Abstract

The Effect of Biofertilizers and Anorganic Fertilizers to Several Soil Chemical Properties and Spinach (Amaranthus tricolor) in Pedungan Village Inceptisol The aim of this study is to know the effect of interaction between biofertilizers and inorganic fertilizers on soil chemical properties and yield of spinach in Pedungan Inceptisol. This research was conducted from October 2017 to February 2018 at the greenhouse. The experiment located in Pedungan Village and the Soil and Environmental Laboratory, Agriculture Faculty of Udayana University. Randomized Block Design (RBD) with 9 treatments and 3 replications were used in this experiment. The data were analyzed by using the Costat program and continued by the LSD test at 5% level if the treatment was significantly different. The variables observed in this study consisted of soil chemical properties and production of plants. The result of this experiment showed that there are no interaction between biofretilizer and inorganic fertilizer on all of parameters observed. The effect of biofertilizers had a significant diferences on all of parameters, while in using inorganic fertilizers only on the half parameters. The treatment of biofertilizers at a dose of 20 mL had a significant effect on the improvement on all parameters, while inorganic fertilizer treatment at half of the recommended dose had the same effect as the treatment of real recomended dose for increasing the total N, K, soil pH, and plant height.

Abstract

ABSTRACT Effect of Various Timing on Bud Cutting as Cuttings Material and Dosage of Rootone F on the Growth of Vanilla Short-Cutting (Vanilla planifolia Andrews) Vanilla (Vanilla planifolia Andrews) can generally be propagated by seed (generative) and vegetative by cuttings. As the multiplication by seed takes longer to flowering, thus the vanilla propagation for commercial purpose is done by cuttings. This study aims to determine the effect of time on bud cutting as cuttings material and dosage of Rootone F as well as its interactions on the growth of vanilla short-cuttings. This study uses two factors factorial design, i.e.: time on bud cutting as cuttings material and dosage of Rootone F. The result of the research showed an interaction between time on bud cutting as cuttings material and dosage of Rootone F provides a very significant effect on the variable of oven dried stems shoots weight, and showed a significant effect on the variable total oven dry weight; while other variables showed no significant effect. Treatment of time on bud cutting as cuttings material (P) showed highly significant effect on the variable of time to sprout, while the other variables showed no significant effect. Dosage of Rootone F (D) showed no significant effect on all variables. The interaction of time on bud cutting as cuttings material 8 days before planting with Rootone F dose of 100 mg / 1 ml of water provides a better growth of vanilla short-cuttings with the highest total oven dry weight is 1,12 grams and has increased 89.83% compared to the bud cut when the cuttings is about to be planted and Rootone F dose of 0 mg / 0 ml of water. Keywords: Vanilla time on shoot-cutting as cuttings material, Rootone F, nurseries, vanilla.

Abstract

ABSTRACT Isolation and Identification of Agrobacterium tumefaciens from  Carrots (Daucus carota L.) Agrobacterium tumefaciens is a plant pathogenic bacterium that is widely used as a vector for inserting foreign genes into a plant chromosome to produce a transgenic plant. This bacterium cause a disease namely crown gall in dicotyledonous plants. This study is aim to isolate A. tumefaciens from local carrot (Daucus carota L.) by using some method such as selection media, characterization of the bacteria, and confirm by Koch’s postulates. The result of this study determined that carrot’s root taken from Bedugul area which showed crown gall symptom, an A. tumefaciens due to its characteristics e.g. shape of colonies, color, and growth of the bacteria in AB minimal medium that was a specific for A. tumefaciens.  By using the Koch’s postulates test showed that the isolated caused convex gall on the surface of carrot slice. DNA of isolated bacterium successfully electroforeted through agarose gel electrophoresis.  These results showed that the bacteria associated with carrot was A. tumefaciens. Keywords : A. tumefaciens, AB medium, Carrot, LB medium, Crown gall

Phytoremediation of Irrigation Water Using Water Hyacinth Plant (Eichornia Crassipes) and Lotus Plant (Nympae sp.) in Subak Sembung Peguyangan Village, North Denpasar Subak Sembung Peguyangan village, north Denpasar has an area of 115 ha. Agrochemical use and land use change threatens the quality of irrigation water This research aimed to determine the quality of irrigation water in which the research had conducted from August to December 2015. The physical, chemical and biological qualities of the irrigation water analyzed through sampling the water gradually then the result was being compared to water quality standard in accordance with regulation Peraturan Gubernur Bali No. 8 Tahun 2007. The result showed that the irrigation water is contaminated with heavy metal Cadmium (Cd). The physical qualities of irrigation water was indicated by total value of TSS (0.60 mg/l) and TDS (270 mg/l), the chemical qualities was indicated by the value of pH (7,21), BOD (3,875 mg/l), COD (9,996 mg/l), and Pb (0,0345 mg/l) and the biological qualities of irrigation was indicated by Total Coliforms (150/100 ml). These three qualities is still below the quality standard. Treatment of water hyacinth and lotus show a downward trend in the value of Cadmium (Cd) and chromium (Cr) and the trend is increasing the value of BOD and COD. The result of using water hyacinth plant (Eichornia Crassipes) produces revealed better irrigation water quality than lotus plant (Nympae sp.).
