Abstract

ABSTRACTPollinator Bat Identification Based on Pollen as Feed Resources at LinggarjatiDistrict-Ciremai National Park, West Jawa. Pollen identification that used for feedresources by six species of frugivorous bats from Ciremai National Park was identifiedunder microscope. The results shown that are known 21 species as important plants forbats and in the contrast possibly for bats as pollinator. By the principle componentanalysis and cluster grouping based on Euclidian distance indicated that Aethalops alecto,Chironax melanocephalus as important polinator role in primary forest and high in elevationlevel, C. brachyotis and Macroglossus sobrinus in secondary or fragmented forest andMegaerops kusnotoi in lower primary forest and Cynopterus tittacheilus is important rolefor intermediate area between primary and secondary forest and at lower elevation range.On the niche overlaps analysis indicated that the highest overlaps were known in C.brachyotis and M. sobrinus (0.881) but the lowest in niche overlaps between M. sobrinusand C. melanocephalus (0.288)Key words: pollen, bats, feed resources

Abstract

Penelitian vegetasi mangrove telah dilakukan di Kalitoko, Mayalibit Bay, Pulau Waigeo, Kabupaten Raja Ampat,Papua Barat Provinsi. Hasil penelitian menunjukkan bahwa keragaman mangrove di Kalitoko relatif tinggi.Ditemukan 65 jenis mangrove (56 marga dan 35 suku), ternyata 21 jenis diantaranya dimasukkan sebagai jenislangka berdasarkan daftar IUCN dengan status rawan (VU) dan kritis (CR). Hasil analisis vegetasi satu transekdengan luas 4.500 m2 di hutan mangrove Kalitoko tercatat 18 jenis tumbuhan mangrove dengan tingkatkepadatan pohon sebesar 549 individu / ha dan basal areanya 18.61 m2/ha. Untuk tingkat belta kepadatan mencapai1.514 individu / ha dan basal areanya 3,86 m2/ha. Sedangkan kerapatan semai mencapai 194.889 individu /ha.Kata Kunci: Mangrov, komposisi, Mayalibit, Waigeo, Raja Ampat

Abstract

ABSTRACTThe study on population dynamic and  roosting trees damage by flying fox was conducted for 16 months (March 2008-June 2009) in Bogor Botanical Garden.  Recording of individual flying fox was carried out by use direct observation in the roosting trees habitat at 9.00-15.00, with one replication every week. The result indicated that in 2008 the total flying fox were 420 individuals and in 2009 were 563 individuals (160 juveniles, 218 adult females and 185 adult males). The natality and mortality rate in this location is 0.0214 and 0.0099 respectively. The flying fox population growth can be predicted using exponential equation is N(t+1)=563(1.8395)t.  The study also indicated that in the KRB there are 9 individual trees which are the main perch, 2 individual trees of Pterodotus alata, Pterocarpus indicus, and Shorea leprosula, and each 1 individual tree of the species  Dipterocarpus cornutus, Ficus drupacea, and Syzygium syzygioides. The extent of damage to the tree where the perch is very dependent on the diameter of the canopy, bat population size in one tree, and time periods per roosting season.  Keywords: Population Dynamic, Flying Fox (Pteropus vampyrus), Bogor Botanical Garden

Abstract

ABSTRACTPattern Activities and Diversity of Grasshopper (Insecta: Orthoptera) in Ciremai MountainNational Park, Kuningan, West Java. The grasshoppers of the Gunung Ciremai National Park,based on collections in 2008 are reported. The grasshoppers were collected by transect methodin three locations: Seda (508 m asl), Palutungan (1076 m dpl) and Cilengkrang (1143 m asl). Atotal, thirty three species of grasshoppers were recognized. The numbers of species/diversityindex Shanon are: 25/2.99, Palutungan 18/2.60, and Cilengkrang 13 species/2.46. The highestactivity of Ciremai grasshoppers was between 11.00-12.00 pm. The Erianthus sp was newrecord from Java, whereas Oxya spp.and Catantops (Acridiidae) spp were only found onSeda. Bibracte sp.(Acridiidae) and Paratrigonidium faseocinctum (Gryllidae) were only foundon Cilengkrang, whereas Oxyrrhepes obtusa (Acridiidae) was only found on Palutungan.Species similarity on location, observation time, activity, and habitat of each grasshopper arediscussed.Kata kunci : Activities, diversity, grasshopper, orthoptera, insecta, Ciremai

Abstract

ABSTRACTSurra is a parasitic disease caused by T. evansi and causing high economic losses in Indonesia. Some isolates have been isolated from several areas experiencing outbreaks of Surra in Indonesia. The isolates have been reported to have a diversity of protein profiles based on SDS PAGE (sodium dodecyl sulphate polyacrilamide gel electrophoresis). The research aims to identify immunogenic protein from each isolate and make the selection of T. evansi isolates which is potential as a source of antigens for immunoassay development. Each isolates were obtained to be purified from blood and then the protein was isolated. The proteins were run onto polyacrylamide gel electrophoresis and visualized by Coomassie blue. Another electrophoresis results were transferred onto a nitrocellulose membrane for immunoblotting. The results showed that the immunogenic proteins that consistently detected among nine T. evansi isolates of Indonesia are 100, 90, 85, 76-80, 70, 65, 55, 49-52, 44-46, 40, 34-36, 31 -33 kDa. Among the nine T. evansi isolates, N372 isolate was selected as a candidate for immunoasay development, especially ELISA. Immunogenic proteins were specifically found on the N372 isolate are 85, 70, 65, 49-52, 44-46, 34-36, 31-33, 24-28, 15-20 kDa.Keywords : Trypanosoma evansi, immunoblotting, protein profiles, immunogenic protein

Abstract

Hylarana celebensis (Peters, 1872) is an endemic frog to Sulawesi, the species being a member of family Ranidae.  The presence of the frog in its habitat is easily detected from its advertisement call; males usually call in a chorus, they call to each other in a large group. Bioacoustics of calls that a typical individual male of H. celebensis has not been described in detail, although it is very easy to find this species in freshwater swamps, permanent ponds, or slow-flowing waters in the lowland areas. The purpose of the bioacoustic analysis on H. celebensis?s calls that were recorded at Bahodopi area is to build a reference collection to be compared with H. celebensis?s bioacoustics to the other regions in Sulawesi. Because of the wide distribution of this frog in Sulawesi; population genetic structure of H. celebensis may also follow the population genetic structure of Ingerophrynus celebensis. Calls of H. celebensis have two types of calls, i.e. pure tone and pulse; however, pure tones have three variation, namely pure tone type 1, pure tone type 2 and pure tone type 3; however pulsed call has only one type. Keywords: Anura, Hylarana celebensis, bioacoustics, Sulawesi.

Abstract

ABSTRACTInsects Community on the Flower of Rafflesia patma Blume (Rafflesiaceae) in its Non NativeHabitat of Bogor Botanical Gardens, Bogor City, Province of West Java, Indonesia. The studywas conducted at the Bogor Botanical Gardens, Bogor, West Java, Indonesia using a bloomingfemale flower of R. patma. The insects were directly counted in the morning, noon, and afternoonon both fresh blooming and rotten R. patma . Twenty three insect species were collectedduring the study belonging to the order Coleoptera (2 families, 2 species, 5 individuals),Diptera (9 families, 18 species, 1176 individuals), and Hymenoptera (2 family, 4 species, 13individuals). Number of individuals of each insect species captured were frequently less than1.35% from total captured. There were specialization of flies visiting fresh opening flower andthe rotten one. Six species, Leiomyza laevigata (Asteiidae), Chrysomya megacephala, andHemipyrellia tagaliana (Calliphoridae), Stegana coleoptrata (Drosophilidae), Heteromyzaoculata and Tephrochlamys rufiventris (Heleomyzidae) were predicted as important pollinatorsof R. patma.Key words: Insect community, flower, Rafflesia patma, non native habitat, Bogor BotanicGarden.

Abstract

ABSTRACTTaro (Colocasia esculenta (L.) Schot.) has been cultivated for a long time in Indonesia. Taro tuber can be used as alternative food for rice substitute to support food diversification program. Taro cv. Bentul is preferable to farmers because it has few buds that are easy to maintain and adaptable in both lowlands and highlands. Its tuber has a delicious taste and a soft texture. Somatic cell manipulation of Indonesian taro through biotechnology may contribute to increase its productivity. One of in vitro technique for somatic cell manipulation is polyploidy. Oryzalin has been able to obtain taro cv. Bentul tetraploid. A total of 17 tetraploid clones were used in this study for clustering. Those clones were obtained from previous research and have stable ploidy level. The objective of this study was to determine cluster Bentul tetraploid of shoot culture using Ward method based on their in vitro growth as an initial selection before further selection in the field. Shoot tips of tetraploid were cultured on MS medium containing 2 mg/l BAP, 1 mg/l thiamine and 2 mg/l adenine for 6 weeks. The observed growth variables were number of shoots, length of petiole, number of leaves and roots. The clusterring was done using Ward and Euclidean Distance method followed by Analysis of Varians and Duncan Multiple Range Tests (DMRT). Out of 17 clones observed resulted in 3 clusters. Cluster 1 consisted of 2 clones, cluster 2 consisted of 9 clones, and cluster 3 consisted of 6 clones respectively. The best cluster was cluster 3 which was significantly different on the average number of shoots and leaves. Abstract

Xylanase extracellular enzyme is produced by various microbes. Hydrolysis of bagasse xylan can produced xylooligosaccharide. The main components of hemicellulose was xylan at bagasse. IdentificationBacillus subtilisXJ28 using primer 16S RNA. The qualitative test used Congo-Red stainning, whereas quantitative test used Dinitrosalicyclic Acid (DNS) methode. The hydrolyzing product was analysed used TLC (Thin Layer Chromatography). This research aims were to characterized xylanase from isolate XJ28 and analyzing the hidrolyzing product from xylan bagasse. Pretreatment process included delignification using sodium hypochlorite 1% and xylans extraction using alkaline (NaOH 15%) as the solvent. The result of the xylans extracted from bagasse was 9,9% xylan and after purification was obtained 3,4% soluble xylan. Xylanase activity has the highest activity at 96 h of incubation time with activity 11,3 U/mL. Xylanase Bacillus subtilis XJ28has the optimum condition at pH 7 and 50 ºC and stable up to 72 hr of  incubation time at room temperature and 4 ºC. The hydrolysis product using xylanase crude enzyme was reducing sugar with molecular weight around  of sucrose and oligosaccharide. Keywords: xylanase, xylan bagasse, hydrolysis, TLC.
