Abstract

ABSTRACTSurra was reported in various types of animals both livestock, pets and wildlife. ELISA (enzyme linked immunosorbentassay) is a sensitive and specific diagnosis technique for Surra detection. The use of speciesspecific conjugate (anti bovine IgG-HRP) has limited while protein A/G can be used for a wide variety of animalspecies. This study aimed to evaluate the initial application of the protein A/G-HRP compared with antibovine IgG-HRP using standard samples using four (4) types of diluent buffer. The standard serum samples (23serum) consist of positive and negative sera from bovine (cattle and buffalo) were reacted with Surra antigenon microplate. Positive and negative serum was diluted with different diluent buffer, namely PBS-Tween20(PBST), RBA (RedBuff A), RBB (RedBuff B) and LC (Low Cross). Results of ELISA using protein A/G-HRPshowed absorbance values reduced 36.16% - 69.30% compared to the anti-bovine IgG-HRP. The percentagereduction of PBST, RBA, RBB and LC, were 51.76%; 56.64%; 36.16% and 69.30% respectively. The use ofprotein A/G-HRP and fourth diluent buffer can reduce antigen - antibody bonding with a weak affinity whichlowers the absorbance value of ELISA Surra.Keywords : Protein A, Protein G, Surra, ELISA, Trypanosoma evansi

Abstract

ABSTRACTThe influence of zooxanthellae isolated from corals on the growth and suwivorshipof giant clams. Giant clam known as marine bivalves (Tridacnidae) which live in coralreef. One important aspect of giant clam biology is the existence of zooxanthellae assymbiotic algae which have important role source of energy In hatchery operationalprocedure zooxanthellae were introduced into larvae. Zooxanthellae were isolated fromadult clams. Since clams are also known as endangered species, it is important to findother sources of zooxanthellae. The objective of this research is to investigate the effectof introduction of zooxanthellae which were isolated from different sources on the survivaland growth of giant clams larvae. The results showed that there is no differences on thesurvivorship and growth of giant clams larvae introduced by zooxanthellae isolated fromseveral corals. This result has open a possibility of using corals as source of zooxanthellaeinstead of using giant clams.Key words : zooxanthellae, giant clams, survivorship, growth

Abstract

Mangifera casturi Kosterm. (Anacardiaceae) merupakan spesies yang dinyatakan punah di alam liar. Spesies ini endemikdi pulau Kalimantan. Saat ini, Mangifera casturi telah dibudidayakan di luar sebaran alaminya, terutama diKabupaten Indragiri Hilir Provinsi Riau, Sumatera. Penelitian dilakukan di dua kecamatan di Kabupaten IndragiriHilir, yaitu Kecamatan Gaung Anak Serka (GAS) dan Kecamatan Gaung. Sebanyak 1.315 individu M. casturiditemukan dalam penelitian yang dilakukan. Keberhasilan konservasi ex situ M. casturi di daerah ini didukung olehsetidaknya tiga stimulus: alami, manfaat, dan stimulus religius. Namun faktor internal dan eksternal milik masyarakatsetempat juga mempengaruhi secara signifikan. Di Kecamatan GAS, konservasi ex situ M. casturi terutamadidukung oleh stimulus manfaat sementara di Kecamatan Gaung upaya konservasi ex situ didukung oleh stimulusreligius. Analisa komponen utama dilakukan untuk menentukan kemungkinan hubungan antara konservasi ex situM. casturi dan variabel pendukung dalam masyarakat. Di Kecamatan GAS, yang upaya konservasi ex situ M. casturisecara signifikan didukung oleh ukuran populasi, panen dan pemasaran serta kegiatan pra tanam. Di KecamatanGaung, konservasi ex situ dari M. casturi tidak hanya didukung oleh ukuran populasi, panen dan kegiatan pasar, tetapijuga oleh kegiatan pra tanam, status dan luas lahan.Kata Kunci: Indragiri Hilir, Konservasi ex situ, Mangifera casturi, Punah di alam liar

Abstract

Propagation of Amorphophallus titanum Becc (Araceae) via In Vitro Technology. Amorphophallus titanum is a unique plant with gigantic inflorescence and radiating corpselike odorl and therefore has become a botanical flagship and attract visitors. Its existence innature may be endangered due to habitat conversion that reduce population size required for cross pollination to occur. Propagation via non-conventional technique is much needed for the resulting planting material to be exchanged among botanical garden or even traders. Mass propagation using in vitro technology has been successful. An improvement of the propagation method has been developed by incorporating high level of cyitokinin BA of 1-2 mg/l in the medium for shoot proliferation and lowering it to 0-0,1 mg/l for shoot elongation. Shoot elongation is effectively influenced by age and the phase of medium whereas the liquid phase induced the enlargement of the shoots significantly without even promoting any hyperhidricity.The plantlets produced survive acclimatization.Key words: Amorphophallus titanum, mass propagation, in vitro, mediurm cair, BA.

Abstract

ABSTRAKPemilihan Tumbuhan Hutan Sebagai Sumber Pakan dan Tempat Bersarang Kuskus kerdil(Strigocuscus celebensis) dan Analisa Nutrien di Pulau Wawoni, Sulawesi Tenggara. Astudy on feed plants selection and nesting site of dwarf cuscus (Strigocuscus celebensis) wasconducted in Wawonii Island, South-East Sulawesi. The study was done in the lowland forestwith the altitude of 1 - 200 m a.s.l. The results showed 45 species of plants consisting of 23families was selected by dwarf cuscus as their feed resources and 11 species of plants as theirnesting site. Parts of the feed plants being consumed were fruit, young leaves, flower, andtopmost of trunk.Keywords: Forest plant, feed resources, nesting site, Strigocuscus celebensis Wawonii Island

Abstract

 ABSTRACTThe objective of study was to explore Enggano?s people live survival strategy ulilizing plant resources .  To enable living in Enggano, local community utilize plant resources with diverse strategy.  The main argument of their article is social and enviromental changes affect on the utilization of plant resources as the main strategy of survival.  There strategies of data collection were applied to verify the local community survival strategy.  Namely: FGD, questionair and interview. So we observed society change their live strategy on plant resources utilization in accordance with social change. Abstract

"Cibinong Science Center LIPI or CSC?s LIPI as an artificial habitat to the bird species conservation, especiallywith their ecological parks. This research was to study the distribution patterns and dynamics of birdcommunities in the CSC?s LIPI areas during 2005 to 2009 and on 2014. Transect Line?s method was used inthis study. The results were 39 species of birds to live in the eight sites observation on 0.27 km2 areas.However, there are no significant effects, especially relationships between Margalef?s index value to area,cotton area closest distance to the river and their habitat situation. According to ?multiple linear regression?analysis show that ?= 3.626-0.69 area -1.31 cotton area closest distance to the river + 0.94 habitat situation,with determinant coefficient value (R2) = 0.161. The densities of nine species were found high. Those specieswere Lonchura punctulata (D=49.37 individuals/ha), Zosterops palpebrosus (D=22.2 individuals/ha),Collocalia linchi (D=15.00 individuals/ha), Orthotomus sutorius (D=12.70 individuals/ha), Lonchura maja(D=12.48 individuals/ha), Passer montanus (D=12.19 individuals/ha), Pycnonotus aurigaster (D=7.37individuals/ha), Dicaeum trochileum (D=6.07 individuals/ha) and Streptopelia chinensis (D=5.85 individuals/ha). The diversity indices value (H?) 1.52 to 2.51 and the evenness indices value (E) 0.52 to 0.86. Thesimilarities indices value (SI) of birds in the CSC?s LIPI will be compare to another places, i.e. the IPBcampus at Darmaga, Bogor or Yogyakarta State University?s campus in Yogyakarta showed no different,relatively. The IS value were 58.82 to 61.54%. Birds in the CSC parks were high dominant to occure in thelower strata (27.78?43.18%) and upper strata (50.43-70.83%), while in the middle strata (1.39-9.15%) tend tobe slightly. Most activities were carried by birds , especially foraging and calls. Although, the birds in the CSCparks also breeding, take a rest etc . The CSC?s LIPI parks at Cibinong could be used as a reference to thebird?s artificial habitat model to others areas. That?s an important if the areas are industries complex or theCenter office of some government buildings. But, the composition and structure of the vegetation will bedeveloped in there should be to produce some natural food resources and safety well for breeding of birds.Keywords: bird species, ecological parks, artificial habitat, distribution patterns, dynamics communities

Abstract

Ethnobotany Contributes to The Understanding Human Relationship with Plant and TheirEnvironment. The scope of ethnobotanical research has been developed to a broader scopefrom the originated word of ethnobotany was coined.. This discipline attempts to explainreciprocal relationships which occur between local societies and its natural world, in extant,between local societies and their cultures that reflected in the archeological records.Ethnobotany is also very closely related to the domestication of plants such as the speciesdomesticated, where these species domesticated, the purpose of domestication, the manner,and the status of the domesticated plants today. Etnobothany also concerns to the role ofplants in ecology, environment and phytogeography as conceived by tradition or by the localcommunities. In addition to its traditional role in economic botany and the exploration ofhuman cognition, ethnobotanical research has been applied to the practical areas such asbiodiversity prospecting and vegetation management. Thus ideally, ethnobotany shouldincludes rules and categorization acknowledged by local communities. Rules and categorizationare use to appropriately facing daily social situations in recognizing, interpreting and utilizingplant resources in their environment. In summary, the scope of research in ethnobotany isinterdisciplinary and ethnoscience as mentioned earlier and these scopes will be the mainfocus of discussion in this article. Abstract

ABSTRACTThe Biodiversity of Fish In Squaq Balimbing and Ketambe Gunung Leuser NationalPark, Sumatera. Gunung Leuser National Park (GLNP) is one of the biggest national park inIndonesia which is the habitat of various flora-fauna. GLNP is a conservation area inSumatera island that lies in the two province, North Sumatera Province and Nanggroe AcehDarussalam Province. Little has reported about fishes inhabiting inland waters of Acehprovince and Sumatera Utara province, because The Fisheries Service of both province andThe University of Syah Kuala and The University of Sumatera Utara are more interested inconducting research on marine fish.Ichthyofauna research was conducted at the two Research Stations of Gunung LeuserNational Park, i.e Suaq Balimbing and Ketambe Research Station. Research activity at Suaq- Balimbing RS which is located in South Aceh Regency was conducted in 1997, whileresearch activity in Ketambe RS which is located in South East Aceh Regency was conductedin 1998. The research activity successfully collected 64 fish species which is belong to 36familia. Abstract

Patients with H5N1 disease have a characteristic complications of pneumonia with acute respiratory syndromedisterss and multi-organ failure associated with cytokine dysregulation, so it is suspected that the clinical forms ofsevere H5N1 disease in humans is caused by dysregulation of cytokine induction of H5N1 virus. Study oncytokines associated with H5N1 influenza virus infection is still limited, while animal studies using virus H5N1/97give conflicting results. Recent studies, showing that in fact the high cytokine is not a common form of the H5N1viruses, so Pathogenecity of H5N1 viruses were not only determined by whether or not the virus can inducehypercytokinemia. In this study we wanted to determine the phenotype of the Indonesia H5N1 virus in inducingcytokine expression. This study compared three Indonesian H5N1 viruses that have different genetic variations ofthe virus including the clade 2.1.1 and two other viruses included in clade 2.1.3 that were infected in Madin DarbyCanine Kidney (MDCK) cell. The MDCK cell that infected virus were observed for 72 hours. We observed theinfection during 72 hours and the cytokines expression were examined using RT-PCR method. The results showedthat the H5N1 influenza viruses used in this study showed the low expression of cytokines IFN-γ, and did notshow any expression of the TNF-α cytokines in MDCK cells infected with the H5N1 virus within 72 hours afterinfection.Keywords : Expression, H5N1 virus, Cytokines, sel Madin Darby Canine Kidney (MDCK)
