Abstract

Irradiation of ??ray at shoot culture of Kentang hitam (Solanostemon rotundifolius) is effective for mutant production. Kentang hitam is sterile and vegetatively propagated andtherefore its genetic diversity is narrow. Mutation is an alternative way to increase genetic heterogeneity. Irradiation of shoot cultures with different doses followed by culturing of the inoculum (leaf, petiole and internodes) from that irradiated culture on a regeneration medium MS containing 5 mg/l BA and 0.1 mg/l NAA has resulted in curvilinear respon of growth, morphogenesis and plant regeneration. The growth variables increase from 0 to 6 Gy and thendecrease to 25 and plateu or increase a little at 35 Gy and growth death wes observed at 50 Gy. Leaf and petiole inocula were more responsive than the internode. Respon of growth of shootregeneration of 50% were obtained at doses of 10-12.5 Gy. However higher level of doses, such as 25 Gy had also been effective for inducing mutant. Morphological and growth different were observed from growth in tissue culture to the field. Abstract

ABSTRACTHerpetofaunal Community Structure of Ciremai Mountain National Park, WestJava. Distribution and broad habitat associations of the herpetofauna on four limitedsites of Ciremai mountain, west Java, Indonesia were studied. The field surveys wereconducted on Apuy-Arban site from 7 until 21 April 2006 and from 6 until 10 March2007, Linggarjati site from 10 until 15 February 2007 and from 19 May until 2 June2007, Seda forest from 18 until 24 February 2007 and Palutungan site from 26 Februaryuntil 4 March 2007. A variety of methods were employed to sample the herpetofauna. Atotal of 43 amphibians and reptiles taxa were recorded, comprising 16 frogs, 18 lizardsand 9 snakes. Two of them were endemic to Java, they are Huia masonii and Microhylaachatina. Unidentified and possibly undescribed species of unusual specimen referred tothe genus Elapoidis. From this study, three habitat were clustered and divided into twomain habitat zones for herpetofauna.Key words: amphibians, reptiles, gunung Ciremai.

Abstract

ABSTRACTGenetic variation and relatedness of two new species of star fruit (Averrhoa leucopetalaRugayah et Sunarti sp nov and A. dolichorpa Rugayah et Sunarti sp nov., Oxalidaceae)based on Random Amplified Polymorphic DNA. Two wild species of Averrhoa from Papuaand Gorontalo respectively has recently been described. These two species were previouslytreated as ‘intermediate species’ between A. carambola and A. blimbi on the basis ofmorphological characters. This present study aimed to assess genetic variation and geneticrelatedness of the two species compared to their relatives (A. carambola and A. blimbi) byusing Random Amplified Polymorphic DNA (RAPD). Five RAPD primers (OPA 9E, OPA 13,OPB 7, OPB 18 dan OPN 12) were used to amplify total DNA genom and produced 31 bands towhich 90.32% were polymorphic. These bands were ranging in size from 300-1700 bp. DNAfingeprints for each species was indicated by differences in RAPD profiles resulted fromamplification of five primers. Clustering analysis was performed based on RAPD profilesusing the UPGMA method. The genetic similarity range between 0.25-1.00 indicating widerange of genetic variations observed. Results also indicated that the two species weregenetically distant from A. carambola and A. blimbi, thus supported the recent morphologicaltreatment.Key words: Averrhoa, RAPD profiles, genetic variations.

Abstract

ABSTRACTPotency of Hypertolerance and Metal Absorption of Some Plant Species Under GoldMine Waste. Degraded mined land is characterized by extreme of alkalinity or acidity, highconcentration of soluble salt and high concentration of heavy metals. Contamination of minedsoil and water affects not only on agriculture system but also on food chains andepidemiological problems. As soil metal can not be biodegraded, remediation of soil heavymetal risk has been a difficult and expensive goal. Remediation of hazardous soil is requiredto reverse the risk to humans and the environment. Recently there are several differentstrategies available for the clean up and restoration of contaminat6d soils i.e conventionalwhich is mainly engineering-base and phytoremediation which is a biological-base method.Phytoremediation is defined as: clean up of pollutants mainly mediated by photosyntheticplants.. In this research four species of plants i.e Mimosa pigra, Crotalaria juncea, Crotalariasp. and lpomoea sp. were studied their potencies as hypertolerant plants on the waste of PTANEKA TAMBANG and public mines of Pongkor and Cigaru. The results showed that theplants were able to grow and showing high tolerance to the contaminated media. The mosttolerant species was lpomoea followed by Crotalaria and Mimosa. The results raised someprospectsfor phytoremediation technology for rehabilitating contaminated mined lands.Keywords: Phytoremediation, hypertolerance

Abstract

Bacterial leaf blight (BLB) is a major disease in Indonesian lowland rice.This research was undertaken to pyramid three BLB resistant genes xa5, Xa7 and Xa21 and one background BLB resistant gene Xa4 into Ciherang and Inpari 13 varieties. The donor parent Code (Xa4+Xa7) was crossed with Angke (Xa4+xa5) while Ciherang and Inpari 13 were crossed with IRBB21 (Xa21). Progenies were selected using marker assisted selection and yield component observation. Foreground selection was conducted using SSR and STS markers linked with the targeted genes in the F1 and DCF1 population. Individuals with triple positives Xa genes were screened for the presence of Xa4 gene as the background. Selected heterozygote plants in F1 Code x Angke, F1 Ciherang x IRBB21 and F1 Inpari 13 x IRBB21 were used to develop DCF1 population. Molecular analysis on DCF1 population through alleles of three BLB resistant genes xa5, Xa7 and Xa21 and one background BLB resistant gene Xa4 resulted 8 (2,6%) in DCF1 Ciherang and 13 (3,5%) in DCF1 Inpari 13. Yield component characters on F1 Code x Angke resulted significant in number of panicle. F1 Ciherang x IRBB21, F1 Inpari 13 x IRBB21 and DCF1 Ciherang resulted significant in weight of empty grain while DCF1 Inpari 13 resulted no significance in all of observed characters. Keywords: Rice, F1 Population, DCF1 Population, molecular marker, Xa gene

Abstract

ABSTRACTStudy on the relationships between vegetation coverage and bird distribution in Moti Island,Ternate, North Moluccas. Research study on relationships between vegetation coverage andbird distribution in Moti Island, Ternate, Moluccas was conducted on May 2010. The objectiveof this research is to understand the bird species occurrence on a vegetation type as anindicator for environment quality determination in small Moti Island. Data on the occurrenceof bird species in the every vegetation type was collected and recorded using explorationmethod. The bird species were identified for the scientific name, local name, their activities,location or coordinate position and their vegetation or habitat. The data then compiled andtabulated for the spatial analyses using Ikonos image and topographic (SRTM) maps data.The data output from the spatial analyses then analyzed using Principle Component Analyses(PCA) to get the most important factors of vegetation cover types that influenced the occurrenceof the bird species. The results showed that about 34 bird species, belong to 20 families and 29genera have occurred in the Moti Island. About 13 vegetation types were recorded as naturalsites of bird species for feeding, playing and breeding grounds. Analyzing data using PCAshowed that at least 3 vegetation types have played as important sites for bird species in thisarea. The sites were mangrove, secondary forest and mixed gardens. The roles of both threeimportant vegetation types and bird species as environment quality indicators were in detaildiscussed in this paper. The discussion also includes how to develop fisherman villages inMoti Island using its own natural resources and biodiversity.Key words: Moti Island, birds distribution, vegetation coverage, spatial analyses

Abstract

Indonesia sebagai negara tropis memiliki biodiversitas yang sangat tinggi. Keanekaragaman hayati ini diperkirakan mencerminkan keanekaragaman kimiawi sekaligus keragaman genetik yang dapat dimanfaatkan untuk mencari biokatalis baru. Enam isolat bakteri yaitu GLB5, LP3, TPIK, MICC, 23A2, dan 23A2 telah diisolasi dari berbagai limbah industri dan mempunyai potensi sebagai pendegradasi nitril.  Pengucilan, identifikasi dan purifikasi gen penyandi enzim nitrilase dari keenam isolat bakteri tersebut  telah dilakukan. Dari kegiatan penelitian ini 3 isolat bakteri unggulan, yaitu GLB5, LP3, dan TPIK teridentifikasi sebagai Rhodococcus pyridinivorans, sedangkan  MICC teridentifikasi sebagai Bacillus substilis, 23A2 teridentifikasi sebagai Brevibacillus brevis, dan 26A2 teridentifikasi sebagai Microbacterium oxydans. Peta untaian basa nukleutida dari gen penyandi enzim nitrilase dari ketiga isolat yaitu GLB5, LP3, dan TPIK telah terpetakan dengan ukuran gen nitrilase sebesar 960 bp. Hasil analisis dengan BLASTN memperlihatkan bahwa fragmen gen nitrilase yang diamplifikasi dengan primer Nit1101F dan Nit1101R mempunyai homologi yang tinggi terhadap Rhodococcus rhodochrous strain tg1-A6 nitrilase gen dengan persentase kesamaan sebesar 96% . Kata Kunci: Gen, isolasi, nitril, degradasi, enzim

Abstract

ABSTRACTCharacterization of Amylase Production by Saccharomyces cerevisiae W303A Recombinants. Cloning of amylase gene from Endomycopsis fibuligera ITB.R.cc.64 into S. cerevisiae W303a can effectively increase the yeast function to digest starch directly into ethanol. Production of amylase by S. cerevisiae W303a recombinants (I and P) were done by growing in yeast peptone starch (YPS) medium. The result showed that the recombinants could be produced of amylase by gave clear zone after staining by iodium vapor. The optimum condition of production of amylase by S. cerevisiae W303a recombinants were pH 7.0, 40?C temperature incubation, and gave maximum activity after 36 hours incubation. Amylase activity of I was higher than P recombinant for these condition respectively.Key words: Characterization, amylase, S. cerevisiae W303a

Abstract

ABSTRACTFood TypeCharacteristic of the Fruit Bats at Urban Area: A Case Study in Bogor BotanicalGarden, West Jawa. Bats have important role on seed dispersal and or plant pollinator. Theidentification of flower and their pollens as the feed resource for bats was conducted in BogorBotanical Gargen. The resultsof this study showed that Eonycteris spelaea male has interestsin with personatus corola type flower, while the female in disk type. Furthermore the male ofMacroglossus sobrinus has interested in rotatus, tubulosus, and perferesence, corola types.Where as the female has interest in campanulatus type. The campanulatus and papilionaceustypes has a potential to be visitd by Cynopterus minutus male and female of C. sphinx;Urceolatus type has important for female of C. titthaheileus and C. brachyotis. The male ofMacroglossus sobrinus and female of Eonycteris spelaea has interests to visit the flower withsuboblate and prolate spheroidal pollen types; prolate pollen type has importance for themale of Eonycteris spelaea; oblate type for the male of C. minutus, C. brachyotis and C.titthaheileus; oblate spheroidal for the female of Rousettus amplexicaudatus and male of C.sphinx. The male of C. titthaheileus and female of Macroglossus sobrinus has interests ingigantic type (>200 ì m), while the female of C. sphinx, C. brachyotis and R. amplexicaudatuslike permagnae type (100-200 ì m).Key words: Fruit bats, pollen identification, urban areaKata kunci: Kelelawar buah, pollen, identifikasi, perkotaan
