Abstract

Indonesia's plant diversity potentially as natural dyes. A general process in dyeing with natural dyes needed mordanting workmanship on the material to be dyed/stamped. The process of mordanting is done by immersing the material into the metal salts, such as aluminum, iron, tin or chrome. Amid fears of negative impacts caused by dyes and synthetic mordant to health and the environment, people begin to look back the use of natural dyes. So far, the material used for mordanting is alum; though Loba tree (Symplocos fasciculata Zoll.) is one type of plant that can be useful as mordant but today not many people knows the role of this plant. This paper aims to determine the S. fasciculata role and its use as mordant for natural dye textiles. The method used was interviews with parties concerned. The results of this research is aknowledge on S. fasciculata role as a mordant as local wisdom in traditional Balinese weaving. Plant parts that may be used are leaves and inner bark. Conservation efforts is eagerly continued by Eka Karya Bali Botanic Gardens-The Indonesian Institute of Science (LIPI). Currently two species (among 250 naturally grow) of Symplocos conserved at Eka Karya Bali Botanic Garden-LIPI.

Abstract

N-acetyl-D-glucosamine (GlcNAc), the monomeric unit of polymer chitin has attracted much attention for their therapeutic activity in osteoarthritis. It is mainly produced by acid hydrolysis of chitin which affect the environmental problem because of its acidic wastes. Therefore, it is need to develop the new method for GlcNAc production. The aim of this experiment is to produce GlcNAc by mean of submerged fermentation of chitin. The preliminary study showed that fermentation of chitin by Aspergillus sp.501 produced higher GlcNAc than that of Saccharomyces sp. It was 1.39 ng/ml and 1.07 ng/ml. Then the Aspergillus sp. 501 was used in optimation of GlcNAc production. The effect of pH and nitrogen course such as bacto peptone, yeast extract, amonium sulfat and urea to GlcNAc production was examined. Then the product of GlcNAc was precipited by vacum evapotated and freeze dryed. The results showed that the highest of production GlcNAc of 2.228 ng/mL was obtained on pH 4 of medium solid state fermentation using urea as Nitrogen source at 10 days incubation.

Abstract

The recent studies were carried out in June-July 1994,And aimed at investigating the distribution of the avifauna in eight main habitat types of Siberut Island. These habitat types were primary dipterocarp forests, primary mixed forests, secondary forests, swamp forests, mangroves, and coastal forests. It appeared that the habitat types tended to determine the bird communities living in the habitats, suggesting that the distribution of bird communities is related to the resources available within the habitats. The results indicate that the conservation measures of avifauna in this island would be better focused on the conservation of their habitats.

Abstract

Microbial groups of entomopathogenic (fungi and bacteria) had been reported causing insect mortality. The aim of the study was to explore and characterized entomopathogenic from various host and locations. Fungal identification at genus and species level was caried out based on conidial morphology, hyphal growth, conidiophore and colony color; whilst for bacterial identification was based on standard Bergey’s manual for determinative bacteria. Sixteen entomopathogenic isolates that consisted of fungal and bacteria have been collected and preserved for further characterization. Of the 16 entomopathogen collected samples, five fungal genera was found i.e. Paecilomyces; Metarhizium, Beauveria, Hirsutella; and Cordyceps. Seven isolates belonging to six fungal isolates, and one bacterial isolate had been identified based upon ITS and 16S rDNA sequences, respectively. We confirmed that 6 fungal isolates belong to species of Paecilomyces reniformis, B. bassiana, M. anisopliae, M. anisopliae var acridum, Hirsutella thomsonii. One isolate of red pigmented bacteria Sm201102 have been identified was belonging to species Seratia marcescence. It was also obtained two fungal isolates from different host (spider and beetle) which confirmed by morphological character belong to Cordyceps sp.

Abstract

Abstract

This study was aimed at studying the effectiveness of alcohol, formaldehyde and their mixture in inhibiting fungal growth. This study contained two step. The first step was fungal isolation from specimen. One isolate was then selected to be inoculated into alcoholat various concentration (15%, 30%, 50% and 70%) with or without formaldehyde mixture. The second was fungal inoculation intoalcohol at various concentration (15%, 30%, 50%, dan 70%) with or without formaldehyde. The effectiveness of these subtances as specimen preservatives for shrimp were studied after six months incubation time. From the fisrt step, only genus group Monascus was isolated from seven specimens(Mabuia multifasciata, Myotis muricola,Chironax melanocephalus,Naya naya sputratrix, Miniopterus schreibersi blepotis, unidentified shrimp, unidentified Coelenterata). From the second step, the best growth of isolate Monascus sp.MM which isolated from the fisrt step was achieved in 70% alcohol. This fungus was not able to grow in both formaldehyde and its mixture with alcohol (1:1, 1:2, 1:3 and 1:4). The results also showed that Monascus growth was observed in alcohol with not sterilized shrimps without inoculation with Monascus sp. MM. It is recommended to apply mixture alcohol 70% with formaldehyde 1% at ratio 4:1 as animal specimen preservative to inhibit fungal growth.

Abstract

Proper stocking density on larval rearing of barred loach (Nemacheilus fasciatus Valenciennes, 1846) is very important in terms of obtaining the best growth and survival rate. This study was aimed to observe the growth and survival rate of barred loach subjected to different larval stocking density. Fish larvae were obtained by artificial spawning from in aquarium. Larvae were stocked on aquariums (50 × 40 × 30 cm) with three stocking density treatments (5, 10, and 15 individuals/L) and three replications. The experimental fish larvae were fed by Artemia and commercial feed (crude protein content: 30%) and feeding frequency twice per day (ad libitum). The length and weight were reassured every 10 days during 70 days of rearing period. Temperature, pH, and dissolved oxygen were measured at each treatment. Data obtained from measurements were analysed to assess the length gain, weight gain, specific growth rate (SGR), biomass gain, and survival rate. The results showed length gain of 3.35 ± 0.01, 2.94 ± 0.10, and 2.82 ± 0.15 cm and weight gain of 0.573 ± 0.023, 0.436 ± 0.018, and 0.417 ± 0.043 g in larval stocking density of 5, 10 and 15 individuals/L, respectively. Specific growth rates and biomass growth were of 6.22 ± 0.41, 5.93 ± 0.21, and 5.68 ± 0.41 %/day and 94.04 ± 5.39, 146.80 ± 3.81, and 167.24 ± 17.48 g in larval stocking density of 5, 10 and 15 individuals/L, respectively. Meanwhile, the survival rates obtained in this study were 89.13 ± 7.25, 93.13 ± 5.13, and 76.50 ± 3.54 % in larval stocking density of 5, 10 and 15 individuals/L, respectively. Increased stocking density has a negative impact on the growth and survival rate of barred loach.

Abstract

Some plant species growing in the contaminated areas, indicated high tolerance and potentially affective in accumulating pollutants. These plants can be utilized as hyper accumulators for cleaning up contaminated sites. This research aims to examine plants that grow under Hg contaminated media and to analyze their potency as hyper accumulators. Two pioneer plant species Enterolobium cyclocarpum (Willd.) Griseb and Centrosema pubescens Bent. that proven tolerant and dominant at contaminated sites of gold mine were examined in this research. The plants were grown in liquid media added with mercury (HgCl2) with different level of concentrations 0 ppm, 10 ppm and 20 ppm. Chelate Ammonium Thiosulphate [(NH4)2S2O3] was added with concentration levels of 0 ppm and 20 ppm. Plant performances, cellular responses, Hg accumulation in the plant were assessed. The results revealed that the growth of both plant species decreased with the increase of Hg contamination level. The presents of chelating agent improved adaptability of the plants, indicated by the increase of biomass production and Hg content in the plants, even at the highest level of Hg concentration (20 ppm). Cellular responses showed at 20 ppm Hg, indicated by the decrease of both the number and the size of epidermis and pith cells. Accumulation of Hg in the plants increased with the increased of Hg concentration in the media. Mercury accumulation in both plant species were more concentrated in the root system rather than the shoot.
