Abstract

Currently, the use of PRP requires a person to visit a skin care clinic by doing a series of treatments directly. Therefore, in order to facilitate the use of PRP, asiaticoside lotion base products containing antioxidants can be used as a PRP product base. asiaticoside antioxidants serve to stabilize PRP when added to the base. This research uses Platelet-Rich Plasma (PRP) and Asiaticosida as active substances, Fancor® Uni-Embase as emollient and emulgator, DMDM hydantoin as preservative, and distilled water as carrier. The stages in this research include preparation of tools and materials, lotion base optimization, lotion preparation formulation, addition of active substances, lotion preparation evaluation, physical stability test, and antioxidant activity test. The results of the PRP lotion stability test with asiaticoside for 10 days showed significant results (p<0.05) which means that for 10 days there is a tendency to be unstable. Antioxidant activity test results obtained IC50 results on Asiaticosida of 57.63 µg/mL, on PRP of 196.1 µg/mL, on the base with the addition of Asiaticosida of 77.19 µg/mL, on F0 of 166.90 µg/mL, on F1 of 96.727 µg/mL, on F2 of 88.395 µg/mL, and on F3 of 82.017 µg/mL.Keywords: Lotion, PRP, asiaticoside, Antioxidant

Abstract

Rational use of drugs aims to give therapy according to patient’s clinical needs. Based on Ni komang's research (2021) there are still inaccuracies in the use of antipsychotic drugs. The purpose of this study was to determine the evaluation of the use of Antipsychotics based on the parameters of right indications, drugs, dosage, and frequency in outpatient schizophrenia patients at the West Java Provincial Mental Hospital. The study used a method with descriptive presentation of data and retrospective data collection. Data obtained period October-December 2022 from 130 patients, this study evaluated the use of Antipsychotics with the parameters of right indications, drugs, dosage, and frequency then the data will be analyzed based on the criteria for drug use from Pharmacotherapy A Pathophysiologic Approach 11th and PIONAS. The results of the study on the use of Antipsychotics in schizophrenic patients at the Java Provincial Mental Hospital based on Pharmacotherapy A Pathophysiologic Approach 11th and PIONAS obtained the right indications as much as (100%), the right drugs (100%), the right dose (98%), and the right frequency (98%).

Abstract

Abstract

Obesity is a global health problem closely associated with endothelial dysfunction, oxidative stress, and renal impairment, where endothelial nitric oxide synthase (eNOS) expression and nitric oxide (NO) levels serve as critical biomarkers for endothelial function. This study aimed to evaluate the effects of turmeric (Curcuma longa) extract, pegagan (Centella asiatica) extract, and their combination on body weight, organ index, fat index, blood biomarkers, and renal eNOS gene expression in obese rats. Male white rats were randomly divided into six groups: normal control, negative control (high-fat diet), positive control (orlistat 10.8 mg/kgBW), turmeric extract (200 mg/kgBW), pegagan extract (200 mg/kgBW), and a combination of turmeric–pegagan extracts (200:100 mg/kgBW). Treatments were administered for 60 days, and parameters assessed included body weight, organ index, fat index, blood biomarkers (ITT, creatinine, NO, elastin), and renal eNOS expression using PCR. Statistical analysis was performed using one-way ANOVA followed by Tukey’s post hoc test. The results demonstrated that the combination of turmeric and pegagan extracts significantly reduced body weight, liver, kidney, and heart indices, as well as visceral fat index, while improving blood biomarkers through the reduction of creatinine levels, along with an increase in NO serum approaching normal compared with the negative control group; the respective mean values were -10.14±0.57%, 5.52±0.73%, 1.23±0.21%, 0.64±0.06%, 1.35±0.05%, 0.33±0.06 mg/dL, dan 5.14±0.58 mmol/L. Renal eNOS expression exhibited an increasing trend in the combination group, although not statistically significant. In conclusion, the combination of turmeric and pegagan extracts exerts synergistic effects in improving endothelial dysfunction and renal impairment in obesity through lipid profile modulation, reduction of organ damage biomarkers, and enhancement of NO bioavailability.
