Abstract

Diversity of fruit flies (Diptera: tephritidae) attracted by me lure in CSG-BG germplasm carambolae plantation. Germplasm Garden (KPN) Cibinong Science Center-Botanical Garden (CSC-BG) is one of the gardens that conserve and utilizes Indonesian local germplasm, one of which is carambolae. The current problem in KPN is the attack of fruit flies (Bactrocera spp.). This study was conducted to determine the diversity of fruit flies in the KPN carambola plantation area. A total of eight traps were placed randomly in the plantation area. Each trap contained 1 mL of methyl eugenol pheromone solution. Identification of fruit flies was carried out based on three morphological differences, they were wings, abdomen, and hind legs. There were 317 fruit flies collected and separated into three species, namely Bactrocera carambolae, B. papayae, dan B. umbrosa, of which B. carambolae had a dominance index of 0.74 and abundance index of 85%. Data on the types of fruit flies can be used as primary data to determine the preparation of pest prevention efforts.

Abstract

Pada perbanyakan tanaman secara in vitro, sitokinin dan hara nitrogen merupakan beberapa komponen utama yang mempengaruhi pertumbuhan dan morfogenesis tanaman. Pada penelitian ini, komponen utama tersebut diuji untuk optimasi pertumbuhan kultur tunas kelor dengan perlakuan berikut: (1) konsentrasi BA yaitu 0.00, 0.05, 0.10, 0.25, 0.50, 1.00 mg L-1, (2) total konsentrasi N dalam medium yakni 20, 40, 60, 80 mM; (3) perlakuan nisbah NO3-:NH4+ yakni 0:1, 1:3, 1:1, 3:1, 1:0. Rancangan yang digunakan adalah acak lengkap dengan 10 ulangan. Hasil penelitian menunjukkan bahwa perlakuan BA 0.25 mg L-1 menghasilkan proliferasi tunas terbaik dengan jumlah buku tertinggi (8.07±3.48). Respon pertumbuhan tunas terhadap perbedaan konsentrasi nitrogen cenderung bersifat parabolik dengan konsentrasi optimum 40–60 mM pada medium standar MS. Perlakuan nisbah nitrat/ amonium menunjukkan pertumbuhan dan biomassa tunas in vitro yang optimal pada nisbah NO3-:NH4+ 3:1. Medium MS dengan modifikasi total konsentrasi nitrogen 40-60 mM dengan nisbah NO3-:NH4+ 3:1 dapat digunakan untuk proliferasi tunas kelor secara in vitro untuk tujuan perbanyakan klonal dari genotipe terpilih. Abstract

Wild bananas are believed to have genes for resistance to biotic and abiotic stress in nature, making them potential genetic resources for creating superior varieties. Wild banana seeds, such as Musa acuminata var. tomentosa are generally difficult to germinate in vivo, so that in vitro embryo culture technique is needed. This study aimed to increase embryo germination and regeneration of wild banana M. acuminata var. tomentosa by soaking the seeds as hydropriming. The treatment comprised of soaking the seeds in sterile distilled water for four periods of time: 0 (control), 1, 4, and 7 days. A total of 45 embryos for each treatment were planted on petri dishes containing MS + 0.5 mg/L BA + 1 mg/L biotin + 1 mg/L proline. The results showed that hydropriming increased the rate of embryo germination and regeneration. Seeds soaked for 1, 4, and 7 days successfully resulted in embryo germination percentages of 87%, 62%, and 62%, respectively, while the control unsoaked seeds germinated with a lower percentage of 42%. One-day soaking treatment was the most optimal treatment to increase the rate of germination and regeneration as well as obtained the best vigor as demonstrated by the highest average height of plantlets, number of leaves, and roots than other treatments. Thus, 1-day seed hydropriming is the best treatment for embryo rescue and regeneration of wild banana M. acuminata var. tomentosa
