan induk matang 100% di luar musim pemijahan. Dengan demikian, pemberian kombinasi suplemen baik spirulina atau kunyit dengan induksi hormon 0,5 mL kg-1 bobot ikan dapat menginduksi 2,2 kali lebih cepat di luar musim pemijahan.

Abstract

The objective of the experiment was to describe the influence of estradiol (E2) and methyltestosterone (MT) in combination with hCG and anti-dopamine (AD) on the percentage of female and quality of ovary development of Indonesian short-finned eel (Anguilla bicolor bicolor). Eel with 200±15 g of body weight in the yellow phase injected with a estradiol solution of 3mg mL-1 + hCG 2mg mL-1 + AD 10 mg mL-1 (hEA), MT 3 mg mL-1 + hCG 2 mg mL-1 + AD 10 mg mL-1 (hTA), estradiol 3 mg mL-1 + AD 10 mg mL-1 (EA), MT 3 mg mL-1 + AD 10 mg mL-1 (TA), AD 10 mg mL-1 (A) and 9 mg mL-1 NaCl control (F). Injection frequency was six times with a period of two weeks. After six weeks of experiment, EA and hTA combination hormone treatments were effective to increase the female percentage 72% and 66%, respectively. The best ovary development was found in the hTA treatment i.e. attain the late-vitellogenesis with GSI value of 4.80%. Abstrak Penelitian ini bertujuan untuk menjelaskan pengaruh estradiol (E2) dan metil testosteron (MT) yang dikombinasikan dengan hCG dan anti dopamin (AD) terhadap persentase jenis kelamin betina dan kualitas perkembangan ovari sidat (Anguilla bicolor bicolor). Sidat dengan bobot 200±15g pada faseyellow diinjeksi dengan larutan estradiol 3 mg mL-1 + hCG, 2 mg mL-1 + AD 10 mg mL-1 (hEA), MT 3 mg mL-1 + hCG 2 mg mL-1 + AD 10 mg mL-1 (hTA), estradiol 3 mg mL-1 + AD 10 mg mL-1 (EA), MT 3 mg mL-1 + AD 10 mg mL-1 (TA), AD 10 mg mL-1 (A), dan kontrol 9 mg mL-1 NaCl (F). Frekuensi injeksi sebanyak enam kali dengan periode dua minggu sekali. Pemberian hormon kombinasi dengan perlakuan hEA dan hTA ternyata efektif meningkatkan persentase jenis kelamin betina berturut-turut sebesar 72% dan 66% setelah enam minggu. Perkembangan gonad betina terbaik ditemukan pada perlakuan hTA yaitu mencapai fase vitellogenesis akhir dengan nilai GSI 4,80%

Abstract

Artificial spawning on stripe catfish has generally carried out by stripping because of the absence in reflex of spawning. Mechanical stripping usually caused stress, decreased quality of gametes and seeds and damage on gonad. Induced spawning without stripping could be used for the process of gonad maturation to stimulate the ovulation of the fish which has the difficulties to spawn in its non natural habitat. Induced spawning without stripping by using hormones combi-nation was conducted in the present study. This research proposed to evaluate the injection of hormones combination between aromatase inhibitor (AI) and oxytocin association with ovaprim and PGF2a for stimulating ovulation and spawning without stripping. The completely randomized design of combined hormones consisted of five treatments such as P1(AI,oxytocin), P2 (AI, oxytocin,ovaprim), P3 (AI, oxytocin, ovaprim, PGF2a), P4 (ovaprim) as positive control, and P5 (NaCl) as negative control. Each treatment was performed using five paires of males and females as individual replicate of about 2-6 kg weight. The results showed that the combination of P3 was the most effectively and successfully induced ovulation with naturally spawning without stripping, treatment P4 ovulation with stripping, while the treatment P5 there was not ovulation. The hormones combination caused decreasing of estradiol-17p concentration and testosterone (p<0.05) as the sign of the final maturation. The average time of ovulation was 12.35±4.05 to 15.20±2.25 minutes. The highest number of eggs about 145.865 from the treatment P3 (AI, oxytocin, ovaprim, PGF2a). Abstrak Pemijahan buatan pada ikan patin dilakukan dengan cara pengurutan karena tidak memiliki kemampuan untuk menge-luarkan telur secara alami. Teknik pengurutan berdampak stres pada induk, kualitas gamet menurun, dan gonad menjadi rusak. Proses pematangan gonad dan pemijahan tanpa pengurutan dapat diinduksi secara hormonal untuk membantu ovulasi ikan yang sulit memijah di luar habitatnya. Penelitian ini bertujuan mengevaluasi penggunaan hormon penghambat aromatase (aromatase inhibitor - AI) dan oksitosin serta kombinasi hormon untuk merangsang ovulasi dan pemijahan pada ikan patin tanpa pengurutan(semi alami). Perlakuan kombinasi hormon terdiri atas P1(AI+Oksitosin), P2 (AI+Oksitosin+Ovaprim), P3 (AI + Oksitosin + Ovaprim + Pgf2a), P4 (ovaprim) sebagai kontrol positif, dan P5 (NaCl) sebagai kontrol negatif. Pada setiap perlakuan digunakan lima induk ikan patin jantan dan lima induk ikan patin betina sebagai ulangan individu dengan bobot berkisar 2-5 kg. Perlakuan diberikan satu kali dengan cara penyuntikan hormon ke dalam jaringan pada bagian otot dibawah sirip punggung (intramuscular). Hasil penelitian menunjukkan perlakuan P3 berhasil menginduksi ikan untuk ovulasi dan memijah tanpa pengurutan, sedangkan pada perlakuan P4 ikan memijah dengan cara pengurutan, dan pada P5 ikan tidak ovulasi dan tidak memijah. Perlakuan kombinasi hormon menyebabkan konsentrasi estradiol-17p dan testosteron plasma menurun yang menunjukkan tahap pematangan akhir. Lama waktu ovulasi tidak berbeda nyata yaitu berkisar antara 12,35±4,05 sampai 15,20±2,25 jam, sedangkan jumlah telur yang diovulasikan tertinggi adalah 145.865butir pada perlakuan kombinasi hormon P3 (AI + Oksitosin + Ovaprim + Pgf2a).
