Abstract

The research was undertaken in two experiments. The first experiment was carried out to find out the germination capacity and vigor of sorghum seeds that consisted of four varieties namely Rio, Mandau, Sangkur and Keris. The second experiment was to determine the response of two sorghum varieties which had good germination capacities and so vigorous from the first experiment by osmoconditioning treatments. The growth media is saline soil with NaCl content 8.61% and pH 8.2. The osmoconditioning treatments were soaking seeds in each Na,SO4 0.2 M, NH4C1 0.2 M, KNO3 0.2 M solution for 48 hours and untreated sorghum seeds served as the control. The results showed that there was no interaction between varieties of sorghum seeds and osmoconditioning treatments on germination percentage but osmoconditioning treatments with Na,SO4 0.2 M and NH4C1 0.2 M could promote germination percentage of sorghum seeds on saline condition.

Abstract

Krisis ekonomi yang terjadi di Indonesia terutama bersumber dari dalam negeri kita sendiri.Pemerintah orde baru, terutama sejak era konglomerasi (1987-1994), terlalu mengabaikan berkembangnya kesenjangan.Pertumbuhan ekonomi ternyata hanya sebuah fatamorgana (Mubyarto,2000). Krisis ekonomi 1997-1998 dan keadaan politik yang tidak menentu, mengakibatkan nilai mata uang rupiah dalam tempo yang amat singkat, menurun drastis. Indonesia pun langsung terpuruk. Kegiatan ekonomi tersendat karena sebagian besarnya sangat tergantung kepada bahan baku impor. Banyak perusahaan yang gulung tikar dan banyak buruh terpaksa di PHK. Jumlah penganggur dan jumlah penduduk miskin meningkat dengan tajam.

Abstract

To increase domestic and international demand of Kencur (Kaempferia galanga L.) makes this plant potentially develop.Is traditionally used to keep the body warm, as analgetic and expectorant. In the attemp of providing adequately and qualitatively uniformed supply, in vitro experiment has been conducted at BB-Biogen (Indonesian Center for Agricultural Biotechnology and Genetik Resources Research and Development).The selected rhizomes was used as explant. The experiment was orthogonally arranged consisting of MS vitamin and B5, and BA ( 0, 3 and 5 mg/1) and thidiazuron 0,1 mg/1. This experiment comprised three activities, they were shoot initiation, shoot multiplication and acclimatization. The result showed that MS + BA 3 mg/1 + thidiazuron 0,2 mg/1 could induce shoot formation. From the applied media, it was shown that the addition of MS vitamin at the MS basic media and BA 3 and 5 mg/1 added with thidiazuron could result the most optimum shoot, leaves and roots and was not significantly different from the addition of B5 vitamin at basic media of MS + BA 3 and 5 mg/1, 6.9 shoot was averagely produced in this media. The shoot could generate such an adequate number of root that it could be directly acclimatized. The acclimatized plantlet in the green house uses the mixture of soil and manure with the ratio of 1:1 can optimally grow.

Abstract

Protoplasts regeneration was studied on 3 cultivars of eggplant (pupel, green, ang white fruits) and the regenerant were tested for their resistance to bacterial wilt. The researh was conducted in different steps, including protoplast isolation, protoplast purification, protoplast culture, cell suspention dilution, shoot regeneration, plantlet aclimatization, and testing for the resistant to bacterial wilt. Observations were made for protoplast number of each cultivar, protoplast cell wal formation, cell division, number of cell colony, microcalli, and shoot, percent of plantlet survival, in acclimatization, and the resistance of regenerants to bacterial wilt. Data was analized by calculating the average, percentage, and standart deviation. Results show that the combination of 0.5% cellulase Onozuka RS, 0.5% macerozyme R-10, 0.05% MES, and 9,1% mannitol in CPW solution was able to isolateds alot of sprotoplast (10 ) in each cultivars. The first culture incubation with out lighty in both (KM8P and VKM) media and reach which 0,2 mg/1 2,4-D, 0,5 mg/1 zeathin, 1 mg/1 IAA, promoted the formation of cell wall and cell division developed to microcalli. Regeneration media of MS, applied with vitamin MW, 0,1 mg/1 NAA, and 2 mg/1 zeathine produced 13 shoots.. From 21 regenerants acclimatized only 15 regenerants survive. There were differences among the regenerants in their resistance to bacterial wilt,3 regenerants resistant, 2 regenerant recovery (tolerant) and 10 regenerants were susceptibel.

Abstract

Kasepuhan ethnic group surrounding Mt. Halimun National Park uses plants traditionally. Based on our research in May 2004 in Cisungsang, Lebak District, Banten province, there are 113 species of plants belongs to 41 families used in many ways. There are 20 local rice varieties, 30 species as vegetables, 30 species as medicinal properties, 28 species as food, 2 species as ornamental plants, 2 species as handycraff and 1 species as for dye. The wisdom in the traditional knowledge of this ethnic group is be discussed.

Abstract

In order to prevent aflatoxin contamination during the production of kecap, an Indonesian soy sauce, a white-spored mutant K-IA strain induced from an aflatoxin-negative koji mold was applied to koji making process of kecap. While kecap koji making process took 9 days by conventional method without inoculum, the use of K-IA strain as a starter shortened the process to 3 days at room temperature. The koji prepared with this starter contained 2.3 x 107 cfu/g of Aspergillus sp. K-IA as the dominant speciesand some contaminants from the environment such as other species of Aspergillus (2.0 x 10 cfu/g) and Mucorales (1.4 x 10s cfu/g). The mash prepared with kecap koji inoculated with this starter contained higher formol nitrogen (FN) and water-soluble nitrogen (WN) than those prepared with koji inoculated without any starter.These results demonstrated that the use of this starter did not only contribute to the prevention of aflatoxin contamination but also improved the conventional kecap fermentation process.

Abstract

One of the interesting topic in Plant-Water Relations is the synthesis and accumulation of solutes by plant cells and within thosecells. This paper is to make a short review upon this topic. It shows the relationship of water potential components of plant cells (a - total water potential, 0n - osmotic potential and 0p-turgor pressure), in accordance with environmental fluctuations (especially water stress), osmotic regulation (osmotic adjustment) and the role of solute synthesis and accumulation in plant cells when drought develop. Proline (among sugars and organic acids), dominates most of the available reports as both an osmotic constituent/ osmotic agent and a compatible solute. Such a reviewed topic that still less available in Indonesian.

Abstract

Phenotypic variance derived from banana [Musa paradisiaca) mutant lines of cv. Barangan were carried out in nursery level which were conducted at CRDIRT-Green House, Pasar Jumat, Jakarta. Several phenotypic performances of mutant lines such as leaf and pseudostem, petiole and leaf length, plant height, leaf width were observed. Number of 150 plants, were then divided into 11 groups based on performance of leaf, pseudostem, and petiole length. Results indicated that 26% was showed for normal performance,while 6,7% was showed for abnormal lamina with normal petiole, 11,3% was showed for abnormal lamina and petiole, 28,7% was given for stunted plant, 3,3% with chocking lamina, 1,3% was showed for abnormal petiole, 2,0% with light green leaf and abnormal petiole, 6,0% was given for reddish pseudostem, 0,7% with cigar leaf, 1,3% was showed for discoloration leaf and 2,7% was showed for elongation leaf and abnormal petiole. From the result of this experiment can be concluded that gamma irradiation with dose 15 Gy produced high phenotypic variance of cv. Barangan.
