Abstract

This study aimed to determine the effect of 0.2 mT magnetic field exposure on metals (Al, Pb, Cd and Cu) containing media, to proteolytic index of Bacillus sp., protease activity and cell morphology of Bacillus sp. This study consisted of three stages. The first stage was a proteolytic test on solid media containing milk. The second stage was the production of protease enzymes in liquid media. The third stage was Scanning Electron Microscopy (SEM) analysis on a culture treatment known as protease enzyme activity. The results showed that the largest proteolytic index on CuCl2 metal ion content of magnetic 0.2 mT magnetic field for 10 minutes increased the value of the largest proteolytic index was 4.33. While in the ion solution containing CdCl2, Bacillus sp. culture did not grow. In the production of enzymes in the liquid medium, the highest enzyme activity (0.140 U/ ml) was produced on a medium containing AlCl3 and exposed to magnetic fields. The SEM analysis also proved that the supplementation of AlCl3 increased cell length by 2.38 and 2.78 times longer than control without magnetic field and for magnet field, respectivelly.

Abstract

Srikandi Tilapia is one of the superior hybridized varieties between black tilapia (Oreochromis niloticus) and blue tilapia (Oreochromis aureus). This fish has fast growth and high salinity tolerance. In its development, efforts are needed to improve and improve its genetic quality. Spawning srikandi tilapia using the parent of male blue tilapia F2 (family selection) is expected to improve performance better than the previous generation.The purpose of this study was to evaluate the growth performance and heterosis value of Srikandi tilapia produced from the extraction of female Nirwana strain tilapia with male blue tilapia F2. Hatchery activities were carried out in freshwater by mass spawning method on the 25 m2 pool. Enlargement activities were carried out at brackish water pond with salinity 25–40 g / L, using net measuring 3×5×1 m3 for 120 days. The stocking density used is 15 fishes / m2. The results of this study indicate that at the final test, the hybridization of Nw ♀x Au (F2)♂ has the best performance with the average of long gain16.60 ± 0.41cm, weight gain 215.33 ± 12.19 g and survival rate 78.50 ± 02.12. Evaluation of heterosis values on the characters of length, weight, survival, and biomass showed a positive value both on the hybridization of Nw ♀x Au (F2)♂ and Au (F2)♀x NW♂. The highest high parent heterosis value for blue tilapia broodstock is shown by population Nw ♀x Au (F2)♂ with values in each character of 6.96% (length), 26.06% (weight), 2.63% (survival), 28.57% (biomass).

Abstract

Endophytic fungus Fusarium sp. CSP-4 was isolated from the rhizome of Curcuma sumatrana Miq. The ethyl acetate extract of CSP-4 was fractionated by chromatographic column with silica as a stationary phase. The fractions were tested for antibacterial activity against S.aureus and E.coli by Thin Layer Chromatography (TLC)- bioautography. Determination of Minimum Inhibitory Concentration (MIC) was carried out by the microdilution method on a 96-microwell plate with the concentrations from 2 to 256 µg/ml. The results showed that fraction 15 and 11 were able to inhibit bacterial growth with the MIC values of 256 and> 256 μg / mL against S.aureus and>256 μg / mL against E.coli, respectively.The antibacterial activity of fractions 15 and 11 was weaker compared to the positive control of chloramphenicol with the MIC value of 4 µg/mL against S.aureus and E.coli. Based on the results obtained showed that the fractions of the methanol extract of endophytic fungus Fusarium sp. CSP-4 have weak antibacterial activity.

Abstract

Odontochilus uniflorus (Blume) H.Ǣ. Pedersen &Ormerod, is a new addition to Flora of Java based on three specimen collected from Mt. Halimun-Salak National Parks. This species was previously known only from Thailand, Malaysia, and Sumatra. Hence, this new finding expands the range of distribution of the species of O. uniflorus as well as adding information for orchids of Java. Identification key of the species of Odontochilus in Java is also presented in this paper.

Abstract

Outbreed may increase genetic diversity and produce better offspring. Meanwhile, genetic diversity can be inferred from phenotypes variability. This study aimed to analyze the diversity of phenotypic characters from four populations of tinfoil barb outbreed, namely Java (J), Kalimantan (K), ♀ Java × ♂ Kalimantan (JK) and ♀ Kalimantan × ♂ Java (KJ).Analysis of the phenotypic diversity was calculated based on truss morphometric on 30 individuals sampled from each populations. The growth and survival rate were measured for 40 days reared. The study was conducted experimentally using a completely randomized design (CDR) with three replications.Rearing four population of tinfoil barb, beginning with acclimatization for two weeks and was continued reared using 12 aquaria40x30x30 cm3. Each aquaria was filled with 10 individual seeds/20 L-1 water with the size of 18,66±1,34 g weight and 8,28±0,34 cm of length. Feeding was done three times a day on ad-libitum using commercial pellet with protein content of31˗33%. The result of the canonical function analysis of truss morfometrik thetinfoil barb on 21 characteris indicated that the individual morphological characterof population of the tinfoil barb outbreed (JK and KJ) interrelated with each other.The highest sharing component value of inter population was obtained on the fish from outbreed of KJ and JK (13,3%), while the highest of intra population was obtained of K (96,7%) and the lowest of JK (86,7%). The highest survival rate (100%) was recorded on the JK population and significantly different (P>0,05) with survival rate of 90% observed in the J population.

Abstract

Orangutans are solitary animals from the Pongidae family. There are three sub-species of Bornean orangutans, i.e. Pongo pygmaeus wurmbii, P. pygmaeus pygmaeus and P. pygmaeus morio, all of which are highly endangered. Orangutans carry out several daily activities such as eating, moving, resting, and nesting in the tree canopy. However, it has yet remains unknown on how their behaviours vary with tree height. This study aimed to determine the frequency of presence, daily activities of orangutans, and vertical space utilization in orangutan activities in the natural habitat of the Panti Branch Research Station, Gunung Palung National Park. This study used the Focal Animal Sampling method with a two minute time interval for ± 12 hours and was studied for over six months. The focal orangutans whose activities were recorded during the study consisted of 12 individuals divided into 3 age groups, namely the adult, adolescent, and juvenile age groups. The results showed that the daily activity of orangutan was observed on 36 species of trees with the greatest frequency found on Garcinia sp. and Syzigium sp. accounted for33% each. The female orangutans were found to be more dominant in feeding activities (3.93 per day), while male orangutans spent more time for resting activities (4.56 hours per day). The utilization of vertical space in young male orangutans took place in tall trees and eating activities occurred at an altitude of 33.6 m. On average, teenage female orangutans choose tree to make their nests in trees 30 m high.

Abstract

The SARS-CoV-2 or COVID-19 pandemic has reached a new height with an unprecedented infection rate and mortality post-world war II history. However, there is no particular designed drug for COVID-19 up to this point. Thus, there exist three strategies for COVID-19 drug design; drug repurposing option, herbal medicine development, and transcriptomics-based drug lead. As the most underutilized option, transcriptomics-based drug lead could be leveraged to deal with SARS-CoV-2 infection. One of the main methods to block the SARS-CoV-2 infection is to inhibit the RNA polymerase enzyme that is responsible to the viral replication. In this regard, the objective of the strategy is to design the anti-sense siRNA drug and lead to inhibit the mRNA of the RNA Polymerase Enzyme (RdRp) gene that encodes the viral RNA Polymerase of the SARS-CoV-2. The Computer-Aided Drug Design (CADD)-based method was leveraged with sequence retrieval of 24 RdRp gene sequences, multiple sequence alignment, phylogenetic tree reconstruction, 2D/3D RNA structure predictions, and RNA-RNA docking. Both the RNAalifold conserved structure from the RdRp genes and the RNAfold structure of the siRNA for blocking the conserved structure are negative or less than 0 kcal/mol. The predicted RNA docking occurred with the best RMSD score of 22.53 Å, which is beyond the accepted threshold of 10-20 Å. Based on the findings, the 2D/3D structures of both the siRNA and mRNA could be elucidated, and the docking between them is feasible. However, this finding should be elucidated in the wet laboratory setting for the final lead validation.

Abstract

Plants from Ixora genus exhibit a variety of pharmacological activities including antioxidant, antibacterial, and antitumor activities. This plants may contain many interesting bioactive compounds, especially phenolics and terpenoids groups. Ixora cumingiana is one of the Ixora species whose pharmacological effect has not been explored comprehensively. This study aimed to evaluate the antibacterial and antioxidant activity of eight extracts from bark and leaf of I. cumingiana. The plant samples were successively extracted using n-hexane, dichloromethane, ethyl acetate and methanol. The antibacterial evaluation was carried out against Staphylococcus aureus and the antioxidant activity was evaluated using a radical scavenging assay against 1,1-diphenyl-2-picrylhydrazyl (DPPH). Initial activity screening was performed using thin layer chromatography-bioautography followed by the determination of minimum inhibitory concentration (MIC) and the determination of antioxidant activity index (AAI) using microdilution technique. The dichloromethane extract of the bark as well as the n-hexane and dichloromethane extract of the leaf of I. cumingiana exhibited moderate antibacterial effect with MIC value of 128, 128, and 256 µg/ml, respectively. The methanol extract from the bark displayed a strong antioxidant activity (AAI = 1.5±0.13) and possessed the highest total phenolic content (43±0.91 mg AGE/ g extract). These experimental results showed that I. cumingiana is potential to be developed as an antioxidant agent rather than as an antibacterial agent.

Abstract

The Bonokeling society is a part of the Javanese indigenous groups who still maintain and practise the local wisdom in their daily lives. Socio-cultural research concerning the Bonokeling religion has been reported and provided useful information to the society, while the ethnoecological studies about Bonokeling’s local knowledge related to the environment have not been reported. The research aimed to provide a comprehensive study about Bonokeling society on the management and utilization of natural resource and environment. This study used qualitative and quantitative approaches through participatory survey, open-ended interviews, and structured interviews with questionnaires. The results showed that the community recognized five environmental categorizations based on functions, namely settlements, home gardens, agricultural land, sacred sites, and irrigation sources. The plant diversity found were consisted of 288 species with 12 categories of uses. Pari is a species of cultivated plant that has the highest cultural importance value. The Bonokeling local knowledge can be scientifically proven has an ecological value. The management and utilization of agricultural land by the Bonokeling society have been carried out not only to optimize the production, but also provide the benefits to the environment. Besides, the management of sacred site has shown that traditional conservation practices have been maintained well for sustainability.

Abstract

Pod shattering is one of the sources of yield losses during soybean cultivation in Indonesia. This research aimed to identify the resistant genotypes to pod shattering and the performance of its agronomic characters. The field study was conducted in the dry land of South Lampung from May to August 2017 using twelve soybean genotypes. The experiment was arranged in a randomized block design with four replicates. The observation of pod shattering was using oven-dry and sun-drying methods. The result showed that a rise temperature of oven increased the pod shattering. The rates of shattering on oven temperature of 30 °C, 40 °C, 50 °C, and 60 °C were of 0, 8.26%, 46.13%, and 51.06%, respectively. The rate of shattering of oven-dry method at 60 °C ranged from 3.62 – 87.23%. On the sun-drying method, the rate of shattering after 15 days treatment was ranged from 0 – 67.36%. The screening for pod shattering by using the oven-dry method has successfully obtained two resistant varieties to pod shattering (Anjasmoro and G511H/Anj-1-3), whereas by using the sun-drying method there were three resistant genotypes (Anjasmoro, G511H/Anj-1-3, G511H/Anjasmoro-1-4). The evaluation of shattering resistance using oven-dry method resulted in a higher pressure than those of by sun-drying method. The Anjasmoro variety and G511H/Anj-1-3  showed consistent resistance to pod shattering by both methods. The G511H/Anj-1-3produced high yield, have large seed size and early days ofmaturity. Thus, the G511H/Anj-1-3was potentially be developed in Indonesiaand other tropical regions. The availability of a high yield and shattering resistant genotype will be beneficial to increase soybean productivity.
