Abstract

Sea grapes (Caulerpa racemosa) are green algae that contain secondary metabolites, including saponins, which have potential as bioactive compounds. This study aimed to determine the saponin content in 70% and 96% ethanol extracts and to analyze the differences in saponin levels between the two solvents using the UV-Vis spectrophotometric method.This research was an experimental laboratory study involving sample preparation, extraction by maceration using 70% and 96% ethanol, qualitative saponin testing, and quantitative determination of saponin levels using the vanillin–sulfuric acid method. Measurements were carried out at a maximum wavelength of 495 nm using a standard calibration curve with concentrations of 20–100 ppm.The results showed that the saponin content in the 70% ethanol extract was 954.06 ± 11.0 mg QE/g, while in the 96% ethanol extract it was 938.06 ± 8.9 mg QE/g. Statistical analysis using an independent t-test indicated no significant difference between the two extracts (p > 0.05).In conclusion, both 70% and 96% ethanol solvents are capable of extracting saponins from Caulerpa racemosa, but there is no significant difference in the saponin content obtained.
