Abstract

ABSTRACTPurifikasi Parsial dan Karakterisasi Enzim Kitin Deasetilase yang Dihasilkan Bacillus thermoleovorans LW-4-11. Kitin deasetilase yang dihasilkan dari bakteri Bacillus thermoleovorans LW-4-11 telah dimurnikan menggunakan amonium sulfat 70% diikuti dengan perlakuan panas (suhu 70°C selama 1 jam). Kemurnian enzim yangdipisahkan dari medium fermentasi meningkat 4.28 kali dengan aktivitas spesifik sekitar 43.7 mUImg. Enzim memiliki suhu dan pH optimum masing-masing 80°C dan 6.0 dalam substrat 0-hydroxyethylated chitin (glycol chitin). Kitindeasetilase ini relatif tahan panas dengan waktu-paruh sekitar 30 menit pada suhu 80 "C. Enzim dihambat oleh ion Li', ZnZ+, MnZ+, Co2' and Ni' pada konsentrasi 1 mM, tetapi diaktifkan oleh EDTA 1 mM.Kata kunci: purifikasi parsial, karakterisasi, kitin deasetilase, B. thermoleovorans LW-4-11

Abstract

Famili Rubiaceae telah lama dikenal sebagai sumber tanaman obat Indonesia. Beberapa spesies seperti Coffea arabica,Morinda citrifolia, Chinchona succirubra telah digunakan sebagai obat rakyat sejak jaman dulu. Penelitian ini bertujuanuntuk mengetahui nilai peroksida dan aktivitas antioksidan pada 10 jenis ekstrak famili Rubiaceae yang lain.Nilai peroksida (POV) diukur menggunakan metode iodometri dan hasil pengukuran menunjukkan POV yang berbedanyata kulit kayu Anthocephalus macrophyllus (69,48), kulit kayu Wendlandia glabrata (67,86), kulit kayu Guettardaspeciosa (73,17) dan daun Paederia foetida (89,14). Aktivitas antioksidan diuji menggunakan metode tiosianat(FTC) pada empat jenis ekstrak yang berpotensi, yaitu kulit kayu A. macrophyllus, kulit kayu W. glabrata, kulit kayuG. speciosa dan daun P. foetida. Nilai absorbansi (A) dari masing-masing jenis adalah 0,294; 0,293; 0,365; dan 0,375,dibandingkan dengan kontrol negatif (0,919) dan kontrol positif (0,31).Kata kunci: Rubiaceae, sifat antioksidan, POV, metode tiosianat (FTC)

Abstract

Association of endangered medicinal plants in some forest areas of national parks in Jawa. The existences of medicinal plants in the forest is decreasing and most of them are becoming scarce. Some species of endangered medicinal plants spread over the area national parks onthe island of Java. The inventory has been widely performed, but the studies of the existence of the species concerned associated with other species as well as with environmental factors are still very rare. This paper presents the results of association analysis interspecies with chisquare test and species-habitat associations using Canoco for windows 4.5. The results show that there is no interspecies association in general, but a positive association multispecies hasoccurred in the area of Meru Betiri NP and negative association multispecies occurred in Alas Purwo NP and Bromo Tengger Semeru NP. Ordination plot show that Eucresta horsfieldii has association with the altitude and has not associated with other rare medicinal plants. While Arcangelisia flava is not associated with environmental factors as well as other species of medicinal plants.Key words: endangered medicinal plants, association analysis

Abstract

ABSTRACTThe Mega Bat (Pteropodidae) Distribution on Several Land Coverage at Surrounding KerinciNational Park. The study of bats was carried out on May 2005 to July 2005. In all 5 major landuse system (primary forest, disturbed forest, old rubber plantation, young rubber plantationand mix garden) survey was conducted using the same standardized mistnetting. Each of theland use system was sampled for 4 days using 4 mist-nets a night. The total of 2512.5 m2 ofmist-nets night area were set during this survey. A total of 156 bat individuals which consist of14 specieses and 5 families (Pteropodidae, Vespertilonidae, Emballonuridae, Hipposideridaeand Rhinolophidae) acquired. Calculation result using the diversity index measurement frombat samples indicate that the highest diversity was in disturbed forest habitat (H?=1.831;E=0,865) and the lowest was in forest habitat (H?=0.5; E=0.561), meanwhile the biggestdominancy reside in the young rubber plantation (?=0.767) and disturbed forest had the lowestdominancy (?=0,138). The most dominant at young rubber plantation habitat is Cynopterusbrachyotis. The result of clustering analysis using SPSS 11.5 showed that habitat type weredivided become 2 clusters which were old rubber and non old forest (forest, disturbed forest,young rubber plantation and mix garden). This division showed that forest and non forest hada distinctively different habitat. According to Canonical Correspondence Analysis (CCA) byusing Canoco for Windows version 4.5 indicated that physical parameter such as temperature,humidity, rainfall and altitude influenced the distribution, commnunity and clustering of batsfor land use systemKey words: Bats, diversity, Kerinci Seblat National Park, land use type, rubber plantation

Abstract

ABSTRACTObjectives of the research were to determine the best source of explant origin, the optimum culture media and effects of various concentrations and types of growth regulators on the induction and multiplication as well as the growth of in vitro shoot of cassava Ubi Kuning genotype. The experiment was arranged in a complete randomized design with three factors, i.e the source of explant origin (from field and greenhouse), the growth media containing of 6-Benzyl Amino Purine (BAP) and Gibberellin (GA3) at the concentration level of 1-2 ppm, and multiplication medium supplemented with BAP, Naphtalenic acetic acid (NAA) and Kinetin at concentration level of 0.1 mg/l and 0.5 mg/l. The results showed that the cuttings explants from the field showed more shoots and the average shoot height of axenic culture in the fourth week ranging from 0.2-1.2 cm compared to those from the field (0.5- 1.3 cm). Axenic culture showed no significant growth in growth media (MS) supplemented with BAP and GA3 (1-2 ppm). In the multiplication medium, the highest number of roots were obtained when plantlets were grown in the treatment medium MS + BAP 0.5 mg / l followed by MS + NAA 0.1 mg / l. The largest number of nodes was produced on plantlets grown in MS + BAP media 0.1 mg / l and MS + NAA 0.5 mg / l (p <0.05). While the longest root growth was obtained in plantlets cultured on MS + Kinetin 0.1 mg / l medium. Plantlets showed high survival (100%) after they were acclimatizedfor 4 weeks in a greenhouse using a medium containing a mixture of compost, coco peat, and ground burn husk with a ratio of 1: 1: 1. Keywords: genotype, ubi kayu, ubi kuning  in vitro

Abstract

Plant ecology study has been conducted in the southern part of Rinjani Mountain National Park on April 2015 to determinethe structure and composition of tree species. Four plots, 30 x 30 m each were established at four locations, those are; Pal-Belanda (1046 m asl), Pengorot-1 (929 m asl), Pengorot-2 (820 m asl), and Sebau (1134 m asl). The results showed thatthere are two types of forest in the sites, namely the secondary forest (Pal-Belanda, Pengorot-1 and Pengorot-2) and primaryforest (Sebau). From all plots (0.36 ha) we found 36 species of trees (trunk diameter > 5 cm) of 449 individuals, 28 genusand 21 families with a total Basal Area of 16.73 m2 (46.47 m2/ha). Shannon Diversity Index (H') in the Pengorot-2 recorded1,15, followed Pengorot-1 (1.13), Pal-Belanda (1.03) and Sebau (0.72). Saurauia pendula recorded as the dominant speciesin all site with Important Value Species (IVS) of 245% followed Dendrocnide microstigma (IVS = 162%), Platea excelsa(IVS=148.2%), Syzygium glomeruliferum (IVS=72%) and Symplocos cochinchinensis (IVS=49.3%). Myrtaceae andEuphorbiaceae were the common families in the sites. Structure and composition of tree species in the Pal-Belanda,Pengorot-1 and Pengorot-2 were relative similar (Similarity Index 70.3% and 69.4%) but different than in Sebau area(Similarity Index 24.9%).Keywords: Structure, composition, tree diversity, Rinjani, Lombok

Abstract

ABSTRACTThe Grouping of Fruit Bats Based on Pollend Type Characterized as Food Resources inBogor Botanical Garden, Indonesia. A study was conducted to identify pollen consummedby fruits bat in Bogor Botanical Garden from March 2008 to June 2009. The types of crown ofthe flower, pollen and pollen size which chosen by fruit bats were analysed by using highdetrended canonical correspondent. The result indicated that there are three major groups ofbats based on the similarities of food type or flower resources consumed by fruit bats. Thegroups were (1) males of Macroglossus sobrinus and the females of Eonycteris spelaea, (2)Cynopterus brachyotis and the females of C. minutus, and (3) males and females of C.titthaheileus, females of C. brachyotis and Macroglossus sobrinus, males of C. sphinx and C.minutus, and females of C. sphinx.Key words: Megachiroptera, pollend, seed distribution,

Abstract

ABSTRACTGrowth of the Groupers (Serranidae) in Floating Net Cages at The Coastal WatersKodek Bay Malaka Village West Lombok. Study of rearing of Bararnundi cod(Cromilileptes altivelis), flowered cod (Ephinephelus fuscogutatus) and estuarygrouper (E. tauvia) at floating net cages was canied out on June to December 2004at the coastal water Kodek Bay MalakaViiage, West Lombok. There was sigdicantdifference in weight growth rate of groupers during observation (X count = 67.420and X2a 0.05 = 2 1.026 or X count > X2a 0.05) during observation, but not in length (X2count = 0.998 and X2a 0.05 = 2 1.026 or X count < X2a 0.05). Observed physicochemicalproperties of experimental set up are water temperature between 27.2 - 27.8 OC, salinitybetween 3 1.8 - 33.8 %o, transparency between 10.3 - 14.5 m, air temperature between26.6 - 27.28 OC and pH about 8.3 - 8.4.Key word: Groupers fish of growth, waters condition

Abstract

First year re-monitoring conducted in 2009 through measuring the entire individuals that have been given numbersand giving new numbers to individuals with stem diameter more than 5 cm. The result of re-monitoring indicatesthat the numbers of populations and species increased, but not significant. Mortality rate observed 7 individualsper hectare (about 1.20 %); whereas natality rate observed 25 individuals per hectare (about 2.52 %), which isregarded as low. Basal area observed in 2008 was approximately 26.55 m2 per hectare and it increased to about27.34 m2 per hectare in 2009. Total number of individuals in 2009 was 595 individuals from 119 species, 86genera, and 44 families with diversity index of 3.60 (Hâ??). Elaeocarpus petiolatus (Elaeocarpaceae) and Planchoniavalida (Sapotaceae) are new additions. Abstract

Irradiation of ??ray at shoot culture of Kentang hitam (Solanostemon rotundifolius) is effective for mutant production. Kentang hitam is sterile and vegetatively propagated andtherefore its genetic diversity is narrow. Mutation is an alternative way to increase genetic heterogeneity. Irradiation of shoot cultures with different doses followed by culturing of the inoculum (leaf, petiole and internodes) from that irradiated culture on a regeneration medium MS containing 5 mg/l BA and 0.1 mg/l NAA has resulted in curvilinear respon of growth, morphogenesis and plant regeneration. The growth variables increase from 0 to 6 Gy and thendecrease to 25 and plateu or increase a little at 35 Gy and growth death wes observed at 50 Gy. Leaf and petiole inocula were more responsive than the internode. Respon of growth of shootregeneration of 50% were obtained at doses of 10-12.5 Gy. However higher level of doses, such as 25 Gy had also been effective for inducing mutant. Morphological and growth different were observed from growth in tissue culture to the field.
