Abstract

Black rice contains higher levels of anthocyanin, fiber, and protein compared to local white rice. The price of black rice is higher compared to local white rice, it is one of prove that black rice has functional and economical values. The main obstacles in the cultivation of black rice are the long harvest age, high plant height, and lack of productivity. One way that can be applied to overcome this dissadvantageis with the utilization of gamma ray irradiation. The purpose of this research is to get the early-season rice, short stem, and higher productivity. This research was conducted in Tempel, Gatak, Sukoharjo District, by planting M3 seed (gamma ray irradiation of 300 Gray dosage on M2). The results of black rice from gamma ray irradiation of 300 gray dosage are M-CI-03-239, M-CI-03-244, M-CI-03-246, M-CI-03-346, M- CI-03-350, M-CI-03-234, M-CI-03-346, M-CI-03-350, M-CI-03-250, M-CI- 03-346, M-CI-03-CI-03-262, M-CI-03-265, M-CI-03-270, M-CI-03-283, and M-CI-03-311 with the agronomical properties of early age, i.e 2 weeks faster (64 days) with control (80 days). There are 125 short-postured plants of gamma ray irradiation result with  99.2% percentage. There are 12 individual plants in the M3 strain (M-CI-03-240, M-CI-03-251, and M-CI-03-261) of gamma ray irradiation with the maturation category, short stems, and high productivity compared with control. The difference of productivity of irradiated crops with control is 57,91%.

Abstract

Black rice Cempo Ireng is a local variety with a long plant life, high plant posture, and low productivity. Plant breeding by gamma irradiation was carried out to correct these weaknesses. The results of the gamma irradiation process included a selection process for each generation and produced potential lines numbers such as 8, 51, and 52. The characterization was based on the NDUS (Novelty, Distinctiveness, Uniformity, and Stability) test. The purpose of this study was to characterize potential lines as one of the requirements for the release of plant varieties. The study was conducted in the paddy fields of Pakahan Village, Jogonalan District, Klaten Regency, from February-July 2022. The method used was a single factor randomized complete block design with three replications. Data were analyzed by T test with 5% level. The results of the study showed that the three potential lines have differences with the parent and the Sembada varieties. Three potential lines can be deemed to be new, dictinct, uniform, and stable.

Abstract

Menoreh durian is an accessions durian fruit originally from Kulonprogo district, Special Region of Yogyakarta, Indonesia. In the context of proposing Menoreh durian to be a new superior variety, it is necessary to have an identity and position in the molecular genetic group. The aim of the study was to obtain the identity and position of the of Menoreh Durian  in the molecular genetic classification. The wet laboratory experiment was using PCR-based RAPD method. Eleven accessions of durian were identified and characterized then grouped based on molecular analysis which were pink Menoreh and 3 Yellow Menoreh accsessions, Petruk, Sunan, Sukun, Sitokong, Montong, Kani and Aspar. The RAPD method was carried out in stages: DNA extraction, DNA quantity and quality test, amplification, electroforesis, visualization and analysis of results. The RAPD classification of 11 durian accessions , resulted in three groups; namely group I which consisted of Menoreh Kuning, Petruk, Aspar Sunan and Menoreh Jambon; Group II consisted of Montong, Sitokong, and Kani from Thailand; and group III consisted of only Sukun accession. Based on this grouping, Menoreh Kuning dan Menoreh Jambon which were separated from other groups at a genetic distance of more than 0.25 indicated that the accession of Menoreh Kuning and Menoreh Jambon only as variety.
